Maturation of spinal-evoked potentials to tibial and ulnar nerve stimulation in clinically normal dogs

Spinal-evoked potentials were recorded from 2 litters of clinically normal mixed-breed dogs between 35 and 300 days of age. Summated responses to tibial nerve stimulation were recorded from percutaneous needle electrodes placed at L7-S1, L4-5, T13-L1, C7-T1, and the cisterna cerebellomedullaris. The ulnar nerve was stimulated with recordings at C7-T1 and the cisterna cerebellomedullaris. Amplitudes did not change significantly with age, but were significantly (P < 0.05) different between various recording sites. On day 35, segmental and overall (L7-cisterna cerebellomedullaris) conduction velocities were less than half of the adult values. Spinal cord conduction velocities increased with age, reaching adult values at approximately 9 months of age. It was determined that quadratic equations best predicted the conduction velocities during maturation.

The study of pigs as a human disease model has been conducted in neuroscience. But the morphological development of pig brain by using MRI is rare. The purpose of this study is to determine whether cerebellum maintains consistent proportion to other brain regions in aging. Clinically healthy sixteen micropigs, 1, 2, 4, and 8 months were studied. The micropigs were anesthetized with isoflorane. MRI was acquired using a 0.3T system. To figure out development of ratio that allowed identification of normal cerebellum size, we measured the area of the cerebellum, brainstem, and forebrain from the mid-sagittal brain images on T1W. Mid-sagittal cross-sectional area (CSA) of total brain, forebrain, brainstem, and cerebellum were expressed as absolute values and also as percentages which were compared between the four age groups of micropigs for the purpose to define the effect of age on brain morphometry. It was found that there was not a significant difference in the percentage of the brain occupied by an individual region between groups although the absolute CSA differed significantly among age groups. There was no effect of age on the ratio between the cerebellum and total brain in 4 age groups. The normal size of cerebellum changes during brain development maintained a consistent ratio to other brain regions in normal micropigs. The ratio of CSA quantified on the mid-sagittal MR images offers a suitable method to detect presence of cerebellar anomalies in micropigs.

Recordings of averaged brain stem auditory-evoked potentials were obtained from 13 Beagle pups of both genders to document the postnatal development of the response from age 1 to 76 days. Responses were recorded between needle electrodes placed on the vertex and the ipsilateral ear, with ground at the interorbital line. Recordings were performed without sedation. Low-amplitude responses to high-intensity stimuli could be recorded from animals prior to opening of the ear canals. Peak latencies did not change after day 20 for peak I, day 30 for peaks II and III, and day 40 for peak V. As a result, the interpeak latencies between peaks I and III did not change after day 30, but continued to decrease until day 40 for peaks III-V and I-V. Peak amplitudes reached plateau values by day 20 (peak I) or day 30 (peaks II, III, and V). All of the measured latency and amplitude values had significant (P <0.001) linear regression lines of latency vs age and amplitude vs age. The brain stem auditory-evoked potential thresholds were mature by day 20.

To examine the postnatal development of equine small intestine, biopsy specimens of jejunal mucosa from 8 ponies, between 6 and 28 weeks old, were subjected to analytical subcellular fractionation and assay of organelle marker enzymes. Fractionation revealed a reduction in the particulate brush border component of beta-galactosidase (lactase) activity between 6 and 28 weeks, and a corresponding increase in soluble activity, although the reduction in mean specific activity was not significant. There also was a decrease in the proportion of brush border to soluble aminopeptidase N activity, a relative loss of brush border gamma-glutamyltransferase activity, and a considerable decrease in the specific activity of alkaline phosphatase throughout the gradient fractions. In contrast, there were marked increases in activities of (alpha-glucosidase (maltase) and sucrase in the older ponies, accompanied by considerable changes in the intracellular distribution of particulate alpha-glucosidase activity, which was predominantly associated with endoplasmic reticulum at 6 weeks, whereas the large increase in activity observed by 28 weeks was clearly associated with the brush border. The modal density of brush borders also increased with age, suggestive of an increase in the glycoprotein-to-lipid ratio of the microvillar membrane. In contrast to these brush border changes, there was relatively little alteration in the activities or density distributions of marker enzymes for endoplasmic reticulum, basolateral membranes, mitochondria, or lysosomes. These findings indicate that maturation of equine intestinal epithelium during the first few months of life results in major changes in the properties and enzyme composition of enterocyte brush borders.

Six foals were deprived of colostrum for the first 36 hours after birth and, instead, received reconstituted powdered milk. Five control foals suckled their dams naturally. Blood samples were obtained from all the foals after birth and at approximately weekly intervals until at least 5.5 months of age. Sera were analyzed for hemolytic complement activity, complement component C3, and correlating IgG concentration. Hemolytic complement (P = 0.0145) and C3 (P = 0.0002) values were significantly higher in colostrum-deprived foals (CDF) than in naturally nursed foals at 2 to 5 days of age. In addition, significantly (P = 0.0149) higher IgG concentration was found in CDF than in naturally nursed foals between 3 and 5.5 months of age. It was concluded that the observed high complement activity in CDF within 2 to 5 days of age may provide an alternative in immune defense for IgG-deprived foals after failure of colostral transfer.

