Purpose. Provide bioinformatic analysis and comparison of target regions of the acetolactate synthase (als) gene in several members of the Poaceae family and, on the basis of the obtained data, explore the possibility of creating a unified genetic construct for als gene editing using the CRISPR-Cas9 system. Methods. The als gene sequences of various members of the Poaceae family were obtained from the NCBI: Nucleotide database. For comparison, a fragment of the imi-2 gene of wheat of the soft line ‘TealIMI11A’ was used in two regions of the 367–390 and 1729–1749 nucleotide sequences. The Sequence Viewer 3.37.0 tool was used to assess the presence of nucleotide substitutions in the working sequence of the imi-2 gene. The dendrogram was built using the “Blast Tree” tool from the NCBI: Blast: Nucleotide resource. Results. A comparative analysis of the nucleotide sequences of seven different species was carried out: soft wheat (Triticum aestivum L.), common wild oat (Avena fatua L.), barley (Hordeum vulgare L.), Asian rice (Oryza sativa L.), maize (Zea mays L.), aleppo grass (Sorghum halepense Pers.) and Tausch’s goatgrass (Aegilops tauschii Coss.). The dendrogram is based on the gene sequence als, showed that all studied genotypes can be divided into two blocks: the first block included maize and aleppo grass, and the second block, a separate branch includes Asian rice and common wild oat, barley, soft wheat and Tausch’s goatgrass. 367–390 nucleotide sequences of soft wheat showed the highest 100% homology to Asian rice, Tausch’s goatgrass and common wild oat. The lowest homology was for maize and aleppo grass at 83.3%. Evaluation of the nucleotide sequence 1729–1749 showed no complete homology at the 100% level. It was the highest for barley and Tausch’s goatgrass – 95.2%, and the lowest for rice, maize and aleppo grass – 80.9% each. Conclusions. The analysis confirms a significant degree of homology of the als gene sequence for various species of the Poaceae family, which allows us to create a universal genetic vector. However, taking into account the high degree of sequence homology for species such as soft wheat, Tausch’s goatgrass, barley, Asian rice and common wild oat, it can be assumed that the corresponding genetic vector can be used with the greatest efficiency to alter the als gene of these genotypes.

