The effect of melatonin on testicular circumference and semen characteristics in non-breeding season in Lori-Bakhtiari ram

BACKGROUND: This study was conducted to evaluate the effect of melatonin on semen quality and quantity changes and increase of reproductive efficiency in Lori-Bakhtiari ram in the non-reproductive season. OBJECTIVES: The purpose of this study was to investigate the effect of melatonin on testicular size and semen qualitative and quantitative changes in the non-breeding season is Lori-Bakhtiari rams. METHODS: This study was designed to evaluate the effects of melatonin on the quantity and quality of semen and testicular diameter in 8 Lori- Bakhtiari rams (4 control rams and 4 treatment rams) for 10 weeks during spring 2013. Each ram in the treatment group was received three tablets of 18 mg implantable melatonin subcutaneously at the base of ear after 5 weeks from the start of the trial. RESULTS: Semen parameters and testicular diameter measurements were evaluated on a weekly basis. No significant differences were observed between 2 groups before melatonin implanting. A significant difference was observed after melatonin implanting between 2 groups in the mean of testicular diameter (treatment: 33.5±0.25 and control: 30.67±0.07 cm), in semen volume (treatment: 1.34±0.03 and control: 0.87± 0.04 ml) and in sperm concentration (treatment: 1.76±0.05 and control: 1.37±0.04 109/ml) (p<0.05). However, other parameters including average of lives sperm percentage, total sperm motility, sperm progressive movement and percentage of abnormal sperm were not significant (p>0.05). CONCLUSIONS: The results of this experiment showed that the use of melatonin in the non-reproductive season resulted in increase of testicular size, semen volume and sperm concentration in Lori-Bakhtiari ram. However, the rest of the semen parameters have not been affected.

BACKGROUND: Heat stress is very important in poultry production. OBJECTIVES: To evaluate the effect of oral elatoninsupplementation and increasing of dark period on biochemical profile of broiler chickens under Heat Stress (HS). ETHODS: A total of 200 day- old broiler (Arian) chicks were randomly allocated in to 4 groups (dark ,melatonin, negative and ositive controls). Melatonin group received 40 mg melatonin/kg diet from 30-40 days of age and chickens of dark group were xposed to a special lighting schedule from 10-40 days of age. All groups (except negative control) were daily exposed to HS (39± 1 c) for 6 hours per day from 35-40 days of age. At the end of 6 weeks, body weight was measured. At 30, 35, and 40 days, serum uric acid, glucose, total protein, albumin, globulin, activity of AST and creatinine were measured. atawas analyzed by ANOVA and Tukey test in SPSS software. RESULTS: In 30 days, there was no significant difference etween groups. In the day 35, only serum uric acid of melatonin group was significantly increased in comparison with egative control. Also, serum uric acid of melatonin group increased significantly in 40 days in comparison with other roups, as melatonin plays an important role in the antioxidant defense system and uric acid is an important antioxidant inchickens. Serum glucose of positive control compared to other groups significantly increased in 40 days. Other biochemical parameters were not significant. In this study,the mean body weight of dark group was significantly decreased. The percentage of mortality rate in negative control, dark, melatonin and positive control treatments was zero, 4 ,5 and 10% respectively. CONCLUSIONS: These results demonstrate that dark period and oral melatonin supplementation candecrease HS.

A dog (5.6 kg, 4-year-old, castrated male, Miniature pinscher) with diffuse symmetric alopecia on the chest and excessive scales was referred to Veterinary Medical Teaching Hospital of Chungnam National University, Korea. On physical examination, diffuse alopecia on ventral aspect of the neck and caudomedial thighs, and severe scales were presented. Dermatologic tests of lesions revealed bacterial infections. Histopathologically, atrophy of hair bulb was observed in the severely alopecic lesion of caudomedial thighs. ACTH stimulation test was performed to differentiate the endocrine disorder related to generalized alopecia. The pre-ACTH serum testosterone and progesterone concentrations were above normal ranges and the post-ACTH serum testosterone concentration was high. The pre- and post-ACTH serum estradiol and cortisol concentrations were within normal ranges. Canine pattern alopecia was diagnosed based on history, physical examination, hormonal assay and dermatohistopathologic examination. After 3 months of melatonin administration, multifocal alopecia on the trunk was improved and general hair regrowth was identified.

OBJECTIVE To determine the effect of orally administered melatonin on the intraocular pressure (IOP) of ophthalmologically normal dogs. ANIMALS 20 ophthalmologically normal dogs (40 eyes). PROCEDURES In a randomized crossover study, each dog received a 7-day regimen of melatonin (0.1 to 0.2 mg/kg, PO, q 12 h) and a placebo (150 mg of lactose powder in a capsule, PO, q 12 h), with a 7-day washout period between treatment regimens. Rebound tonometry was used to measure the IOP in both eyes of each dog 5 times at 2-hour intervals on days 0 (before administration of the first dose), 2, 4, and 7 (after administration of the last dose) of each treatment period. Repeated-measures ANOVA was used to evaluate the effects of treatment, day, and IOP measurement time within day on IOP. RESULTS Intraocular pressure was not significantly associated with treatment but was associated with day and the interaction between day and IOP measurement time within day. The mean ± SD IOP was 14.26 ± 2.95 and 14.34 ± 2.69 mm Hg for the melatonin and placebo regimens, respectively. Within each treatment period, the mean IOP tended to decrease from day 0 to 7 as well as within each day, which was attributed to the dogs becoming acclimated to the study protocol and natural diurnal variations in IOP. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that oral administration of melatonin (0.1 to 0.2 mg/kg, q 12 h for 7 d) did not significantly affect the IOP of ophthalmologically normal dogs.

