In the genus Paradiplozoon, few hard structures are present therefore most of the taxonomic studies have focused on the unique ultrastructure of the sclerites. Alcohol-fixed specimens were transferred to BSA-saline for 5 min. before staining indefinitely with 5 µl WGA-TRX, 5 µl SYTO 9 and 5 µl of Cal co fluor White M2R. Rhodamine, Fluorescein and DAPI bandpass filters on the microscope enabled selective light wavelength illumination of the three flourochromes by a mercury light source. This method provided an easy and rapid methodology to show the internal sclerites of attachment clamps. It is suitable for alcohol preserved specimens and may have additional applications in other helminth organisms.

The objective of this study was to determine the species spectrum of ixodid ticks infesting birds in Free State Province, South Africa. To this end a large number of birds belonging to several species were examined for ticks and a total of 180 birds belonging to 39 species at 17 localities were infested, and ticks belonging to eight species were recovered. The immature stages of only two, namely Amblyomma marmoreum and Hyalomma marginatum rufipes, were sufficiently prevalent and numerous to safely assume that they regularly use birds as hosts. Helmeted guineafowls, Numida meleagris, were the most heavily infested and one harboured a total of 319 larvae and four nymphs. Amongst the other species an eastern clapper lark, Mirafra fasciolata, was infested with 69 larvae and a nymph, but no other bird harboured more than 40 ticks. The larvae and nymphs of H. m. rufipes were most numerous on birds from April to August.

An endoparasite survey of a small pride of African lions (Panthera leo) was conducted at Intu Afrika Kalahari Game Reserve, southwestern Namibia, during winter and summer of 2003 and 2004, respectively. Overall, 23 fresh lion scats were collected opportunistically during fieldwork trials. A flotation technique was employed for the diagnosis of parasites. Three nematodes, Ancylostoma braziliense, Gnathostoma spinigerum and Uncinaria stenocephala and two coccidians, Toxoplasma gondii and Isospora felis were recorded. By using the McMaster method for quantification, a maximum number of 14 866 oocysts per gram of faeces was obtained for I. felis during winter 2003. Endoparasite taxa carried by the different individuals in the pride were found to be related to their levels of association. Rates of infection were relatively low as a result of the habitat, semi-captive conditions and earlier sporadic deworming.

An Ehrlichia ruminantium culture system was utilized for the anti-rickettsial evaluation of two ethnoveterinary plants, Elephantorrhiza elephantina and Aloe marlothii. Well-established E. ruminantium cultures were incubated with the plant leaf acetone extracts and compared to oxytetracycline and untreated controls. Effectivity was established by comparing the percentage parasitised cells and the calculation of both EC50 and extrapolated EC90 in µg/ml. The plant extracts were also screened for antibacterial activity using bioautography. Elephantorrhiza elephantina and A. marlothii demonstrated anti-ehrlichial activity with an EC50 of 111.4 and 64.5 µg/ml and EC 90 of 228.9 and 129.9 µg/ml, respectively. The corresponding EC50 and EC90 for oxytetracycline was 0.29 and 0.08 µg/ml. Both plants appeared to produce their inhibitory activity by a similar mechanism, unrelated to that of the tetracyclines. Both the plant acetone extracts demonstrated antibacterial activity against Escherichia coli and Staphylococcus aureus (ATCC strains).

