Duck virus enteritis (duck plague) outbreak in an Australian black swan (Cygnus atratus) flock at safari park in Bangladesh: A case report

Objective: Duck virus enteritis is a severe viral disease that kills ducks and swans worldwide. The clinical manifestations, gross pathology, molecular detection, and characterization of the duck virus enteritis virus (DVEV) in Australian black swans at a safari park in Bangladesh were described in this case report.Materials and Methods: On a safari park in Bangladesh, an Australian black swan flock exhibited clinical signs of anorexia, greenish watery diarrhea, increased thirst, partial paralysis, and death. Postmortem examinations of deceased swans revealed extensive pathological abnormalities in the trachea, liver, and spleen. To isolate DVEV, a viral inoculum produced from the liver and spleen of dead swans was implanted into 913-day-old embryonated duck eggs via the chorioallantoic membrane (CAM) route. DVEV was confirmed using a polymerase chain reaction (PCR) assay. Phylogenetic analysis was used to determine the genetic relationship between the DVEV isolates from Australian black swans, and 16 DVEV isolates previously described in the GenBank. Results: Hemorrhage was noted in the annular ring of the trachea, as well as an enlarged and hemorrhagic liver and spleen. The PCR assay amplified a 446-bp fragment of the DVEV DNA polymerase gene in the liver, spleen, and CAM homogenates. The phylogenetic analysis found that the DVEV isolates from swans were comparable to those from Bangladesh, India, Vietnam, China, Germany, the USA, and Egypt. Conclusion: According to the findings of this study, the DVEV was the cause of illness and mortality in an Australian black swan flock. [J Adv Vet Anim Res 2021; 8(4.000): 557-562]

Zika virus, a member of Flaviviridae is the etiology of Zika or Zika fever or Zika virus (ZIKV) disease characterized by mild symptoms similar to very mild form of Dengue or Chikungunya. The virus transmits through Aedes mosquitoes, particularly by Aedes aegypti. The most dangerous effect of ZIKV infection is the ability of the virus to cause microcephaly and congenital malformation to the newborn baby if the mother is infected. The neurological disorders including Guillain-Barre syndrome might be associated with adults and children due to ZIKV infections. Zika has emerged as a serious global public health problem as it has been found in 87 countries, particularly in Africa, America, and Asia and has no vaccine and treatment so far. Bangladesh is at a high risk of ZIKV infection and we consider ZIKV as a possible emerging threat for Bangladesh. This short review summarizes the insights of ZIKV infection, present status of the disease in Bangladesh and its neighboring countries, and recommendations for necessary preparations and strategies to be taken for effective controlling of the ZIKV infection in Bangladesh before getting any havoc. Objective: The morphometric and meristic variations of Xenentodon cancila was studied based on the landmark-based truss network system to assess their phenotypic variations among four different freshwater stocks, viz. Boluhorpur baor, Jhenaidah (BBJ) (n = 29); Bhairab River, Jashore (BRJ) (n = 34); Arial Khan River, Madaripur (AKRM) (n = 28), and Bohnni baor, Gopalganj (BBG) (n = 25) in Bangladesh.Materials and methods: Seven meristic characters were counted by using a needle. Eight morphometrics and 28 truss measurements were measured by using tpsDigV.2.1 software. In meristic characters, KruskalWallis test was performed to determine any significant differences, whereas, in morphometrics and truss measurements, univariate statistics and discriminant function analy-ses were carried out by using SPSS 22 version. Results: Significant differences were observed in four meristic characters among seven meristic characters in the KruskalWallis test. In univariate statistics, only nine characters were observed significantly different among eight morphometrics and 28 truss measurements. The contribution of three discriminant function analyses (DFA), in which first DFA showed 49.2%, second DFA showed 33%, and third DFA showed 17.8% on behalf of both morphometric and truss measurements. In discriminant space, the four stocks were clearly separated. Two clusters were formed among four stocks, where BBG formed a single cluster, whereas BBJ and BRJ aggregately formed another cluster. Additionally, AKRM formed a sub-cluster with BBJ.Conclusion: The preliminary information generated from the current study would be beneficial for further genetic studies and in the assessment of ecological impacts on X. cancila stocks in Bangladesh. Objective: There is scarce literature regarding hematology profile of fat-tailed sheep (Dhumba). The study was conducted to determine reference intervals for their hematology profile in the context of Bangladesh.Materials and methods: Blood samples were collected from 32 healthy fat-tailed sheep from Dhaka, Bangladesh, during SeptemberOctober, 2015. Hemoglobin (Hb), packed cell volume (PCV), erythrocyte sedimentation rate (ESR), total leukocyte count (TLC), total erythrocyte count (TEC), lymphocyte, monocyte, eosinophil, basophil, and