Genetic diversity, virulence profile of Campylobacter coli and Campylobacter jejuni isolated from poultry and human in Assiut governorate, Egypt

Thermotolerant Campylobacter genus is one of the most prevalent causes of gastroenteritis in humans, especially C. coli and C. jejuni. Despite the importance of Campylobacter diagnosis to public health, many laboratories continue to adopt the slow, inaccurate conventional culturing approach, which leads to false-negative/positive results. The origin, transmission, pathogenicity, and pathophysiology of Campylobacter spp. diseases are poorly understood. Therefore, in this study, the samples were collected over a period from August 2021 to September 2022; about 100 poultry samples and 43 stool specimens from children were collected. According to conventional culturing techniques, the overall prevalence of the Campylobacter genus in both poultry and humans was determined to be 31.5%, whereas PCR analysis of poultry (30) and human specimens (43) for Campylobacter genus revealed a 35.6% isolation rate. While C. coli was the only species detected in poultry-positive Campylobacter genus samples demonstrated by 27.3%, the human-positive Campylobacter’s isolates were C. coli with 33.3%, C. jejuni and mixed infection with 6.7%. Shannon and Simpson biodiversity indexes quantify genetic diversity; assuming that Campylobacter species express virulence genes differently, we found that C. coli had a higher Shannon diversity index (0.8487) and Simpson index (0.4938), while C. jejuni had (0.6931) for Shannon and (0.5) for Simpson index. Regarding host-virulence genes diversity, human-derived strains had a higher Shannon diversity index (1.474) and Simpson index (0.75) than poultry. This study provided evidence that the genetic profiles of circulating species of Campylobacter differ depending on the origin, highlighting the need for genetic diversity knowledge for effective management and prevention strategies.

Meat contamination is a problem that must be addressed since it can affect consumers' health. Standard acceptable limits for heavy metals in meat and organs have been established by many international organizations. Livestock including sheep, cattle, camels, and goats side by side with other poultry species are the most significant sources of meat for protein. Heavy metal contamination in animal products may be harmful to people's health. The earth's crust naturally contains heavy metals, but uncontrolled human activity has significantly changed their geochemical cycles and biological equilibrium. This causes a buildup of metals in animal tissue that contain secondary metabolites, which gives rise to a specific pharmacological action. Humans can have negative health effects from prolonged exposure to heavy metals as lead, cadmium, arsenic, and mercury. This review showed that most of the studies conducted in different countries on heavy metals were mostly in red meat, and the research was few in the cases of domesticated birds and very rare in game birds, although it is considered a good example for investigating heavy metals in the environment

Up to now, there has been limited information regarding the circulation of pathogenic Listeria species in poultry and their contact handlers. The present study was, therefore, planned to evaluate the potential existence of Listeria spp. in different poultry species and their products as well as their contact workers at Dakahlia Governorate, Egypt and to address the antimicrobial profile of Listeria isolates using classical microbiological techniques and multiplex PCR targeting three virulence-associated genes (iap, hlyA, and actA). The study included cloacal swabs from different poultry species (n =200), poultry carcasses (n = 25), poultry products (25 samples each of chicken luncheon and frozen chicken breast fillets), chicken eggs (n = 50) and stool specimens from the contact workers (n = 25). Findings showed that 60 samples out of 350 (17.14%) were found to be positive to Listeria spp., which were identified as follow: L. innocua (28.3%), L. ivanovii (28.3%), L. monocytogenes (15%) and L. grayi (15%), L. seeligeri (10%) and L. welshimeri (3.3%). L. monocytogenes showed high resistance to nalidixic acid (100%) followed by neomycin (85.1%), streptomycin (80%), cefotaxime (65%) and penicillin g (58.3%). Results could likely suggest that poultry and poultry products had a significant role in dissemination and transmission of virulent and multidrug resistant L. monocytogenes which represent a potential risk particularly in the absence of strict hygienic practices and preventive measures. A constant monitoring of pathogenic L. monocytogenes is thus urgently needed to avoid the dissemination of such pathogenic strains in food production chains.

