Studying the potential of the initial potato material with the aim of breeding for drought resistance

Purpose. To evaluate and select hybrids, varieties of the parental nursery of the breeding process, biotechnological lines and wild species of potato for physiological parameters of drought resistance. Methods. Physiological and biochemical, selection ones, statistical data processing. Results. The data is given concerning the evaluation of the water retaining and water regeneration capacity of potato leaves of promising hybrids of competitive and ecological test, varieties of the parental nursery, biotechnological lines and wild species and their integral indicator of drought resistance. The studied samples were grown in the nurseries of field selection crop rotation. Accordingly, the initial material with the highest drought resistance value has been defined. Among the evaluated material, eight hybrids of the competitive and ecological test have been selected (drought resistance coefficient was ranging from 59.4% to 84.8%) and five biotechnological lines of ‘Hlazurna’ and ‘Dorohin’ varieties (drought resistance coefficient was in the range of 55.5% to 67.5%). As for wild species, almost half of the samples (47.8%) were characterized by a high coefficient of drought resistance (from 55 to 78%). Selected samples with high values of drought resistance were recommended to use as a source and drought resistance donors when creating new potato varieties. Conclusions. The initial potato material (hybrids, varieties, biotechnological lines and wild species) with high values of drought resistance (55.0–84.8%) has been selected. These samples are recommended to use in the breeding process when creating new drought resistance potato varieties.

Purpose. To reveal breeding and genetic peculiarities of modern spring barley varieties for the “number of grains per main ear” trait and identify genetic sources of increased combining ability for involving in hybridization. Methods. Investigations were carried out at the V. M. Remeslo Myronivka Institute of Wheat of NAAS of Ukraine. Modern varieties of domestic (‘Virazh’, ‘Talisman Myronivskyi’, ‘Komandor’) and foreign (‘KWS Aliciana’, ‘KWS Bambina’, ‘Zhana’, ‘Explorer’) breeding were involved in crossing for a full (7´7) diallel scheme. Parents and F1 were studied in field conditions during 2014–2016. Results. The analysis of variance of combining ability has shown a significant advantage in varying of general combining ability (GCA) effects. The mean square of specific combining ability (SCA) was significantly less than the GCA, but reliable throughout the years. The reciprocal effect was reliable only in 2014. Stably high effects of GCA during all years of investigations were noted in the varieties ‘KWS Aliciana’ (1.18–1.62) and ‘Virazh’ (1.33–1.48). The variety ‘KWS Bambina’ was characterized by lower but reliable positive effects of GCA (0.43–0.99) as compared to mentioned above. Non-allelic gene interaction was not found, that allowed to calculate the basic parameters of genetic variation. During all years of investigations, dominant effects of genes (H1 and H2) prevailed over the additive (D) ones in phenotypic expression of grain number per main ear. Mean degree of dominance in the experiment (H1/D) has shown overdominance. The same pattern was also distinctive for the index of mean degree of dominance in the loci . The dominance was reliably directed. Dominant effects of genes increased grain content, and recessive ones reduced it. At least 3–4 genes (groups of genes) have been revealed which determined the effects of dominance. At the same time, recessive genes (F<0) or gene effects were prevailed quantitatively in the varieties investigated. A high coefficient of heritability in broad sense (H2 = 0.98) has shown a significant determination of phenotypic variability with genetic factors. The coefficient of heritability in narrow sense (h2 = 0.66–0.68) confirmed that despite the advantage of dominant effects over the additive ones, the contribution of the latter was also significant. Conclusions. The prevalence of dominant effects of genes in the phenotypic expression of the number of grains per main ear causes the need for sufficient sample size of hybrid material and points to the expediency of conducting a more “rigid” selection for phenotype in later generations. At the same time, the considerable contribution of additive effects and high values of heritability indices give reason to predict the efficiency of selections aimed at increasing the trait in created hybrid material. The varieties ‘Virazh’, ‘KWS Aliciana’, ‘KWS Bambina’ should be used as effective genetic sources to increase grain content in combination breeding.

