Accumulation of pollutants in nestlings of an endangered avian scavenger related to territory urbanization and physiological biomarkers

We monitor pollutant accumulation and investigate associated changes at the physiological level within the population of an obligate avian scavenger, the Egyptian Vulture (Neophron percnopterus), from Catalonia (NE Spain). This population is expanding its range, presumably because of the use of human waste disposal sites as food resource. We hypothesized that habitat urbanization, presumably associated with feeding from human wastes, could influence the accumulation of persistent organic pollutants and metals. The aim of this study was to explore the relationship between accumulated pollutants and biochemical parameters in nestling blood. We used the proportion of urban surface within an 8 km radius of each nest as a proxy to study the relationship between anthropic influence and pollutant accumulation. Observed blood levels of metals, organochlorine pesticides, polychlorinated biphenyls (PCBs), per- and polyfluoroalkylated substances (PFAS) and polybrominated diphenyl ethers (PBDEs) were relatively low, as expected for nestling individuals due to short-term exposures. CB-180 and PBDEs were associated with variations in blood biochemistry parameters; hexa-BDEs appeared positively associated with activities of the enzymes aspartate aminotransferase and lactate dehydrogenase, whereas CB-180 accumulation was associated with an increased activity of creatine phosphokinase and elevated glutathione levels. Increased CB-180 levels were also related to decreased blood concentrations of calcium, cholesterol, α-tocopherol and lutein. A proportion of urban surfaces as low as 6.56% within a radius of 8 km around the nest appears related to the accumulation of CB-180, the majority of analysed PFAS and of PBDE congeners 99 and 209, and increased urbanization was also associated with decreased plasma levels of α-tocopherol and carotenoids. These associations suggest that changes in blood profiles of vitamins, carotenoids or other analytes, despite related to increased plasma levels of CB-180, would be consequence of exploitation of artificial food sources rather than of a direct effect of the pollutants.

Although mercury bio-amplifies through the food chain and accumulates in top predators, mercury concentrations in tissues of the wandering albatross are greater than in any other vertebrate, including closely related species. In order to explore the alternative explanations for this pattern, we measured total mercury concentrations in feathers, plasma and blood cells of wandering albatrosses of known age, sex and breeding status sampled at South Georgia. Mercury concentrations were low in feathers and blood components of chicks, and higher in the feathers of young pre-breeders than in feathers or blood of older pre-breeders and breeding adults. There was no effect of sex on mercury concentrations in the feathers of pre-breeders or breeding adults, whereas levels were significantly higher in blood cells of breeding females than males. The high feather mercury concentrations of young pre-breeders compared with older birds suggest an increase in moult frequency as birds approach maturity.

Environmental pollution by metals has repercussions on wildlife health. It is known that some metals can have an influence on the neuroendocrine stress response, and at the same time, metals have pro-oxidant effects that can overwhelm the antioxidant system and cause oxidative stress. This study evaluates the association of metals with neuroendocrine stress activity and biomarkers of oxidative stress in 42 captive female Morelet's crocodiles (Crocodylus moreletii). We measured five metals of ecotoxicological importance (Hg, Cd, Pb, Cu and Zn), and three biomarkers of the oxidative stress response in the liver: glutathione (GSH) and glutathione disulfide (GSSG) as markers for antioxidant system and thiobarbituric acid reactive substances (TBARS) for oxidative damage. We also measured one biomarker of the neuroendocrine response to stress: corticosterone (B) in blood plasma. The mean ± SD concentrations of metals in the liver expressed in μg/g (dw) were: Cd: 0.004 ± 0.003, Hg: 0.014 ± 0.019, Cu: 0.017 ± 0.013, Zn: 0.043 ± 0.035, Pb: 0.16 ± 0.256. The mean ± SD of GSH was 0.42 ± 0.35 nmol/mg protein, the mean ± SD of GSSG was 0.24 ± 0.20 nmol/mg protein, the mean ± SD concentrations of TBARS were 0.36 ± 0.21 nmol/mg protein, and the mean ± SD of B was 393.57 ± 405.14 pg/mL. Hg presented a significant negative relationship with corticosterone. Cd had a negative relationship with both GSH and GSSG; meanwhile, Zn showed a negative relationship with TBARS levels, could be a protective element against hepatic oxidative damage. Finally, B had negative relationship with oxidative damage. The connection found between Hg and the neuroendocrine stress response, as well as the correlations of Cd and Zn with oxidative damage and antioxidant activity should be studied further, given their toxicological importance and implications for the conservation of C. moreletii and other crocodilians.

