A total of 774 pigs were enrolled in 13 cohorts across 7 commercial farms to examine the effect of nursery feeding program on serum haptoglobin, subsequent growth performance, and carcass characteristics of finishing pigs. Pigs were weaned [23.4 ± 0.2 days of age and 5.3 ± 1.5 kg body weight (BW)] and randomly assigned to either a complex (HC: highly digestible sources of proteins, including animal proteins; N = 346) or simple (LC: corn- and soybean meal-based; N = 340) diet for 37.7 ± 1.7 days over 3 phases (1 pen per diet per cohort; pen was the experimental unit; n = 13); sex was balanced between treatments. Thereafter, pigs received common grower-finisher diets. At a targeted BW of 118 kg, pigs (subset: n = 275 and 258 for HC and LC, respectively) were processed to evaluate carcass characteristics. Nursery feeding program did not influence BW, BW gain, or serum haptoglobin at any stage of production. Feed cost was reduced by $2.82 per pig during the nursery period for the LC-fed pigs (P < 0.001). Loin eye depth, back fat depth, carcass weight, percent lean yield, and carcass gross revenue at slaughter were not influenced by nursery feeding program. Feeding LC nursery diets on commercial farms is a feasible way to reduce feed cost without negatively impacting serum haptoglobin, growth performance during or after the nursery period, or carcass value.

OBJECTIVE To develop a reliable method for injection of the tarsometatarsal (TMT) joint of horses through a medial approach and to characterize anatomy of the synovial recesses of the TMT joint with CT. SAMPLE 13 pairs of fresh cadaveric equine tarsi. PROCEDURES TMT joints were assessed with radiography and CT. Twelve pairs of TMT joints were assigned to an experienced veterinarian (8 joints) or veterinary student (16 joints) for injection with contrast medium through a medial or a lateral approach. Every TMT joint was then reexamined radiographically and with CT to assess diffusion of contrast medium. The number of attempts for the lateral and medial approaches between and within investigators was evaluated, which included evaluation of the diffusion of contrast solution in relation to the approach used. RESULTS All 24 TMT joints were successfully injected, as confirmed with radiography and CT. There was no significant difference in the number of attempts between the medial and lateral approaches and between investigators. No significant difference was detected in the distribution of contrast medium in each synovial recess between the medial and lateral approaches. CONCLUSIONS AND CLINICAL RELEVANCE The medial approach described here for injection of the TMT joint of horses was highly reliable. It involved use of an easily palpable anatomic landmark on the mediodistal aspect of the TMT joint. An established alternative method for injection of the TMT joint may be useful in clinical practice for diagnostic and therapeutic purposes, although a medial approach may increase risk of injury to veterinarians.

OBJECTIVE To determine effects of hydrocortisone administration on serum leptin and adiponectin concentrations, abdominal fat distribution, and mRNA expression of leptin and adiponectin in abdominal adipose tissue of dogs. ANIMALS 12 healthy dogs. PROCEDURES Dogs received hydrocortisone (8.5 mg/kg; n = 6) or a placebo (6) orally every 12 hours for 90 days. Serum leptin and adiponectin concentrations were measured with a canine-specific ELISA on the day before (day 0; baseline) and during (days 1, 3, 7, 30, 60, and 90) administration. On days 0, 30, 60, and 90, abdominal fat mass was quantified with CT, and mRNA expression of leptin and adiponectin in abdominal fat was analyzed by use of a PCR assay. RESULTS Hydrocortisone administration resulted in an increase in visceral fat mass on days 60 and 90, compared with the mass at baseline. Visceral fat mass at the level of L3 increased during hydrocortisone administration. Serum leptin concentration began to increase on day 1 and was significantly higher than the baseline concentration on days 30 and 60. Serum adiponectin concentration on days 30, 60, and 90 was significantly lower than the baseline concentration. Leptin and adiponectin mRNA expression in abdominal fat was greater on day 30, compared with expression at baseline, but lower on days 60 and 90, compared with expression on day 30. Serum leptin concentration and visceral fat mass were correlated. CONCLUSIONS AND CLINICAL RELEVANCE Hydrocortisone administration affected abdominal fat distribution and serum leptin and adiponectin concentrations through dysregulation of leptin and adiponectin expression.