A study was carried out to detect and identify the presence of coccidia oocysts in the faeces of village chicken from the district of Pasir Putih, Kelantan, West Malaysia. A total of 135 fecal samples were collected from 15 areas in the Pasir PutihDistrict. The faecal samples were examined by direct smear method (qualitative study). A pinch of the faeces was put onto the glassslide with 1-2 drops of normal saline and cover slip, which was then observed under the compound microscope to detect thecoccidia oocysts. The presence of coccidia oocyst was then identified by its size and shape. Results showed that ten out of 135 samples were positive for coccidia oocysts, and classified as Eimeria maxima and Eimeria mitis, both of which are from two locations at Kampung Chap Banir, Pasir Putih, Kelantan. The remaining 125 samples were observed to be negative. This may suggest that the chickens reared in the backyard (extensive)are less susceptible to the coccidia infection due to their environment with lower stocking density (mostly free ranging chicken), and no damp/wet litter as bedding which canfacilitate sporulation of the coccicia oocyst thereby spreading the infection. Further studies need to be done to elucididate other factors which may affect coccidial infections in free range chicken such as the availability of medications in feed or genetic hardiness and tolerance to field infections. The localvillage chicken industry is an up and coming facet of the poultry industry and needs concerted efforts to boost it.

Data over a period of eleven years was analysed for Infectious Bursal Disease (IBD) virus isolated from chicken samples submit ted to the Regional Veterinary Laboratory at Bukit Tengah, Malaysia (RVLBT) for diagnosis. A total of 247 suspect IBD cases were tested by Virology Section, RVLBT between years of 2006 to 2016. IBD virus has been isolated by using Agar Gel Precipitation Test (AGPT), a bursal homogenate which has been used as an antigen against a known positive antiserum. About 27 cases (11%) from a total of 247 suspect cases in chickens were positive for the presence of IBD. The rate of IBD may be influenced by age of chickens with an increase in the possibility of IBD occurring in chicken older than 3 weeks. Apart from that, both broiler and local chickens are highly susceptible to this disease. Therefore, awareness on the existing IBD cases indicates the importance of strict management procedures, proper management programmes, vaccination and immunisation for chickens in Malaysia.
Mostrar más [+] Menos [-]Towards an improved estimate of antimicrobial use in animals: Adjusting the "population correction unit" calculation

International comparisons of animal antimicrobial use (AMU) have typically been based on total national estimates of antimicrobials sales standardized by the national animal biomass calculated as the population correction unit (PCU). The objective of this paper was to compare the currently accepted PCU calculation with that of the adjusted population correction unit (APCU), which re-evaluates the standard animal weights used in the calculation and accounts for animal lifespan. The APCU calculation resulted in substantial changes to the 2009 national biomass estimates for cattle, pigs, and poultry in 8 European countries and Canada. The estimated national biomass for cattle increased 35% to 43%, while the estimated national biomass of pigs and poultry typically decreased by approximately 51% and 87%, respectively. Among the 9 countries, the total national APCU ranged from an increase of 1% to a decrease of 40% relative to PCU, and these differences were statistically significant. Adjusted population correction unit is preferred over PCU in comparing and contrasting AMU among animals with different lifespans because it is more transparently derived and is a reasonable approximation of the animal biomass at risk of antimicrobial treatment.

Avian influenza viruses (AIV) have been isolated from a wide range of domestic and wild birds. Wild birds, predominantly ducks, geese and gulls form the reservoir of AIV in nature. The viruses in wild bird populations are a potential source of widespread infections in poultry. Active surveillance for AIV infection provides information regarding AIV distribution, and global AIV surveillance can play a key role in the early recognition of highly pathogenic avian influenza (HPAI). Since 2003 in Korea, there have been four H5N1 HPAI outbreaks caused by clade 2.5, 2.2 and 2.3.2. Therefore, improvement of AIV surveillance strategy is required to detect HPAI viruses effectively. This article deals with the major events establishing the role of wild birds in the natural history of influenza in Korea. We highlighted the need for continuous surveillance in wild birds and characterization of these viruses to understand AIV epidemiology and host ecology in Korea.

