Bioaccumulation of per- and polyfluoroalkyl substance in fish from an urban river: Occurrence, patterns and investigation of potential ecological drivers Texto completo

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous in aquatic environments and a recent shift toward emerging PFAS is calling for new data on their occurrence and fate. In particular, understanding the determinants of their bioaccumulation is fundamental for risk assessment purposes. However, very few studies have addressed the combined influence of potential ecological drivers of PFAS bioaccumulation in fish such as age, sex or trophic ecology. Thus, this work aimed to fill these knowledge gaps by performing a field study in the Seine River basin (France). Composite sediment and fish (European chub, Squalius Cephalus) samples were collected from four sites along a longitudinal transect to investigate the occurrence of 36 PFAS. Sediment molecular patterns were dominated by fluorotelomer sulfonamidoalkyl betaines (i.e. 6:2 and 8:2 FTAB, 46% of ∑PFAS on average), highlighting the non-negligible contribution of PFAS of emerging concern. C₉–C₁₄ perfluoroalkyl carboxylic acids, perfluorooctane sulfonic acid (PFOS), perfluorooctane sulfonamide (FOSA) and 10:2 fluorotelomer sulfonate (10:2 FTSA) were detected in all fish samples. Conversely, 8:2 FTAB was detected in a few fish from the furthest downstream station only, suggesting the low bioaccessibility or the biotransformation of FTABs. ∑PFAS in fish was in the range 0.22–3.8 ng g⁻¹ wet weight (ww) and 11–140 ng g⁻¹ ww for muscle and liver, respectively. Fish collected upstream of Paris were significantly less contaminated than those collected downstream, pointing to urban and industrial inputs. The influence of trophic ecology and biometry on the interindividual variability of PFAS burden in fish was examined through analyses of covariance (ANCOVAs), with sampling site considered as a categorical variable. While the latter was highly significant, diet was also influential; carbon sources and trophic level (i.e. estimated using C and N stable isotope ratios, respectively) equally explained the variability of PFAS levels in fish.

The number of published studies evaluating the effects of microplastics (MPs) in fish has increased in the last decade. However, of the available studies, few have explored the long-term effects of MPs on fish growth and reproduction and have resorted to MPs in the form of μm-sized beads/microspheres. In this study, 6-10 day-old post-hatch medaka (Oryzias latipes) fish were exposed to 50 (i.e. 1X) and 500 (i.e. 10X) μg of heterogeneously sized and irregularly shaped virgin polystyrene (PS) MP particles (200-μm range)/L for 150 days. These concentrations corresponded to respective daily mean values of 247 and 3087 particles/L administered through the diet. The PS MPs dietary exposure resulted in body burdens of 114 and 440 particles/g fish on day 50, and of 78 and 173 particles/g fish on day 100 since the respective exposures to the 1X and the 10X treatments started. The biometric analyses found no incidence of PS MPs ingestion on overall fish growth and development. The histological survey in the 10X group did not reveal alterations in gills or in the digestive tract. Mild alterations in other organs were seen and included increased fluid material in the peritoneal cavity, glomerular and tubular alterations in kidneys, and differences in the diameter of the thyroid follicles and thickness of the follicular epithelial cells. The initial days of the reproductive phase revealed MP-related differences in the number of gravid females, fecundity, and fertilization rates. Overall, these values reverted to normal rates throughout the succeeding days. No significant effects of PS MPs exposure were evidenced on offspring success. The 150-day PS MPs dietary exposure used in this study provided clues of histological effects and a reproduction delay. However, it did not seem to compromise overall growth/thriving and the ongoing reproduction.

A multinational demersal longline survey was conducted on the Gulf of Mexico continental shelf over the years 2015 and 2016 to generate a Gulf-wide baseline of polycyclic aromatic hydrocarbon (PAH) concentrations in demersal fishes. Tilefish (Lopholatilus chamaeleonticeps) were sampled in all regions of the Gulf of Mexico for biometrics, bile, and liver. Tilefish liver was also obtained from surveys in the northwest Atlantic Ocean for comparison. Liver tissues (n = 305) were analyzed for PAHs and select alkylated homologs using QuEChERS extractions and gas chromatography tandem mass spectrometry. Bile samples (n = 225) were analyzed for biliary PAH metabolites using high-performance liquid chromatography with fluorescence detection. Spatial comparisons indicate the highest levels of PAH exposure and hepatic accumulation in the north central Gulf of Mexico, with decreasing concentrations moving from the north central Gulf counterclockwise, and an increase on the Yucatán Shelf. Hepatic PAH concentrations were similar between the Gulf of Mexico and the northwest Atlantic, however, Tilefish from the northwest Atlantic had higher concentrations and more frequent detection of carcinogenic high molecular weight PAHs. Overall, results demonstrate that PAH pollution was ubiquitous within the study regions, with recent exposure and hepatic accumulation observed in Tilefish from both the Gulf of Mexico and northwest Atlantic.

