Evaluation of stability and potency of live attenuated rinderpest vaccine of lapinized-avianized tissue culture strain origin for the establishment of expiration period

In this study, we evaluated the stability and potency of live attenuated rinderpest vaccines of lapinized-avianized tissue culture strain origin, which had been produced annually from 2005 to 2008. When immune responses to the vaccines were evaluated using two Holstein calves weighing 100~150 kg, neutralizinng antibody titer of 1 : 16 was induced at 21 days post vaccination. When calves were also inoculated with vaccines lots that had been stored for 39 months at 4℃, same level of antibody titer was observed. Using the virus titer test, we found that all batches of the vaccine that had been kept for 3, 10, 15, 22, 27, 34, 39, and 45 months showed no significant loss of titers, and fulfilled the requirement necessary (greater-than or equal to 3 logTCID∧50) to be used as the national rinderpest vaccine reserve in Korea. In this study, we demonstrated that stability and potency of the rinderpest vaccines were maintained over three years when kept at 4℃ storage. This indicates that it maybe feasible to extend the expiration period of this vaccine from one year to three years.

The level of antibody by ELISA,immunsosuppressive effect baes on the response of birds to Newcastle disease vaccination and damage to bursa of Fabricius by IBD vaccination were studied. The efficacy of six different IBD vaccination schedules were studied using intermediate and intermediate plus strains of vaccines either alone or in combination. In vaccinated groups, the sero-conversion of the vaccine virus was noticed during fourth week, reaching to the peak between eight to twelve weeks of age in different groups. Afterwards, there was a gradual decrease in the titres, by the end of 20th week (maximum period tested). There was no significant difference in the titres of different treatment groups. However, all the groups showed titres above protective level during the entire period of study. There was significant difference in bursa body weight (B-BW) ratios of vaccinated groups in comparison with control group. Histopathological studies of bursal sections revealed depletion of lymphoid follicles, presence of cystic spaces, edema and hemorrhages. The birds vaccinated with hot strain of IBD vaccine showed metaplastic changes, presence of foam cells with pronounced interfollicular fibrosis. The bursal scores were maximum in the groups vaccinated with hot strains of IBD vaccines.

Test vaccines comprised of inactivated water-in-oil emulsions containing various antigen levels were prepared using a non-pathogenic H5N1 avian influenza (AI) virus, A/duck/Hokkaido/Vac-1/04 (H5N1). The potencies of these test vaccines were evaluated by two experiments. In the first experiment, the triangular relationship among the antigen levels of test vaccines, the hemagglutination inhibition (HI) antibody response, and the protective effect against challenge with a highly pathogenic avian influenza (HPAI) virus, A/chicken/Yamaguchi/7/04 (H5N1), was confirmed. Then lasting immunity of chickens after a single-shot vaccination was confirmed in the second experiment. As a result, complete protection after the challenge was observed in chickens immunized by test vaccines with an antigen level of 160 HA units/dose or higher. Thus, it was ascertained that the minimum antigen level in the AI vaccine was 160 HA units/dose, and the minimum HI antibody titer that could protect chickens from HPAI virus infection-related death was considered to be 1:16. Dose-dependent HI antibody responses were observed in chickens after the vaccination. Thus, 640 HA units/dose were thought to be similar to the optimal antigen level. Alternatively, the HI antibody titers of chickens, injected with the vaccine containing 640 HA units/dose, were maintained at 1:181 or higher for 100 weeks after the single-shot vaccination.

Production examination of efficiency of concentrated vaccine against swine erysipelas from matrix Konev of trial series, control of its immunizing power and production experiments of biological preparation were realized in the conditions of the Republic of Belarus. Research results showed that the presented vaccine had more expressed immunogenic properties in comparison with its production analogue and provided piglets protection against infection. There was analyzed the immunological efficiency of the vaccine: determination of quantity of erythrocytes; leukocytes; content of hemoglobin, level of lysozyme activity, bacterial growth-inhibitory activity, blood serum, phagocytic activity, neutrophilic leukocytes, content of total protein and protein fractions, titre of erysipelas antibodies, and preventive properties of blood serum. Efficiency of biological preparation was determined on the basis of analysis of disease incidence rate, loss of immunized swine caused by swine erysipelas

The realized studies of an experimental series of inactivated vaccine showed that the developed biological preparation made it possible to achieve high preventive efficiency in the conditions of cows heavy with calves for the creation in newborn calves the expressing immunity after milk feeding in the first hours of their life. As a result realized in the conditions of the Republic of Belarus production experiment of emulsified vaccine against rotavirus infection and colibacillosis of cattle in animal breeding farms of different regions of the Republic the prophylactic efficacy of a developed vaccine made up 97,9-98,9%. Research results showed that the developed inactivated vaccine caused at the immunizated animals the formation of specific antibodies at a higher level in comparison with other analyzed national and foreign analogue preparations.

