Genetic characterisation of virulence genes associated with adherence, invasion and cytotoxicity in Campylobacter spp. isolated from commercial chickens and human clinical cases Texto completo

Virulence-associated genes have been recognised and detected in Campylobacter species. The majority of them have been proven to be associated with pathogenicity. This study aimed to detect the presence of virulence genes associated with pathogenicity and responsible for invasion, expression of adherence, colonisation and production of the cytolethal distending toxin (cdt) in Campylobacter jejuni and Campylobacter coli. Commercial chicken faecal samples were randomly sampled from chicken farms within the Durban metropolitan area in South Africa. Furthermore, human clinical Campylobacter spp. isolates were randomly sampled from a private pathology laboratory in South Africa. Out of a total of 100 chicken faecal samples, 78% (n = 78) were positive for Campylobacter growth on modified charcoal cefoperazone deoxycholate and from the random laboratory collection of 100 human clinical isolates, 83% (n = 83) demonstrated positive Campylobacter spp. growth following culturing methods. These samples were screened for the presence of the following virulence genes: cadF, hipO, asp, ciaB, dnaJ, pldA, cdtA, cdtB and cdtC. As expected, the cadF gene was present in 100% of poultry (n = 78) and human clinical isolates (n = 83). Campylobacter jejuni was the main species detected in both poultry and human clinical isolates, whilst C. coli were detected at a significantly lower percentage (p < 0.05). Eight per cent of the C. jejuni from human clinical isolates had all virulence genes that were investigated. Only one C. coli isolate demonstrated the presence of all the virulence genes investigated; however, the pldA virulence gene was detected in 100% of the C. coli isolates in poultry and a high percentage (71%) in human clinical C. coli isolates as well. The detection of cdt genes was found at higher frequency in poultry than human clinical isolates. The high prevalence rates of virulence genes detected in poultry and human clinical isolates demonstrate their significance in the pathogenicity of Campylobacter species.

The aim of this study was to investigate the immunogenicity of a plasmid deoxyribonucleic acid (DNA) vaccine encoding the G1 epitope of bovine ephemeral fever virus (BEFV) G glycoprotein in mice. A plasmid DNA carrying the G1 gene was constructed and designated as pcDNA3.1-G1. The expression of the target gene was confirmed in human embryonic kidney 293 (HEK 293) cells transfected with pcDNA3.1-G1 by indirect immunofluorescent staining. Immunisation experiments were intramuscularly carried out by vaccinating 6-week-old female mice in four groups, including the pcDNA3.1-G1 construct, pcDNA3.1 (+) plasmid alone, BEF-inactivated vaccine and phosphate-buffered saline (PBS) (1X) three times with 2-week intervals. Fourteen days after the last immunisation, the animals were bled and the resulting sera were tested for anti-G1-specific antibodies by immunoblotting analysis, indirect enzyme-linked immunosorbent assay (ELISA) and virus neutralisation (VN) test. Serological assays showed that the pcDNA3.1-G1 construct expressing G1 protein was able to elicit specific antibodies against this antigen. Virus neutralisation test showed that pcDNA3.1-G1 could induce anti-BEFV-neutralising antibodies in mice. Our findings indicated that a new dimension can be added to vaccine studies for bovine ephemeral fever (BEF) using eukaryotic expression plasmids encoding the G1 antigen in the future.