Сиквенсы гена als различных представителей семейства Злаковых были получены из базы данных NCBI: Nucleotide. Для сравнения был использован фрагмент гена imi-2 пшеницы мягкой линии ‘TealIMI11A’ в двух участках сиквенса 367–390 и 1729–1749 нуклеотидов. Для оценки наличия нуклеотидных замен в рабочих сиквенсах гена imi-2 использовали инструмент ‘Sequence Viewer 3.37.0’. Дендрограмму строили с использованием инструмента “Blast Tree” с ресурса NCBI: Blast: Nucleotide. Результаты. Был проведен сравнительный анализ нуклеотидных последовательностей семи различных видов: пшеницы мягкой (Triticum aestivum L.), овсюга обыкновенного (Avena fatua L.), ячменя обыкновенного (Hordeum vulgare L.), риса посевного (Oryza sativa L.), кукурузы (Zea mays L.), сорго алепского (Sorghum halepense Pers.) и эгилопса Тауша (Aegilops tauschii Coss.). На основе сравнительного анализа сиквенса участков гена іmi-2 для 7-ми генотипов построили филогенетическое дерево, которое показало, что исследованные виды можно разделить на два блока. В первый блок вошли кукуруза и сорго алеппское, а ко второму блоку – рис посевной, овсюг обыкновенный, ячмень обыкновенный, пшеница мягкая и эгилопс Тауша. Определение степени гомологии между последовательностью 367–390 нуклеотида пшеницы мягкой и другими видами показала, что абсолютной была гомология с соответствующими последовательностями риса посевного, эгилопса Тауша и овсюга обыкновенного (100%). Наименьшим нуклеотидное родство оказалось для кукурузы и сорго алеппского – по 83,3%. На участке 1729–1749 нуклеотидов гена іmi-2 никакой из 6 сиквенсов не показал 100% гомологии с последовательностью пшеницы мягкой. Самой высокой она была для ячменя обыкновенного и эгилопса Тауша – 95,2%, а наименьшей для риса посевного, кукурузы и сорго алеппского – по 80,9%.Выводы. Проведенный анализ подтверждает значительную степень гомологии последовательности гена als для различных видов семейства Злаковых. Это позволяет допустить возможность создания универсальной генетической конструкции, с помощью которой можно осуществлять редактирование генома и получения растений, устойчивых к гербициду разных представителей этой семьи. Учитывая высокую степень гомологии последовательностей для таких видов как пшеница мягкая, эгилопс Тауша, ячмень обыкновенный, рис посевной и овсюг обыкновенный, можно предположить, что с наибольшей эффективностью соответствующая генетическая конструкция может быть использована для редактирования гена als именно этих генотипов. | Провести біоінформатичний аналіз та порівняти цільові ділянки гена ацетолактат синтази (als) у декількох представників родини Злакових і на основі отриманих даних дослідити можливість створення уніфікованої генетичної конструкції для направленого редагування гена als за допомогою системи CRISPR-Cas9. Методи. Сиквенси гена als різних представників родини Злакових було отримано з бази даних NCBI: Nucleotide. Для порівняння було використано фрагмент гена іmi-2 пшениці м’якої лінії ‘TealIMI11A’ у двох ділянках сиквенсу: 367–390 та 1729–1749 нуклеотидів. Для оцінювання наявності нуклеотидних замін в робочих сиквенсах гена іmi-2 використовували інструмент “SequenceViewer 3.37.0”. Дендрограму будували з використанням інструменту ‘BlastTree’ з ресурсу NCBI: Blast: Nucleotide. Результати. Було проаналізовано нуклеотидні послідовності сімох різних видів: пшениці м’якої (TriticumaestivumL.), вівсюга звичайного (AvenafatuaL.), ячменю звичайного (HordeumvulgareL.), рису посівного (OryzasativaL.), кукурудзи (ZeamaysL.), сорго алепського (SorghumhalepensePers.) та егілопса Тауша (AegilopstauschiiCoss.). На основі порівняльного аналізу сиквенсу ділянок гена іmi-2 для 7-ми генотипів було побудовано філогенетичне дерево, яке показало, що досліджені види можна поділити на два блоки. До одного з них увійшли кукурудза та сорго алепське, а до другого – рис посівний, вівсюг звичайний, ячмінь звичайний, пшениця м’яка та егілопс Тауша. Визначення ступеня гомології між послідовністю 367–390 нуклеотиду пшениці м’якої та іншими видами показало, що абсолютною була гомологія з відповідними послідовностями рису посівного, егілопса Тауша та вісюга звичайного (100%). Найменшою нуклеотидна спорідненість виявилась для кукурудзи та сорго алепського – 83,3%. У ділянці 1729–1749 нуклеотидів гена іmi-2 жоден з 6 сиквенсів не показав 100% гомології з послідовністю пшениці м’якої. Найвищою вона була до ячменю звичайного та егілопсу Тауша – 95,2%, а найменшою для рису посівного, кукурудзи та сорго алепського – по 80,9%. Висновки. Проведений аналіз підтверджує значний ступінь гомології послідовності гена als для різних видів родини Злакових. Це дозволяє зробити припущення про можливість створення універсальної генетичної конструкції, за допомогою якої можна редагувати геном та отримувати рослини, стійкі до гербіцидів різних представників цієї родини. Зважаючи на вищий ступінь гомології послідовностей для таких видів як пшениця м’яка, егілопс Тауша, ячмінь звичайний, рис посівний та вівсюг звичайний, можна припустити, що ефективнішою відповідна генетична конструкція буде для редагування гена als саме цих генотипів.

