A Molecular and Serological Study of Toxoplasma gondii Infection in Slaughtered Sheep in Mashhad Area

BACEKGROUND: Toxoplasmosisis one of the most important zoonotic diseases in Iran and the world. OBJCTIVES: Due to the high consumption of lamb meat and the high frequency of Toxoplasma infection in sheep in Iran, the aim of study was to determine frequency of Toxoplasma infection in the slaughtered sheep of Mashhad area. METHODS: In order to do this study, from summer 2015 to spring 2016, 25 blood and 25 heart muscle samples were seasonally collected from Torghabae slaughterhouse in Mashhad area. The samples were transferred to parasitology laboratory. First, the blood samples were centrifuged and the serum samples were isolated, then a portion of the heart muscles sample was taken for PCR examination. The sera and muscles samples were kept at -20 ºC in freezer until examination time. The sera samples were examined to detect antibody against T.gondii by ELISA method. DNA of heart muscle was extracted by commercial extraction kit and was examined to detect Toxoplasma DNA by nested –PCR. RESULTS: In the present study, of 100 sampled sheep, only 1 (1%) of the serum samples was seropositive, while 22 (22%) of the DNA samples were PCR positive. In this study, the highest frequency of Toxoplsma PCR-Positive was seen in spring and the lowest in summer in sheep. Also, the result of this study showed that the agreement between the molecular and EISA method was “fair”. CONCLUSIONS: Based on the high frequency of Toxoplasma infection in heart muscle of sheep, it seems that the risk of transmission of Toxoplasma infection from sheep meat is high.

BACKGROUND: Congenital Toxoplasma gondii infection during pregnancy due to reduced maternal immune system could increase the risk of a child's autism disorder. OBJECTIVES: The aim of this study was to compare the Toxoplasma gondii infection in children with autism and normal children. METHODS: The research method was a case control study. The participants included 50 autistic children and 50 normal (N=100) children, aged between 3 and 12 years-old. They were matched according to age, socio-economic status, severity of the disorder, lack of physical and mental illness and other criteria considered in this research. From each group 5ml blood samples were collected to assess the prevalence of antibody against Toxoplasma gondii. To investigate the research hypotheses, the data were analyzed by Chi-square test. RESULTS: The results showed that children with autism had a higher level of frequency contamination with Toxoplasma gondii parasite infection than normal group (p < /em><0.001). CONCLUSIONS: Infection rate of Toxoplasma gondii in children with autism was significantly higher than in normal children.

BACKGROUND: Toxoplasma gondii is an obligate intracellular protozoon with worldwide distribution that infects human and a wide spectrum of warm- blooded animals. OBJECTIVES: The aim of the study is to survey the prevalence of T.gondii antibody in slaughtered livestock in slaughterhouse with modified agglutination test in Sanandaj in 2015. Methods: Totally 343 serum samples were collected from 130 cows, 111 sheep and 102 goats from May to July 2015 in Sanandaj slaughterhouse. Sera were examined with modified agglutination test (MAT) for T.gondii antibody and 1.40 titer antibody and above was considered as positive. Results: Antibodies of T.gondii were found in sera of 51 out of 343 (15.45%) samples. From 51 seropositive sera, 13 cows (23.84), 13 goats (12.74%) and 9 sheep (8.1%) were infected. The differences among species were statistically significant (P≥.002). Conclusions: Results indicate that T. gondii in sheep, goats and especially cows is prevalent, so consumption of undercooked meats should be avoided, also the prevention methods of infection should be taught to the people and due to the possibility of limitations in the results of serological tests, repetition of the study using molecular techniques is recommended

Characterization of Toxoplasma and Trichinella isolates from muscles of black bears in Pennsylvania.