neutrophil counts were measured.Results: The levels showed a wide range and variation based on age and sex. Objective: Chicken infectious anemia virus (CIAV) is an economically important emerging infec¬tion of poultry as it causes immunosuppression and reduces egg production. Although it is world¬wide distributed and first reported (single case) in Bangladesh in 2002, no epidemiological and serological investigations have been conducted. The current study aimed to conduct a serologi¬cal investigation on the prevalence of CIAV infection in broiler breeder and layer farms in some selected areas of Bangladesh. Materials and Methods: A total number of 460 sera samples were randomly collected from unvac¬cinated broiler breeder and layer flocks, of which 276 were from 11 broiler breeder farms and 184 from 12 layer farms. The sera samples were subjected to a commercially available enzyme-linked immunosorbent assay kit to observe antibodies induced by CIAV. Results: Results demonstrated that the overall prevalence of CIAV was 83.6% among a total of 460 samples. In broiler breeder birds, the prevalence was 89.9%, whereas it was 78.3% in layer birds. A higher number of female birds was found to be seropositive than male birds. However, chickens of all age groups were found to be susceptible to the virus. Conclusions: These results indicate the presence of CIAV in Bangladesh, which may be the sequel of naturally occurring either vertical or horizontal infection in all bird flocks tested without clinical symptoms of the disease. A further epidemiological investigation will be required, followed by molecular isolation and characterization of the virus for suitable vaccine candidate selection and/ or preparation. Objectives: The present study estimated the seroprevalence of avian reovirus (ARV) infections in backyard chickens of the Mymensingh district in Bangladesh. Materials and Methods: Considering several risk factors, a total of 460 serum samples were col¬lected from backyard chickens from eight Upazilas of the Mymensingh district in Bangladesh. Blood samples were taken from the wing vein using 3-ml sterile syringes and kept at room tem¬perature for clotting in a slanting position and then transported to the laboratory maintaining the cool chain. Subsequently, the prepared sera were harvested and stored at −20°C until used. Finally, an indirect enzyme-linked immunosorbent assay (ELISA) was performed to detect ARV-specific antibodies using a commercial ARV antibody detection ELISA test kit. Results: The results revealed high prevalence rates of ARV antibodies, with a total seroprevalence of 69.78% (321/460). Area-wise, 74.55% (82/110) seroprevalence was recorded as the highest in Mymensingh Sadar, whereas 64% (32/50) was the lowest in Gauripur Upazila. With regard to sex, female chickens showed a significantly higher (p < 0.05) seroprevalence as 90.33% (271/300) compared to male chickens 31.25% (50/160). With regard to age groups, the seroprevalence of ARV infection was 59.33% (89/150) within 28 weeks, 82% (205/250) within 916 weeks, and 45% (27/60) within 1720 weeks, respectively. Based on hygienic conditions, the highest sero¬prevalence of ARV was noted in backyard chickens housed in poor conditions 80% (120/150) than good conditions 50% (40/80). Backyard chickens reared in free-ranging conditions exhibited a significantly higher seroprevalence 73.33% (220/300) of ARV antibodies compared to rearing in separate houses 63.12% (101/160). The seroprevalence of ARV was higher in crossbreeds 71.67% (43/60), brought from market 76% (38/50), and unhealthy 78.57% (55/70) backyard chickens than non-descriptive indigenous 69.5% (278/400), home-reared 69.02% (283/410), and healthy chick¬ens 68.21% (266/390). Conclusion: The high prevalence of ARV antibodies revealed in the current study indicates an extensive exposure of ARV to backyard chickens in Bangladesh that may be transmitted naturally to other chickens, ultimately leading to ominous economic effects on the poultry sector. Immediate after the official declaration of COVID-19 in Bangladesh on 8 March 2020, it has created public panic which results in price plummeting of the capital market and price hike of many essential commodities. Worldwide, the outbreak of COVID-19 has declared a pandemic. In response, the Government of Bangladesh has initiated some strict measures such as stopping the entry of passengers from Europe, stopping on-arrival visas and self-quarantine for 2 weeks for all passengers return from abroad. Still, many loopholes exist at the entry points of Bangladesh. Most of the people of Bangladesh are yet to aware of the consequences of COVID-19. In this backdrop, this article has attempted to create public awareness about COVID-19, providing some guidelines to restrict this deadly disease, enlisting current challenges of this disease in Bangladesh. Objective: The objective of the study was to investigate Foot and Mouth Disease virus (FMDV) outbreak in cattle in the Sarankhola Upazila under Bagerhat district of Bangladesh with isolation, identification, and molecular characterization of FMDV during April 2018. Materials and Methods: This Upazila is located at southern border of