OBJECTIVE To evaluate the cardiovascular effects of atipamezole administered at half the volume or the same volume as dexmedetomidine to isoflurane-anesthetized cats. ANIMALS 6 adult (1 to 2 years old) domestic shorthair cats (body weight, 3 to 6 kg). PROCEDURES Each cat was anesthetized with isoflurane and rocuronium 3 times; there was a 1-week washout period between successive anesthetic procedures. For each anesthetic procedure, dexmedetomidine (5 μg/kg) was administered IV. Five minutes after dexmedetomidine was administered, atipamezole (25 or 50 μg/kg) or saline (0.9% NaCl) solution was administered IM. Pulse rate, mean arterial blood pressure (MAP), cardiac output (CO), and systemic vascular resistance (SVR) were measured during anesthesia before dexmedetomidine administration (baseline), after dexmedetomidine administration, and 15, 30, 60, and 120 minutes after administration of atipamezole or saline solution. Pulse rate and MAP were also recorded when MAP was at its lowest value. Hemodynamic variables were compared among treatments at baseline, after dexmedetomidine administration, and after administration of atipamezole or saline solution. Effects of treatment and time on all variables were assessed with mixed-effects models. RESULTS Both doses of atipamezole resulted in a significantly lower MAP than did saline solution. Pulse rate, CO, and SVR were not significantly different among treatments after atipamezole or saline solution were administered. CONCLUSIONS AND CLINICAL RELEVANCE Atipamezole administered IM at half the volume or the same volume as dexmedetomidine was ineffective at increasing pulse rate or CO in anesthetized cats that received dexmedetomidine. However, atipamezole caused short-lasting but severe arterial hypotension.

The efficacy of a porcine reproductive and respiratory syndrome (PRRS) subunit vaccine was evaluated and compared with a modified-live virus (MLV) vaccine under field conditions. Three farms were selected based on their history of respiratory diseases caused by co-infection with both PRRSV-1 and PRRSV-2. In each farm, 60 pigs were randomly allocated to 2 vaccinated and 1 unvaccinated groups (20 pigs per group). One group of pigs were administered the PRRS subunit vaccine at 21 and 42 days of age and another group administered the PRRS MLV vaccine at 21 days of age. The subunit vaccine had similar efficacy and, in some instances, performed even better than the MLV vaccine. Vaccination of pigs with either of the PRRS vaccines resulted in significantly improved growth performance in Farm B but not in Farm C. In Farm A, pigs vaccinated with the PRRS subunit vaccine had a better growth performance statistically compared to those vaccinated with the PRRS MLV vaccine. At the peak of PRRSV-1 and PRRSV-2 viremia, neutralizing antibodies and T-cell responses against PRRSV-1 and PRRSV-2 were at low levels suggesting that either vaccine is only able to provide a partial protection against co-circulating PRRSV-1 and PRRSV-2.

The efficacy of 4 commercial porcine reproductive and respiratory syndrome virus (PRRSV) modified-live vaccines (MLV), against PRRSV-1 and PRRSV-2 was evaluated and compared in growing pigs. Two of the vaccines were based on PRRSV-1 and two on PRRSV-2. There were no significant differences between each of the two PRRSV-1 MLV vaccines and the two PRRSV-2 MLV vaccines respectively based on virology, immunological, and pathological evaluations. Vaccination with either of the PRRSV-1 MLV vaccines resulted in reduced PRRSV-1 but not PRRSV-2 viremia. Additionally, vaccination with either of the PRRSV-1 MLV vaccines resulted in reduction of lung lesions and PRRSV-1 positive cells in PRRSV-1 challenged pigs but had no significant effect in PRRSV-2 challenged pigs. In contrast, vaccination with either of the two PRRSV-2 MLV vaccines resulted in the reduction of both PRRSV-1 and PRRSV-2 viremia. The PRRSV-2 MLV vaccines were also able to effectively reduce lung lesions and PRRSV positive cells after challenge with either PRRSV-1 or PRRSV-2. Our data suggest that while vaccination with PRRSV-1 MLV vaccines can be effective against PRRSV-1, only PRRSV-2 MLV vaccines can protect against both Korean PRRSV-1 and PRRSV-2 challenges in this study.

Bilateral deafness with concurrent vestibular dysfunction was first reported in the Doberman pinscher in 1980. Here, we identify a coding mutation in the MYO7A gene that is perfectly associated with the disorder. The lack of visual deficits in affected dogs suggests that, like rodents but unlike humans, MYO7A is not required for retinal function. DNA testing of the mutation will enable dog breeders to manage the incidence of this genetic defect.