Drug-metabolizing enzyme activities were measured in livers from calves fed commercial milk replacer (nonfunctioning rumen [veal]), and those fed milk replacer supplemented with whole grain and hay from the first week of age (functioning rumen [ruminating calves]). After birth, cytochrome P450 and its NADPH-dependent reductase activities remained unchanged in veal calves; in ruminating calves they increased almost 50%. Cytochrome P450-mediated reactions, such as aniline hydroxylase activity, tripled in ruminating calves, but remained unchanged in veal calves. In both groups of calves, coumarin hydroxylase and 7-ethoxycoumarin 0-deethylase activities increased after birth, but maturation rates and activity values in ruminating calves were considerably greater than those of veal calves. The aminopyrine N-demethylase activity for veal calves was equal to that of calves with functioning rumen. Uridine diphosphoglucuronic acid glucuronyl transferase and glutathione-S-transferase activities also were higher in calves with functioning rumen than in veal calves. This increased activity in calves with functioning rumen probably represents a response to environmental exposure to xenobiotics. Compared with rumen-functional calves, bob veal (0 to 3 weeks old) and fancy veal (15 to 19 weeks old) calves fed commercial milk replacer have a significantly (P = 0.05) diminished capacity for metabolizing drugs and other xenobiotics. From a regulatory perspective, the variance in drug-metabolizing enzyme activities within these different market classes of calves suggests that specific studies designed to determine drug residue-depletion times in veal calves may be needed.

The F waves evoked by supramaximal stimulation of distal tibial nerve were evaluated in chickens aged 2 to 15 weeks. Latency of these potentials increased from mean +/- SD 11.4 +/- 0.12 ms at week 2 to mean 12.88 +/- 0.65 ms at week 15. The F-wave latency increased linearly with age. When this latency was collected for a standard distance to compensate for the increasing limb length with age, latency decreased with maturity.

The development and availability of follicles is an indicator to predict which of the follicle sizes are suitable to recover the oocytes assisted by means of ultrasonography of transvaginal oocyte retrieval (OPU). Thus, the study was done in order to characterize the follicular recruitment and distribution in response to the repeated removal of follicles, and thus to determine the availability of follicles and immature oocytes harvested repeatedlyfor two consecutive days of OPU in beef cows. Results indicated that 24-h OPU showed significantly greater numbers of medium and large follicles than small categories (P<0.05). However the 48-hr of OPU does not showed any differences of follicles categories (P>0.05). The mean total number of follicles and immature oocytes recovered were higher (P<0.05) in 24-hr OPU (13.76±1.2 and 7.38 ± 1.7) compared to 48-hr OPU (9.08 ± 1.5 and 3.54 ± 1.00) with the oocyte retrieval rate of 51.22% and 38.17%, respectively. The morphological classification indicated the 24-hr oocyte retrieval produced 62% of suitable immature oocytes that can be used for in vitro embryo production. In conclusion, the repeated removal of two consecutive days of OPU has averted the development of dominant follicle, and thus, gave an atmosphere to the subordinate follicles to continue growth relatively to an equal proportion of small, medium and large categories of follicles. Due to the reduction of follicle and recovery rate at 48-hr it is suggested that OPU be carried out later than 48 hour so that the follicle has more time to increase the diameter size.

Extrusion of Na, K-ATPase and transferrin receptor with lipid raft-associated proteins in different populations of exosomes during reticulocyte maturation in dogs

The present study characterizes canine reticulocyte exosomes. Exosomes are small membrane vesicles involved in membrane remodeling that are released from reticulocytes during the final maturation step of red blood cells. The vesicles collected from reticulocyte culture supernatants by differential centrifugation contained major exosomal proteins including heat shock protein cognate 70 (Hsc70) and transferrin receptors (TfR), consistent with the definition of the exosome. In addition, the Na,K-ATPase alpha-subunit and stomatin, a lipid raft-associated protein, were extruded by the exosome pathway, possibly leading to the absence of these proteins in erythrocytes, while the major protein constituents of erythrocyte membranes, spectrin and band 3 were retained in reticulocytes and not expelled into exosomes. The Na,K-ATPase alpha-subunit, as well as TfR and about half of the stomatin contained in exosomes, was obtained in a detergent-soluble fraction that was distinct from the lipid raft micro domain. Moreover, Na,K-ATPase and a portion of stomatin were distributed differently to Hsc70, TfR, stomatin, and ganglioside Gsub(M1) in vesicles separated with sucrose density gradient centrifugation. These results demonstrate that a heterogeneous group of exosomes participates in the loss of Na,K-ATPase and membrane remodeling during reticulocyte maturation in dogs.
