An epidemiological study on giardiasis in cattle and humans at Beni-Suef Governorate

The present study was conducted to assess the prevalence of Giardia species infection in cattle and human. One hundred of animal fecal samples and 139 human stool samples were collected from different veterinary clinics and its related hospitals respectively. All samples were undergone to microscopically examination by; direct smears in 0.90% Na Cl solution, Lugol's iodine stain for cyst detection and formol-ether concentration. 9 (28.1%) calves from 32 were positive in microscopic examination by the used techniques. 25% of the examined fecal samples of cattle (17/68) were containing cysts of Giardia species by microscope. 39 of 139 (28.1%) of human stool samples were found infected by this protozoon. Regarding the sex of human cases, 26.30% of examined males were positive while 30.20% of females were positive. The age factor in human infection was clear; the age group of 11 to 20 years were the more infected than the other group (1- 10ys). There is no relation between form of human stool and infection rate. ELISA kits confirmed that 6 % of animal cases and 15.8% of human were positive. The epidemiological aspects were discussed in the study.

This study was conducted to study the UV protective effect against Haemonchus contortus infection in goats. Sixteen male goats were divided into 5 groups, control infected, control uninfected and UV 30minutes; UV 60minutes and UV booster 60minutes exposure. The UV groups were exposed to UV irradiation at wave length 254nm for 30 and 60 minutes. The UV booster 60min was administrated 2 doses of exposed larvae with an interval of one month. All groups except the control negative one were challenged for 42 days from the beginning. In UV booster 60min had reduction in egg count per gram feces and worm burden (93% & 34 % respectively). These parameters were similar in control infected, UV 30min and UV 60min groups. Increases in levels of antibodies were found in goats of UV booster 60min group the other groups. Finally, 2 doses of UV 60min exposure could protect goats from H. contortus.

Buxtonella sulcata (Jameson, 1926) is an intestinal protozoan of large ruminants, with scanty information and contradictious reports about its pathogenesity. This work aimed to investigate the prevalence rate of B. sulcata in Egypt. Forty eight cases collected from cattle (n= 29) and buffaloes (n= 19) from El-Mahalla El-Kubra area, Al-Gharbiya province. Samples were examined for the presence B. sulcata. The overall infection rate was 41.6% (20/48), in cattle 48.2% (14/29) and in buffalo 31.5% (6/19). This is the first study on B. sulcata in Egyptian ruminants also highlights the situation of intestinal ciliates of ruminant animals and provides basic information for the future work of intestinal ciliates of animals and man

The distribution of interstitial cells of Cajal (ICC), the probable pacemakers in gastrointestinal motility, was investigated using an antigenic marker of gastric ICC known as C-Kit. Antiserum raised against the general neuronal marker protein gene peptide 9.5 (PGP) as well as the nitrergic neuronal marker neuronal nitric oxide synthase (nNOS) were used to investigate the distribution of gastric nerves. Polyclonal goat anti-human C-Kit was reliable in labelling ICC in the stomach. Two classes of ICC were identified according to their distribution: ICC-MY distributed around the periphery of myenteric ganglia and ICC-IM in the circular and longitudinal muscle layers. The neuronal marker PGP was reliably consistent in revealing the density and distribution of the enteric nervous system. Density of nerve fibres was higher in circular smooth muscle than in longitudinal smooth muscle. From nNOS immunohistochemistry, it is evident that inhibitory (nitrergic) nerves constitute a substantial fraction of the enteric nervous system.

The attenuated <em>Salmonella typhimurium χ</em>4550 strain was used to harbour a reconstructed bicistronic DNA vaccine against porcine rotavirus, which carried the rotavirus nonstructural protein 4 (<em>NSP4</em>) and VP7 genes simultaneously. Using a balanced lethal system, the kanamycin resistance gene of expressing eukaryotic plasmids pVAX1 and pVAXD were replaced by the aspartate β-semialdehyde dehydrogenase (<em>asd</em>) gene. The NSP4 cleavage product (259–525) of rotavirus OSU strain and VP7 full-length genes were amplified by reverse transcription polymerase chain reaction and then inserted into the eukaryotic single-expression plasmid, pVAX1-asd, and the eukaryotic dual-expression plasmid, pVAXD-asd, respectively. The recombinant plasmids pVAX1-asd-NSP4, pVAX1-asd-VP7 and pVAXD-asd-NSP4-VP7 were transformed into the attenuated <em>S. typhimurium χ</em>4550 strain by electrotransformation. An indirect immunofluorescence assay of the expressed COS-7 cell suggested that the recombinant <em>S. typhimurium χ</em>4550 strain was constructed successfully. The recombinant <em>S. typhimurium χ</em>4550 strain was orally administered to BALB/c mice. The group immunised with dual- expression plasmids produced a significantly higher level of serum Immunoglobulin G (IgG) and intestinal Immunoglobulin A (IgA) than the group immunised with single-expression plasmids. These results indicated that eukaryotic bicistronic plasmid DNA vaccines could be successfully constructed to enhance humoural, mucosal and cellular immune response against rotavirus infection.

