Ralstonia eutropha Q2-8 reduces wheat plant above-ground tissue cadmium and arsenic uptake and increases the expression of the plant root cell wall organization and biosynthesis-related proteins

In this study, the molecular mechanisms involved in Ralstonia eutropha Q2-8-induced increased biomass and reduced cadmium (Cd) and arsenic (As) uptake in wheat plants (Triticum aestivum cv. Yangmai 16) were investigated in growth chambers. Strain Q2-8 significantly increased plant biomass (22–75%) without and with Cd (5 μM) + As (10 μM) stress and reduced plant above-ground tissue Cd (37%) and As (34%) contents compared to those in the controls. Strain Q2-8 significantly increased the proportions of Cd and As in wheat root cell walls. Under Cd and As stress, 109 root proteins were differentially expressed among which those involved in metabolisms, stress and defence, and energy were dominant in the presence of strain Q2-8. Furthermore, energy-, defence-, and cell wall biosynthesis-related proteins were found to be up-regulated. Notably, differentially expressed cell wall biosynthesis-related proteins in roots were only found in bacteria-inoculated plants under Cd and As stress. The results suggest that strain Q2-8 can alleviate Cd and As toxicity to wheat plant seedlings and reduce above-ground tissue Cd and As uptake by increasing the efficiency of root energy metabolism, defence, and cell wall biosynthesis under Cd and As stress.

Emission of hazardous trace elements (HTEs) from energy production is receiving much attention due to concerns about the toxicity to the ecosystem and human health. This study presented new field measurement data on the HTEs partitioning behavior and size-segregated elemental compositions of gaseous particular matter (PM) generated from a commercial circulating fluidized bed (CFB) power plant. Mineralogical and morphological characteristics of combustion ash and PM2.5 (particle diameter less than 2.5 μm) were determined by X-ray diffractometer (XRD) and scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM/EDS). Functional groups alteration during CFB combustion was characterized by Fourier transform infrared spectroscopy (FTIR). The presence of aliphatic hydrogen at 2910 cm−1 and 2847 cm−1 in the PM2.5 suggested that the aliphatic carbon-rich volatiles were absorbed on the fine particles with large surface area. Fine fly ash (PM2.5) occurred as irregular glass particles or/and as unburned carbon. The typical irregular particles were mainly composed of Al-Si-Ca or Al-Si-Fe phases. The enrichment behavior of HTEs was determined for the airborne size-segregated particular matter. Elemental occurrences, combustion temperature, unburnt carbon, and limestone additives during CFB combustion were critical in the transformation behavior of HTEs. The total potentially mobile pollutants that exit the CFB power plant every year were estimated as follows: 0.22 tons of Cr, 0.12 tons of Co, 0.73 tons of Ni, 0.04 tons of As, 0.07 tons of Se, 3.95 kg of Cd, and 3.34 kg of Sb.

An experiment was performed to explore the interactive impacts of zinc oxide nanoparticles (ZnO NPs) and cadmium (Cd) on growth, yield, antioxidant enzymes, Cd and zinc (Zn) concentrations in wheat (Triticum aestivum). The ZnO NPs were applied both in Cd-contaminated soil and foliar spray (in separate studies) on wheat at different intervals and plants were harvested after physiological maturity. Results depicted that ZnO NPs enhanced the growth, photosynthesis, and grain yield, whereas Cd and Zn concentrations decreased and increased respectively in wheat shoots, roots and grains. The Cd concentrations in the grains were decreased by 30–77%, and 16–78% with foliar and soil application of NPs as compared to the control, respectively. The ZnO NPs reduced the electrolyte leakage while increased SOD and POD activities in leaves of wheat. It can be concluded that ZnO NPs (levels used in the study) could effectively reduce the toxicity and concentration of Cd in wheat whereas increase the Zn concentration in wheat. Thus, ZnO NPs might be helpful in decreasing Cd and increasing Zn biofortification in cereals which might be effective to reduce the hidden hunger in humans owing the deficiency of Zn in cereals.