Anthrax, a neglected zoonotic disease that is transmitted by a spore-forming, rod-shaped bacterium, Bacillus anthracis, has reached endemic proportions in the Western Province of Zambia. Transmission of anthrax from the environment as well as between cattle has been observed to be partly because of entrenched beliefs, perceptions and traditional practices among cattle farmers in the known outbreak areas. This study was aimed at exploring lay perceptions, beliefs and practices that influence anthrax transmission in cattle of the Western Province. A mixed-methods study was conducted from August to December 2015. Quantitative data were collected using a cross-sectional survey. Qualitative data were generated by interviewing professional staff and community members. Five focus group discussions and five key informant interviews were conducted. Thematic analysis of interview data was performed using NVivo software. The findings suggested that cattle anthrax was biologically as well as culturally maintained. Cattle farmers were reluctant to have their livestock vaccinated against anthrax because of perceived low efficacy of the vaccine. Also, the cattle farmers did not trust professional staff and their technical interventions. Popular cultural practices that involved exchange of animals between herds contributed to uncontrolled cattle movements between herds and subsequent transmission of anthrax. These findings imply the need for professional staff to be culturally competent in handling socio-cultural issues that are known to be barriers for disease control in animals. There is a need to develop a policy framework that will foster integrated control of anthrax across disciplines.

The prevalence of coccidiosis was determined and Eimeria species were identified in farms at different locations in the Bejaia region, Algeria. The study was conducted from February to December 2016. Unvaccinated birds were selected randomly. Samples from litter and faeces were collected randomly (147 and 109, respectively). Necropsy and parasitological examinations were carried out using standard methods. Of the samples examined, 93 out of the 147 litter samples and 78 out of the 109 intestinal content samples were infected with Eimeria oocysts (63.26% and 71.55%, respectively). Mixed infections with Eimeria spp. were observed in some of the positive farms, with an overall prevalence of 54.28%. Five species of Eimeria (viz. E. acervulina, E. tenella, E. maxima, E. brunetti and E. mitis) were identified with different indices. Eimeria acervulina followed by E. tenella were the predominant species infecting chickens at the farms visited (32.05% and 26.92%, respectively). Statistically, the most prevalent Eimeria spp. was E. Acervulina (p < 0.05). This study demonstrated that coccidiosis is an omnipresent parasitic intestinal disease. It could strongly decrease production performance in broiler chickens.

Shiga toxin-producing Escherichia coli (STEC) O157 and non-O157 are food-borne pathogens and contaminants of foods of animal origin. This study was conducted to investigate the presence of virulence and integrase genes in STEC isolates from diarrhoeic calves in Fars Province, Iran. Five hundred and forty diarrheic neonatal calves were randomly selected for sampling. Rectal swabs were collected and cultured for isolation and identification of E. coli following standard methods. The isolates were analysed for the presence of class 1 integrons and bacterial virulence factors using polymerase chain reaction (PCR). Antimicrobial susceptibility testing was performed using the Kirby-Bauer disc diffusion method. Out of 540 diarrhoeic faecal samples, 312 (57.7%) harboured E. coli and 71 (22.7%) of them were identified as STEC: 41(69.5%) carried the stx2 gene, 21 (35.6%) carried the stx1 gene and 3 (5%) carried both. Twenty-six (44%) of the isolates showed the eae gene. Among the STEC isolates examined for susceptibility to eight antimicrobial agents, erythromycin and penicillin (96.8%) resistance were most commonly observed, followed by resistances to ampicillin (71.8%), tetracycline (62.5%) and trimethoprim/sulfamethoxazole (39%). Integrons were detected by PCR in 36% of the STEC tested isolates, 57 (89%) of which showed resistance to at least three antimicrobial agents. Our findings should raise awareness about antibiotic resistance in diarrhoeic calves in Fars Province, Iran. Class 1 integrons facilitate the emergence and dissemination of multidrug-resistance (MDR) among STEC strains recovered from food animals.

The occurrence of veterinary drug residues in chicken meat originating from 320 small and medium scale chicken slaughterhouses in Peninsular Malaysia was determined. 637 chicken meat samples were examined for tetracycline (TCs), sulphonamide (SAs) and quinolone residues using a microbiological inhibition test and was further confirmed using liquid chromatography mass spectrometry (LCMS/MS). The presence of TC residues were confirmed in 10 (1.6%) samples, and 1 (0.2%)sample was confirmed in compliance to the established maximum residue limit (MRL) for residues of quinolone. A total of 6 (0.9%) samples were above the MRL for TC. The samples were from Pulau Pinang, Terengganu and Kelantan. Among those tested in compliance, the main analytes found for TC and quinolone werechlortetracyclines (CTC), enrofloxacin and mixture of chlortetracycline (CTC) and oxytetracycline (OTC). No samples were found to contain sulfonamides residues.