Objective: To evaluate the pharmacokinetics of nalbuphine decanoate after IM administration to Hispaniolan Amazon parrots (Amazona ventralis). Animals: 9 healthy adult Hispaniolan Amazon parrots of unknown sex. Procedures: Nalbuphine decanoate (37.5 mg/kg) was administered IM to all birds. Plasma samples were obtained from blood collected before (time 0) and 0.25, 1, 2, 3, 6, 12, 24, 48, and 96 hours after drug administration. Plasma samples were used for measurement of nalbuphine concentrations via liquid chromatography–tandem mass spectrometry. Pharmacokinetic parameters were estimated with computer software. Results: Plasma concentrations of nalbuphine increased rapidly after IM administration, with a mean concentration of 46.1 ng/mL at 0.25 hours after administration. Plasma concentrations of nalbuphine remained > 20 ng/mL for at least 24 hours in all birds. The maximum plasma concentration was 109.4 ng/mL at 2.15 hours. The mean terminal half-life was 20.4 hours. Conclusions and Clinical Relevance: In Hispaniolan Amazon parrots, plasma concentrations of nalbuphine were prolonged after IM administration of nalbuphine decanoate, compared with previously reported results after administration of nalbuphine hydrochloride. Plasma concentrations that could be associated with antinociception were maintained for 24 hours after IM administration of 37.5 mg of nalbuphine decanoate/kg. Safety and analgesic efficacy of nalbuphine treatments in this species require further investigation to determine the potential for clinical use in pain management in psittacine species.

Objective: To compare the effect of extracorporeal shock wave therapy (ESWT) on expression of fibroblast growth factor-7 (FGF-7), transforming growth factor-β1 (TGF-β1), insulin-like growth factor-1 (IGF-1), platelet-derived growth factor-A (PDGF), and vascular endothelial growth factor-A (VEGF) in skin with surgically created skin wounds and intact skin in horses. Animals: 14 healthy horses. Procedure: 8 horses were treated with ESWT at 6 locations along the neck at 36, 24, 12, 6, 2, or 1 hour prior to collection of full-thickness biopsy specimens from each location; a control specimen was collected from a sham-treated location. In 6 horses, 5 full-thickness wounds were created in each forelimb. Wounds in 1 forelimb/horse received ESWT immediately after creation and subsequently on days 7, 14, and 21; wounds in the contralateral forelimb remained untreated. Biopsy specimens were collected from 1 wound on each forelimb on days 7, 14, 21, 28, and 35. Expression levels of FGF-7, TGF-β1, IGF-1, PDGF, and VEGF were assessed in tissue samples from the horses' necks and forelimbs. Results: In surgically created wounds, ESWT treatment was associated with reduced TGF-β1 expression, compared with expression in control wounds, during the entire study period. At 28 days following wound creation, IGF-1 expression was significantly increased for treated and untreated wounds, compared with findings on days 7, 14, 21, and 35. There was no significant effect of treatment on FGF-7, TGF-β1, IGF-1, PDGF, or VEGF expression in intact skin. Conclusions and Clinical Relevance: Intervention with ESWT to suppress TGF-β1 may decrease granulation tissue production, resulting in improved wound healing on the distal portion of horses' limbs.

