A new species of Cooperia, for which the name Cooperia pigachei n. sp. is proposed, was recovered from a mountain reedbuck, Redunca fulvorufula, from the Sterkfontein Dam Nature Reserve, Free State Province, and is described and illustrated. It is close to Cooperia neitzi Mönnig, 1932 and the South African race of Cooperia rotundispiculum in having more than 14 longitudinal cuticular ridges and in that the lateral cervical synlophe is of the closed type. The new species differs from all the other species of the genus in that the lateral branches of the dorsal ray are large and T-shaped. The spicules are robust, over 0.3 mm long and have large, curved shoes on their tips.

The influence of feeding urea-molasses blocks (UMB) on growth and gastrointestinal (GI) nematode parasitism of weaner goats grazing the same pasture was investigated on a farm in Nyandarua District, Kenya. Thirty female Small East African goat kids at an average age of 5 months were initially treated with albendazole orally (5 mg kg-1 body mass) and randomly assigned into one of two groups: group I were fed UMB prepared using a cold process and group II kids (controls) received no block supplementation (NBS). The UMB were given in the evening when the animals returned from grazing and were consumed during the night at a rate of 95.0 g head-1 day-1. Supplementation was undertaken for 3 consecutive months from July to September 2001 and January to March 2002. Body mass of the kids and faecal egg counts were measured monthly and larval cultures were performed on positive faecal samples of kids of each group. Five goats from each group were randomly selected for slaughter and total counts and identification of worms at the end of June 2002. Significant differences (P < 0.05) were found in cumulative mass gains of kids in group I from September compared with those in group II. On termination of the study kids in group I had gained an average of (+ SD) 20.4 ± 1.4 kg while those in group II had gained 11.8 + 1.1 kg. From January 2002, faecal egg counts of the kids in the UMB group differed significantly (P < 0.05) from those of the NBS group and at slaughter, the mean (+ SD) worm counts for the UMB group was 482 + 299 while that of the NBS group was 1 302 + 410. In all the goats, Haemonchus contortus was the predominant nematode recovered. These results indicate that UMB had significant effects in the control of GI nematode parasitism and enhanced growth of the young goats.

A crossbred calf (3 months old) obtained from a farm where regular control of ticks was practised and found to be free of blood parasites was inoculated with 20 ml pooled blood collected from four field cattle which had very low Trypanosoma theileri parasitaemias (one parasite per 70 µl blood as determined by the haematocrit centrifugation technique). Trypanosoma theileri was present in the blood 6 days after injection and a peak parasitaemia of 42 parasites per 70 µl blood was recorded by day 12. Hyalomma anatolicum anatolicum nymphs were applied on the ears of the calf on day 8 and they dropped engorged by days 13 and 14. The resulting adult ticks were examined for the presence of T. theileri by severing a leg and making a smear of the clear haemolymph which exuded from the wound. The smear was fixed in methanol and stained with Giemsa stain. The infection rate with T. theileri in the ticks was 43.3 % (26 out of 60). The intensity of infection was very high and various developmental stages of the flagellates were observed (epimastigotes, sphaeromastigotes, trypomastigotes and other intermediate stages). The haemolymph from 12 ticks was also collected in tissue culture medium and the trypanosomes survived for 25 weeks before eventually dying. The results demonstrated unequivocally the high vectorial capacity of the tick H. a. anatolicum for T. theileri.

The mass of residual yolk sac expressed as a percentage of initial mass of the egg from which the chick hatched decreased sharply in the first 2 days post-hatching. A gradual reduction occurred between 3 and 10 days after which a sharp decline was noted between 11 and 13 days post-hatching. The highest number of chicks with unabsorbed yolk sac was noted on day 5 post-hatching followed by days 6 and 7. Chick mortality followed the same pattern. The dynamics, causes and clinical consequences of yolk sac utilization are discussed.