Freshwater ecosystems are facing increasing contamination by major ions. The Multi-Ion Toxicity (MIT) model, a new tool for risk assessment and regulation, predicts major ion toxicity to aquatic organisms by relating it to a critical disturbance of the trans-epithelial potential (TEP) across the gills, as predicted by electrochemical theory. The model is based on unproven assumptions. We tested some of these by directly measuring the acute TEP responses to a geometric series of 10 different single salts (NaCl, Na₂SO₄, KCl, K₂SO₄, CaCl₂, CaSO₄, MgCl₂, MgSO₄, NaHCO₃, KHCO₃) in the euryhaline rainbow trout (Oncorhynchus mykiss) and the stenohaline goldfish (Carassius auratus) acclimated to very soft, ion-poor water (hardness 10 mg CaCO₃/L). Results were compared to 24-h and 96-h LC50 data from the literature, mainly from fathead minnow (Pimephales promelas). All salts caused concentration-dependent increases in TEP to less negative/more positive values, in patterns well-described by the Michaelis-Menten equation, or a modified version incorporating substrate inhibition. The ΔTEP above baseline became close to a maximum at the 96-h LC50, except for the HCO₃⁻ salts. Furthermore, the range of ΔTEP values at the LC50 within one species was much more consistent (1.6- to 2.1-fold variation) than the molar concentrations of the different salts at the LC50 (19- to 25-fold variation). ΔTEP responses were related to cation rather than anion concentrations. Overall patterns were qualitatively similar between trout and goldfish, with some quantitative differences, and also in general accord with recently published data on three other species in harder water where ΔTEP responses were much smaller. Blood plasma Na⁺ and K⁺ concentrations were minimally affected by the exposures. The results are in accord with most but not all of the assumptions of the MIT model and support its further development as a predictive tool.

Trace element profiling in the incrementally formed dentine of mammalian teeth can be applied to reconstruct temporal variation of incorporation of these elements into the tissue. Using an electron microprobe, this study analysed fluoride distribution in dentine of first and third mandibular molars of free-ranging eastern grey kangaroos inhabiting a high-fluoride area, to assess temporal variation in fluoride uptake of the animals. Fluoride content in the early-formed dentine of first molars was significantly lower than in the late-formed dentine of these teeth, and was also lower than in both, the early and the late-formed dentine of third molars. As early dentine formation in M1 takes place prior to weaning, this finding indicates a lower dentinal fluoride uptake during the pre-weaning compared to the post-weaning period. This is hypothetically attributed to the action of a partial barrier to fluoride transfer from blood to milk in lactating females and a low bioavailability of fluoride ingested together with milk. Another factor contributing to lower plasma fluoride levels in juveniles compared to adults is the rapid clearance of fluoride from blood plasma in the former due to their intense skeletal growth. The combined action of these mechanisms is considered to explain why in kangaroos from high-fluoride areas, the (early-formed) first molars are not affected by dental fluorosis while the (later-formed) third and fourth molars regularly exhibit marked to severe fluorotic lesions.

This study investigates concentrations of 18 perfluoroalkyl substances (PFAS) in blood plasma of adult lactating Weddell seals (Leptonychotes weddellii) (n = 10) from McMurdo Sound, Antarctica. Perfluoroundecanoic acid (PFUnDA) was detected in all samples at concentrations ranging from 0.08 to 0.23 ng/ml. Perfluorooctane sulfonate (PFOS), perfluorohexanoate (PFHxA) and perfluorotridecanoate (PFTriDA) were sporadically detected, while the remaining compounds were below the limit of detection. This is the first report of detectible concentrations of PFASs in an endemic Antarctic marine mammal species. We suggest that the pollutants have been subjected to long range atmospheric transportation and/or derive from a local source. A review of these and published data indicate that perfluoroalkyl carboxylates (PFCAs) dominate in biotic PFAS patterns in species feeding south of the Antarctic Circumpolar Current (ACC), whereas PFOS was the major PFAS detected in species feeding predominantly north of the current.