Children can be exposed to organophosphate and carbamate mixtures, which pose additive health effects via soil exposure. However, only 23 countries have soil standard values for organophosphate and carbamate pesticides, and most regulatory jurisdictions do not consider the cumulative exposure. This study derived proposed soil standards for organophosphates and carbamates by introducing the relative potency quotient approach (RPQ). The probabilistic cumulative risk assessment was also applied to evaluate current soil standards of pesticide mixtures. The U.S. Environmental Protection Agency (EPA) have soil standards of 19 organophosphates and five carbamates. However, these standards cannot protect population health via chronic exposure in conservative and semi-conservative scenarios based on the probabilistic risk assessment because the U.S.EPA simplified the regulatory process for the cumulative exposure to pesticide mixtures and omitted the soil allocation factor, which should be set for aggregate exposure. The analysis of proposed soil standards developed by the RPQ approach indicates that some human behavior variables, such as soil intake rate and exposure duration, have stronger impacts on the proposed soil standards than human biometric variables like body weight. This study may be helpful to develop regulatory standards and a framework for pesticide mixtures having additive health effects.

The Subantarctic Kerguelen Islands (49°S, 70°E) contain freshwater ecosystems among the most isolated in the world. Concentrations of polychlorinated biphenyls (PCBs) were assessed in the muscle of 48 brook trout and 38 brown trout caught during summer and spring 2006 in the rivers, lakes and ponds of Kerguelen. The sum of 29 PCBs averaged 404 and 358ngg⁻¹ lipid, and dioxin-like PCB was 19 and 69ngg⁻¹ lipid, in brook and brown trout, respectively. The values showed a high variability and some fish accumulated PCBs at levels similar to those of fish from impacted areas. While inter-sex differences were limited, the season and the morphotype appeared to have the most influence. Fish captured in summer had muscle PCB concentrations about three times higher than those caught in spring and the ‘river’ morphotype of brook trout showed the highest PCB levels.

Current approaches in chemical hazard assessment face significant challenges because they rely on live animal testing, which is time-consuming, expensive, and ethically questionable. These concerns serve as an impetus to develop new approach methodologies (NAMs) that do not rely on live animal tests. This study explored a molecular benchmark dose (BMD) approach using a 7-day embryo-larval fathead minnow (FHM) assay to derive transcriptomic points-of-departure (tPODs) to predict apical BMDs of fluoxetine (FLX), a highly prescribed and potent selective serotonin reuptake inhibitor frequently detected in surface waters. Fertilized FHM embryos were exposed to graded concentrations of FLX (confirmed at < LOD, 0.19, 0.74, 3.38, 10.2, 47.5 μg/L) for 32 days. Subsets of fish were subjected to omics and locomotor analyses at 7 days post-fertilization (dpf) and to histological and biometric measurements at 32 dpf. Enrichment analyses of transcriptomics and proteomics data revealed significant perturbations in gene sets associated with serotonergic and axonal functions. BMD analysis resulted in tPOD values of 0.56 μg/L (median of the 20 most sensitive gene-level BMDs), 5.0 μg/L (tenth percentile of all gene-level BMDs), 7.51 μg/L (mode of the first peak of all gene-level BMDs), and 5.66 μg/L (pathway-level BMD). These tPODs were protective of locomotor and reduced body weight effects (LOEC of 10.2 μg/L) observed in this study and were reflective of chronic apical BMDs of FLX reported in the literature. Furthermore, the distribution of gene-level BMDs followed a bimodal pattern, revealing disruption of sensitive neurotoxic pathways at low concentrations and metabolic pathway perturbations at higher concentrations. This is one of the first studies to derive protective tPODs for FLX using a short-term embryo assay at a life stage not considered to be a live animal under current legislations.

This study evaluated the chronic toxicity (30 days) of different sizes of polyethylene terephthalate (PET) microplastics (60–3000 μm) provided alone or in combination with acid rain, on garden cress (Lepidium sativum). Both biometrical and physiological traits have been evaluated: i) percentage inhibition of seed germination, plant height, leaf number and fresh biomass production; ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione production); iii) impairment in photosynthetic machinery in term of pigments production; iv) aminolevulinic acid and proline production. Results highlighted that different sizes of PET, alone or in combination with acid rain, are able to negatively affect both biometrical and physiological plant traits. In particular, the lower size of microplastics is able to negatively affect growth and development, as well as to trigger the oxidative burst. Regarding the pigments production, PET coupled with acid rain, induced a higher production of Chl-b, and an inhibition of aminolevulinic acid.

