Prevalence and drug resistance of Salmonella in dogs and cats in Xuzhou, China

Salmonellosis is a zoonotic disease, and Salmonella spp. can sometimes be found in dogs and cats, posing a risk to human health. In this study, the prevalence and antimicrobial susceptibility of faecal Salmonella were investigated in pet dogs and cats in Xuzhou, Jiangsu Province, China. Faecal samples from 243 dogs and 113 cats, at seven pet clinics, were tested between March 2018 and May 2019. Each Salmonella isolate was characterised using serotyping and antimicrobial susceptibility tests. The prevalence of Salmonella was 9.47% in dogs and 1.77% in cats. Among the 25 isolates, eight serotypes of Salmonella enterica subsp. enterica were detected, S. Kentucky (n = 11), S. Indiana (n = 5), and S. Typhimurium (n = 4) predominating. S. Derby, S. Toucra, S. Sandiego, S. Newport, and S. Saintpaul all occurred singly. The 23 Salmonella strains found in dogs were from seven different serovars, while the two strains in cats were from two. The highest resistance rates were found for tetracycline (92%), azithromycin (88%), cefazolin (84%), nalidixic acid (80%), ampicillin (80%), ceftriaxone (80%), and streptomycin (76%). Resistance to three or more antimicrobial agents was detected in 24 (96%) isolates. Most of the S. Kentucky and S. Indiana isolates were multi-drug resistant to more than 11 agents. The carriage rate was far higher in dogs than in cats from Xuzhou. Some isolated strains were highly resistant to antimicrobials used to treat infections in humans and pets, which may raise the risk of humans being infected with multi-drug resistant Salmonella via close contact with pets.

Introduction: Laryngeal swab samples collected from three waterfowl slaughterhouses in central Taiwan were cultured and suspected isolates of Riemerella anatipestifer were identified by API 20NE and 16S rDNA PCR. Material and Methods: Serum agglutination was used for serotyping, and antimicrobial susceptibility was tested. Results: Seventy-six R. anatipestifer isolates were detected, and the prevalences in the ducks and geese were 12.3% (46/375) and 8.0% (30/375), respectively. The positive isolation rates were 65.6% for all arriving waterfowl, 76.0% for birds in the holding area, 1.6% for defeathered carcasses, but zero for degummed carcasses. A PCR examination detected R. anatipestifer in the slaughtering area frequently. Serotype B was dominant in both duck (34.8%) and goose (46.7%) isolates, but the wide serotype distribution may very well impede vaccination development. All isolates were resistant to colistin, and 79.7% were resistant to more than three common antibiotics. Conclusion: The results proved that most ducks had encountered antibiotic-resistant R. anatipestifer in rearing, which suggests that the bacterium circulates in asymptomatic waterfowl. It is worth noting that most waterfowl farms were found to harbour R. anatipestifer, and contaminated slaughterhouses are a major risk factor in its spread. Effective prevention and containment measures should be established there to interrupt the transmission chain of R. anatipestifer.

Avian pathogenicEscherichia coli (APEC) causes serious colibacillosis and significant economic losses. Data on profiles of virulence factors and antibiotic resistances among APEC strains are crucial to the control of infection. In this study, strains were isolated from eastern China, and the prevalence of virulence factors and distribution of antibiotic resistance were determined. APEC strains were isolated and characterised by PCR for O serogroups, virulence factor genes, antibiotic resistance, and phylogenetic groups. O78 was the most prevalent serogroup and type A was the most frequent phylogenetic group. ThefimH,feoB, andiron genes were the most prevalent among the isolates. All isolates were multiresistant, and all strains were resistant to ampicillin and tetracycline, which are widely used in the poultry industry in China. This study provided important data on the presence of virulence genes and antibiotic resistance profiles of APEC from poultry farms in eastern China.

Introduction: The toxinotyping and antimicrobial susceptibility of Clostridium perfringens strains isolated from processed chicken meat were determined. Material and Methods: Two hundred processed chicken meat samples from luncheon meats, nuggets, burgers, and sausages were screened for Clostridium perfringens by multiplex PCR assay for the presence of alpha (cpa), beta (cpb), epsilon (etx), iota (ia), and enterotoxin toxin (cpe) genes. The C. perfringens isolates were examined in vitro against eight antibiotics (streptomycin, amoxicillin, ampicillin, ciprofloxacin, lincomycin, cefotaxime, rifampicin, and trimethoprim-sulfamethoxazole) Results: An overall of 32 C. perfringens strains (16%) were isolated from 200 processed chicken meat samples tested. The prevalence of C. perfringens was significantly dependent on the type of toxin genes detected (P = 0.0), being the highest in sausages (32%), followed by luncheon meats (24%), burgers (6%), and nuggets (2%). C. perfringens type A was the most frequently present toxinotype (24/32; 75%), followed by type D (21.9 %) and type E (3.1%). Of the 32 C. perfringens strains tested, only 9 (28%) were enterotoxin gene carriers, with most representing type A (n = 6). C. perfringens strains differed in their resistance/susceptibility to commonly used antibiotics. Most of the strains tested were sensitive to ampicillin (97%) and amoxicillin (94%), with 100% of the strains being resistant to streptomycin and lincomycin. It is noteworthy that the nine isolates with enterotoxigenic potential had a higher resistance than the non-enterotoxigenic ones. Conclusion: The considerably high C. perfringens isolation rates from processed chicken meat samples and resistance to some of the commonly used antibiotics indicate a potential public health risk. Recent information about the isolation of enterotoxigenic C. perfringens type E from chicken sausage has been reported.