Purpose. To define spring barley breeding lines with an optimal combination of yielding capacity and stability under different weather conditions in the Central Forest-Steppe zone of Ukraine. Methods. Field studies, ANOVA, AMMI, GGE biplot analysis. Results. Hydrothermal regime during interphase periods of spring barley vegetation under conditions of The V. M. Remeslo Myronivka Institute of Wheat of NAAS in 2012–2014 was characterized by significant variability, that facilitated detailed evaluation of the lines for productivity, stability, and resistance to abiotic and biotic factors. In the performance of an experiment, the highest average yielding capacity (5.87 t/ha) was noted in 2012, the lowest one (3.50 t/ha) was in 2013. As for these years, average yield of 4.63 t/ha was obtained in 2014. By applying AMMI and GGE biplot analysis, significant differences in response of the studied lines to variability of weather conditions was revealed. Using AMMI model, additive components of the main effects of the breeding lines and years of testing as well as multiplicative components of their interaction were characterized. GGE biplot genotypes ranking in relation to a hypothetical “ideal” genotype showed an absolute advantage of breeding line ‘Nutans 4540’ for yielding capacity and stability. In addition, breeding lines ‘Nutans 4241’ and ‘Nutans 4120’ were close to ideatype. Selected breeding lines were characterized by resistance and moderate resistance to leaf diseases and lodging. Conclusions. Use of AMMI and GGE biplot analysis to evaluate breeding lines at the final stages of breeding process allows to describe them thoroughly and graphically as well as differentiate them not only for average yielding capacity but also for their interaction with changing conditions during the years of testing. Breeding lines ‘Nutans 4540’, ‘Nutans 4241’, and ‘Nutans 4120’ to be identified for the stability of yielding capacity display in combination with other agronomic characters were transferred to the State variety testing as new spring barley varieties ‘MIP Myrnyi’, ‘MIP Saliut’, and ‘MIP Sotnyk’ respectively.

Purpose. To study the effect of AKM plant growth regulator on growth, development and yield formation of sunflower (Helianthus annuus L.) hybrids in hybridization plots under the conditions of the Southern Steppe zone of Ukraine. Methods. Laboratory tests, field study, statistical evaluation. Results. The results of studies devoted to determining the optimal AKM concentration for the treatment of seeds of the maternal and paternal lines, the effect of AKM on field germination, biometric parameters of sunflower plants, seed quality (F1) and yield are presented. Three hybrids of the Ukrainian selection, such as ‘Alpha’, ‘Logos’ and ‘Persei’ were studied during 2014–2016. Optimal concentration of AKM (0.0015 g/l) was defined. The vigor of seeds processed by AKM was higher than in check variety by 0.8–12.8 р.р. (♂); 0.4–10.7 p.p. (♀), laboratory germination – by 2.3–6.1 p.p. (♂); 3.5–6.2 p.p. (♀). In 2016, the sunflower plant height for all variants exceeded this parameter to be obtained for other years of the study. This could be explained by the fact that HTC in 2016 for the BBCH 00–39 period was 1.4 times higher than in 2015. In general, hybrids as the studied factor considerably influenced sunflower yield, and the share of the hybrid (factor A) influence is 33%. This should be considered when selecting hybrids for sunflower cultivation technologies in the Steppe zone of Ukraine. Conclusions. Hydrothermal conditions of the year had the maximum impact on the formation of seed quality and yield of sunflower plants of the hybrids under study, but when using AKM growth regulator for presowing seed treatment, this negative impact was reduced by an average of 23%.

Purpose. To identify and describe the basic criteria for studying and assessing decorative and economic-and-biological characters of the genus Heuchera L. varieties in the context of introduction. Methods. Introduction procedure, analytical approach, morphological analysis and biometric data evaluation. Results. Aided by the comparative morphological analysis of introduced varieties of the genus Heuchera, significant differences in their characters were revealed, including plant height, height and width of the basal rosette of leaves, generative shoot height. There was a significant variation of such parameters as the number of generative shoots within one plant, the number of flowers on a single generative shoot, flowering abundance. Based on investigation findings, 17 groups of varieties were identified for the dominant color of adaxial surface of the leaf blade and 7 groups – for inflorescence color. According to phenological observations, Heuchera varieties groups were determined and the dates of commencement and duration of plants flowering were registered in the context of introduction. The results of these investigations are the necessary basis for the study and evaluation of this culture assortment, they are important in breeding and landscaping. Conclusions. Based on the results of investigations of morphological features of introduced species of the genus Heuchera, traits and parameters for varieties grouping were defined as well as decorative and economic-and-biological traits that should be evaluated.