During the period between January 1999 and December 2000, the distribution and seasonal patterns of Schistosoma mattheei infections in cattle in the highveld and lowveld communal grazing areas of Zimbabwe were determined through monthly coprological examination. Faecal samples of cattle were collected from 12 and nine dipping sites in the highveld and lowveld communal grazing areas, respectively. Patterns of distribution and seasonal fluctuations of the intermediate host-snail populations and the climatic factors influencing the distribution were also determined at monthly intervals from November 1998 to October 2000, a period of 24 months, in six dams and six streams in the highveld and nine dams in the lowveld communal grazing areas. Monthly, each site was sampled for relative snail density, the vegetation cover and type, and physical and chemical properties of the water. Mean monthly rainfall and temperature were recorded. Snails collected at the same time were individually examined for shedding of cercariae of S. mattheei and Schistosoma haematobium. A total of 16 264 (5 418 calves, 5 461 weaners and 5 385 adults) faecal samples were collected during the entire period of study and 734 (4.5 %) were positive for S. mattheei eggs. Significantly higher prevalences were found in the highveld compared to the lowveld (P < 0.001), calves compared to adult cattle (P < 0.01) and the wet season compared to the dry season (P < 0.01). Faecal egg output peaked from October/ November to March / April for both years of the study. Bulinus globosus, the snail intermediate host of S. mattheei was recorded from the study sites with the highveld having a significantly higher abundance of the snails than the lowveld (P < 0.01). Monthly densities of B. globosus did not show a clearcut pattern although there were peaks between March / May and September / November. The mean num ber of snails collected was positively correlated with the water plants Nymphaea caerulea and Typha species. Overall, 2.5 % of B. globosus were shedding Schistosoma cercariae. In the highveld, 2.8 % of B. globosus were infected with schistosome cercariae and 1.5 % in the lowveld, with the figures at individual sites ranging from 0-18.8 % in the highveld and from 0-4.5 % in the lowveld. The cercariae recorded here were a mixture of S. mattheei and S. haematobium since they share the same intermediate host. The transmission of Schistosoma cercariae exhibited a marked seasonal pattern, being more intensive during the hot, dry season (September / November).

Patterns of distribution and seasonal fluctuations of the intermediate host-snail populations and the climatic factors influencing the distribution were also determined by sampling at monthly intervals for a period of 24 months (November 1998 to October 2000) in six dams and six streams in the highveld and in nine dams in the lowveld communal grazing areas. Each site was sampled for relative snail density and the vegetation cover and type, physical and chemical properties of water, and mean monthly rainfall and temperature were recorded. Aquatic vegetation and grass samples 0-1 m from the edges of the snail habitats were collected monthly to determine the presence or absence of F. gigantica metacercariae. Snails collected at the same time were individually checked for the emergence of larval stages of F. gigantica. A total of 16 264 (calves 5 418; weaners 5 461 and adults 5 385) faecal samples were collected during the entire period of the study and 2 500 (15.4 %) of the samples were positive for F. gigantica eggs. Significantly higher prevalences were found in the highveld compared to the lowveld (P < 0.001), for adult cattle than calves ( P < 0.01) and in the wet season over the dry season (P < 0.01). Faecal egg output peaked from August / September to March / April for both years of the study. Lymnaea natalensis, the snail intermediate host of F. gigantica was recorded from the study sites with the highveld having a significantly higher abundance of the snail species than the lowveld (P < 0.01). The snail population was low between December and March and started to increase in April reaching a peak in September / October. The number of juvenile snails peaked between April and August. The mean number of snails collected was negatively correlated with rainfall and positively correlated with temperature. Mean number of snails collected was also positively correlated with Potamogeton plant species and negatively correlated with Cyperus plant species. However, none of the L. natalensis collected from the habitats were found shedding Fasciola cercariae. Metacercariae were found on herbage from the fringes of the snail habitats between February and August for both years, with most of the metacercariae concentrated on herbage 0-1 m from the banks of the habitats. Based on the findings of this study, anthelmintic treatment should be administered in December / January to control chronic and mature fasciolosis. A second treatment should be given in April / May to reduce pasture contamination and subsequently snail infection, as this is the time the snail population starts to build up. To control acute fasciolosis due to the immature liver flukes a third treatment should be given in August. The first application of molluscicides to control the snail intermediate hosts can be done in June the time when the snail is harbouring the parasite and a second application in September in order to kill new generations of infected snails.

