BACKGROUND: Obtaining an appropriate anesthetic drug for rapid anesthesia with long term and safe recovery has always been the concern of fisheries science researchers. OBJECTIVES: In this study the anesthetic strength of Lavendula officinalis essence oil on histopathological and blood biochemical factors of Hypophthalmichthys molitrix has been investigated. METHODS: 260 juvenile silver carp were divided into four groups with a mean weight and length of about 23.55±1.5 g and 15.6±1 cm, respectively. Three groups were anesthetized with concentrations of 200, 300 and 400 ppm Lavender essence oil and 4 groups were considered as control group. The time needed to reach different stages of anesthesia was recorded and in two times (0 and 24h after anesthesia), hematology tests were conducted. After removing plasma serum by centrifugation, ALP, AST and ALT amounts were measured, venesection histological (sampling of liver, kidney and gills) and conserved for histology. RESULTS: Lavender essence oil anaesthetizes Hypophthalmichthys molitrix in less than 3 minutes and recovery time was also less than 3 minutes. Moreover, anesthesia with different concentrations of lavander had no effect on hematological profile and no significant changes in the AST, ALT and ALP were observed (p<0.05). Histopathological analysis showed the optimum concentration of 300 ppm with no side effects and indicated that the concentration power of essences can be used without fear of damage. CONCLUSIONS: It is advised that Lavender essence oil be used as a standard medicine to anesthetize and supersede prevalent chemicals to create anesthesia and sedation in fish.

BACKGROUND: Rumen health in ruminants has a great importance because its function has a great deal with milk, meat and fetus production. Many factors can affect the rumen health and destruct its function and performance. OBJECTIVES: this research investigates histopathology lesions resulted from ruminants in the sheep. METHODS: for this purpose, macroscopic examination in 1562 rumens of the slaughtered sheep Sanandaj was done. Among these, 23 rumens has lesions placed in drums of formalin 10% for macroscopic evaluations by means of Hematoxylin and Eosin coloring were referred into the pathobiology lab. RESULTS: in Macroscopic observations, swelling (65.22%), swelling and focal congestion (30.43%) and looming papillary (4.35%) were detected. The highest microscopic signs in the rumen lesions included: sub mucosal edema (32.69%), ruminate eosinophilic (23.07%), parakeratosis (13.46%), granulomatous ruminate (7.69%), watery degeneration and acute purulent ruminate (5.76%). CONCLUSIONS: pathologic lesions of rumen can be due to hyperplasia change in the result of mechanical agitation, stress, parasitic diseases, and mature woody forage.

Epstein–Barr virus (EBV) is a γ-herpesvirus associated with various neoplasms in humans and is a probable aetiological agent in breast cancer; however, a causal relationship has not yet been established. Because of the epidemiological and clinicopathological similarities between breast cancer and canine mammary tumours, dogs have been proposed as a valid model for breast cancer. A total of 47 canine mammary gland tumour tissues were processed by routine histopathological technique with haematoxylin-eosin staining and classified according to the type of neoplasm. DNA was extracted from paraffin-embedded tissues and the EBNA-1 gene and the BamHI-W region specific for EBV were evaluated by nested PCR. The histopathological evaluation revealed 2 benign neoplasms, and many carcinomas: 2 in situ, 9 simple, 3 solid, 10 complex, and 21 mixed. One sample was positive for the EBNA-1 gene, while all were negative for the BamHI-W region. No association was found between EBV and mammary tumours in dogs. However, here we report for the first time the presence of an EBV gene sequence in a canine mammary tumour. It is likely that detection of EBV might be affected by the quality and quantity of DNA extracted from paraffin-embedded tissues. Additional studies are necessary to establish any association of EBV with mammary gland cancer in humans and in dogs, which could eventually lead to better public health prevention and control.