Purpose. To establish the reasons of submitting not highly decorative lilac hybrids to state variety testing so that unpromising varieties and look-alikes can be sometimes included in the world collection, and focus on solving this situation. Methods. Analytical approach. Results. It was established why and how unoriginal hybrids were submitted to state variety testing that for several reasons can allow them to obtain the status of variety. Conclusions. In order to prevent obtaining the status of variety by some low decorative lilac hybrids, it is necessary that both a breeder and testing stations employer should use known presentable collections of identified varieties for evaluation of real value of a new hybrid. In Ukraine, there is such a collection at N. N. Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine where more than 130 varieties and 21 species of the genus Syringa occupies the area of 2.35 ha. It is desirable to compare not only with the check variety that the breeder selected for his hybrid among such, but, if it is possible, with all varieties in the collection close to it by decorative effect. The problem is that the author of a hybrid, sometimes ignorantly or for some other reason, selects a check variety for his hybrid that is not the most decorative one among such, against which a candidate for variety status will be more attractive during its evaluation. In such a case, the breeder should change a check variety that permits to estimate really the decorative effect of a submitted hybrid.

Purpose. To review publications relating to the key point of the genotyping technology that is competitive allele-specific polymerase chain reaction (which is called now Kompetitive Allele Specific PCR, KASPTM) and its use in various genetic-breeding researching (a study of maize). Results. The essence of KASP-genotyping, its advantages are highlighted. The requirements for matrix DNA are presented, since the success of the KASP-analysis depends on its quality and quantity. Examples of global projects of plant breeding for increasing crop yields using the KASP genotyping technology are given. The results of KASP genotyping and their introduction into breeding and seed production, in particular, for determining genetic identity, genetic purity, origin check, marker-assisted selection, etc. are presented using maize as an example. It is demonstrated how genomic selection according to KASP genotyping technology can lead to rapid genetic enhancement of drought resistance in maize. Comparison of the effectiveness of creating lines with certain traits (for example, combination of high grain yield and drought resistance) using traditional breeding approaches (phenotype selection) and molecular genetic methods (selection by markers) was proved that it takes four seasons (two years in case of greenhouses) in order to unlock the potential of the plant genotype using traditional self-pollination, test-crossing and definitions), while using markers, the population was enriched with target alleles during one season. At the same time, there was no need for a stress factor. Conclusions. KASP genotyping technology is a high-precision and effective tool for modern genetics and breeding, which is successfully used to study genetic diversity, genetic relationship, population structure, genetic identity, genetic purity, origin check, quantitative locus mapping, allele mapping, marker-assisted selection, marker-assisted breeding. It is expedient and timely to introduce KASP genotyping technology in our country to solve a wide range of modern genetics, breeding, seed production tasks.

Purpose. To determine the effect of the basic organo-mineral fertilizer on the formation and stability of grain quality of winter wheat varieties in multiple crop rotation after black fallow as a predecessor. Methods. Field experiments were based on a multifactorial scheme using split-plot method with due regard to all requirements of the field experiment procedure, analysis of variance was used for statistical processing of the obtained results. Results. Investigation data was given concerning determination of grain quality indices in winter wheat varieties of different ecotypes after black fallow as a predecessor depending on organo-mineral fertilizer application in the Left-Bank Forest-Steppe zone of Ukraine. In average for the period of investigation (2011–2015), the highest protein content in winter wheat grains was formed in no treatment variant [in such varieties as ‘Doridna’ (14.1%), ‘Dykanka’ (14.3%) and ‘Levada’ (14.2%)] and in case of organo-mineral fertilizer application [in the varieties ‘Hordovyta’ (14.0%), ‘Kalyta’ (14.0%), ‘Dykanka’ (14.7%) and ‘Levada’ (14.6%)]. The highest content of crude gluten in grains, without regard for the variant of the experiment, was found in the following varieties as ‘Dykanka’ (24.9–25.1%) and ‘Levada’ (23.7–25.4%). Conclusions. It was established that the content of protein and crude gluten in grains as well as the falling number of winter wheat was highly dependent on such factors as the variety and the year of cultivation as compared to the fertilizer background. The following varieties as ‘Hordovyta’, ‘Mulan’, Dykanka’ and ‘Levada’ were very sensitive to the application of organo-mineral fertilizer for the protein content, while ‘Hordovyta’ (2.4%), ‘Levada’ (1.7%), ‘Borvii’ (1.2%) and ‘Mulan’ (1.1%) – for the crude gluten content.

