Pharmacokinetic interaction of tulathromycin with Flunixin meglumine after intravenous injection in goats Texto completo

The pharmacokinetic aspects of tulathromycin (2.5 mg/kg b.w.) were studied following intravenous administration alone and in combination with flunixin meglumine (2.2 mg/kg b.w) in apparently healthy goats. Tulathromycin concentrations in serum were determined by microbiological assay technique using Bacillus subtiles (ATCC 66343) as test organism. The half-lives of distribution and elimination (t0.5(a)and to.5(p)) were 0.071, 0.046 and 6.43, and 5.05 h. following intravenous injection of tulathromycin alone and in combination with flunixin, respectively. Volume of distribution at steady state (Vdss) was 0.249 and 0.96l/kg., mean residence time (MRT) was 6.27 and 5.99 h and total body clearance (ClB) was 0.046 and 0.17 l/kg/hr., respectively. It was concluded that flunixin significantly altered the pharmacokinetics of tulathromycin by increase its distribution and accelerate its elimination from body. Therefore care should be taken during use of tulathromycin in goats concurrently with flunixin.

The present study was carried out in six provinces in both Egypt and Libya throughout the period from January to October 2014. These areas were (Beni-Suef, Ismailia, and Matrouh district) in Egypt and (Tripoli, Zliten, and Zawia district) in Libya. To assess the hygienic quality of underground water sources intended for animal and human drinking and detect the source of pollution. The physicochemical parameters as pH, alkalinity (mg/l), electrical conductivity (μS/cm), total hardness (mg/l), hardness Ca+2 (mg/l), hardness Mg+2 (mg/l), ammonia (mg/l), nitrite (NO2-) (mg/l), nitrate (NO3-) (mg/l) and some heavy metals (Pb, Fe, Cu, Cd, Cr, Zn and Mn) in the underground water were determined. A total of 60 water samples of the underground water were collected from dug wells. Samples were investigated for assessment the physicochemical quality of water destined for human and/or livestock consumption using appropriate instruments for the estimation of metals in the underground water using atomic absorption spectrometer. It has been revealed that a significant increase in mean values of alkalinity (P<0.001) in Ismailia district, Egypt and Zawia district, Libya. Meanwhile, the total hardness showed a significant increase in Tripoli, Zliten and Zawia districts of Libya (367.8±23.73, 345±17.20 and 330±20.19 mg/l, respectively). Mean values of lead (Pb) were higher in Tripoli and Zliten districts, Libya and Matrouh and Ismailia districts, Egypt (0.03±0.1, 0.02±0.3, 0.02±0.07 and 0.02±0.04, respectively). Meanwhile, mean values of cadmium (Cd) were the highest in the three Libyan districts (0.24± 0.003, 0.22±0.07 and 0.012±0.006 mg/l, respectively). In conclusion, the absence of unified system to monitor physicochemical parameters in ground water in the studied areas represented an important task in the evaluation of such water sources and subsequently causing an environmental risk for both animals and humans health.

Peste-des-petits-ruminants virus (PPRV) is a highly contagious, fatal and economically important viral disease of small ruminants that is still endemic and militates against the production of sheep and goats in endemic areas of the world. The aim of this study was to describe the viral strains within the country. This was carried out by collecting tissue and swab samples from sheep and goats in various agro-ecological zones of Nigeria. The phylogeny of archived PPRV strains or isolates and those circulating and causing recent outbreaks was determined by sequencing of the nucleoprotein (N)-gene. Twenty tissue and swab samples from apparently healthy and sick sheep and goats were collected randomly from 18 states, namely 3 states in each of the 6 agro-ecological zones visited. A total of 360 samples were collected. A total of 35 samples of 360 (9.7%) tested positive by reverse transcriptase–polymerase chain reaction, of which 25 were from oculo-nasal swabs and 10 were from tissue samples. Neighbour-joining phylogenetic analysis using Phylogenetic Analysis Using Parsimony (PAUP) identified four different lineages, that is, lineages I, II, III and IV. Interestingly, the Nigerian strains described in this study grouped in two separate major lineages, that is, lineages II and IV. Strains from Sokoto, Oyo, Plateau and Ondo states grouped according to the historical distribution of PPRV together with the Nigerian 75/1 strain of lineage II, while other strains from Sokoto, Oyo, Plateau, Akwa-Ibom, Adamawa, Kaduna, Lagos, Bauchi, Niger and Kano states grouped together with the East African and Asian strains of lineage IV. This finding confirms that both lineage II and IV strains of PPRV are circulating in Nigeria. Previously, only strains of lineage II were found to be present in the country.