In China, the cadmium (Cd) levels in paddy fields have increased, which has led to the excessive uptake of Cd into rice grains. In this study, we determined the physicochemical properties of soil samples, including the pH, soil organic matter (SOM) content, cation exchange capacity (CEC), and total Cd content (Cdsoil) in order to establish a quadratic discriminant analysis (QDA) model for assessing the risk of Cd in rice and to calculate its prior probability. Decision tree and logistic regression models were also established for comparison. The results showed that the accuracy rate was 74% with QDA, which was significantly higher than that obtained using the decision tree (67%) and logistic regression (68%) models. The correlation coefficients between the soil pH and the other three factors (CEC, SOM, and Cdsoil) were higher in the inaccurate set than the accurate set, whereas the correlation coefficients were smaller in the inaccurate set than the accurate set.

Metal concentrations (As, Cd, Pb, Cr, Ba, Co, Ni, Cu, and Zn) in conventional and organic produce were assessed, specifically, five most-consumed vegetables from the US including potato, lettuce, tomato, carrot and onion. They were from four representative supermarkets in a college town in Florida. All vegetables contained detectable metals, while As, Cd, Pb, Cr, and Ba are toxic metals, Co, Ni, Cu, and Zn are nutrients for humans. The mean concentrations of As, Cd, Pb, Cr and Ba in five vegetables were 7.86, 9.17, 12.1, 44.8 and 410 μg/kg for organic produce, slightly lower than conventional produce at 7.29, 15.3, 17.9, 46.3 and 423 μg/kg. The mean concentrations of Co, Ni, Cu, and Zn in five vegetables were 3.86, 58.5, 632, and 2528 μg/kg for organic produce, comparable to conventional produce at 5.94, 68.2, 577, and 2354 μg/kg. For toxic metals, the order followed tomato < lettuce < onion < carrot < potato, with root vegetables being the highest. All metals in vegetables were lower than the allowable concentrations by FAO/WHO. Health risks associated with vegetable consumption based on daily intake and non-carcinogenic risk based on hazard quotient were lower than allowable limits. For the five most-consumed vegetables in the US, metal contents in conventional produce were slightly greater than organic produce, especially for Cd and Pb.

A national scale survey was conducted to determine an array of inorganic and organic contaminants in agricultural soils from two cultivation modes (greenhouse vs. open field) in 20 provinces across China. The investigated contaminants include organochlorine pesticides (OCPs), phthalate esters (PAEs), polycyclic aromatic hydrocarbons (PAHs), lead (Pb), zinc (Zn), copper (Cu) and cadmium (Cd). The large amounts of agrochemicals used and special cultivation mode in greenhouse caused substantial soil pollution and deterioration of soil quality. Mean concentrations of both OCPs and PAEs in greenhouse soil were approximately 100% higher than those in open field. The pH values were 6.85 ± 1.04 and 7.34 ± 0.84 for greenhouse and open field, respectively (p > 0.05). The soil microbial community was predicted to be affected by pollution in greenhouse through the PICRUSt analysis of 16s rRNA sequences. The 12 variables including various chemicals and soil properties together explained 15% of the observed variation in the community composition. In the studied variables, PAEs and lead were the primary factors affecting microbial diversity in greenhouse soils, while pH had the greatest impact on the microbial community in open field soils. These findings enhanced our understanding of the environmental impact and contamination management of greenhouses worldwide.

Mineral phosphorus fertilizers are regularly applied to agricultural sites, but their uranium (U) content is potentially hazardous to humans and the environment. Fertilizer-derived U can accumulate in the soil, but might also leach to ground-, spring and surface waters. We sampled 19 mineral fertilizers from the canton of Bern and soils of three arable and one forest reference sites at each of four locations with elevated U concentrations (7–28 μg L⁻¹) in nearby drinking water wells. The total U concentrations of the fertilizers were measured. The soils were analysed at three depth intervals down to 1 m for general soil parameters, total Cd, P, U and NaHCO₃-extractable U concentrations, and ²³⁴/²³⁸U activity ratios (AR). The U concentrations and AR values of the drinking water samples were also measured. A theoretical assessment showed that fertilizer-derived U may cause high U concentrations in leaching waters (up to approx. 25 μg L⁻¹), but normally contributes only a small amount (approx. 0–3 μg L⁻¹). The arable soils investigated showed no significant U accumulation compared to the forest sites. The close positive correlation of AR with NaHCO₃-extractable U (R = 0.7, p < 0.001) indicates that application of fertilizer can increase the extractable U pool. The lack of depth gradients in the soil U concentrations (1.5–2.7 mg kg⁻¹) and AR (0.90–1.06) ratios are inconsistent with the accumulation of U in the surface soil, and might indicate some leaching of fertilizer-derived U. The AR values in the water samples were close to 1, possibly suggesting an influence of fertilizer-derived U. However, based on findings from the literature and considering the heterogeneity of the catchment area, the agricultural practices, and the comparatively long distance to the groundwater, we conclude that fertilizer-derived U makes only a minor contribution to the elevated U concentrations in the water samples.