Purpose. To define the level of impact of such factors as rate, time and methods of sowing on seed productivity and certified seeds, and the individual response of winter wheat varieties to these factors if used the ecological system of seed farming, that would allow to form and obtain genetically determined potential of variety productivity. Methods. Weight measurement for determining seed yield; statistical, variance and regression analysis for identifying the reliability of the experiment results. Results. During the research period, weather conditions were greatly differed both by temperature regime and precipitation, but stability of grain and seeds yield on an annual basis was the main requirement to varieties. During the study of «grain productivity» value and quantitative indices of winter wheat yield structure, the results from 684 plots were obtained which were grouped and analyzed for impact factors for complete certainty, and NIR0,05 was calculated. Conclusions. Thus, in case of shifting sowing time from optimum to later period, efficiency of water use by plants was decreasing to a greater extent during years with insufficient precipitation. The shift of sowing time to later period providing optimal seeding rate and row seeding method did not reduce yield. The yield of winter wheat varieties to be studied when sowing in usual manner with seeding rate of 5,5 million seeds/ha in the period from September 15 and October 5 was the highest. Varieties ‘Slavna’ and ‘Chorniava’ provided the highest grain productivity for seeding rate of 5,5 million seeds/ha and using row seeding method, with slight impact of sowing time factor. Grain yield of studied varieties showed negative response during the experiment when seeding rate decreased up to 2,5–3,0 million seeds/ha. ‘Astarta’ variety provided the highest productivity for certified seeds yield as compared to the control (from 1,59 to 3,38 t/ha). The variant of the experiment when sowing on October, 5 in usual manner with seeding rate of 5,5 million seeds/ha showed the highest productivity of 7,02 t/ha (+3,38 t/ha, or +92,9%). Shifting of sowing time for October, 5 resulted in a significant decrease in yields of certified seeds as compared to sowing time on September 15 and 25. Seed yield of studied varieties when sowing on October, 5 was higher than in case of sowing on September, 25. The highest percentage of certified seeds output in the studied varieties was observed for a seeding rate of 2,5–3,0 million seeds/ha. For seeding rate of 5,5 million seeds/ha for varieties with seeding time between September 15 and 25, the difference in grain yield was negligible. The winter wheat variety ‘Astarta’ provided the highest yield of seeds during the research years – 9,36 t/ha, the standard variety ‘Bogdan’ – the lowest one in the best variant of 5,55 t/ha.

Purpose. To study the dynamics of soft winter wheat lectin activity in caryopsis and at the early stages of ontogenesis of varieties developed by Poltava State Agrarian Academy as well as investigate varietal differences depending on dynamic pH medium. Methods. Laboratory analysis, method of hemagglutination reaction, analytical approach, statistical evaluation. Results. A change in lectin activity was determined in samples of 14 soft winter wheat varieties in caryopsis and at the early stages of ontogenesis (on the third, seventh, tenth and twelfth day of seedlings emergence). High lectin activity was observed in samples of seven and ten days seedlings. This could be evidence of active participation of proteins in physiological and biochemical processes at the time of emerging wheat tillering node that can be important in forming adaptive reactions to stress. Wheat lectin activity variation for рН ranging between 4,0 and 8,0 that depended on a variety was defined for the first time. Conclusions. According to hemagglutination ability of wheat lectins, all varieties were divided into three groups: of high ability – 4 varieties, middle – 9 varieties, low – 1 variety. Varietal variability for the investigated trait can be used in breeding programs as marker trait. In most varieties variation of hemagglutination was observed at dilutions ranging between 1:2 and 1:256. Availability of two lectin agglutination peaks in acidic and alkaline media for seedlings was fixed.

