Multiple-stressor effects of ocean acidification, warming and predation risk cues on the early ontogeny of a rocky-shore keystone gastropod

To understand how climate change stressors might affect marine organisms and support adequate projections it is important to know how multiple stressors may be modulated by the presence of other species. We evaluated the direct effects of ocean warming (OW) and ocean acidification (OA) together with non-consumptive effects (NCEs) of the predatory crab Acanthocyclus hassleri on early ontogeny fitness-related traits of the commercially important rocky-shore keystone gastropod Concholepas concholepas. We measured the response of nine traits to these stressors at either the organismal level (survival, growth, feeding rates, tenacity, metabolic rate, calcification rate) or sub-organismal level (nutritional status, ATP-supplying capacity, stress condition). C. concholepas survival was not affected by any of the stressors. Feeding rates were not affected by OW or OA; however, they were reduced in the presence of crab NCEs compared with control conditions. Horizontal tenacity was affected by the OA × NCEs interaction; in the presence of NCEs, OA reduced tenacity. The routine metabolic rate, measured by oxygen consumption, increased significantly with OW. Nutritional status assessment determined that carbohydrate content was not affected by any of the stressors. However, protein content was affected by the OA × NCEs interaction; in the absence of NCEs, OA reduced protein levels. ATP-supplying capacity, measured by citrate synthase (CS) activity, and cellular stress condition (HSP70 expression) were reduced by OA, with reduction in CS activity found particularly at the high temperature. Our results indicate C. concholepas traits are affected by OA and OW and the effects are modulated by predator risk (NCEs). We conclude that some C. concholepas traits are resilient to climate stressors (survival, growth, horizontal tenacity and nutritional status) but others are affected by OW (metabolic rate), OA (ATP-supplying capacity, stress condition), and NCEs (feeding rate). The results suggest that these negative effects can adversely affect the associated community.

The adverse effects of plastic waste and nanoplastics on the water environment have become a focus of global attention in recent years. In the present study, using adult Chinese mitten crabs (Eriocheir sinensis) as an animal model, the bioaccumulation and the in vivo and in vitro toxicity of polystyrene nanoplastics (PS NPs), alone or in combination with the bacteria, were investigated. This study aimed to investigate the effects of PS NPs on apoptosis and glucose metabolism in Chinese mitten crabs, and whether PS NPs could synergistically affect the antibacterial immunity of crabs. We observed that NPs were endocytosed by hemocytes, which are immune cells in crustaceans and are involved in innate immunity. The RNA sequencing data showed that after hemocytes endocytosed NPs, apoptosis and glucose metabolism-related gene expression was significantly induced, resulting in abnormal cell apoptosis and a glucose metabolism disorder. In addition, exposure to NPs resulted in changes in the antimicrobial immunity of crabs, including changes in antimicrobial peptide expression, survival, and bacterial clearance. In summary, NPs could be endocytosed by crab hemocytes, which adversely affected the cell apoptosis, glucose metabolism, and antibacterial immunity of Eriocheir sinensis. This study revealed the effects of NPs on crab immunity and lays the foundation for further exploration of the synergistic effect of NPs and bacteria.

Aquatic animals live in an acoustic world in which they often rely on sound detection and recognition for various aspects of life that may affect survival and reproduction. Human exploitation of marine resources leads to increasing amounts of anthropogenic sound underwater, which may affect marine life negatively. Marine mammals and fishes are known to use sounds and to be affected by anthropogenic noise, but relatively little is known about invertebrates such as decapod crustaceans. We conducted experimental trials in the natural conditions of a quiet cove. We attracted shore crabs (Carcinus maenas) and common shrimps (Crangon crangon) with an experimentally fixed food item and compared trials in which we started playback of a broadband artificial sound to trials without exposure. During trials with sound exposure, the cumulative count of crabs that aggregated at the food item was lower, while variation in cumulative shrimp count could be explained by a negative correlation with crabs. These results suggest that crabs may be negatively affected by artificially elevated noise levels, but that shrimps may indirectly benefit by competitive release. Eating activity for the animals present was not affected by the sound treatment in either species. Our results show that moderate changes in acoustic conditions due to human activities can affect foraging interactions at the base of the marine food chain.