Long-term trends of sediment compositions are important for assessing the impact of human activities on the sediment and protecting the sediment environment. In this study, based on the contents of heavy metals and the Pb isotope ratios in lake sediments, atmospheric dustfall and soil in Yixing, China, the representative heavy metals (Zn, Pb, Cr and Cd) in lake sediments from western Taihu Lake were studied. The evolution history of heavy metals in the local environment was constructed for the past 100 years. From 1892 to the 1990s, the anthropogenic fluxes of the representative heavy metals were negligible, indicating minimal anthropogenic emissions of heavy metals. Since the 1990s, anthropogenic fluxes of the representative heavy metals began to increase, concurrent with the economic growth and development in the western Taihu Lake Basin after the Chinese economic reform. The maximum flux percentage of the heavy metals in the sediments, caused by human activities, is 23.0% for Zn, 31.6% for Pb, 39.5% for Cr and 85.3% for Cd, indicating that most of the Cd comes from human activities. The Cd content in the western Taihu Lake Basin was significantly higher than that in the other areas, and the rapid development of the industry in the western Taihu Lake Basin and ceramics in Yixing led to the enrichment of heavy metals in local sediments. Since the 21st century, measures have been taken to control the pollution of heavy metals, including the increase in local government attention and the deployment of environmental monitoring technology. However, heavy metal content remains high, and the Pb content is still increasing. The ratios of Pb isotopes show that the main sources of heavy metals in the western Taihu Lake sediments, the local soil of Yixing and the atmospheric dustfall are coal combustion, leaded gasoline combustion, industrial wastewater and domestic sewage.

Sediment samples from three coastal sites - two beach resorts (Beach 1 and Beach 2 sites) and an area lying between an oil refinery and a river estuary (Estuarine site) - were analyzed for antibiotic- and heavy metal (HM)-resistant enterococci.A total of 123 enterococci, 36 E. faecium, 34 E. casseliflavus, 33 E. hirae, 5 E. faecalis, 3 E. durans, 3 E. gallinarum, and 9 Enterococcus spp, were recovered. Strains resistant to erythromycin, tetracycline and quinupristin/dalfopristin (Q/D) were recovered from all sites, whereas multidrug-resistant isolates were recovered only from “Beach 2” (14%) and “Estuarine” (3.7%). As regards HM resistance, the strains showed a high frequency (68%) of cadmium and/or copper resistance and uniform susceptibility to mercury. The prevalence of cadmium-resistant strains was significantly higher among erythromycin-resistant than among erythromycin-susceptible strains. A significant association between cadmium or copper resistance and Q/D resistance was also observed at “Estuarine” site. The levels of the two HMs in sediment from all sites were fairly low, ranging from 0.070 to 0.126 μg/g, for cadmium and from 1.00 to 7.64 μg/g for copper. Mercury was always undetectable. These findings are consistent with reports that low HM concentrations may contribute to co-selection of antibiotic-resistant bacterial strains, including enterococci.

Natural levels of heavy metals (HM) have increased during the industrial era to the point of posing a serious threat to the environment. The use of tree species to record contamination is a well-known practice. The objective of the study was to compare HM levels under different pollution conditions: a) soil pollution due to mining waste; b) atmospheric pollution due to coal-fired power plant emissions. We report significant HM enrichment in Pinus halepensis tissues. Near a burning power plant, Pb content in a tree wood was 2.5-fold higher that in natural areas (no pollution; NP). In mining areas, Cd content was 25-fold higher than NP. The hypothesis that HM contents in tree rings should register pollution is debatable. HM uptake by pines from soil, detoxification mechanisms and resuspended local soil dust is involved in HM contents in wood and bark.