A description of a method of chemical inactivation of Aujeszky's disease virus by teotropin was studied in the conditions of the Republic of Belarus. The developed inactivated viral suspension was tested for its antigenic activity. On the basis of realized experiments there were determined the optimal parameters of inactivation of Aujeszky's disease virus y means of 0,1% solution of teotropin under temperature of 37 deg C, pH 7,8 and duration of exposition of 24 hours. Immunomorphological changes in young hens vaccinated with an associated inactivated emulsion vaccine against bursal disease, infectious bronchitis and Newcasle disease was realized in the conditions of the Republic of Belarus. Replacement young hens of 130-158 days old were divided into 2 groups with 200 head in each according the analogue principle. At the age of 130 days old the first experimental group of chickens was immunized against bursal disease, infectious bronchitis and Newcasle disease with a liquid inactive emulsion vaccine (produced in the republic of Belarus) in dose of 0,5 ml intramuscularly. In course of the experiment chickens from each group were killed on the third, seventh, fourteenth, twenty-first and twenty-eighth days. For the analysis of immunomorphological reaction there were tested tissue pieces from the places of vaccine administration, as well as thymus, bursa of Fabritius, lien, cecal tonsils and Garder's gland. Research results showed that in tissues in places of the vaccine administration there were developed hemorrhages, serous inflammatory edema, necrobiosis and necrosis of muscular fibers, inflammatory cellular infiltration, and moderate plasmocytic reaction. At a later stage at the places of damaged structures there was noted the development of granulation tissue with presence of some lymphoid nodules. In response to vaccination in the central organs of poultry immunity there was stated the sinuated changes of morphological indexes. Immunization of chickens against infectious bursal disease, infectious bronchitis and Newcasle disease promoted the increasing of absolute weight of lien, activation of blastic and plasmocytic reaction in bursa of Fabritius and lien and did not render substantial influence on immunomorphological indexes of cecal tonsils and Garder's gland

Morphological indexes in immune system organs of piglets which were vaccinated which were immunized by SPS vaccine (against salmonellosis (Salmonella), pasteurellosis (Pasteurella) and streptococcus) in combination with immunostimulative drugs and without them were studied in the conditions of the Republic of Belarus. Piglets of the first group were vaccinated against salmonellosis, pasteurellosis and streptoccocusis by SPS vaccine. Piglets of the second group were vaccinated by SPS vaccine in combination with vitamin C. Piglets of the third group were vaccinated by SPS vaccine with immunotstimulant sodium thiosulphate. The fourth (control) group of animals was administrated with normal saline. Animals were immunized twice, intramuscularly with 7 day interval and in dose 4 ml (primary), 5 ml (secondary). Vitamin С was added in dose 0,05g per head. Research results showed that immunization by SPS vaccine with sodium thiosulphate and Vitamin С activated the morphological reaction of immune system. Application of Vitamin С and sodium thiosulphate (30% concentration) in combination with SPS vaccine promoted the increasing of lymphoid nudels in lymphodnudus and lien, increasing of plasma cells quantity in 1,3-2,2 times, activation of micro- and macrophage reaction in 1,5-2,8 times in comparison with vaccinated animals

Experimental application of a vaccine against salmonellosis, pasteurellosis, and streptococcosis (SPS) for piglets of 30-35 days was realized in the conditions of the Republic of Belarus. Research results showed that administrationof sodium thyosulphate and vitamin С in combination with SPS vaccine promoted the statistically-valid increasing of preventive properties of blood serum of piglets and increasing of titers of specific antibodies to agents of pasteurellosis and a salmonellosis in 1,3-2 times, as well as increasing in blood serum of quantity of immunoglobulins and to reduction of quantity of globulins.