Urine sample tests are one of the most common methods of estimating human exposure to polycyclic aromatic hydrocarbons (PAHs) and assessing population health risks. To evaluate the reliability of the urine test and the impact of other PAH elimination routes on the health risk estimated by this test, we proposed a backward modeling framework integrating other common elimination routes of PAH metabolites to calculate the overall intake rate of the parent PAH based on the levels of corresponding main metabolites in urine. Due to limited biotransformation data, we selected naphthalene as an example to evaluate model performance and collected urine samples from 234 random adults in Shenzhen. The overall intake rates of naphthalene were then simulated and compared to current literature data. The simulated intake rates of naphthalene ranged from 3.70 × 10⁻³ mg d⁻¹ to 1.95 mg d⁻¹ and followed a lognormal distribution with a median value of 6.51 × 10⁻² mg d⁻¹. The results indicated that, if naphthalene exposure occurred only via food for the population of Shenzhen, the literature data fell within the most frequent interval [3.70 × 10⁻³, 4.45 × 10⁻²] but were lower than the simulated median value. However, if other exposure routes were considered, the allocation factor-adjusted literature data were close to the simulated median values. In addition, under normal physiological conditions, the simulated results were more sensitive to 1-hydroxynaphthalene (1-OHN) and 2-hydroxynaphthalene (2-OHN) levels in urine than other biometric variables, which is due to the limited load of 1-OHN and 2-OHN in human elimination routes. Furthermore, the suggested safety levels of 1-OHN and 2-OHN in urine to protect 99% of the general population of Shenzhen were 6.40 × 10⁻⁶ and 3.75 × 10⁻⁵ mg L⁻¹, which could be used as regulatory indicators based on the high reliability of the model.

This study provided a comprehensive characterization on ingestion of different typologies of microplastics in several fish and invertebrate species from the Adriatic Sea, considered as a preferential area of plastic accumulation in the Mediterranean. Almost 500 organisms were sampled in the three sectors of Northern, Central and Southern Adriatic, testing the hypothesis that area of collection, habitat and feeding strategy might influence the occurrence of plastic particles in biota. In this study, the overall characterization considered separately plastic microparticles (MPs) from textile microfibers (MFs) which also included natural and semi-synthetic ones. Ingestion of MPs was a widespread phenomenon, but their number (typically 1 or 2) did not reveal any significant relationship with biometric values, geographical areas or ecological features of the species. Conversely, the frequency of ingestion, ranging from 13 to 35% of organisms containing MPs, appeared a more reliable index to highlight such differences, revealing higher values in species from Central and Southern basins compared to the Northern one, as well as in benthopelagic compared to benthic or pelagic organisms. Geographical differences also occurred in terms of size and typology of ingested particles, suggesting the importance of local river runoffs and surface currents dynamics. Textile microfibers (MFs) were also abundant in Adriatic food webs occurring in all the analyzed species with average numbers (3–10) and frequencies (40–70%) higher than those reported for MPs; further, an elevated percentage of MFs (>80%) was of natural or semi-synthetic origin.Overall, this study provided general insights toward the harmonization of a common biomonitoring strategy, as in the context of MSFD, including the suggestion of a frequency-based index and of a multi-species approach to increase the ecological relevance of assessment, as well as the comparability between different areas and trophic webs.

The aim of this study was to determine the effect of persistent organic pollutants (POPs) and biometric variables on circulating levels of steroid hormones (androgens, estrogens and progestagens) in male polar bears (Ursus maritimus) from Svalbard, Norway (n = 23). Levels of pregnenolone (PRE), progesterone (PRO), androstenedione (AN), dehydroepiandrosterone (DHEA), testosterone (TS), dihydrotestosterone (DHT), estrone (E1), 17α-estradiol (αE2) and 17β-estradiol (βE2) were quantified in polar bear serum by gas chromatography tandem mass spectrometry (GC-MS/MS), while POPs were measured in plasma. Subsequently, associations between hormone concentrations (9 steroids), POPs (21 polychlorinated biphenyls (PCBs), 8 OH-PCBs, 8 organochlorine pesticides (OCPs) and OCP metabolites, and 2 polybrominated diphenyl ethers (PBDEs)) and biological variables (age, head length, body mass, girth, body condition index), capture date, location (latitude and longitude), lipid content and cholesterol levels were examined using principal component analysis (PCA) and orthogonal projections to latent structures (OPLS) modelling. Average concentrations of androgens, estrogens and progestagens were in the range of 0.57–83.7 (0.57–12.4 for subadults, 1.02–83.7 for adults), 0.09–2.69 and 0.57–2.44 nmol/L, respectively. The steroid profiles suggest that sex steroids were mainly synthesized through the Δ-4 pathway in male polar bears. The ratio between androgens and estrogens significantly depended on sexual maturity with androgen/estrogen ratios being approximately 60 times higher in adult males than in subadult males. PCA plots and OPLS models indicated that TS was positively related to biometrics, such as body condition index in male polar bears. A negative relationship was also observed between POPs and DHT. Consequently, POPs and body condition may potentially affect the endocrinological function of steroids, including development of reproductive tissues and sex organs and the general condition of the male polar bears.