Veterinarians use flumequine (FLU) widely but its toxicological effects are still unclear. FLU doses of 53, 200, or 750 mg/kg were administered orally for six weeks to pubertal male rats for evaluation of their toxicity. Weight gain was poorer after seven days of exposure to FLU 750, but relative weights of the brain, adrenal and thyroid glands, and testes were notably higher. Haematological and lipid profile parameters, cardiac markers, and inorganic phosphate significantly increased in the FLU 750 group. Blood glucose, oestradiol and serum concentrations of immunoglobulins G (IgG) and E (IgE) significantly decreased after treatment. The levels of interleukins 10 (IL-10) and 6 (IL-6) fell significantly in the FLU 200 and FLU 750 groups. Cytochrome P450, family 1, subfamily A, polypeptide 1 (CYP1A1) and cyclooxygenase-2 (Cox-2) expression amplified after treatment. Serum levels of free triiodothyronine (fT3) and free thyroxine (fT4) reduced in the FLU 200 and FLU 750 groups without changes in total T3 or T4 level. All doses of FLU significantly depressed concentrations of thyroid-stimulating hormone (TSH) and testosterone. Histopathology of thyroid glands from rats treated with FLU 750 showed degeneration and depletion of thyroid follicular epithelial cells. Expression of 8-hydroxydeoxyguanosine (8-OHdG) was increased in a dose-dependent manner in the brain, but decreased in the testes. Expression of CYP1A1 increased in the adrenal and pituitary glands. The results of this study suggest that the toxicity of FLU in rats is an effect of its disruptive influence on the pituitary-thyroid hormonal system and on the dysfunction of the immune system.

Nerium oleander is a plant of the Apocynaceae family toxic to humans, animals, and insects. This study was performed to determine the cardiac and neurotoxicity of the plant extract by oral administration in Wistar rats and Balb/c mice and to compare the susceptibility of these animal models to oleander toxicity. Four groups of eight mice and eight rats received N. oleander extract orally while a fifth group was the control. Serum concentrations of the biochemical toxicity indicators, namely troponin and creatine kinase (CK), were determined and histopathological evaluation of the heart and brain was performed. In mice, CK and troponin concentrations were respectively 1.5 and 7 times higher than in the control group (P < 0.05), while in rats, they were 6–7 and 11 times higher. Hyperaemia, haemorrhage, and myofibrolysis, without infiltration of inflammatory cells, were observed in the heart. In the brain the authors observed hyperaemia associated with perivascular and perineuronal oedema, and in higher-dosed rats multifocal haemorrhagic and liquefactive necrotic lesions. Oleander can affect serum levels of CK and troponin due to nervous and cardiac injuries. Rats showed more severe changes in the biochemical indicators and histopathological lesions than mice. Therefore, biochemical and pathological findings indicate that Wistar rats are more susceptible to the cardiac toxicity and neurotoxicity effects of N. oleander poisoning than Balb/c mice.

In recent years, the use of bone scaffolds as bone tissue substitutes, especially the use of such as hydroxyapatite and tricalcium phosphate, has been very popular. Today, the use of modern engineering techniques and advances in nanotechnology have expanded the use of nanomaterials as bone scaffolds for bone tissue applications. This study was performed on 60 adult male New Zealand rabbits divided into four experimental groups: the control group without any treatment, the second group receiving hydroxyapatite, the third group treated with β-tricalcium phosphate, and the fourth group receiving nanocomposite polycaprolactone (PCL) scaffold. In a surgical procedure, a defect 6 mm in diameter was made in a hind limb femur. Four indexes were used to assess histopathology, which were union index, spongiosa index, cortex index, and bone marrow. The results showed that nanocomposite PCL and control groups always had the respective highest and lowest values among all the groups at all time intervals. The histopathological assessment demonstrated that the quantity of newly formed lamellar bone in the nanocomposite PCL group was higher than in other groups. All these data suggest that PCL had positive effects on the bone healing process, which could have great potential in tissue engineering and clinical applications.

This study consisted in histopathological and immunohistochemical examinations of the central nervous system of 15 sheep suspected of infection with Coenurus cerebralis. The sheep displayed compulsive circling and were submitted for necropsy in 2012–2016. Species identification was made on the basis of the PCR analysis and parasitological examination of the cysts. Coenurus cerebralis cysts were detected only in the cerebral tissue of 13 sheep and in the cerebral and cerebellar tissues of 2 animals. Out of the 33 parasite cysts, most (21.21%) were located in the right and left frontal lobes of the cerebrum. The largest cyst measured 6 × 5 cm and the smallest cyst was 2 × 2 cm in size. The highest and lowest numbers of scolices were 55 and 21, and the number of rostellar hooks ranged between 22 and 30. Histopathological examination revealed the presence of typical parasitic granulomatous inflammatory foci. Immunohistochemical staining showed that most common in the periphery of the parasite cysts were, in descending order by cell number, GFAP, CD163, CD3, and CD79α-positive cells. The study confirms the role of cellular defence mechanisms in the pathogenesis of Coenurus cerebralis infection in sheep.