OBJECTIVE To evaluate the mRNA expression of T helper (Th)1, Th2, and Th17 cell–associated inflammatory mediators in cells of bronchoalveolar lavage fluid samples collected from healthy horses exposed to hyperbaric oxygen (HBO) and to monitor blood oxygen concentration during and following HBO therapy. ANIMALS 8 healthy horses. PROCEDURES In a randomized controlled crossover design study, each horse was exposed (beginning day 1) to 100% oxygen at a maximum of 3 atmospheres absolute (304 kPa) daily for 10 days or ambient air at atmospheric pressure in the HBO chamber for an equivalent amount of time (control). Bronchoalveolar lavage fluid samples were collected on days 0 and 10. After validation of candidate reference genes, relative mRNA expressions of various innate inflammatory, Th1 cell–derived, Th2 cell–derived (including eotaxin-2), Th17 cell–derived, and regulatory cytokines were measured by quantitative PCR assays. For 3 horses, arterial blood samples were collected for blood gas analysis during a separate HBO session. RESULTS The optimal combination of reference genes was glyceraldehyde-3-phosphate dehydrogenase, hypoxanthine ribosyltransferase, and ribosomal protein L32. Compared with day 0 findings, expression of eotaxin-2 mRNA was significantly lower (0.12-fold reduction) and the percentage of neutrophils in bronchoalveolar lavage fluid samples was significantly lower on day 10 when horses received HBO therapy. Values of Pao2 rapidly increased (> 800 mm Hg) but immediately decreased to pretreatment values when HBO sessions ended. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that HBO therapy does not increase mRNA expression of inflammatory cytokines, but reduces eotaxin-2 mRNA transcription. The Pao2 increase was transient with no cumulative effects of HBO.

OBJECTIVE To evaluate circadian and postprandial variations in plasma citrulline concentration in healthy dogs. ANIMALS 8 healthy Beagles. PROCEDURES Blood samples were collected from dogs after 12 hours of food withholding (0 hours; 8:00 am) and then every 2 hours for 12 hours (until 8:00 pm) and again at 24 hours (8:00 am the next day). The same protocol was repeated, with the only difference being that a meal was given immediately after the 0-hour sample collection point. Plasma citrulline concentration was measured by ion exchange chromatography. RESULTS No significant difference in plasma citrulline concentration was identified among measurement points when food was withheld. Mean ± SD plasma citrulline concentration at 4 hours (72.2 ± 12.7 μmol/L) and 24 hours (56.1 ± 12.5 μmol/L) after dogs were fed was significantly different from that at 0 hours (64.4 ± 12.7 μmol/L). CONCLUSIONS AND CLINICAL RELEVANCE Plasma citrulline concentration had no circadian variation in unfed dogs but increased significantly in fed dogs 4 hours after a meal. Therefore, food should be withheld from dogs for 8 to 12 hours before blood sample collection for measurement of citrulline concentration.

