Antibiotics control in aquaculture requires more than antibiotic-free feeds: A tilapia farming case

Public concern over the health implications of antimicrobials employed in aquaculture has resulted in adoption of strict regulations for their use. This study employed a high-throughput protocol covering 86 compounds in six pharmaceutical groups to screen feed and sediments from 20 tilapia ponds randomly in 18 farms of an aquacultural unit in southern China, one of important tilapia fillet suppliers in the world. Seventeen samples of commercial feeds from manufacturer-sealed bags in the farms were tetracyclines-free but not antibiotic-free. All the sealed-bag feeds contained quinolones and two feeds had sulfonamides (up to 140 μg kg⁻¹). Meanwhile, seven leftover-feeds in opened bags contained added antimicrobials: tetracyclines (570–2790 μg kg⁻¹) in all and florfenicol (20–294 μg kg⁻¹) in four. All the feeds regardless sealed or not had large amounts (221–2642 μg kg⁻¹) of salicylic acid (possible antimicrobial substitute) and caffeine, and one sealed-bag feed even was quantified with medroxyprogesterone. Surface sediments (0–10 cm) from the ponds were detected with 36 compounds and sublayer sediments (10–20) with 8 compounds. Large amounts of salicylic acid were present in both surface and sublayer sediments accounting up to 10% of total pharmaceutical residues. Surface sediments were dominated by antibiotics (5.2–172 μg kg⁻¹), mainly sulfonamides and quinolones, contributing 68% of the total quantitative compound mass. Sublayer sediments were also enriched in quinolones (up to 260 μg kg⁻¹). Surprisingly, all sediments contained progesterone (up to 8.0 μg kg⁻¹) in coincidence to the feed with medroxyprogesterone, perhaps arising from endocrine abuses or cross-contamination. Although high levels of other pharmacologic residues (caffeine) in sediment posed greater than medium ecological risks, antibiotic residues contributed only 2–35% to the risk. These findings suggest that antibiotic-free feed may be insufficient to control antibiotic abuse in aquaculture and that additional regulatory actions may be necessary, such as veterinary prescription as human antibiotic uses.

Many types of contaminants of emerging concern (CECs), including per- and poly-fluoroalkyl substances (PFAS), have been found in leachate of operating municipal landfills. However, there is only limited information on CECs presence in leachate of historic landfills (≥3 decades since closure, often lacking engineered liners or leachate collection systems) at concentrations that may pose a risk to nearby wells and surface water ecosystems. In this study, 48 samples of leachate-impacted groundwater were collected from 20 historic landfills in Ontario, Canada. The CECs measured included artificial sweeteners (ASs), PFAS, organophosphate esters (OPE), pharmaceuticals, bisphenols, sulfamic acid, perchlorate, and substituted phenols. The common presence of the AS saccharin, a known indicator of old landfill leachate, combined with mostly negligible levels of the AS acesulfame, an indicator of modern wastewater, revealed that most samples were strongly influenced by leachate and not cross-contaminated by wastewater (which can contain these same CECs). Several landfills, including ones closed in the 1960s, had total PFAS concentrations similar to those previously measured at modern landfills, with a maximum observed here of 12.7 μg/L. Notably elevated concentrations of several OPE, sulfamic acid, cotinine, and bisphenols A and S were found at many 30-60 year-old landfills. There was little indication of declining concentrations with landfill age, suggesting historic landfills can be long-term sources of CECs to groundwater and that certain CECs may be useful tracers for historic landfill leachate. These findings provide guidance on which CECs may require monitoring at historic landfill sites and wastewater treatment plants receiving their effluent.

