Historical review was done concerning the formation and development of scientific research of the Experimental Station of Medicinal Plants over the period 1916–2016. The main stages of experimenting in the direction of “medicinal plant cultivation’’ in Ukraine were covered. Role and importance of the Experimental Station of Medicinal Plants in the scientific and methodological support of such scientific directions as breeding, seed production, cultivation technologies, pharmacognosy in medicinal plant cultivation were shown. For the needs of the branch, over 100 species of introduced medicinal plants were studied at the Station and agricultural techniques of their growing were developed, in addition, more than 50 varieties of medicinal plants were created, a number of machines and devices for growing, harvesting and initial treatment of raw materials and seeds of the most important medicinal plants was designed. Other scientific achievements of the Station were mentioned and promising areas of medicinal plant research in Ukraine were outlined.

High-throughput sequencing (HTS) enables the generation of large amounts of genome sequence data at a reasonable cost. Organisms in mixed microbial communities can now be sequenced and identified in a culture-independent way, usually using amplicon sequencing of a DNA barcode. Bulk RNA-seq (metatranscriptomics) has several advantages over DNA-based amplicon sequencing: it is less susceptible to amplification biases, it captures only living organisms, and it enables a larger set of genes to be used for taxonomic identification. Using a model mock community comprising 17 fungal isolates, we evaluated whether metatranscriptomics can accurately identify fungal species and subspecies in mixed communities. Overall, 72.9% of the RNA transcripts were classified, from which the vast majority (99.5%) were correctly identified at the species level. Of the 15 species sequenced, 13 were retrieved and identified correctly. We also detected strain-level variation within the Cryptococcus species complexes: 99.3% of transcripts assigned to Cryptococcus were classified as one of the four strains used in the mock community. Laboratory contaminants and/or misclassifications were diverse, but represented only 0.44% of the transcripts. Hence, these results show that it is possible to obtain accurate species- and strain-level fungal identification from metatranscriptome data as long as taxa identified at low abundance are discarded to avoid false-positives derived from contamination or misclassifications. This study highlights both the advantages and current challenges in the application of metatranscriptomics in clinical mycology and ecological studies.

Purpose. To study morphometric parameters of fruits (cypselae) of purple coneflower (Echinacea purpurea (L.) Moench) of ‘Zirka Mykoly Vavylova’ cultivar and pale coneflower (Echinacea pallida (Nutt.) Nutt) of ‘Krasunia Prerii’ cultivar and determine the impact of agro-meteorological factors on their performance according to long-term data. Methods. Laboratory, Mathematics and Statistics. Results. It was found that the parameters of cypselae and its weight in various types of coneflowers varied considerably from year to year. In this context the indicators of cypselae width and thickness had a larger variability as compared with the length. Mean length of Echinacea purpurea ranged from 4,57 to 6,16 mm, width – 2,30–2,97 mm, thickness – 1,74–2,28 mm. Weight of a cypselae made up 4,40–6,50 mg. Length of a fruit of Echinacea pallida was 4,57–5,74 mm, width – 2,51–3,18 mm, thickness – 1,76–2,37 mm, and a fruit weight varied from 5,65 to 7,70 mg. The correlation analysis revealed reliable connection of agro-climatic parameters and the parameters of the fruit. Conclusions. Using long-term data, the morphology of cypselaes of two Echinacea species introduced to Ukraine were studied, their basic parameters and the variability of indicators, connection with agro-climatic factors that is necessary to consider in crop growing for seeds .