OBJECTIVE To assess feasibility of flexible endoscopic evaluation of swallowing (FEES) in awake dogs, determine whether specific variables associated with the oropharyngeal phase of swallowing can be recognized, and evaluate the safety and tolerability of FEES. ANIMALS 6 healthy client-owned large- and giant-breed adult dogs. PROCEDURES A topical anesthetic was applied to the nasal passage of each dog, and a fiberoptic endoscope was passed transnasally until the tip of the scope was positioned in the oropharynx. All dogs voluntarily drank colored water followed by consumption of a commercial canned diet and then a kibble diet mixed with food color. During each swallow, laryngeal and pharyngeal anatomic structures were evaluated and depth of bolus flow prior to the pharyngeal phase of swallowing was assessed. Evidence of bolus retention in the vallecula or pyriform sinuses and laryngeal penetration of the bolus were recorded. RESULTS FEES was completed without major adverse events and was tolerated well by all 6 dogs. Mild, self-limiting epistaxis was noted for 2 dogs. The nasopharynx, oropharynx, and hypopharynx were observed in all dogs; movement of food boluses through the esophagus was observed in 2 dogs, and food boluses in the stomach were visible in 1 dog. Pharyngeal and laryngeal function was considered physiologically normal in all dogs. CONCLUSIONS AND CLINICAL RELEVANCE FEES appeared to be a feasible diagnostic tool for use in large- and giant-breed dogs. Studies are warranted in dogs with oropharyngeal dysphagia to determine whether FEES can be tolerated and whether it can augment videofluoroscopy findings.

OBJECTIVE To evaluate the biomechanical properties of 4 methods for fusion of the centrodistal and tarsometatarsal joints in horses and compare them among each other and with control tarsi. SAMPLE 24 sets of paired tarsi without substantial signs of osteoarthritis harvested from equine cadavers. PROCEDURES Test constructs (n = 6/type) were prepared from 1 tarsus from each pair to represent surgical drilling; 2 medially to laterally placed kerf-cut cylinders (MLKCs); a single large, dorsally applied kerf-cut cylinder (DKC); and a dorsomedially applied locking compression plate (DMLCP). Constructs and their contralateral control tarsi were evaluated in 4-point bending in the dorsoplantar, lateromedial, and mediolateral directions; internal and external rotation; and axial compression. Bending, torsional, and axial stiffness values were calculated. RESULTS Mean stiffness values were consistently lower for surgical drilling constructs than for contralateral control tarsi. Over all biomechanical testing, surgical drilling significantly reduced joint stability. The MLKC constructs had superior biomechanical properties to those of control tarsi for 4-point bending but inferior properties for external and internal rotation. The DMLCP and DKC constructs were superior to control tarsi in dorsoplantar, rotational, and axial compression directions only; DMLCP constructs had no superior stiffness in lateromedial or mediolateral directions. Only the DKC constructs had greater stiffness in the mediolateral direction than did control tarsi. Over all biomechanical testing, DMLCP and DKC constructs were superior to the other constructs. CONCLUSIONS AND CLINICAL RELEVANCE These biomechanical results suggested that a surgical drilling approach to joint fusion may reduce tarsal stability in horses without clinical osteoarthritis, compared with stability with no intervention, whereas the DMLCP and DKC approaches may significantly enhance stability.

OBJECTIVE To determine the diagnostic accuracy of a rapid immunoassay (RIA) for point-of-care detection of urinary tract infection (UTI) of dogs, compared with criterion-referenced diagnosis with bacterial culture. SAMPLE 200 urine samples obtained from dogs and submitted to a veterinary microbiology diagnostic laboratory for routine bacterial culture and antimicrobial susceptibility determination. PROCEDURES Samples were evaluated by use of quantitative bacterial culture and the RIA. Sensitivity, specificity, and positive and negative predictive values of the RIA were calculated; results of bacterial culture were the criterion-referenced outcome. A κ statistic was calculated to determine agreement between bacterial culture and RIA results. RESULTS 56 of 200 (28%) urine samples had positive results for bacterial growth by use of culture methods; there were 38 (19%) positive results likely to be associated with bacterial UTI on the basis of sample collection method and bacterial concentration. Sensitivity and specificity of the RIA for detecting samples likely to be associated with UTI (≥ 1,000 CFUs/mL) were 97.4% and 98.8%, respectively. The positive and negative predictive values of the RIA for bacterial cultures with likely UTI were 0.949 and 0.994, respectively. Agreement between bacterial culture and RIA outcome for UTI was substantial (weighted κ, 0.718). CONCLUSIONS AND CLINICAL RELEVANCE The RIA test evaluated in this study accurately detected UTI of dogs, compared with detection with the criterion-referenced bacterial culture method. Use of this point-of-care RIA could allow clinicians to diagnose UTI at the time of a patient visit and provide information useful for immediately initiating empirical antimicrobial treatment.