Bangladesh and surrounded by mangrove forest Sundarban. The outbreak investigation team collected epidemiological data from outbreak location. In addition, the team collected a total of 30 (15 calves, 15 adult) tongue epithelial tissue samples from a clinically FMD-affected cattle. The confirmation of FMDV and its three serotypes (A, O, and Asia-1) was performed by Reverse Transcriptase Polymerase Chain Reaction (RT-PCR). An amplified product of the VP1 region of FMDV genome was sequenced by Sanger sequencing method after cultivation and reconfirmation of FMDV into the BHK21 cell line. Genetic variability was studied by constructing a phylogenetic tree. Results: The investigation survey was carried out in overall 8,393 (8,393/15,580; 53.89%) cases including 3,050 (3,050/8,393; 36.34%) cases in calf and 5,343 (5,343/8,393; 59.77%) cases in adult cattle. The overall case fatality rate (CFR) was recorded as 2.27% (354/15,580) with significantly highest CFR in the calf (71.46%; 253/354) compared to an adult. The collected all 30 samples found with FMDV positive and mixed infection of all samples with serotype Asia-1 and serotype O were observed. In BHK 21 cell line, the eight FMDV positive samples showed a typical cytopathic effect during the third passage. Finally, DNA sequence data of two isolates found closely related with the isolates of bordering country India and Myanmar. Conclusion: The investigation identified the risk factors involved in an outbreak of FMDV, namely, sharing the common paddy land after harvesting, no FMD vaccination, the interaction between cattle and wildlife, and cross bordering movement. Objective: The objective of the study was to explore the outbreak situation in terms of animal, place, and time towards minimizing the risk of animal infection at the source in future and subsequent spillover in human in the endemic rural settings.Methodology: An outbreak investigation team from the Department of Livestock Services visited in each of the outbreak sites to explore the event towards strengthening the control program in the future. Meat samples of the infected slaughtered animals were collected to confirm the causal agent of the animal outbreak using polychrome methylene blue microscopic examination technique. Participatory epidemiology tool such as semi-structured interview had been used in these investigations to realize the knowledge and practices of local people/cattle keepers on anthrax control and prevention in animal and human as well.Results: All identified affected human cases had been confirmed as a history of contact with the animal carcasses or handling/processing with infected meat. The level of awareness at the community level was not satisfactory for the prevention and control of anthrax at the source and further spillover in human. The infected slaughtered animals found to be in non-vaccinated status during the outbreak investigation and uncontrolled animal movement is considered to be responsible for new outbreaks in a vaccinated zone where enforcement of veterinary legislation is inadequate. Conclusion: A comprehensive preparedness and response strategy is to be obligatory for prevention, control and respond on anthrax in Bangladesh. Maximum vaccination coverage in the animal, increase community awareness of animal and human anthrax are also demanded for transmission of anthrax from animal to human. Objective: The present study was conducted in New Sandwip island (Jahajerchar) of the Meghna estuary, Noakhali to explore animal species are being reared, their disease status, and also to finding out the constraints and opportunitities for keeping livestock in the island. Materials: Considering local knowledge as an epidemiological intelligence, the livestock species and their disease profile as well as livestock keeping problems were assessed through two focus group discussion with informants’ interview using Participatory Disease Searching (PDS) tools. The interview was conducted predominantly using semi-structured questionnaire (SSI) and other participatory epidemiology(PE) tools such as simple ranking (SR), pair-wise ranking (PWR), proportional pilling (PP) and transect work (visualization) were applied with flexibility to collect information.Results: PDS tools identified sheep (69%) is the main livestock species followed by cattle (16%) and buffalo (15%) in the island. The Peste des Petits Ruminant (PPR) in sheep (90%), anthrax in cattle (52%) and Haemorrgic Septicemia (HS) in buffalo (59%) were confirmed as top important species-wise diseases via the study. This place is prospect for sheep and buffalo raising rather than cattle. Lack of of shelter (Khella) for keeping livestock during adverse climatic condition, no access of portable water along with inadequate veterinary health care facilities were recognized as constraints for livestock raising in this island.Conclusion: This place is potential for sheep and buffalo raising rather than cattle. The study has validated the significance of accepting participatory disease searching tool in order to capture voluntarily submitted epidemiological data towards establishing a cost effective, unique national disease surveillance system in Bangladesh.