Blue crabs from a contaminated estuary (Hackensack Meadowlands-HM) and a cleaner reference site (Tuckerton-TK) were analyzed for Cr, Cu, Hg, Pb, and Zn in muscle and hepatopancreas. Crabs from each site were taken into the laboratory and fed food from the other site, or in another experiment, transplanted to the other site for eight weeks. All crabs were analyzed for metals. Overall, tissue concentrations reflected environmental conditions. Tissue differences were found for Cu, Pb and Zn (all higher in hepatopancreas), and Hg (higher in muscle). HM muscle had more Hg than TK muscle, but did not decrease after transplanting or consuming clean food. HM crabs lost Cu, Pb and Zn in hepatopancreas after being fed clean food or transplanted. TK crabs increased Hg in muscle and Cr and Zn in hepatopancreas after transplantation or being fed contaminated (HM) food. Concentrations were variable, suggesting that blue crabs may not be fully reliable bioindicators of polluted systems. The accumulation of metals within the muscle and hepatopancreas of blue crabs was highly variable, but often followed environmental concentrations.

A series of European Marine Sites has been designated as Special Areas of Conservation (SAC) in England. The aim of this study was to develop a practical methodology to assess the condition of SACs by applying a suite of biomarkers. Biomarkers were applied to the blue mussel Mytilus edulis and the shore crab Carcinus maenas from the Fal and Helford SAC (Cornwall). Individual biomarkers provided useful diagnostic information on the activity of certain classes of contaminants and an integrated Biomarker Response Index (BRI) was used to achieve a more holistic understanding of the condition of the SAC. The BRI indicated that the general health of both organisms was impacted in the upper part of the SAC (Fal Estuary) which correlated well with known chemical hotspots and sources of contamination. The BRI allows a pragmatic way to prioritise SAC sites that may require further investigative studies. A suite of biomarkers was successfully used to create a Biomarker Response Index to assess the health of aquatic organisms from European Marine Sites.

Aquaculture ecosystem is a hot-spot for antibiotic resistance genes (ARGs). Rice-crayfish co-culture was considered an eco-friendly aquaculture model and has been widely adopted in China. However, it is unclear whether rice-crayfish co-culture is one of the most eco-friendly models from the perspective of antibiotic resistance profiles. In this study, we evaluated the eco-friendliness of rice-crayfish co-culture, and compared this model with other aquaculture models, from the perspectives of antibiotics and ARG patterns, based on multi-omics and antibiotic profiles. Results showed that the nutrient levels, antibiotic concentrations, dominant microbial genera and ARG patterns in the rice-crayfish co-culture model were profoundly different from the other three aquaculture models (crab only aquaculture model, crayfish only aquaculture model, and crab-crayfish co-culture models). Specifically, the rice-crayfish co-culture model has significantly lower diversity of ARGs and lower potential risks of ARGs when compared to the other aquaculture models. Nutrient and antibiotic concentrations were the important environmental factors for shaping ARG patterns, but compared with environmental factors, the effects of mobile genes and bacteria community on the proliferation and transmission of ARGs were stronger. This study has deepened our understanding of ARGs in freshwater aquaculture ecosystem, and suggested that rice-crayfish co-culture model is a relatively eco-friendly aquaculture model when compared with the other aquaculture models.

As the two most commonly used organophosphorus herbicides, glyphosate (Gly) and glufosinate-ammonium (Glu) have unique properties for weed control and algae removal in aquaculture. However, the occurrences and health risks of Gly and Glu in aquaculture ponds are rare known. This study aimed to investigate the occurrences of Gly, AMPA (primary metabolity of Gly) and Glu in surface water, sediment and aquatic products from the grass carp (ctenopharyngodon idella), crayfish (procambarus clarkii) and crab (eriocheir sinensis) ponds around Lake Honghu, the largest freshwater lake in Hubei province, China where aquaculture has become the local pillar industry. Three age groups (children, young adults, middle-aged and elderly) exposure to these compounds through edible aquatic products (muscle) consumption were also assessed by target hazard quotient (THQ) method. The results indicated that Gly, AMPA and Glu were widely occurred in surface water, sediment and organisms in the fish, crayfish and crab ponds. AMPA was more likely to accumulate in the intestine of aquatic products than Gly and Glu. According to the total THQ value (1.04>1), muscle consumption of grass carp may pose potential risk to children.

