Secretome improves testosterone and androgen-binding protein production in testicular dysfunction rats induced by cisplatin

Objective: This study was determined to see the effects of secretome on cellular production of testosterone and androgen-binding protein (ABP) using immunohistochemistry in testicle dysfunction due to cisplatin. Materials and Methods: Forty-eight rats were divided into four groups: the healthy group, the group with testicular dysfunction, the secretome-treated group with 0.2 ml/kg body weight (BW), and the secretome-treated group with 0.3 ml/kg BW. The immunohistochemistry staining method is used to find out testosterone and ABP reactivity in tissue organs. Results: Very strong testosterone and ABP immunoreactivity were found in Leydig cells of normal testes. While in the Leydig cells of cisplatin-induced testicles, testosterone and ABP immunore activity were not observed. Testosterone and ABP were observed 1 week after the second secretome injection. The number of testosterone-immunoreactive cells in the low dose group from week 1 to 3 was 0, 19, and 32, respectively. From week 1 until week 4, the high dose group was 0, 29, 33, and 65, respectively. The number of ABP-immunoreactive cells from the first week until the third week in the low dose group was 0, 28, and 34, respectively. The high dose group from the first week until the fourth week was 0, 26, 58, and 83, respectively. The number of cells that produce testosterone and ABP increased from week 2 to 4. Conclusion: The administration of secretome could increase the number of immunoreactive cells toward testosterone and ABP in testicular dysfunction. [J Adv Vet Anim Res 2021; 8(4.000): 687-694]

Objective-To quantitatively assess distraction-induced bone formation in a crural lengthening model in dogs by use of delayed-image bone scintigraphy. Animals-12 mature Labrador Retrievers. Procedure-Dogs were randomly allocated to 1 of 3 groups. A circular external skeletal fixation system was mounted on the right crus of each dog. Osteotomy of the distal portion of the tibia and fibula was performed in groups 1 and 2 and was followed by a lengthening procedure of 10 mm in the first group only. The third group served as sham-operated controls. Delayed-image bone scintigraphy with technetium-99m hydroxy methylene diphosphonate was performed 2, 4, and 6 weeks after surgery. Delayed-image-to-region-of-interest, delayed-image-to-crural, and delayed-image-to-femoral scintigraphic activity ratios were calculated. New bone formation was quantified by use of densitometric image analysis, and values for the scintigraphic ratios were compared. Results-In the distraction and osteotomy groups, delayed-image-to-region-of-interest and delayed-image-to-crural ratios increased significantly. Although densitometric image analysis revealed increased bone formation after distraction, the region-of-interest ratios and crural ratios were similar in both groups. All dogs had increased delayed-image-to-femoral ratios. Conclusions and Clinical Relevance-Delayed-image bone scintigraphy ratios were not effective at differentiating between the amounts of distraction-induced bone and osteotomy-induced bone. Metabolic bone activity in the adjacent femur was increased as a consequence of circular external skeletal fixator placement. Delayed-image bone scintigraphy was not adequately sensitive to quantitatively monitor bone formation but may be useful as an early predictor of bone healing.

Study of the regeneration processes and raising the efficiency of rape (Brassica napus) organs and tissues cultivation in vitro methods, as well as the achievement of a stable yield of proper regenerants were realized in the conditions of the Republic of Belarus. The obtained data specified the possibility of generation of rape finished regenerants in vitro culture without additional passages and transfers by means of effectively selected concentrations of phytohormones and biologically active substances, which made it possible to realize principles of totipotency and polarity of crop vegetative organs. On the explant part which was submerged into culture medium there was formed a root, and on opposite one - the morphogen structures. The possibility of obtaining rape regenerants from hypocotyls segments by straight regeneration was studied. Application of the analyzed substances considerably simplified and reduced the price of process of full regenerants formation, and made it possible to receive a wide spectrum of various forms that was important for selection process due to the opportunity to select valuable (in protein content) samples for the further studies and their inclusion into production process