Introduction: Methicillin-resistant Staphylococcus aureus (MRSA) is a highly resistant and difficult to cure zoonotic microorganism, which makes up a large part of food toxic infections and has shown high prevalence among pig population all over the world. The aim of the study was to establish the occurrence of MRSA in slaughterhouses, evaluate its antimicrobial resistance, and verify whether there are any differences or similarities with reference to other European countries. Material and Methods: A total of 100 pigs, 105 carcasses, 19 workers, and 24 samples from the environment of several slaughterhouses were examined by conventional microbial and molecular methods. Results: In total, 78 MRSA isolates were found. MRSA prevalence in slaughtered pigs varied from 8.0% to 88.6% depending on the slaughterhouse, reaching higher prevalence in slaughterhouses with higher slaughter capacity. In total, 21.1% of all workers were carriers of MRSA and 6.7% of carcasses were contaminated with MRSA. The 98.2% of MRSA isolates were resistant to penicillin, 89.1% to tetracycline, 60.1% to erythromycin, 65.5% to gentamycin, and 15 different spa types were found, among which spa type t01333 was most widespread. Conclusion: The study indicated that MRSA prevalence and spa types differed according to slaughterhouse slaughter capacity and good hygiene practices. Quite high MRSA occurrence among slaughterhouse workers is one of the main factors which increase pork contamination risk.

Introduction: In the present study, some of the commercial fish farms located in the Black Sea region of Turkey, were screened for bacteria between 2012 and 2014.Material and Methods: The bacterial agents isolated from fish were identified by classical biochemical tests and the rapid diagnostic tests (API 20 E and API 20 Strep). All strains were further identified by sequencing of the 16S rRNA genes. The strains were also investigated for resistance to different antimicrobials by the disc diffusion method. Antibiotic resistance genes, including tetracycline (B), β-lactam (ampC, blaTEM, blaPSE), florfenicol (floR), erythromycine (ereA, ereB), sulphonamide (sulI, sulII), and trimethoprim (dhfr1) genes, were determined by the PCR method.Results:Vibrio anguillarum, Vibrio fluvialis, Photobacterium damselae subsp. piscicida, Pseudomonas luteola, Lactococcus garvieae, Streptococcus iniae, Aeromonas hydrophila, and Yersinia ruckeri were isolated from marine and freshwater cultured fish. According to the results of disc diffusion, all isolates were sensitive to florfenicol, trimethoprim+sulfamethoxazole, oxitetracycline, and enrofloxacin, and resistant to lincomycin, penicillin G, and amoxicillin. Also, sulI, sulII, and floR resistance genes were detected in the bacteria.Conclusion: The results of the study open up the opportunity to perform further investigations which could determine the possible role of ARGs in fish pathogens.