This study was conducted to investigate biochemical properties and O group serotypes of pathogenic 203 Escherichia (E.) coli isolates from poultry with collibacillosis in Korea during the period from April 2003 to December 2005. Biochemical and fermentative properties of 203 isolates of E. coli tested were in accordance with Cowan and Steel's classification standard. One hundred and forty one isolates (69.5%) could be classified into a total of 20 O serotypes. Among them, the predominant O groups were O78 (32.5%), O88 (7.8%), O15 (6.8%), O141 (6.4%), and O158 (3.0%) in decreased order. Other infrequently encountered serogroups included: O8 (2%), O161 (2%), O20 (1.5%), O125 (1.5%), O2 (1%). And O6, O18, O24, O46, O76, O109, O119, O138, O139 and O148 had a frequency of 0.5%, respectively. Sixty two isolates (30.5%) were non-typeable with standard 173 O antisera used in this study.

Antimicrobial drugs are widely used in poultry industry as growth promoters or to control infectious diseases. However, this practice is reported to have caused high resistance to antimicrobial drugs in normal chicken flora and pathogens. Antimicrobial resistance to Escherichia coli (E. coli) from chicken has been mainly reported in normal flora, but rare in pathogenic organism in Korea, recently. Therefore, this study was conducted to investigate prevalence of antimicrobials resistance, transfer of R plasmid, and association between antimicrobial drug resistance and O serotype of 203 pathogenic E. coli from poultry in Korea during the period from April 2003 to December 2005. These isolate showed a high resistance to tetracycline (Tc, 92.6%), streptomycin (Sm. 81.8%), ampicillin (Ap, 77.3%), ciprofloxacin (Ci, 70.9%), sulfisoxazole (Su, 66.5%), and trimethoprim (Tp, 58.1%). Two hundred-one (99.0%) of the isolates were resistant to one or more drugs. They showed 57 different resistant patterns, and the most prevalent resistant pattern among them was Tc, Sm, Su, Ap, Tp, Ci, Na. Sixty-eight (33.8%) of the isolates transferred all or a part of their antimicrobial resistant pattern to the recipient strain by R plasmid. The most common antinicrobial resistant pattern was Tc, Sm, Su, Ap, Tp, Ci, Na in serotype O78, O88 and O15, respectively. These results exhibit high individual and multiple resistance to antimicrobials of pathogenic E. coli from poultry in Korea. They also suggest the needs for surveillance to monitor antimicrobial resistance in pathogenic bacteria that can be potentially transmitted to humans from food animals and to regulate the abuse of antimicrobials in food-producing animals in Korea.

The B1 strain of Newcastle disease virus (NDV-B1), which is nonpathogenic for newly hatched chickens, killed embryos when it was used to inoculate chicken eggs at embryonation day 18. Treatment of NDV-B1 with an alkylating agent, ethylmethane sulfonate (EMS) markedly reduced the pathogenicity of the virus for 18-day-old chicken embryos. Eggs inoculated with the modified virus (NDV-B1-EMS) hatched, and the virus was isolated from lungs and spleen of 1-day-old chickens. The hatched chickens developed antibody to NDV and were protected against challenge exposure (at 4 weeks of age) with a highly virulent GB-Texas strain of NDV. Presence of maternal antibody to NDV in embryonating eggs did not influence the protective ability of NDV-B1-EMS, which also induced protective immunity when administered to 4-week-old chickens. The 50% protective dose of NDV-B1-EMS in maternal antibody-negative and -positive embryos was calculated to be 10.77 and 17.70 embryo 50% lethal doses, respectively. Results of the study indicated that NDV-B1-EMS may be used as an embryo vaccine to protect chickens against Newcastle disease.
