Characterisation of porcine enteropathogenic Escherichia coli isolated in northeastern India

Enteropathogenic Escherichia coli (EPEC) is one of the main pathotypes causing gastroenteritis, particularly in young immunocompromised hosts. The study reports the prevalence, characterisation, and molecular epidemiology of EPEC from piglets in northeastern India. A total of 457 faecal samples were collected, from which 1,286 E. coli strains were isolated and screened by PCR. The resultant EPEC strains were serotyped and phenotypically characterised for resistance against 15 antimicrobials. Also, the phylogenetic sequence was analysed for 11 selected strains. A total of 42 strains (3.26%) belonged to atypical EPEC, of which, 15 (35.71%, and 2.29% of the 654 strains from this farm type) were isolated from organised and 27 (64.29%, and 4.27% of the 632 strains from this farm type) from unorganised farms; further, 5 (11.90% of the EPEC strains and 1.51% of the 330 strains from this breed) were isolated from the indigenous breeds and 37 (88.10%, and 3.87% of the 956 strains from this breed) from crossbred piglets. Serogroups O111 (11.9%) and O118 (7.14%) were the most prevalent of the 10 present. Sequence analysis of a length of the eaeA gene of 11 isolates of the region showed them to have 100% homology with each other and their identity ranged from 99.4% to 99.7% with GenBank reference sequences. All the EPEC isolates were multi-drug resistant, showing the highest resistance to amoxicillin (80.9%) and cephalexin (76.19%). The study highlighted the association of EPEC with piglet’s diarrhoea in northeastern India. EPEC isolates belonged to many serotypes and phenotypically all were multi-drug resistant with close genetic homology.

Contamination by Staphylococcus aureus of food produced from animal sources may have diverse and multifactorial causes that depend on geographical distribution. The goal of this study was to isolate and characterise S. aureus strains from contaminated fermented pork sausage, which is a local food of northeastern Thailand. S. aureus strains were isolated from local pork sausage, and the presence of classical enterotoxins was determined by PCR and reversed passive latex agglutination. These results were compared with strains derived from hospitalised patients and healthy carriers. Additionally, production of extracellular enzymes and haemolysin, biofilm formation, and antibiotic susceptibility were assessed. S. aureus was identified in 36 sausage isolates (60%). The strains positive for staphylococcal enterotoxin A were more frequently found in isolates from sausage and healthy carriers than in those from patients. All tested S. aureus strains were positive for DNase, lipase, proteinase, haemolysin, and biofilm formation; notably, strains isolated from food and healthy carriers displayed similar values. Most isolates were resistant to penicillin and ampicillin, while none were to methicillin. Thai fermented pork sausages are associated with a high risk of staphylococcal food poisoning, which may be linked to contamination caused by carriers. Dissemination of knowledge regarding best practices in sanitation and hygiene is important in local communities.

Objective: The objectives of this study were to determine the biofilm-forming capability and anti¬microbial susceptibility of Escherichia coli recovered from bovine endometritis samples. Materials and Methods: A total of 120 uterine specimens were collected from cows suffering from endometritis for bacteriological examination. Antimicrobial susceptibility testing was carried out for all isolated E. coli by using the disc diffusion method. The isolates were phenotypically studied for biofilm-forming ability by cultivation on yeast extract -casamino acids Congo red agar (CRA). Some randomly selected isolates were chosen for the molecular identification of some virulence and resistance genes. Results: A total of 58(48.3%) E. coli isolates could be isolated from the 120 samples. Antimicrobial susceptibility testing exhibited that 91.4%, 79.3%, 79.3%, 74.1%, and 58.6% of the isolates were sensitive to gentamicin, amoxicillin-clavulanic acid, ciprofloxacin, cephalexin, and sulfamethox¬azole-trimethoprim, respectively. On the other hand, 91.4% and 70.7% isolates were resistant to cefotaxime and doxycycline, respectively. Cultivation on CRA revealed that 46.6% of isolates were biofilm producers. The molecular detection of resistance and virulence genes declared that all iso¬lates harbored blaTEM, sul1, tetA, qnrS, blaCTX-M, and fimH with a percentage of 100%, papC (40%), and hlyA (10%). FimH was the most prevalent biofilm-associated gene. Conclusion: The present study highlights the high prevalence of multi-drug- resistant E. coli asso¬ciated with bovine endometritis. The detection of the fimH gene is circumstantial evidenced that this gene has a crucial role in biofilm formation in intrauterine pathogenic E. coli. [J Adv Vet Anim Res 2020; 7(3.000): 442-451]

