Infectious bovine keratoconjunctivitis epizootic associated with area-wide emergence of a new Moraxella bovis pilus type.

Pilus-mediated adherence is a virulence attribute of Moraxella bovis. Several pilus types have been shown to exist among strains of this bacterium, but correlation between pilus type and specific field cases of the disease has not been done. During the summer of 1987, an epizootic of infectious bovine keratoconjunctivitis was reported in 7 Iowa counties. Eight isolates of M bovis were secured from 12 episodes studied. All 8 of the isolates were nearly homogeneous in biochemical properties and had the same plasmid biotype. Pilus typing performed by immunofluorescence and immunogold electron microscopy identified a single new pilus type among 5 of the 8 isolates. This pilus type was identified in field cases that developed within a narrow time frame and over large distances. The implication of these findings is that infectious bovine keratoconjunctivitis epizootics may be associated with emergence of a novel pilus type, and that rapid dissemination over wide distances can occur, presumably by transportation of carrier cattle.

Influence of time after the removal of nocodazole from nuclear donors on the development of reconstituted embryos in bovine nuclear transplantation.

The present study examined the influence of post-cleavage time of nuclear donors on the development of reconstituted embryos in bovine nuclear transfer. Blastomeres of 16-cell stage embryos derived from in vitro-maturation, fertilization and culture were used as nuclear donor source. They were treated with 10 mu-M nocodazole for 12 hr. Blastomeres that cleaved within 3 hr after the removal of nocodazole were used-for the study. Metaphase II (M-II) oocytes were used as recipient cytoplasm. IN experiment 1, donor blastomeres at 6, 11 and 15 hr after the removal of nocodazole and donor blastomeres no treated with nocodazole were transferred into ethanol-exposed and enucleated oocytes. The reconstituted embryos produced by donor blastomeres oat 6 hr after the removal of nocodazole had a significantly higher developmental rate to the blastocyst stage than those at 15 hr and the untreated groups (P<0.01). In experiment 2, blastomeres at 6 hr after the removal of nocodazole used as nuclear donors were transferred into ethanol-exposed and enucleated M-II oocytes. The reconstituted embryos with ethanol-exposed and enucleated oocytes as recipient cytoplasm had a significantly higher rate of initial-cleavage (P<0.05) and development to the blastocyst stage (P<0.01) than non ethanol-exposed and enucleated M-II oocytes. These results demonstrate that the development of reconstituted embryos was improved when cleaved donor blastomeres after the removal of nocodazole were immediately transferred (at 3-6 hr post-cleavage) into activated enucleated oocytes by exposure to ethanol.

The number and distribution of retinal ganglion cells in a Korean native cattle.

The number and distribution of the retinal ganglion cells in the 2 years old Korean native cattle was determined from whole flat mounted preparation stained with methylene blue and thionin. The total number of retinal ganglion cells was estimated to be 3,085,200 in the bovine retina ranging from 2.214mm** (2) in total area. Visual streak was recognized at the area 2.5mm superior to the optic disc and ganglion cell density drops off rapidly to the direction superior to and inferior to the visual streak. Area centralis (6,800 cells/mm** (2)) was located at the area 10mm temporally from the point of 3mm superior to the optic disc. The number of alpha-type ganglion cells (above 15 micro) was 57,000 in the bovine retina and alpha-type ganglion cells constituted 18.5 % of the total cells. The relative frequency of alpha-type ganglion cells was higher in the peripheral regions than in the visual streak, especially higher in the superior-temporal quadrant than in other region of the bovine retina.

This study was undertaken to obtain basic data of the sternal development in Korean native cattle from the earliest sternal formation to the ossification using histological and histochemical methods. Thirty three sterna were collected from a series of embryos and fetuses ranging from 11 to 225mm (estimated age 37-120 days) in crown rump length. The bilateral sternal bars were observed in the 2nd group (CRL 21-30mm) of Korean cattle embryos. Those bars initiated to be fused in the 3rd group (CRL 31-40mm) and completed in the 7th group (CRL 71-80mm). The ossification centers were detected in the 8th group (CRL 81-90mm) also bilateral ossification centers were found in the same group. The typical epiphyseal plates, endochondral bone and calcium deposit were found in the 9th group (CRL 91-100mm). Osteocytes, osteoblasts, osteoclasts and myeloid cells appeared in ossification centers in the 10th group (more than CRL 101mm). The alcianophility responded markedly in the 9th group that was decreased and showed slightly positive reaction in territorial matrix of the 10th group. Marked positive reaction to PAS was observed in bony trabeculae in the 10th group. The positive reaction to calcium deposit by trichrome stain was observed initially in the hypertrophied zone of epiphyseal plate in the 9th group and was conspicuous in the calcified zone of epiphyseal plate in the 10th group. The 1st positive reaction to the von Kossa stain was observed in the 9th group.

Each of twenty blood samples taken from apparently healthy Korean cows was used to produce five different mixtures of autologous plasma and blood corpuscles such that their values of packed cell volume(PCV) lay between 10 to 50ml/100ml. The measurement of erythrocyte sedimentation rate(ESR) using 45 degree-angled Wintrobe hematocrit tube, 3mm bore, (45deg C-Wintrobe-ESR) was practised for the blood of various levels of PCV lowered. Correlation of the ESR to PCV showed curvilinear regression each in three levels of PCV under the ambient temperature of 10deg C, 20deg C and 30deg C. Correlation of the ESR to the ambient temperature showed linear regression each in five levels of PCV. The ESR increased with ascending ambient temperature, and magnitude of the increase of the ESR became greater as the level of PCV lowered. Correlation of the ESR to PCV showed curvilinear regression each in three levels of the ambient temperature, and the ESR was increased with decreasing PCV. The data were statistically analysed and a list of relative anticipated 45deg C-Wintrobe-ESR values for PCV and ambient temperature was presented.

For the comparison of surface fine structures in the proliferative forms of two major protozoan parasites, Toxoplasma gondii and Sarcocystis species in mammalian hosts, isolated from the artificially infected mice and from the naturally infected cattle, respectively, an SEM(Hitachi S-570) was applied to the fixed, dried and coated with gold ion on the microslide glasses. The tachyzoites of T. gondii from the peritoneal cavity of the mouse showed the crescent-like feature and measured as 5.57 micro m in length and 2.33 micro m in width, while the bradyzoites of Sarcocystis species from the heart muscle of slaughtered cattle was banana-shaped and measured 14.18 micro m in length and 2.85 micro m in width. On the surface of Sarcocystis species bradyzoite, a distinct elliptical micropore was identified in the high magnification observation of 60,000X, and it measured as 0.35 micro m in length and 0.18 micro m in width.