This study aimed to evaluate the effect of melatonin and melatonin-loaded chitosan nanoparticle (CMN) supplementation to maturation media on buffalo oocyte maturation rate and relative expression of genes: growth differentiation factor 9 (GDF9), bone morphogenetic protein 15 (BMP15), B Cell Lymphoma 2 (BCL2), Associated X protein (BAX) and superoxide dismutase 1 (SOD1). Buffalo ovaries were heaved from Al-Mounib slaughterhouse, cumulus-oocyte complexes (COCs) were in vitro matured in three different media, TCM-199 medium (control), TCM-199 with melatonin 10-9M, and TCM-199 with CMN 10-9M. The assessment of the nuclear maturation rate was carried out through the presence of the first polar body. In addition, the mature buffalo oocytes were stored on RNA later for genetic analysis of GDF9, BMP15, SOD1, BCL2, and BAX genes using quantitative real-time PCR (qRT-PCR). The results were reported that buffalo oocytes supplemented with melatonin-loaded chitosan nanoparticle (CMN) or melatonin have a significant effect on nuclear maturation rate 94.04±0.65 and 88.74±0.77 respectively when compared with buffalo oocytes matured with basic media (control) 79.67±1.35. Furthermore, buffalo oocytes supplemented with melatonin-loaded chitosan nanoparticle (CMN) or melatonin showed significant upregulation of GDF9, BMP15, SOD1, and BCL2 genes and significant downregulation of BAX gene when compared with oocyte matured with basic media (control). In conclusion, the results of nuclear maturation rate and relative expression pattern of GDF9, BMP15, SOD1, BCL2, and BAX reflect that melatonin-loaded chitosan nanoparticle (CMN) and melatonin` may play an important role in the buffalo oocytes developmental competence.

Objective—To investigate the in vitro effect of the combination of lignan enterolactone (ENL) or lignan enterodiol (END) with melatonin on steroid hormone secretion and cellular aromatase content in human adrenal carcinoma cells. Sample—Human adrenocortical carcinoma cells. Procedures—Melatonin plus ENL or END was added to cell culture medium along with cAMP (100μM); control cells received cAMP alone. Medium and cell lysates were collected after 24 and 48 hours of cultivation. Samples of medium were analyzed for progesterone, 17-hydroxyprogesterone, androstenedione, aldosterone, estradiol, and cortisol concentration by use of radioimmunoassays. Cell lysates were used for western blot analysis of aromatase content. Results—The addition of ENL or END with melatonin to cAMP-stimulated cells (treated cells) resulted in significant decreases in estradiol, androstenedione, and cortisol concentrations at 24 and 48 hours, compared with concentrations in cells stimulated with cAMP alone (cAMP control cells). The addition of these compounds to cAMP-stimulated cells also resulted in higher progesterone and 17-hydroxyprogesterone concentrations than in cAMP control cells; aldosterone concentration was not affected by treatments. Compared with the content in cAMP control cells, aromatase content in treated cells was significantly lower. Conclusions and Clinical Relevance—The combination of lignan and melatonin affected steroid hormone secretion by acting directly on adrenal tumor cells. Results supported the concept that this combination may yield similar effects on steroid hormone secretion by the adrenal glands in dogs with typical and atypical hyperadrenocorticism.

Melatonin, the principal hormone of the vertebral pineal gland, participates in the regulation of cardiovascular system in vitro and in vivo. However, the effects of melatonin on vascular tissues are still vague. The aim of this study was to assess the relationship between phospholipase C (PLC) and nitric oxide synthase (NOS)/cyclic guanosine 3',5'-monophosphate (cGMP) signaling cascade in the relaxatory action of melatonin in isolated rat aorta. Melatonin induced a concentration-dependent relaxation in phenylephrine (PE)- and KCl-precontraceted endothelium intact (+E) aortic rings.

Melatonin, the principal hormone of the vertebral pineal gland, participates in the regulation of cardiovascular system in vitro and in vivo. Lithium inhibits both inositol polyphosphate phosphatase (IPPase) and inositol monophosphatase (IMPase), which are involved in a wide range of signal transduction pathways. The aim of the present study was to assess the effect of lithium on endothelial-dependent relaxation to melatonin and on the melatonin-induced inhibition of contraction by phenylephrine (PE) in isolated rat aorta. Melatonin induced a concentration-dependent relaxation in PE-precontracted in endothelium-intact (+E) aortic rings.

The effects of melatonin and Light treatments on the reproductive performance of yearling Suffolk rams were investigated. The two groups (A, n = 8; B, n = 8) were given a priming period of long days (18 h Light (L): 6 h dark (D) during 1st February to 14th March. During 14th March to 9th September, group A was exposed to local light + melatonin Implants (Regulin) at 5 weekly intervals, group B was exposed to local light + melatonin Fed (M- 5250) alongwith the feed. The group (C, n = 8) in the local light during 1st February to 14th March and during 14th March to 9th September remained on local light + melatonin implants (Regulin) at 5-weekly intervals. However, the group (D, n=8) was treated as control and kept on local light environment (Lat. 51 degree 43'N) throughout the experiment. Testis diameter, sexual behaviours and semen quality were used to assess reproductive performances. The treatment to group A & B resulted in a significant advancement in high reproductive performance. The treatment to group C indicated that the abrupt application of melatonin in mid March resulted in a weak response in terms of all the criteria of assessment. The investigation indicated that seasonal fluctuations in reproductive behaviour and semen quality may be rephrased by the application of light and melatonin treatments and that these treatments may be considered for the preparation of rams for out of season breeding.