Purpose. To make comprehensive assessment of introduced oat samples of various eco-geographical origin under the conditions of the Forest-Steppe zone of Ukraine for the set of productivity and adaptability indices in order to define the most valuable ones and describe them. Methods. Field study, laboratory analysis, generalization. Results. The authors presented results of the study, evaluation and description of 35 new samples of oats from 5 countries for productivity and adaptability traits during 2011–2015 in Ustymivka Experimental Station for Plant Production to be run by the V. Ya. Yuriev Institute for Plant Production of NAAS. In field and laboratory conditions, the following indicators as yielding, productivity, thousand-kernel weight, early ripening, height of plants and length of panicle, lodging resistance were studied. Conclusions. A set of studies of new oat samples under various weather conditions allowed to identify material with increased parameters of economic and biological characters. During studies, it was found that ‘Laima’, ‘Stendskaia Darta’ (Latvia), ‘Borowiak’ (Poland), ‘Faks’ (Belarus) were the high-yielding varieties. Panicles in such oats samples as ‘Cwal’, ‘Borowiak’ (Poland), ‘Iakov’, ‘Mutika 1077’, ‘Mutika 990’, ‘Irtysh 22’, ‘Pokrovskyі 9’ (Russia), ‘Faks’ (Belarus), ‘Stendskaia Darta’, ‘Stendskaia Mara’ (Latvia) contained a large mass of grains (more than 2,5 g), panicle productivity in these samples was rather high both due to increased amount of grains in it and at the expense of thousand-kernel weight. Analysis of the study results showed that introduced oat samples of various eco-geographical origin were adapted to the Southern Forest-Steppe zone and can be recommended as an initial material in breeding for increasing productive and adaptive capacity.

Purpose. To investigate the variability of plant biometrics of cutting lettuce (Lactuca sativa L.) varieties depending on the concentration of microfertilizer Avatar-1. Methods. Field study, biometric technique, comparative approach, statistical evaluation, generalization. Results. Dependence of the plant height, the diameter of the leaf rosette, the number of leaves per plant, the leaf area of plants on the concentration of microfertilizer Avatar-1 was defined. Investigations of cutting lettuce ‘Afitsyon’ and ‘Concord’ varieties by Dutch breeding were conducted in 2016–2017 in the collection sites of the department of vegetable growing in the scientific-experimental field “Fruit and vegetable garden” of the National University of Life and Environmental Sciences of Ukraine. In order to determine dependence of plant biometrics of cutting lettuce varieties on the concentration of complex microfertilizer Avatar-1, the following scheme was used for the both varieties: variant 1 – water (control); variant 2 – 0,10% solution; variant 3 – 0,25% solution; variant 4 – 0,50% solution. Plants were treated with microfertilizer three times during the vegetative period. Alterations of plant biometrics of cutting lettuce ‘Afitsyon’ and ‘Concord’ varieties depending on the concentration of complex microfertilizer Avatar-1 was studied. Conclusions. It was found that in case of three-time plant treatment with complex microfertilizer Avatar-1 at the time of harvesting, the best plant biometrics was registered in variant 3 (concentration 0,25%) for the both ‘Afitsyon’ and ‘Concord’ varieties of cutting lettuce. The height of plants of the cutting lettuce in ‘Afitsyon’ variety exceeded this figure in ‘Concord variety by 1,1–1,4 cm. The concentration of microfertilizer had no significant effect on the diameter of the leaf rosette of ‘Concord’ variety (25,1–25,9 см). The diameter of the leaf rosette of ‘Afitsyon’ variety was 26,0–28,7 cm. In cutting lettuce, the largest leaf area per plant was registered in ‘Afitsyon’ variety (3516.5 cm2/plant ) in case of plants treatment with 0,25% solution of complex microfertilizer Avatar-1 that exceeded this figure in ‘Concord’ variety by 660 cm2/plant. The best plant biometrics of cutting lettuce of the studied varieties and optimal concentration of complex microfertilizer Avatar-1 (0.25% solution) was defined that is important for improving agricultural cultivation techniques.