OBJECTIVE To compare mitochondrial oxygen consumption rate (OCR) of fibroblasts from Doberman Pinschers with and without dilated cardiomyopathy (DCM) and mutation of the gene for pyruvate dehydrogenase kinase isozyme 4 (PDK4) and to evaluate in vitro whether treatment with adeno-associated virus (AAV) vector (ie, gene therapy) would alter metabolic efficiency. ANIMALS 10 Doberman Pinschers screened for DCM and PDK4 mutation. PROCEDURES Fibroblasts were harvested from skin biopsy specimens obtained from Doberman Pinschers, and dogs were classified as without DCM or PDK4 mutation (n = 3) or with occult DCM and heterozygous (4) or homozygous (3) for PDK4 mutation. Fibroblasts were or were not treated with tyrosine mutant AAV type 2 vector containing PDK4 at multiplicities of infection of 1,000. Mitochondrial OCR was measured to evaluate mitochondrial metabolism. The OCR was compared among dog groups and between untreated and treated fibroblasts within groups. RESULTS Mean ± SD basal OCR of fibroblasts from heterozygous (74 ± 8 pmol of O2/min) and homozygous (58 ± 12 pmol of O2/min) dogs was significantly lower than that for dogs without PDK4 mutation (115 ± 9 pmol of O2/min). After AAV transduction, OCR did not increase significantly in any group (mutation-free group, 121 ± 26 pmol of O2/min; heterozygous group, 88 ± 6 pmol of O2/min; homozygous group, 59 ± 3 pmol of O2/min). CONCLUSIONS AND CLINICAL RELEVANCE Mitochondrial function was altered in skin fibroblasts of Doberman Pinschers with DCM and PDK4 mutation. Change in mitochondrial function after in vitro gene therapy at the multiplicities of infection used in this study was not significant.

OBJECTIVE To determine the temporal effects on tear flow measurements obtained by use of a Schirmer tear test (STT) I after IM administration of various doses of medetomidine or xylazine to healthy dogs. ANIMALS 5 healthy purpose-bred male Beagles. PROCEDURES Each dog received IM injections of 2.0 mL of physiologic saline (0.9% NaCl) solution (control treatment); 0.1% medetomidine hydrochloride (5, 10, 20, and 40 μg/kg), and 2.0% xylazine hydrochloride (0.5, 1.0, 2.0, and 4.0 mg/kg). Treatments were injected into the semimembranosus muscles; there was at least a 1-week interval between successive injections. Order of treatments was determined via a randomized Latin square crossover design. The STT I was performed on both eyes before (baseline) and 0.25, 0.50, 0.75, 1, 2, 3, 4, 5, 6, 7, 8, and 24 hours after each injection.RESULTS STT I values decreased significantly within 45 minutes after injection of medetomidine or xylazine, which was followed by gradual recovery. The lowest mean STT I value was < 10 mm/min for all sedation treatments, except when dogs received 5 μg of medetomidine/kg. Linear regression of the area under the curve for the 8 hours after administration yielded significant effects for all sedation treatments. CONCLUSIONS AND CLINICAL RELEVANCE IM administration of medetomidine or xylazine to dogs reduced tear flow in a dose-related manner. Artificial tear solution or ophthalmic ointment should be used to protect the ocular surface when these drugs are administered to dogs.