During the hunting season of 1992, 322 black bears from Pennsylvania were examined for Toxoplasma gondii- and Trichinella spp-induced infections. Toxoplasma gondii antibodies were found in 79.8% of 322 bears--titer < 1:25 in 65 (20.2%), 1:25 in 18 (5.6%), 1:50 in 11 (34.5%) and 1:500 in 128 (38.7%) bears--by use of the modified agglutination test. Muscle tissues from 89 of these bears were bioassayed for T gondii parasites. Muscles from 64 bears, including heart from 1 bear, and heart alone from another bear, were digested in pepsin, and the digested samples were bioassayed in mice. Toxoplasma gondii was isolated from 5 bears; from the heart of 1, heart and skeletal muscles of 1, and skeletal muscles of 3. The T gondii antibody titers for the 5 bears with detectable T gondii were: greater than or equal to 1:25 in all 5 bears by use of the modified agglutination test; < 1:10 (3 bears, considered Toxoplasma-negative), 1:20 and 1:320 by use of the Sabin-Feldman dye test; < 1:64 (3 bears, considered Toxoplasma-negative), 1:128, 1:512 by use of the indirect hemagglutination test, and < 1:16 (2 bears, considered Toxoplasma-negative), 1:32, 1:64, and 1:512 by use of the latex agglutination test. Toxoplasma gondii was not isolated from feces of 5 cats fed muscles from the remaining 25 bears with T gondii antibody titer < 1:25. Tissue cysts of the 4 T gondii isolates from bears were rendered noninfective by freezing at -13 C. Antibodies against Trichinella spp were found in 6 (1.8%) of 319 bear sera; Trichinella spp larvae were detected in muscle digests of 2 of 63 bears, and in histologic sections of muscles from 3 of 162 bears. Genetic typing indicated that the 2 Trichinella isolates from bears were a sylvatic genotype and were not the species found in domestic pigs.

Toxoplasma gondii is a major neglected parasitic infection occurring in settings of extreme poverty in Africa. Apart from causing reproductive failure in animals it is also a significant zoonotic concern. The objective of this study was to determine the seroprevalence and associated risk factors of T. gondii infection in cats, chickens, goats, sheep and pigs in the southeast of South Africa, of which little is known. Sera was obtained from 601 domestic animals including 109 cats, 137 chickens, 128 goats, 121 sheep and 106 pigs managed under different production systems in different agro-ecological regions and evaluated by the Toxoreagent, a latex agglutination test for T. gondii antibody detection. Household-level and animal-level data were collected by interviewing animal owners and/or herders using a closed-ended questionnaire. The study revealed an overall farm seroprevalence of 83.33% (125/150 farms) with the highest rate of infection for the parasite found in sheep with 64.46% (78/121), followed by goats with 53.91% (69/128), pigs with 33.96% (36/106), cats with 32.11% (35/109 cats) and chickens with 33.58% (46/137). The risk factors that were found to be statistically significant (p < 0.05) to different species of seropositivites were age, location, climate, animal production system, rodent control, seropositive cat, cat-feed access and cat faecal disposal. The relatively high seroprevalence of T. gondii detected in this region suggests that domestic animals may pose a substantial public health risk through the consumption of T. gondii-infected raw meat as well as via contact with cat faeces.

Toxoplasma gondii is one of the most prevalent zoonotic protozoan parasites worldwide and affects the vast majority of warm-blooded animal species, including humans. Postnatal infection in humans occurs through the ingestion of sporulated T. gondii oocysts or via the oral intake of parasite tissue cysts during the consumption of raw or undercooked meat. In this regard, given their high exposure to oocysts, chickens (Gallus domesticus) raised on the ground constitute a potential source of T. gondii.

This is the first report of Toxoplasma gondii infection in black vultures (Coragyps atratus), which are obligate scavengers found throughout the Americas. Serum samples from 121 wild black vultures caught in urban areas of the city of Sao Paulo, Brazil, were tested for the presence of T. gondii antibodies using the modified agglutination test (MAT; cutoff point 1:5). T. gondii antibodies were found in 16 birds (13.2%), with titers of 1:5 (6 birds), 1:10 (8 birds), and 1:20 (2 birds).

The maned wolf (Chrysocyon brachyurus), an inhabitant of the Brazilian savanna, is considered the largest canid of South America and is classified as a near-threatened species by the International Union for Conservation of Nature (IUNC). The purpose of this study was to investigate the presence of antibodies against Toxoplasma gondii, -Neospora caninum, -Leishmania spp., and -Ehrlichia canis in free-ranging maned wolves in the northeastern region of the state of São Paulo, Brazil. Of the 17 serum samples tested by the indirect fluorescent antibody test (IFAT), 88.2% (15/17), 17.6% (3/17) and 52.9% (9/17) showed anti-T. gondii, -Leishmania spp., and -E. canis antibodies, respectively. None of the studied maned wolves tested positive for N. caninum antibodies. Our results indicated the exposure of free-ranging maned wolves to the agents in question. The presence of industrial complexes, extensive agriculture and habitat fragmentation in the northeastern region of the state of São Paulo puts these wild animals in proximity to urban areas, possibly contributing to the transmission of diseases between wild and domestic animals and human beings.