Mean oceanic CO₂ values have already risen and are expected to rise further on a global scale. Elevated pCO₂ (eCO₂) changes the bacterial community in seawater. However, the ecological association of seawater microbiota and related geochemical functions are largely unknown. We provide the first evidence that eCO₂ alters the interaction patterns and functional potentials of microbiota in rearing seawater of the swimming crab, Portunus trituberculatus. Network analysis showed that eCO₂ induced a simpler and more modular bacterial network in rearing seawater, with increased negative associations and distinct keystone taxa. Using the quantitative microbial element cycling method, nitrogen (N) and phosphorus (P) cycling genes exhibited the highest increase after one week of eCO₂ stress and were significantly associated with keystone taxa. However, the functional potential of seawater bacteria was decoupled from their taxonomic composition and strongly coupled with eCO₂ levels. The changed functional potential of seawater bacteria contributed to seawater N and P chemistry, which was highlighted by markedly decreased NH₃, NH₄⁺-N, and PO₄³⁻-P levels and increased NO₂⁻-N and NO₃⁻-N levels. This study suggests that eCO₂ alters the interaction patterns and functional potentials of seawater microbiota, which lead to the changes of seawater chemical parameters. Our findings provide new insights into the mechanisms underlying the effects of eCO₂ on marine animals from the microbial ecological perspective.

This study presents evidence for microplastic contamination in two resident species of brachyuran crab from the Thames Estuary: the native shore crab, Carcinus maenas (Linneaus, 1758) and the invasive Chinese mitten crab, Eriocheir sinensis (H. Milne Edwards, 1853). The gills, gastric mill and intestine of 94 C. maenas and 41 E. sinensis were examined. Crabs were sampled periodically (ca. every three months) between December 2018 and October 2019. A total of 874 plastics were recovered, ranging 34 μm–34 mm in length. Overall, 71.3% and 100% of C. maenas and E. sinensis, respectively, contained at least one item (fibre, film, fragment or tangle of fibres) in the gill chamber, gastric mill or gastrointestinal tract. The most common items were fibres (78.5%) but in some cases, particularly in the gastric mill, these were aggregated into tangles (7.8%). Almost all E. sinensis contained tangles of fibres (95.1%), whereas, relatively few C. maenas contained similar tangles (10.6%).

As emerging pollutants, the occurrence and risks of organophosphate esters (OPEs) in mariculture farms should be concerned; however, information is limited. Beibu Gulf is one of the essential mariculture zones in China. This study aimed to investigate the occurrence of OPEs in mariculture farms of the Beibu Gulf, their phase distribution and bioaccumulation among sediment, organisms (shrimp, crab, and oyster), water, and feed. Human exposure to OPEs through seafood consumption was also assessed. The total concentrations of the 11 target OPEs (∑₁₁OPEs) in the water samples ranged 32.9–227 ng L⁻¹. It was significantly higher in water from the culture ponds (mean 122 ng L⁻¹) than in water from the estuaries and nearshore areas (mean 51.1 ng L⁻¹) (nonparametric test, p < 0.05). ∑₁₁OPEs in the feeds averaged 46.0 (range 21.7–84.5) ng g⁻¹ dw, which is similar to the level in the organism samples (mean 55.5, range 21.3–138 ng g⁻¹ dw) and 4.4 times higher than that in the sediment (mean 10.9, range 35–22.1 ng g⁻¹ dw). The ∑₁₁OPEs released from the feeds to the culture ponds was estimated to be 49 μg m⁻² per three-month period. In the aquaculture ponds, the sediment-water distribution coefficient (log KOC), and the bioaccumulation factors from the water (log BWAFs) or the feed (log BFAFs) to the organisms, depend linearly on the hydrophobicity (log KOW) of OPEs. The log BWAFs and log BFAFs increased with increasing log KOW within the log KOW range of 1–7. The human exposure to OPEs through consumption of shrimp, crab, and oysters from the mariculture farms does not pose a health risk at present.
