BACKGROUND: Obtaining an appropriate anesthetic drug for rapid anesthesia with long term and safe recovery has always been the concern of fisheries science researchers. OBJECTIVES: In this study the anesthetic strength of Lavendula officinalis essence oil on histopathological and blood biochemical factors of Hypophthalmichthys molitrix has been investigated. METHODS: 260 juvenile silver carp were divided into four groups with a mean weight and length of about 23.55±1.5 g and 15.6±1 cm, respectively. Three groups were anesthetized with concentrations of 200, 300 and 400 ppm Lavender essence oil and 4 groups were considered as control group. The time needed to reach different stages of anesthesia was recorded and in two times (0 and 24h after anesthesia), hematology tests were conducted. After removing plasma serum by centrifugation, ALP, AST and ALT amounts were measured, venesection histological (sampling of liver, kidney and gills) and conserved for histology. RESULTS: Lavender essence oil anaesthetizes Hypophthalmichthys molitrix in less than 3 minutes and recovery time was also less than 3 minutes. Moreover, anesthesia with different concentrations of lavander had no effect on hematological profile and no significant changes in the AST, ALT and ALP were observed (p<0.05). Histopathological analysis showed the optimum concentration of 300 ppm with no side effects and indicated that the concentration power of essences can be used without fear of damage. CONCLUSIONS: It is advised that Lavender essence oil be used as a standard medicine to anesthetize and supersede prevalent chemicals to create anesthesia and sedation in fish.

Xylazine, a type of α₂-adrenoceptors, is a commonly used drug in veterinary medicine. Xylazine-induced changes in the content of amino acid neurotransmitters – glycine (Gly) and aspartic acid (Asp), in different brain regions and neurons were studied. Wistar rats were administered 50 mg/kg or 70 mg/kg of xylazine by intraperitoneal injection. In addition, in vitro experiments were conducted, in which neurons were treated with 15 μg/mL, 25 μg/mL, 35μg/mL, and 45 μg/mL of xylazine. Test methods were based on the enzyme-linked immunosorbent assays (ELISA). During anaesthesia, Asp levels in each brain area were significantly lower compared to the control group. Except for the cerebrum, levels of Gly in other brain areas were significantly increased during the anaesthesia period. In vitro, xylazine-related neuron secretion of Gly increased significantly compared to the control group at 60 min and 90 min. Moreover, xylazine caused a significant decrease in the levels of Asp secreted by neurons at 20 min, but gradually returned to the level of the control group. The data showed that during anaesthesia the overall levels of Asp decreased and overall levels of Gly increased. In addition, the inhibitory effect of xylazine on Asp and the promotion of Gly were dose-dependent. Our data showed that different effects of xylazine on excitatory and inhibitory neurotransmitters provided a theoretical basis for the mechanism of xylazine activity in clinical anaesthesia.

Electrical cardioversion is a therapeutic procedure used to convert various types of arrhythmias back to sinus rhythm. It is used to restore the sinus rhythm in dogs with atrial fibrillation. The effect of the electrical energy used during cardioversion on red blood cells (RBC) is not fully understood. Studies on humans reported lysis of RBC following electrical cardioversion. Similar studies have not been carried out on dogs. The aim of the study was to assess the effect of electrical cardioversion on chosen RBC parameters. The study was carried out on 14 large and giant breed dogs weighing from 30 to 84 kg with lone atrial fibrillation (lone AF). Electrical cardioversion was carried out under general anaesthesia by biphasic shock with 70–360 J of energy. Blood was collected at T0 – during atrial fibrillation, prior to cardioversion, and at T1 – 30 min after electrical cardioversion. Complete blood counts as well as total and direct bilirubin concentrations were evaluated. A maximum output of 360 J was used. In all cases, electrical cardioversion was effective, and no significant changes in the number of RBC and RBC indices were noted. Similarly, there were no statistically significant differences in the levels of total and direct bilirubin. Electrical cardioversion in dogs led neither to statistically nor clinically significant RBC lysis.