When creating Triticum aestivum L. varieties in V. M. Remeslo Myronivka Institute of Wheat, intervarietal crossing was used as the basis for classical method of hybridization. Parental pairs were selected according to environmental and geographical principles. Crossing of lines selected by certain adaptive characteristics among themselves and with the local breeding varieties are widely used. The method of induced mutagenesis is one of the main factor of the evolutionary process which increases hundreds of times the frequency of altered forms occurrence. Thermal mutagenesis – selection of winter forms from spring varieties – is based on low temperature effect on spring wheat varieties (it was developed by V. M. Remeslo, the member of the Academy of Sciences). Method of intervarietal selection in winter wheat breeding provides creation of new varieties of this crop through mass selection of the best plants from local populations. At present, 10 varieties of winter wheat are included into the State Register of Plant Varieties Suitable for Dissemination in Ukraine, one variety – into the State Register of Breeding Achievements of the Russian Federation, seven varieties are passing state quality testing in these countries.

When creating Triticum aestivum L. varieties in V. M. Remeslo Myronivka Institute of Wheat, intervarietal crossing was used as the basis for classical method of hybridization. The article highlights the results of studying the soft winter wheat response to the duration of enforced winter dormancy and the time of vegetation recommencing, their impact on growth, development and the survival of the crops. It is found that the impact of those factors in the conditions of central Forest-Steppe of Ukraine is essential, which is to be taken into consideration when scheduling the measures of spring and summer care over the cultivated crops, in particular, where the spring extra nutrition takes place, pesticides and growth regulators are applied, the spaced planting resowing or partial resowing issues are to be settled down. The ecological effect of spring vegetation recommencing dates does not expose annually, therefore it is not always possible to predict the plant development type, but it is possible, nevertheless, to influence the processes of growth, development and survival of plants throughout spring-summer period and the development of their production capacity by means of introducing the intense technologies, optimization of mineral nutrition and the use of plant growth regulators, protection from rogues, diseases, pests.

The article highlights the results of studying the soft winter wheat response to the duration of enforced winter dormancy and the time of vegetation recommencing, their impact on growth, development and the survival of the crops. It is concluded by investigations that wheat crossing of various development types between themselves cause increase of formbuilding process in hybrid progeny, promoting the selection of practically valuable recombinats. The genotypes which present the practical valuable by complex of adaptive traits and properties have been selected by phenotype stability in the breeding process. The new bread winter wheat variety Pamyati Remesla developed with participation of spring wheat variety Hja 22139 (Finland) has been proposed for including it into the Register of Plant varieties of Ukraine adapted for use in Steppe, Forest- Steppe and Woodland of Ukraine since 2010.

The study of impact of complex application of biological compositions based on fixing nitrogen by microorganisms and plant lectin and nitric fertilizers on productivity and quality of grain of winter wheat on the dark-grey soil in the field terms was conducted. Purpose. Theoretical and experimental substantiation of introduction of “Romantica” roses originated by Meilland company under the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. Methods. Biometrical, comparative-morphological, statistic ones. Results. Viability, growth and development indices as well as ornamental qualities (tolerance for winter conditions, frost hardiness, drought resistance, complex disease and pest resistance, habit maintenance, shoot formation, flowering, coloration, shape of flowers, petal number etc.) were analyzed for 12 rose cultivars of “Romantica” series planted on the collection and exhibition plots of the National Dendrological Park «Sofiivka» NAS of Ukraine. The investigations were conducted within 2012–2015. The most of the cultivars got the mark of introduction success 41–45 points of 50; so, the cultivars are totally promising for introduction into the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. ‘Leonardo da Vinci’ and ‘Yves Piaget’ cultivars got point number 35–38 and they can be considered promising ones. The acclimatization number for the most of the investigated cultivars was 85–90 points of 100 that means good adaptation of the plants in the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. The adaptation of cv. ‘Leonardo da Vinci’ and cv. ‘Yves Piaget’ roses (point number 75) can be considered satisfactory. The total evaluation of “Romantica” roses ornamental qualities made 76–87 points of 100; so, all the cultivars display high ornamental qualities and deserve to be introduced to the production. Conclusions. The garden roses of “Romantica” series originated by Meilland company being introduced into the conditions of the Right-Bank Forest-Steppe Zone of Ukraine are adapted well in the introduction region and they are promising for the subsequent introduction.