This study aimed to determine the effect of a single injection of paracetamol on the sevoflurane minimum alveolar concentration (MAC) response to noxious mechanical stimulation. Seven healthy adult beagles were enrolled in a prospective, randomized, blinded, crossover experimental study. Anesthesia was induced with propofol [11.6 ± 2.4 mg/kg body weight (BW)] and maintained with sevoflurane. The MAC was determined before (MAC-1) and after (MAC-2) treatment with 15 mg/kg BW of intravenous (IV) paracetamol or saline over 15 minutes. Samples for plasma paracetamol determination were collected immediately after IV treatment administration and following MAC-2 determination (123 ± 27 minutes after starting paracetamol administration). The MAC-1 was similar between treatments (1.7% ± 0.4%). There were no differences between control and paracetamol groups at MAC-2 (2.0% ± 0.4% and 1.7% ± 0.5%, respectively; P = 0.285). Paracetamol plasma concentrations after paracetamol administration were 34.5 ± 9.9 μg/mL, decreasing at the end of the procedure (8.5 ± 4.2 μg/mL). In conclusion, 15 mg/kg BW of IV paracetamol did not significantly reduce sevoflurane MAC in healthy dogs.

Vaccination of domestic ruminants is considered to be an effective strategy for protecting these animals against Rift Valley fever (RVF), but available vaccines have limitations. Therefore, the aim of this study was to determine the safety and immunogenicity of RVF virus (RVFV) mutagenesis passage 12 (MP-12) and arMP-12&#916;NSm21/384 vaccine candidates in goats (Capra aegagrus hircus) in Tanzania. Goats were vaccinated intramuscularly with RVFV MP-12 or arMP-12&#916;NSm21/384, and then on Day 87 post-vaccination (PV) all animals were revaccinated using the RVFV MP-12 vaccine candidate. Serum samples were collected from the animals before and after vaccination at various intervals to test for RVFV using a Vero cell culture assay and reverse transcription polymerase chain reaction and for RVFV-neutralising antibody using a plaque reduction neutralisation assay. Serum samples collected before vaccination on Days -14 and 0, and on Days 3, 4 and 5 PV were negative for RVFV and neutralising antibody. All animals remained healthy, and viremia was not detected in any of the animals. Rift Valley fever virus antibody was first detected on Day 5 PV at a 1:10 dilution in five of five animals vaccinated with the MP-12 vaccine and in five of eight animals vaccinated with arMP-12&#916;NSm21/384. Titres then increased and were sustained at 1:40 to 1:640 through to Day 87 PV. All animals that were revaccinated on Day 87 PV with MP-12 developed antibody titres ranging from 1:160 to as high as 1:10 240 on Days 14 and 21 PV. Although the antibody titres for goats vaccinated with RVF MP-12 were slightly higher than titres elicited by the arMP-12&#916;NSm21/384 vaccine, these findings demonstrated that both vaccines are promising candidates for the prevention of RVF among Tansanian goats.

Shrubs represent the most affordable and accessible form of feed that livestock can rely on to acquire both essential and non-essential elements of life. In addition to their inherent toxins, they contain endogenous substances commonly referred to as 'antinutritive factors' (ANFs) that often interfere with the utilisation of nutrients. Their abundance may lead to severe clinical trauma. Hence, the objective of the study was to investigate the effects of different extraction techniques on Nerium oleander L. and animal feeds as well as to quantify oxalates. Organic (hexane, acetone and methanol) sequential and aqueous (infusion and decoction) extractions were explored. Qualitative and quantitative analyses were conducted to determine the presence of various phytochemicals and oxalate contents as putative ANFs, respectively. The results showed higher extraction yields of 22.6% and 43.1% in the decoction and infusion of N. oleander, respectively. The quantification methods were validated for linearity, accuracy and precision. Oxalate contents of 6.76 ± 0.245 (0.65%) mg/g and 5.74 ± 0.236 mg/g dry weight (0.55%) were obtained in N. oleander and feeds, respectively. This difference was statistically significant with p < 0.05. Percentage recoveries of 98.5 (percent relative standard deviation &#91;% RSD&#93; = 2.3), 85.7 (% RSD = 1.03) and 80.3 (% RSD = 1.22) at 76%, 95% and 112% fortifications were obtained, respectively. Relative standard deviation for precision was 0.99% and 1.13% at 0.33 mg and 0.39 mg fortifications, respectively, while reproducibility showed 2.21% RSD. Therefore, these methods can be used to provide a valuable basis for qualitative determination of ANFs, particularly in shrub foliage.