Objective: The objective of this study was to assess the effects of anesthetic drugs on heart rate (HR) and heart rate variability (HRV) in dogs.Materials and Methods: Twelve healthy client-owned dogs of various breeds, including five females and seven males were used for elective surgery in this study. The dogs were pre-medicated with four protocols; (1) alfaxalone [at 3 mg/kg body weight (bwt)], (2) zolazepam + tiletamine (Zoletil) (at 5 mg/kg bwt), (3) diazepam (at 0.3 mg/kg bwt) + ketamine (at 5 mg/kg bwt), and (4) diazepam (at 0.3 mg/kg bwt) + propofol (at 5 mg/kg bwt). The HR and HRV of 12 dogs were recorded 20 min before and after the administration of the anesthetic drugs. Doppler was used to obtain systolic, diastolic, and mean blood pressures.Results: After anesthetic drug administration, the dogs pre-medicated and inducted with alfaxalone had the lowest HR values as compared with those of other protocols. The HRV low frequency and high frequency power ratio decreased in the dogs pre-medicated and intubated with alfaxalone.Conclusion: This study demonstrates that alfaxalone preserves the cardiovascular function; and hence, is considered as safe to use for the surgical applicability in dogs. Objective: The scope of anesthesiology is extended immensely. The aim of this study was to know the farmers understanding and scope of veterinary anesthesiology. Materials and method: A total of 155 data were collected from different Upazilla Livestock Office and Veterinary Hospitals during the period from July to December 2017 through interview with a pretested questionnaire. Farmers age, sex, educational status and occupation were considered during interview. OBJECTIVE To determine effects of 3 plasma concentrations of fentanyl on the minimum alveolar concentration of isoflurane (MACiso) and cardiovascular variables in Hispaniolan Amazon parrots (Amazona ventralis). ANIMALS 6 adult parrots. PROCEDURES In phase 1, anesthesia was induced and maintained with isoflurane; intermittent positive-pressure ventilation was provided. The MACiso was determined for each bird by use of a bracketing method and supramaximal electrical stimulus. Fentanyl (20 μg/kg) was administered IV, and blood samples were collected over time to measure plasma fentanyl concentrations for pharmacokinetic calculations. In phase 2, pharmacokinetic values for individual birds were used for administration of fentanyl to achieve target plasma concentrations of 8, 16, and 32 ng/mL. At each concentration, MACiso and cardiovascular variables were determined. Data were analyzed with mixed-effects multilevel linear regression analysis. RESULTS Mean ± SD fentanyl plasma concentrations were 0 ng/mL, 5.01 ± 1.53 ng/mL, 12.12 ± 3.58 ng/mL, and 24.93 ± 4.13 ng/mL, and MACiso values were 2.09 ± 0.17%, 1.45 ± 0.32%, 1.34 ± 0.31%, and 0.95 ± 0.14% for fentanyl target concentrations of 0, 8, 16, and 32 ng/mL, respectively. Fentanyl significantly decreased MACiso in a dose-dependent manner. Heart rate and blood pressure significantly decreased at all fentanyl doses, compared with values for MACiso at 0 ng of fentanyl/mL. CONCLUSIONS AND CLINICAL RELEVANCE Fentanyl significantly decreased the MACiso in healthy Hispaniolan Amazon parrots, but this was accompanied by a depressive effect on heart rate and blood pressure that would need to be considered for application of this technique in clinical settings.

OBJECTIVE To investigate use of the plethysmographic variability index (PVI) and perfusion index (PI) for evaluating changes in arterial blood pressure in anesthetized tigers (Panthera tigris). ANIMALS 8 adult tigers. PROCEDURES Each tiger was anesthetized once with a combination of ketamine, midazolam, medetomidine, and isoflurane. Anesthetic monitoring included assessment of PI, PVI, direct blood pressure measurements, anesthetic gas concentrations, esophageal temperature, and results of capnography and ECG. Mean arterial blood pressure (MAP) was maintained for at least 20 minutes at each of the following blood pressure conditions: hypotensive (MAP = 50 ± 5 mm Hg), normotensive (MAP = 70 ± 5 mm Hg), and hypertensive (MAP = 90 ± 5 mm Hg). Arterial blood gas analysis was performed at the beginning of anesthesia and at each blood pressure condition. RESULTS Mean ± SD PI values were 1.82 ± 2.38%, 1.17 ± 0.77%, and 1.71 ± 1.51% and mean PVI values were 16.00 ± 5.07%, 10.44 ± 3.55%, and 8.17 ± 3.49% for hypotensive, normotensive, and hypertensive conditions, respectively. The PI values did not differ significantly among blood pressure conditions. The PVI value for the hypotensive condition differed significantly from values for the normotensive and hypertensive conditions. The PVI values were significantly correlated with MAP (r = −0.657). The OR of hypotension to nonhypotension for PVI values ≥ 18% was 43.6. CONCLUSIONS AND CLINICAL RELEVANCE PVI was a clinically applicable variable determined by use of noninvasive methods in anesthetized tigers. Values of PVI ≥ 18% may indicate hypotension.