Objectives: This study was performed to probe the antimicrobial resistance and virulence genes profiling in Pseudomonas aeruginosa recovered from the cases of pericarditis in broiler chickens. Materials and Methods: The samples (n = 250) collected from the cases of pericarditis in broiler chickens were bacteriologically examined. Antimicrobial susceptibility was tested by disc diffusion technique. The isolates were genotypically studied for the presence of antimicrobial resistance and virulence gene traits. Finally, the nucleotide sequence of representative resistance gene (mexR gene) and virulence genes (toxA and lasI genes) was analyzed. Results: P. aeruginosa was isolated from 45 samples (18%). Antimicrobial susceptibility testing revealed multidrug resistance in most of the recovered P. aeruginosa isolates, whereas colistin and imipenem were the furthermost in vitro-sensitive antibiotics. Antimicrobial resistance genes, such as blaCTX, fox, and mexR, were prevalent in 100%, 80%, and 100% of the isolates, respectively. PCR confirmed virulence genes such as toxA, exoY, lasB, and lasI in 100%, 60%, 80%, and 80% of the isolates, respectively. Nucleotide sequence analysis of representative resistance gene (mexR gene) and virulence genes (toxA and lasI genes) revealed a high correlation between P. aeruginosa recovered from pericarditis in broiler chickens in the present study with PAO1 (reference strain) and with other sequences published on the GenBank representing different localities worldwide. Conclusion: It could be concluded that P. aeruginosa recovered from pericarditis in broiler chick¬ens in the current study is highly virulent bacteria, resisting most of the therapeutic agents which not only bear hazards for poultry industry but also represent a public health concern. Objective: This study investigated the prevalence of Listeria monocytogenes in retail poultry shops, characterized the antibiotic resistance profile, and detected the genotypic pattern of vir¬ulence genes. Materials and Methods: Broiler meat (n = 90), intestinal content (n = 40), and environmental samples (n = 95) were collected for this study. Besides, hand swabs (n = 20) were obtained from the poultry shop workers and stool samples (n = 40) were collected from the outpatient clinics of Beni-Suef University Hospital, Egypt. The samples were subjected to isolation and identification of L. monocytogenes by conventional bacteriological examinations and biochemical tests, followed by confirmatory identification by the polymerase chain reaction. Results: Among the collected samples (n = 285), L. monocytogenes could be detected in 14.4% (n = 41/285) of the samples, where 30.0% (n = 12/40) of the intestinal content was positive. Similarly, 10.0% (n = 9/90), 15.0% (n = 3/20), and 12.5% (n = 5/40) of the samples of meat, hand swabs, and stools were found positive for L. monocytogenes, respectively. A total of 12 (12.6%) out of 95 environmental samples were positive for L. monocytogenes. Based on the antimicrobial sensitiv¬ity profile, most of the recovered isolates were multidrug-resistant against most commonly used antibiotics. Conclusion: The findings conclude that poultry shops play a vital role in transmitting L. monocy¬togenes to the consumers. Asymptomatic poultry shop workers should draw attention to their potentials for spreading the infection to the consumers through the contaminated carcasses. The objective of this study was to compare the virulence of 3 major Korean porcine circovirus type 2 (PCV2) genotypes in terms of clinical signs, PCV2 viremia and antibody titers, lymphoid lesions, and PCV2-antigen within lymphoid lesions in experimentally infected pigs. Pigs were infected at 7 weeks with PCV2a, PCV2b, and PCV2d strains and necropsied at 28 days post-infection. No statistical differences were observed in clinical signs, PCV2 viremia and antibody titers, lymphoid lesions scores, and numbers of PCV2 antigens among the 3 major Korean PCV2 genotypes. The results of this study indicate that the 3 major Korean PCV2 genotypes, PCV2a, PCV2b, and PCV2d, have similar virulence.

Salmonella enterica serovar Typhimurium (S. Typhimurium) is one of the most significant zoonotic pathogens that poses a threat to humans. Previous studies have identified that Salmonella-secreted effector K3 (SseK3) is a novel translated and secreted protein of S. Typhimurium. The objective of this study was to determine whether deletion of the sseK3 gene can attenuate the virulence of S. Typhimurium. To do this, we constructed an sseK3 deletion mutant using the double-exchange allele of the suicide plasmid pRE112ΔsseK3 and assessed the virulence and intracellular proliferation of the mutant. The sseK3 deletion mutant exhibited adhesion and invasion properties similar to those of wild-type (WT) S. Typhimurium, although the virulence and intracellular proliferation of the mutant were significantly reduced compared to that of the WT strain. Furthermore, the observed increase in the median lethal dose (LD(50)) reflects a decrease in the pathogenicity of the sseK3 deletion mutant in a murine model. In summary, we concluded that disruption of sseK3 can attenuate the intracellular proliferation and reduce the virulence of S. Typhimurium.

A prospective study of 65 research beagles kept in a rabies-free environment was undertaken to determine the duration of immunity after they received licensed rabies vaccines. The eventual goal was to extend mandated rabies booster intervals to 5 or 7 years and help reduce the risk of vaccine-associated adverse events. Three groups of dogs were vaccinated with 1 of 2 commercial rabies vaccines or saline at 12 and 15 weeks of age. Beginning 5 years 5 months later, vaccinated and unvaccinated dogs were challenged with virulent rabies virus and observed for 90 days over a series of 3 trials. Humoral and cellular immune responses were examined by serology and flow cytometry. Brain tissue from all challenged dogs was tested for rabies virus. Challenge trial 1 was confounded due to insufficiently virulent virus. In trials 2 and 3 virulent challenge provided 100% mortality in controls. Vaccinate survival was 80% (4/5) after 6 years 7 months, 50% (6/12) after 7 years 1 month, and 20% (1/5) after 8years 0 months. Antibody responses 12 days post-challenge correlated strongly with survival. In a separate non-challenge trial, administration of either a recombinant or a killed rabies vaccine demonstrated memory antibody responses 6 years 1 month after initial vaccination compared with unvaccinated controls. Our data demonstrated that i) duration of immunity to rabies in vaccinated dogs extends beyond 3 years; ii) immunologic memory exists even in vaccinated dogs with serum antibody titer < 0.1 IU/mL; and iii) non-adjuvanted recombinant rabies vaccine induces excellent antibody responses in previously vaccinated dogs 14 days after administration.