The presence of triclosan, a widely-used antibacterial chemical, is currently unknown in higher trophic-level species such as marine mammals. Blood plasma collected from wild bottlenose dolphins (Tursiops truncatus) in Charleston, SC (CHS) (n = 13) and Indian River Lagoon, FL (IRL) (n = 13) in 2005 was analyzed for triclosan. Plasma concentrations in CHS dolphins ranged from 0.12 to 0.27 ng/g wet weight (mean 0.18 ng/g), with 31% of the sampled individuals having detectable triclosan. The mean IRL dolphin plasma concentrations were 0.072 ng/g wet weight (range 0.025-0.11 ng/g); 23% of the samples having detectable triclosan. In the CHS area, triclosan effluent values from two WWTP were both 190 ng/L and primary influents were 2800 ng/L and 3400 ng/L. Triclosan values in CHS estuarine surface water samples averaged 7.5 ng/L (n = 18) ranging from 4.9 to 14 ng/L. This is the first study to report bioaccumulation of anthropogenic triclosan in a marine mammal highlighting the need for further monitoring and assessment. Animal studies have suggested that phthalate exposure alters the fatty acid composition of blood plasma. Therefore, we conducted an epidemiological study to examine whether urinary concentrations of phthalates are correlated with circulating fatty acids in the general US population. The 2003–2004 and 2011–2012 National Health and Nutrition Examination Survey were used in this study. Ten urinary phthalate metabolites and 23 fatty acids were measured. Fatty acid patterns were identified using principal component analysis (PCA) with an eigenvalue greater than 1. A two-step analysis was performed. We first performed multivariable linear regressions to evaluate whether urinary phthalate metabolites were related to the PCA-derived components of blood fatty acid levels. Then we performed multivariable linear regressions to investigate each of the fatty acids that were suggestively correlated with some of the phthalates in PCA. There were 994 participants (51.91% women). As for men, after adjustments for potential confounding factors, MECPP, MEHHP, and ∑DEHP were all positively correlated with gamma-linolenic, myristoleic, and myristic acids; both MEHHP and ∑DEHP were positively correlated with stearic acid; MMP was positively correlated with docosahexaenoic acid. As for women, MMP was negatively correlated with docosanoic, lignoceric, and arachidic acids; MBzP was negatively correlated with docosahexaenoic acid; both MEHP and MCPP were negatively correlated with docosatetraenoic acid; MEHP was negatively correlated with arachidonic acid, and MCPP was negatively correlated with docosapentaenoic-6 acid. Our findings support that phthalates may be correlated with circulating fatty acids.

We examined relationships between hepatic concentrations of halogenated organic contaminants and ethoxyresorufin O-deethylase (EROD) activity and retinoid (vitamin A) concentrations in livers, as well as retinol and thyroid hormone (TT₃, TT₄) levels in blood plasma, of northern fulmars at two breeding colonies in the Canadian High Arctic. Biomarker levels or responses did not differ significantly between males and females at either colony, nor was there any significant difference between colonies. No significant relationships were found between thyroid hormone or hepatic retinoid concentrations and any of the dioxin-like compounds or their Toxic Equivalents (TEQs) although significant positive correlations were found with plasma retinol (p < 0.03). EROD activity was significantly correlated with hepatic dioxin-like compounds and their TEQs (p < 0.001) as well as total PCBs (p < 0.01), which suggests that EROD induction occurs in northern fulmars at environmentally-relevant concentrations.

The temporal, inter-site, and sex-based variation of 19 organochlorine pesticides (OCPs) in blood plasma samples collected from blue-footed boobies of two islands in Sinaloa, Mexico, was evaluated. The effect of OCPs was evaluated with the heterophil/lymphocyte ratio, micronucleated erythrocyte frequency, and scaled mass index. The OCP-group levels decreased as the breeding season progressed, and interannual (but not inter-colony) differences were detected. Intra-annual variation in OCP levels seemed to reflect run-off inputs, although other environmental processes may better explain the variation between years. Sex-based differences in OCP levels were likely related to ecological and physiological processes linked to breeding (e.g., egg-laying and use of lipid reserves). No correlations between OCP-group levels and biomarkers were detected. Small pelagic fishes are the main prey sources of blue-footed boobies and the targets of regional industrial fisheries, and thus blue-footed booby OCP levels could reflect ecosystem health and indicate potential risks for human consumers.