To obtain updated data on and assess the contribution of trypanosomosis to the disease burden of cattle kept at the edge of the Hluhluwe-iMfolozi Park, a survey was conducted at Mvutshini Dip. Use was made of a purposeful sampling strategy by restricting sampling to animals that the livestock owner considered to be in poor condition. Of a total of 76 blood samples collected, 26 were parasitologically positive and 46 were positive on PCR / RFLP. Almost all infections were due to Trypanosoma congolense savannah subgroup. A total of 63 animals had a PCV < 24 % and were considered to be anaemic. Results from the survey show that trypanosome infections contribute significantly to the overall burden of disease in the area. Further research is required to develop appropriate control methods.

The ascaridoid nematodes Dujardinascaris madagascariensis Chabaud & Caballero, 1966, Dujardinascaris dujardini (Travassos, 1920), Gedoelstascaris vandenbrandeni (Baylis, 1929) Sprent, 1978 and Multicaecum agile (Wedl, 1861) Baylis, 1923 were recovered from the stomach contents of Crocodylus niloticus Laurenti, 1768 from the Okavango River, Botswana, together with Eustrongylides sp., a dioctophymatoid nematode usually parasitizing piscivorous birds. Dujardinascaris madagascariensis was present in most of the infected hosts, while the remaining species were mostly represented in single collections in one to three hosts. All four ascaridoid nematodes represent new geographic records.

Based on published records and own data a summary is given of the geographical distribution of the currently known species of pentastomid parasites infecting crocodiles and alligators, as well as freshwater chelonians. A brief generic diagnosis is provided for each genus. Fourteen out of the currently 23 living crocodilian species have been recorded as being host to one or more pentastomes. Out of the 32 pentastome species six are considered species inquirendae. Presently, six genera of crocodilian pentastomes, Agema, Alofia, Leiperia, Sebekia, Selfia and Subtriquetra are recognized. African crocodiles harbour eight pentastome species, six of which have been recorded from the Nile crocodile, Crocodylus niloticus. Three species belong to the genus Sebekia, Alofia being represented by two and Leiperia by only one species. Two species, Alofia parva and Agema silvaepalustris, occur in the dwarf crocodile, Osteolaemus tetraspis, and the slender-snouted crocodile, Crocodylus cataphractus, exclusively, but a single Sebekia species is shared with the Nile crocodile. The genus Agema is endemic to the African region. Infective stages of the pentastome Sub triquetra rileyi, thought to utilize Nile crocodiles as final hosts, have been recovered only from fishes. The largest number of pentastome species is found in the Australasian region. Of these, the Indo-Pacific croc odile, Crocodylus porosus, harbours seven, representing the genera Alofia, Sebekia, Lei peria and Selfia. Selfia is exclusive to the latter host. The genus Subtriquetra has been reported from "Indian crocodiles", a term possibly referring to either Crocodylus palustris, Crocodylus porosus or Gavialis gangeticus. Ten species of pentastomes parasitizing the crocodilian genera Alligator, Caiman, Crocodylus and Melanosuchus have been recorded from the Neotropical region including the southern states of the North American continent. The two most wide-spread pentastome genera, Alofia and Sebekia, have been recorded together with representatives of the genus Sub triquetra and immature and larval forms of Leiperia. To date the two monospecific genera, Pelonia, from two terrapin species, Pelusios sinuatus and Pelomedusa subrufa, in South Africa, and Diesingia from Hydraspis geoffroyana and Hydromedusa tectifera in South America, are the only chelonian pentastomes recovered world-wide. A possible exception is the crocodilian pentastome Sebekia mississippiensis which can reach maturity in exper imentally infected terrapins.

Experimental transmissions of cloned Theileria parva in cattle with Rhipicephalus appendiculatus ticks were compared to transmissions with uncloned T. parva during studies on the potential for genetic recombination during syngamy of Theileria to produce antigenic diversity for evasion of bovine immunity. Prevalence and abundance of T. parva infection in adult ticks, which resulted from the feeding of nymphs on the calves, were significantly higher in the uncloned compared to the cloned T. parva. Development of sporoblasts of T. parva in the ticks to produce infective sporozoites was similar. There was no statistically significant difference in the clinical course of infection in cattle between cloned and uncloned T. parva. It was concluded that cloned T. parva has characteristics that reduce its viability during the tick stages of its life cycle.