Objective: Salmonella is a widely-reported zoonotic bacterial pathogen and human infection is mostly attributed through direct or indirect contact with chickens. Salmonella Kentucky (S. Kentucky) is one of the motile serovars which has recently been identified from both poultry and human samples in Bangladesh. This study was conducted to assess its pathogenic potentials and shedding probability in backyard chicken. Materials and methods: We infected 22 backyard chickens orally, each with 106 cfu of Salmonella Kentuckey, which were then observed for 23 days to enlist clinical signs, gross and histo-pathological changes. Polymerase chain reaction (PCR) for Salmonella was applied on some representative samples to identify the presence of Salmonella. Results: Four chickens were sacrificed and the internal organs were examined to observe gross and microscopic tissue changes. Some reactive changes were seen in spleen during prolonged course of infection. The probability of S. Kentucky shedding was 77% (95%; CI 54-90%) on DPI 2, 41% (95%; CI 21-60%) on DPI 12 and 13% (95%; CI 3-31%) on DPI 21. The survival probability of the infected chickens was 50% (95%; CI 28-68%) on DPI 6, 32% (95%; CI 14-51%) on DPI 15 and 14% (95%; CI 3-31%) on DPI 23. Conclusion: Zoonotic S. Kentucky strain of human non-typhoidal clinical cases of gastroenteritis has potentials to produce clinical signs such as reduced feed uptake, watery or pasty fecal droppings and lesions, such as catarrhal enteritis and typhlitis. Objective: This study aimed to observe the changes in anatomical pathology, histopathology, and histomorphometry of uterine horn in Aceh cattle with repeat breeding (RB).Materials and methods: In this study, five uterine horns were collected from fertile Aceh cattle (N1, N2, N3, N4, and N5) and Aceh cattle with RB (RB1, RB2, RB3, RB4, and RB5). Changes in the anatomical pathology of uterine horn were observed prior to histopathological preparations. The results were analyzed descriptively.Results: The observation of anatomical pathology revealed that the uterus of fertile cattle was yellowish white with horn-like shape consisting of hollow muscles, while RB cattle were reddish-pale. Furthermore, uterine horn swelling with purulent exudate was observed in the uterine horn of RB1 and RB4, while swelling with serous exudates was found in RB2, RB3, and RB5. Thin-walled uterine horn with caruncular atrophy was observed only in RB3. Histopathological observations showed erosion and hyperplasia of endometrial columnar epithelial and uterine glands in RB1, RB2, RB4, and RB5. Atrophy of uterine gland was found in RB3 and edema of caruncular was observed in RB1 and RB4. In addition, hemorrhage and inflammatory cell infiltration (neutrophils, lymphocytes, and macrophages) were found in all RB cattle. Histologically, cow uterus is divided into three layers, endometrium, myometrium, and perimetrium with the uterine gland found in the endometrium. Histomorphometric measurements found that the uterine horn wall (endometrial, myometrial, and perimetrium) of RB cattle were thicker than of fertile Aceh cattle, 208.06 ± 39.90 vs. 187.39 ± 29.09 μm, 400.138 ± 51.96 vs. 277.91 ± 42.88 μm, and 23.59 ± 9.67 vs. 18.53 ± 4.40 μm. However, the endometrial gland diameter of RB cattle is smaller than that of fertile Aceh cattle, 4.04 ± 0.88 vs. 4.99 ± 1.37 μm.Conclusion: The changes in anatomical pathology and histopathology of the uterine horn of Aceh cattle with RB indicate endometritis which caused by subacute and chronic bacterial infections. Endometritis causes disrupted blood circulation, which is characterized by hemorrhage, edema, and thickening of the endometrium, myometrium, and perimetrium, resulting in an increased diameter of the endometrial gland in RB Aceh cattle. Objective: Sarcocystis spp. are common parasites and in terms of economics and pathogenicity in domestic animals is important. The purpose of this work was to define the rate of contamination of slaughtered carcasses of cattle to Sarcocystis using digestive and histopathological methods in southeast Iran.Material and Methods: In this descriptive laboratory study for 1 year, 500 carcasses were examined and isolated bradyzoites of Sarcocystis with the digest method. Also, tissue samples from the esophagus and diaphragm were considered for pathologic studies and stained with hematoxylin and eosin of sections of histopathological.Results: The results showed that the highest contaminations were in imported male animals aged 23 years old in the spring. There was a significant difference (p < 0.05) in the prevalence rate with the sex and race of cattle but no significant difference (p > 0.05) in the prevalence rate with age and season.Conclusion: Infection with Sarcocystis is common in oxen in this region. The imported cattle are more infected. It seems that racing and the environmental condition affect the prevalence of Sarcocystosis. [J Adv Vet Anim Res 2018; 5(4.000): 381-387]