Objective: This study evaluated the occurrence and antimicrobial susceptibility profile of Salmonella species isolated from various poultry products including chicken meat, poultry eggs, poultry bird's drinking water, and poultry feed. Materials and methods: A total of 79 samples comprising of chicken meat (n=20), egg shell (n=15), poultry egg contents (n=18), drinking water (n=14), and poultry feed (n=12) were bacteriologically and microscopically analyzed for the isolation of Salmonella species. Results: Overall, this study reported a high prevalence of Salmonella species (62%) from various poultry products especially in poultry (chicken) meat and poultry egg contents where the percentage occurrence of Salmonella species was 100% and 20.4% respectively. The antibiogram conducted on the Salmonella species isolated from the various poultry samples reveal that all the isolates were multi-drug resistant to more than 50% of the tested antibiotics especially to tetracycline, gentamicin, tobramycin, nitrofurantoin and imipenem. However, most of the Salmonella species were also found to be highly susceptible to ceftriaxone, cefotaxime, ertapenem and ceftazidime. It was also observed in this study that the highest level of resistance to the tested antibiotics was recorded in Salmonella species isolated from poultry meat samples. Conclusion: Salmonellosis due to the consumption of contaminated or infected poultry products could pose serious public health problem to the general public if allowed. Thus, poultry farms and other poultry product outlets should be operated under sanitized conditions that ward-off the incidence of foodborne pathogens such as Salmonella. The use of antibiotics as growth promoting agents and prophylaxis in the production of poultry birds in this region should be discouraged and ndash; since such practices allowed drug-resistant bacteria to emerge and spread in the community. Objective: Phuchka is one of the most common street foods in Bangladesh. It is served with salad, sweet and sour tamarind dispersed water, and minced eggs as topping at places where people usually gather. This makes these foods susceptible to bacterial contamination. Therefore, assessing the bacterial load and antimicrobial profile of organisms isolated from phuchka and other foodstuffs served with it was the focus of this study. Materials and Methods: Bacterial isolates were isolated and identified from the samples after the bacterial loads were assessed as total viable count (TVC), total coliform count (TCC), and total staphylococcal count (TSC). The antibiotic resistance profile of the isolates was obtained using the disk diffusion method. Molecular detection of Escherichia coli isolates and the presence of gene responsible for tetracycline resistance was confirmed by polymerase chain reaction. Results: According to the recommendations, the TVC value of 70% phuchka and egg samples was safe, whereas TSC value illustrated that 80% of both phuchka and egg samples were at safety level. For the TCC value, 80% egg and 70% phuchka samples were found to be safe for consumption. Among all the samples, the microbial loads of the vendors hand wash were least in the safety level. Antibiotic sensitivity tests revealed that both Staphylococcus spp. and E. coli isolates were sensitive to gentamicin and ciprofloxacin but showed resistance to ampicillin. Conclusion: The data of this study indicate that phuchka can pose a public health problem as foodborne bacterial isolates which are antibiotic-resistant are found in it. Objectives: The present study was carried out to assess the antibiotic resistance and to identify the resistance genes in Escherichia coli from captive Bengal tigers at two Safari parks in Bangladesh. Materials and Methods: A number of 24 environmental fecal swab samples of Bengal tigers were collected from two different Safari parks in Bangladesh. For the isolation of E. coli, samples were submitted to a number of bacteriological screening and biochemical tests. The antibiotic susceptibility of E. coli isolates was determined by disk diffusion method. Results: Results demonstrated that 18 environmental fecal samples were positive to E. coli in bacteriological screening and biochemical test. The overall prevalence of E. coli in Bengal tiger was 75% (n = 18/24). The antibiogram study unveiled that all the isolates were resistant to ampicillin. Sulfamethoxazole-trimethoprim, nalidixic acid, and tetracycline were 89% (n = 16/18) resistant. On the contrary, 100% (n = 18/18) of the isolates were sensitive to colistin sulfate. blaTEM was detected in 78% (n = 14/18) ampicillin-resistant isolates, whereas sul2 was found in 31% (n = 5/16) of the sulfamethoxazole-trimethoprim-resistant isolates. Conclusion: This study, first time in Bangladesh, highlights a significant proportion of environmental fecal samples from captive Bengal tigers at Safari parks harboring antibiotic resistant E. coli. Transmission of resistant E. coli from Bengal tigers to humans and the environment could pose a public health risk at Safari parks in Bangladesh. [J Adv Vet Anim Res 2019; 6(3.000): 341-345]

Objectives: The objective of this study was to isolate and identify Staphylococcus aureus and Escherichia coli from raw milk samples of cattle and buffalo, and to evaluate the antibiotic sensitivity pattern. Materials and methods: A total of 34 milk samples were collected twice from 17 different healthy cattle (n=14) and buffaloes (n=3) at one-month interval, and analyzed in laboratory by staining, cultural and biochemical characteristics followed by polymerase chain reaction targeting nuc gene of S. aureus and 16 S rRNA of E. coli. Antibiotic sensitivity pattern of the isolated bacteria was assessed using the disc diffusion method. Results: Confirmation of the isolates as S. aureus and E. coli were carried out by PCR using nuc gene, 16S rRNA gene specific primers specific for S. aureus and E. coli respectively. A total of 12 samples (35.29%; 11 from cattle, 1 from buffalo) were found to be positive for S. aureus; 5 and 7 during first and second month, respectively. The E. coli were found in three samples (2 from cattle, 1 from buffaloe); one in first month and two in the second month. The antibiotic sensitivity test using 4 commonly used antibiotics indicated that the most of the isolates were resistant to Gatifloxacin and one isolate showed intermediate resistance to Ofloxacin while sensitive to Ciprofloxacin and Levofloxacin.Conclusion: Two different species of bacteria i.e., S. aureus and E. coli are contaminating with milk samples. The pathogenic bacteria can be controlled effectively by using Ciprofloxacin and Levofloxacin in the case of mastitis in cattle and buffaloes in Bangladesh. [J Adv Vet Anim Res 2016; 3(1.000): 62-67]