The effects of dexamethasone and promethazine on the amelioration of pulmonary oedema in East Coast fever were investigated. The clinical effects of these drugs were further investigated when used in conjunction with the antitheilerial drug, buparvaquone. In the first experiment, 15 crossbred (Friesian x Zebu) steers were divided into four groups. With the exception of the animals in group IV, that served as a control group all the others were infected with Theileria parva sporozoites. On the second day of the febrile reaction, the steers in groups I and II were treated with dexamethasone (0.1 mg/kg) and promethazine (1 mg/kg), respectively. Group III steers served as the infected untreated controls. On the fifth day of the febrile reaction the animals in groups I, II and III were infused intravenously with tattoo ink suspension and 1 h later sacrificed for post-mortem examination and tissue sampling. The clinical picture indicated that both drugs significantly mitigated dyspnoea and the post mortem examination revealed a significant reduction in morphological changes. Tattoo ink particle count reflected a significant (P < 0.01) reduction in vascular leakage in the treated animals, with promethazine being significantly (P < 0.05) more effective than dexamethasone in this respect. In the second experiment, 18 steers were infected with T. parva sporozoites, and then were randomly allotted into three groups each of which contained six animals. After the onset of ECF clinical signs, the animals in the first two groups were treated with buparvaquone in combination with either dexamethasone (group I) or promethazine (group II), and the third group was treated with buparvaquone alone. The results indicated that all the animals in groups I, II and III recovered well and no significant differences were observed in clinical disposition between the groups. Two months later, serum samples were collected from the refractory animals and demonstrated the presence of antibodies against T. parva. When the animals were subsequently artificially challenged with T. parva, none of them succumbed to clinical disease. The same T. parva stabilate stock was used in both experiments and it proved to be infective in a separate batch of steers.

The objective of this study was to determine certain aspects of the biology of Rhipicephalus (Boophilus) decoloratus larvae under laboratory and field conditions. Larvae allowed 48 h to select a vertical questing substrate preferred 90 cm rods in length to those of 60 or 30 cm, while in a separate experiment migration from rods 5 cm or 25 cm in length to rods 45 cm in length continued between 48 h and 72 h after larval release. Hatching of the larval progeny of engorged female ticks exposed to ambient field temperatures during the period June to August, occurred synchronously during the third or fourth week of November. With a single exception, larvae that hatched during November and between April and July survived for 38 days or longer, while those that hatched from December to March survived for 31 days or less. Questing larvae were present on vegetation throughout the year, with most being recovered during January and February. Parasitic larvae were present on cattle from October to May with most being collected during January and February.

Psittacine beak and feather disease (PBFD) is a common disease of the psittacine species and is caused by the psittacine beak and feather disease virus (PBFDV). In this study the occurrence of the disease in ring-neck parakeets and budgerigars in South Africa suffering from feathering problems, using polymerase chain reaction as a diagnostic test was investigated. The genetic variation between viral isolates was also studied. Results indicate that PBFDV can be attributed to being the cause of feathering problems in some of the ring-neck parakeets and budgerigars in South Africa. Genetic variation of isolates occurs between species and individuals. A cheap and easy to use method of blood sample collection on filter paper for diagnostic purposes was also evaluated. It proved to be less stressful to the birds and did not inhibit further processes.

Fleas were collected from 61 wild carnivores belonging to 13 species in various nature reserves and on farms, two feral domestic cats in a nature reserve and a domestic dog in the city of Johannesburg. Eleven flea species, including two subspecies of one of these, belonging to six genera were recovered. Amongst these only Ctenocephalides felis felis and Ctenocephalides felis strongylus are considered specific parasites of carnivores. The remaining ten species normally infest the prey animals of the various carnivores.

The crude prevalence of antibodies to Babesia bovis infection in cattle was estimated by serology using indirect ELISA during the period January to April, 1999. Sera were obtained from 1 395 dairy cattle (of all ages, sexes and breeds) on smallholder farms, the majority being kept under a zero grazing regime. The crude prevalence of antibodies to Babesia bovis was 6 % for Tanga and 12% for Iringa. The forces of infection based on the age sero-prevalence profile, were estimated at six for Iringa and four for Tanga per 100 cattle years-risk, respectively. Using random effect logistic regression as the analytical method, the factors (variables) of age, source of animals and geographic location were hypothesised to be associated with sero-positivity of Babesia bovis in the two regions.

Assisted reproductive techniques might prove themselves useful tools in producing buffaloes free of specific diseases (BFSD), which are in demand in South Africa. Freezing protocols for African buffalo semen must not only result in good post-thaw qualities, but must also be practical. Epididymal sperm from six mature African buffalo bulls was collected, diluted with three different semen extenders and frozen. Pre-freezing equilibration times between 2 and 9 h were tested. Total and progressive motility, longevity and acrosomal integrity were measured and compared. The use of TriladylTM proved to result in better post-thaw parameters than the other two diluents. Equilibration times of between 4 and 9 h did not influence post-thaw sperm qualities significantly. For some of the treatments, exposure to semen extenders before freezing for less than 4 h resulted in inferior post-thaw semen parameters.