Malaysia has experienced four waves of H5N1 outbreak but no humandeaths were recorded which is in 2004, 2006, 2007 and latest outbreak in 2017 at Kelantan. The objective of this paper is tocollect, analyse and summarise the data of HPAI cases from the outbreak from 1 March to 22 June 2017. A total of 1,634 cases, 8,544 samples were received at the Registration Unit and Virology Section, Regional Veterinary Laboratory in Kota Bharu (RVLKB) for diagnosis and surveillance of H5N1 during this period. The samples received were pooled organs and intestines from post-mortem (41 cases) and cloaca swabs in tryptose phosphate buffer from surveillance (1,593 cases), which were sent to RVL, Kota Bharu and Veterinary Research Institute, Ipoh, Perak (VRI). They were processed and diagnosed using quantitative real-time reverse transcriptase PCR (RT qPCR) technique to detect and subtyping of the virus. Total positive cases for H5N1 were 53 out of 1,634 with 18 diagnostic cases and 35 surveillance cases. All positive cases during outbreak were detected from 43 locations from six districts of Kota Bharu, Bachok, Pasir Puteh, Pasir Mas, Tumpat and Tanah Merah, with 19 locations in Kota Bharu which is thehighest affected by H5N1, seven locations in Bachok, four in Pasir Putih, six in PasirMas, six in Tumpat and one in Tanah Merah. Many factors could have led to the acute spread of the virus between the districts like chicken smuggling, legal and illegal poultry trade, migration of infected wild birds and others. Understanding the source of outbreak and how it spread is important to control, eradicate and prevent the spread of the disease as it is zoonotic and infects human.

Biogas plant is an anaerobic digester that produces biogas fromorganic materials. The general purpose of implementing biogas plant in an animal farm is to obtain biogas from animal manure and the effluent of biogas digester after fermentation process can be used as organic fertiliser. There are 15 biogas plants based on livestock waste that have been developed throughout Malaysia until 2018. Two more biogas plants are planned to be built in Segamat and Keningau. All biogas plants built were originally aimed for domestic use which is as source of energyused for cooking and incinerator as well as to generate electricity for farm use. Out of 15 biogas plants, 7 plants were built on cattle farms, 4 plants on pig farms, 3 plantson poultry farms and only 1 plant on buffalo farm. Total construction cost for each biogas plant is between MYR4,500 to MYR30 million. Based on the survey conducted, it revealed that only 40% of the plants are still functional. However, 60% were dormant or nonfunctional due to poor maintenance, design errors, lack of technical knowledge to run the system consistently, improper planning and monitoring or insufficient livestock waste to continuously producing biogas.

The livestock industry has been relying merely on chemically synthesised antibiotic for eye infections as sprays and ointment. A natural remedy from Curcuma spp. has been tested for efficacy in curing keratoconjunctivitis and uveitis. A severe case of uveitis was cured within 7 days, with impaired vision restored. These results were observations of a preliminary study conducted in a goat with uveitis.

An adult female Sambar Deer (Cervus unicolor) at Wildlife Conservation Centre, Sungkai, Perak was found dead in the paddock after abortion. The dead foetus was found on the paddock the day before this incident. The workers at the conservation centre also reported that the doe was attacked few times prior to death by a bull of the same group. The paddock in the Sungkai Conservation Centre was under construction and it was suspected that the noise from heavy machiner y and animals being transferred into different paddocks caused further stress to the affected doe which led to abortion and death. Post-mortem lesions showed generalised congestion and haemorrhage of all vital organs including liver, heart, lungs, kidneys and spleen. Histopathology revealed all organs have evidence of generalised congestion. Cause of death in this case is failure of the body system due to generalised congestion of the vital organs due to stress factor.