OBJECTIVE To determine plasma concentrations of lidocaine after laryngeal administration or laryngeal and intratesticular administration in cats. ANIMALS 14 healthy adult sexually intact male cats (7 cats/treatment). PROCEDURES Cats were randomly allocated to receive 0.1 mL of 2% or 10% lidocaine hydrochloride solution (treatments L2 and L10, respectively) sprayed on the larynx for laryngeal desensitization, followed by endotracheal intubation and isoflurane anesthesia. After a 7-day washout period, cats were again randomly allocated to receive treatment L2 or L10, and castration was performed under isoflurane anesthesia following intratesticular administration of 2% lidocaine solution (0.1 mL/kg). In both experiments, a blood sample for measurement of plasma lidocaine concentration was obtained before (0 minutes) and 3, 5, 10, 15, 20, 30, 45, 60, and 75 minutes after laryngeal administration of lidocaine solution. Anesthesia was discontinued at 60 minutes. Plasma lidocaine concentrations were measured with high-performance liquid chromatography. RESULTS After treatments L2 and L10, median maximum plasma lidocaine concentrations were 34.1 ng/mL (range, 0 to 279.4 ng/mL) and 93.6 ng/mL (range, 79.3 to 182.2 ng/mL), respectively. Time to maximum plasma concentration was 10 minutes (range, 0 to 20 minutes) for each treatment. When cats received intratesticular lidocaine administration following L2 or L10 treatment, median maximum plasma concentration was 181.0 ng/mL (range, 103.7 to 600.2 ng/mL) and 301.2 ng/mL (range, 265.8 to 1,770.0 ng/mL), respectively. CONCLUSIONS AND CLINICAL RELEVANCE On the basis of these data, combined laryngeal and intratesticular administration of lidocaine solution at a total dose of approximately 5 mg/kg appears to be safe for use in healthy adult cats.

OBJECTIVE To compare the disposition of fentanyl citrate after a single IV injection in isoflurane-anesthetized red-tailed hawks (Buteo jamaicensis) and Hispaniolan Amazon parrots (Amazona ventralis). ANIMALS 6 adult red-tailed hawks and 6 adult Hispaniolan Amazon parrots. PROCEDURES Anesthesia was induced and maintained with isoflurane; intermittent positive-pressure ventilation was provided. The minimum alveolar concentration of isoflurane was determined for each bird by use of the bracketing method and a supramaximal electrical stimulus. Fentanyl (20 μg/kg) was administered IV. Arterial (red-tailed hawks) or jugular venous (Hispaniolan Amazon parrots) blood samples were obtained immediately before and 1, 2, 4, 8, 15, 30, 60, 120, 180, 240, and 480 minutes (red-tailed hawks) and 1, 5, 10, 15, 30, 60, 120, and 180 minutes (Hispaniolan Amazon parrots) after fentanyl administration. RESULTS A 3-compartment and a 2-compartment model best described fentanyl pharmacokinetics in red-tailed hawks and Hispaniolan Amazon parrots, respectively. Median apparent volume of the central compartment and volume of distribution at steady state were 222 mL/kg and 987 mL/kg, respectively, for the red-tailed hawks and 5,108 mL/kg and 13,079 mL/kg, respectively, for the Hispaniolan Amazon parrots. Median clearance and elimination half-life were 8.9 mL/min/kg and 90.22 minutes, respectively, for the red-tailed hawks and 198.8 mL/min/kg and 51.18 minutes, respectively, for the Hispaniolan Amazon parrots. CONCLUSIONS AND CLINICAL RELEVANCE Pharmacokinetic results for fentanyl in isoflurane-anesthetized red-tailed hawks and Hispaniolan Amazon parrots indicated large differences and should strongly discourage extrapolation of doses between these 2 species.

OBJECTIVE To quantify the effect of time and recumbency on CT measurements of lung volume and attenuation in healthy cats under general anesthesia. ANIMALS 8 healthy research cats. PROCEDURES Anesthetized cats were positioned in sternal recumbency for 20 minutes and then in left, right, and left lateral recumbency (40 minutes/position). Expiratory helical CT scan of the thorax was performed at 0 and 20 minutes in sternal recumbency and at 0, 5, 10, 20, 30, and 40 minutes in each lateral recumbent position. For each lung, CT measurements of lung volume and attenuation and the extent of lung areas that were hyperaerated (−1,000 to −901 Hounsfield units [HU]), normoaerated (−900 to −501 HU), poorly aerated (−500 to −101 HU), or nonaerated (−100 to +100 HU [indicative of atelectasis]) were determined with a semiautomatic threshold-based technique. A restricted maximum likelihood analysis was performed. RESULTS In lateral recumbency, the dependent lung had significantly greater attenuation and a lower volume than the nondependent lung. Within the dependent lung, there was a significantly higher percentage of poorly aerated lung tissue, compared with that in the nondependent lung. These changes were detected immediately after positioning the cats in lateral recumbency and remained static with no further significant time-related change. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that once anesthetized healthy cats were positioned in lateral recumbency, the dependent lung lobes underwent a rapid reduction in lung volume and increase in lung attenuation that did not progress over time, predominantly attributable to an increase in poorly aerated lung tissue.