Objective: To identify matrix metalloproteinase (MMP)-2 and -9 in CSF from dogs with intracranial tumors. Sample: CSF from 55 dogs with intracranial tumors and 37 control dogs. Procedures: Latent and active MMP-2 and -9 were identified by use of gelatin zymography. The presence of MMPs in the CSF of dogs with intracranial tumors was compared with control dogs that were clinically normal and with dogs that had idiopathic or cryptogenic epilepsy or peripheral vestibular disease. Relationships between MMP-9 and CSF cell counts and protein were also investigated. Results: Latent MMP-2 was found in CSF samples from all dogs, although active MMP-2 was not detected in any sample. Latent MMP-9 was detected in a subset of dogs with histologically documented intracranial tumors, including meningiomas (2/10), gliomas (3/10), pituitary tumors (1/2), choroid plexus tumors (5/6), and lymphoma (4/4), but was not detected in any control samples. Dogs with tumors were significantly more likely than those without to have detectable MMP-9 in the CSF, and the presence of MMP-9 was associated with higher CSF nucleated cell counts and protein concentration. Conclusions and Clinical Relevance: Latent MMP-9 was detected in most dogs with choroid plexus tumors or lymphoma but in a smaller percentage of dogs with meningiomas, gliomas, or pituitary tumors. Detection of MMP in CSF may prove useful as a marker of intracranial neoplasia or possibly to monitor response of tumors to therapeutic intervention.

Objective: To compare data obtained with an inertial sensor system with results of subjective lameness examinations performed by 3 experienced equine veterinarians for evaluation of lameness in horses. Animals: 106 horses. Procedures: Horses were evaluated for lameness with a body-mounted inertial sensor system during trotting in a straight line and via subjective evaluation by 3 experienced equine practitioners who performed complete lameness examinations including lunging in a circle and limb flexion tests. Agreement among evaluators regarding results of subjective evaluations and correlations and agreements between various inertial sensor measures and results of subjective lameness evaluations were determined via calculation of Fleiss’ κ statistic, regression analysis, and calculation of 95% prediction intervals. Results: Evaluators agreed on classification of horses into 3 mutually exclusive lameness categories (right limb lameness severity greater than left limb lameness severity, left limb lameness severity greater than right limb lameness severity, or equal right and left limb lameness severity) for 58.8% (κ = 0.37) and 54.7% (κ = 0.31) of horses for forelimb and hind limb lameness, respectively. All inertial sensor measures for forelimb and hind limb lameness were positively and significantly correlated with results of subjective evaluations. Agreement between inertial sensors measures and results of subjective evaluations was fair to moderate for forelimb lameness and slight to fair for hind limb lameness. Conclusions and Clinical Relevance: Results of lameness evaluation of horses with an inertial sensor system and via subjective lameness examinations were significantly correlated but did not have strong agreement. Inertial sensor-based evaluation may augment but not replace subjective lameness examination of horses.

Objective: To compare echocardiographic measurements of left ventricular (LV) volume obtained via a modified Simpson or Teichholz method with those obtained via dual-source CT (DSCT). Animals: 7 healthy Beagles. Procedures: Each dog was anesthetized for DSCT; LV volume was determined from contrast-enhanced images of the LV lumen during all phases of contraction. Echocardiography was performed with dogs awake and anesthetized. End-diastolic volume (EDV), end-systolic volume (ESV), stroke volume, and ejection fraction were measured via a modified Simpson method and Teichholz method. Each dog was anesthetized twice with a 1-week interval between anesthetic sessions. Results: Results obtained while dogs were anesthetized revealed that the modified Simpson method underestimated LV volume (mean ± SD EDV, 24.82 ± 2.38 mL; ESV, 12.24 ± 1.77 mL), compared with that estimated by the Teichholz method (EDV, 32.57 ± 2.85 mL; ESV, 14.87 ± 2.09 mL) or DSCT (EDV, 34.14 ± 1.57 mL; ESV, 16.71 ± 0.76 mL). Ejection fraction (modified Simpson method, 48.53% ± 4.24%; Teichholz method, 54.33% ± 4.26%; DSCT, 51.00% ± 2.71%) differed significantly among the 3 methods. Echocardiographic results obtained while dogs were awake revealed that EDV, ESV, and stroke volume differed significantly between the modified Simpson and Teichholz methods. Conclusions and Clinical Relevance: LV volume determined via the Teichholz method was more similar to that determined via DSCT than was the LV volume determined via the modified Simpson method. The modified Simpson method underestimated LV volume, compared with that obtained via the Teichholz method, in both anesthetized and awake dogs.
