Detailed information on specific species of non-aureus staphylococci (NAS) has become a necessity for effective udder health control programs in South Africa. The main objective of this preliminary study was to identify the different NAS species and strains present in dairy herds in South Africa using a cost-effective method. A further objective was to investigate the effects of cow risk factors and farming systems on the NAS isolates identified. A total of 214 NAS, isolated from milk collected from 17 South African dairy herds, were identified using three diagnostic tests (API Staph test, MALDI-TOF and 16s rRNA). There was a good observed agreement between the MALDI-TOF and 16S rRNA sequencing (92.2%) and a poor observed agreement between the MALDI-TOF and API Staph (25.7%). The genetic relatedness within species was investigated in 128 of these isolates using random polymorphic amplified deoxyribonucleic acid (DNA) (RAPD), verified by multilocus sequence typing (MLST), and phylogenetic analysis and cow risk factors were investigated on species level. The main NAS species isolated were Staphylococcus chromogenes (75.2%), Staphylococcus epidermidis (9.4%) and Staphylococcus haemolyticus (8.9%). The RAPD test identified 34 Staphylococcus chromogenes, 13 Staphylococcus epidermidis and nine Staphylococcus haemolyticus strains, indicating genetic diversity amongst strains and herds. The presence of NAS intramammary infections was found to be significantly related to the farming systems, composite cow milk somatic cell count (SCC), parity and days in milk (DIM). Significantly more NAS were isolated from primiparous and from older cows. This knowledge could assist with the management of NAS on dairy farms.

Dairy cows may infrequently give milk tinged with blood after calving, which is a condition termed haemolactia. Economic losses for dairy farmers are caused by cases of haemolactia because of the condemnation of such milk, potential contamination of good bulk tank milk with haemolactic milk, and need for veterinarian intervention. This study was performed to elucidate the oxidative status of dairy cows with haemolactia during the peripartum period.

This study was performed to elucidate the oxidative status of dairy cows with haemolactia during the peripartum period. Plasma glutathione peroxidase, malondialdehyde (MDA) and superoxide dismutase concentrations along with serum vitamin A, C and E concentrations were determined as indices of oxidative stress. The sampled dairy cows comprised two haemolactic (n = 11 and n = 6) and two non-haemolactic (n = 11 and n = 6) groups. On the first day when haemolactia was identified in colostrum (at mean 2.1 days after parturition), a significantly increased concentration of plasma MDA was noted in the haemolactic group. During the prepartum period, low levels of serum vitamin E were continuously observed from prepartum week 4 to the parturition day but only in the haemolactic group. These results demonstrate that continuous low levels of serum vitamin E in the prepartum period may play a pivotal role as a requisite factor in the onset of haemolactia after calving.

The changes in the expression profile of the angiogenic genes and serum biomarkers were investigated in the cows with the retained placenta after normal parturition. Retained fetal membranes (RFM) is considered one of the main reproductive disorders in dairy cattle. A total of 16 cows were allocated into two groups: group 1 (n = 8) the cows that have a normal loosed placenta, and group 2 (n = 8). The cows that have retained fetal membranes for more than 24 h after parturition. Blood with EDTA was collected from the tail vein for studying the expression of angiogenic proteins including vascular endothelial growth factor (VEGF), vascular endothelial growth factor receptor1 (VEGFR1), and RANTES genes. Serum was collected at a one-week interval before/after parturition for the study of biochemical changes. The VEGF expression decreased ten folds in cows with retained than normally dropped placentae (p ˂ 0.0001). The vascular endothelial growth factor receptor2 (VEGFR2) showed a threefold increase in cows with normal loosed than retained placentae (p˂0.01). The RANTES was increased in cows with retained rather than normal placentae by about 7 folds (p˂0.01). Phosphorus and total proteins were significantly lower (p˂0.05) in RFM cows one week after parturition. The serum albumin, TG, HDL, and cholesterol were significantly lower (p˂0.05) in RFM before, at, and after parturition. CPK was significantly higher in RFM before, at, and after parturition (p˂0.05). Increased values of MDA and decreased values of CAT, SOD, and R-GSH were detected in the blood of cows suffering from RFM (p˂0.05). In conclusion, the translation of VEGF and its receptor and RANTES mRNA at the time of parturition in dairy cows could be proxy biomarkers for the prediction of retained fetal membranes (RFM). In the same line, the activity of total proteins, serum albumin, TG, HDL, cholesterol, and CPK would be useful in the prediction of the RFM in dairy cows.