Infections caused by carbapenem-resistant Enterobacteriaceae are a growing concern worldwide. Raoultella ornithinolytica is a species in the Enterobacteriaceae family which can cause hospital-acquired infections and is sporadically reported as carbapenem-resistant from human and environmental sources. In this study, we firstly report on an NDM-1-producing R. ornithinolytica, Rao166, isolated from drinking water in an animal cultivation area in China. In addition to carbapenem-resistance, Rao166 was resistant to several other antibiotics including gentamicin, sulfamethoxazole-trimethoprim, tetracycline and fosfomycin. Rao166 carried a novel IncFIC-type megaplasmid, 382,325 bp in length (pRAO166a). A multidrug resistance region, 60,600 bp in length, was identified in the plasmid containing an aac(3)-IId-like gene, aac(6′)-Ib-cr, blaDHA₋₁, blaTEM₋₁B, blaCTX₋M₋₃, blaOXA₋₁, blaNDM₋₁, qnrB4, catB3, arr-3, sul1, and tet(D). Results from virulence assays implied that Rao166 has considerable pathogenic potential. Although pRAO166a was found to be non-transmissible, dissemination of the NDM-1 producing strain may occur from well water to humans or animals through cross-contamination during food preparation or directly via drinking water, and potentially lead to difficult-to-treat infections. Thus, contamination of well water by this carbapenem-resistant and presumptively virulent strain of R. ornithinolytica should be considered a potential public health risk.

In microplastics (MPs) research, there is an urgent need to critically reconsider methodological approaches and results published, since public opinion and political decisions might be based on studies using debatable methods and reporting questionable results. For instance, recent studies claim that MPs induce intestinal damage and that relatively large MPs are transferred to, e.g., livers in fish. However, there is methodological criticism and considerable concern whether MP transfer to surrounding tissues is plausible. Likewise, there is an ongoing discussion in MP research if MPs act as vectors for adsorbed hazardous chemicals. In this study, effects of very small (4–6 μm) and very large (125–500 μm) benzo(a) pyrene (BaP)-spiked polyethylene (PE) particles administered via different uptake routes (food chain vs. direct uptake) were compared in a 21-day zebrafish (Danio rerio) feeding experiment. Particular care was taken to prevent cross-contamination of MPs during dissection and histological sample preparation. In contrast to numerous reports in literature describing similar approaches, independent of exposure route and MP size, no adverse effects could be detected. Likewise, no BaP accumulation could be documented, and MPs were exclusively seen in the lumen of the intestinal tract, which, however, did not induce any histopathological effects. Results indicate that in fish MPs are taken up, pass along the intestinal lumen and are excreted without any symptoms of adverse effects.

Face masks have been adopted as an essential measure to prevent transmission and spread of the virus infection during the pandemic of Covid-19. The present study evaluates the potential microfibers transfer from face masks to other recipients and the potential cross-contamination of samples by microfibers released from masks worn during the analysis of microlitter ingestion by fish. Results indicated that masks could easily transfer endogenous (originated from the mask tissue itself) and exogenous microfibers (with a different origin than the mask tissue itself) to other recipients (adhesive tape and air in our experiment). Exogenous fibers may be carried from everywhere and potentially released everywhere. Microfibers are also released into the air, driven by the airflow generated by breathing, and can be transferred to blanks and samples. Microfiber contamination by facial masks increases the risk of samples cross-contamination and raises concerns about the results reliability of the microlitter analysis on marine biota.

Oxidant treatment of ballast water (BW) is commonly used in BW systems in order to minimize the transport of alien species. The release of disinfection by-products (DBPs) associated to the treatment of BW and cross-contamination of butyltin (BT) compounds through BW discharge is a topic of environmental concern. A chemical port baseline survey has been conducted in seven ports of the Adriatic Sea. Analysis have been performed on transplanted mussels, surface sediment, seawater, BW. Results showed an evidence of BT contamination, particularly in sediments, probably related to their illegal usage or to intensive shipping activities. Therefore, BW may act as a vector and contribute to re-buildup of BT contamination in the coastal regions.A baseline set of data concerning DBPs is provided, showing the preferential distribution of these compounds in the marine environment that will be useful for future considerations on monitoring and assessment of chemical contamination associated with BW.

Ground-cover vegetation attracts and harbors beneficial insects to the agrosystem, playing an important role in conservation biological control. Integrated pest management (IPM) program guidelines recommend the implantation of sowed or resident wild covers in perennial crops. Given the high-quality fruit requirements, even in IPM programs, insecticides can be required in citrus crops. This study presents, over a year, the levels of neonicotinoids (thiamethoxam and imidacloprid) in not-target ground-cover wildflowers growing spontaneously in citrus orchards after foliar treatment of citrus trees. The presence and persistence of these neonicotinoids in different wildflower species were studied. Concentrations of thiamethoxam and imidacloprid in whole wildflowers ranged from < method quantification limit (MQL) to 52.9 ng g⁻¹ and from < MQL to 98.6 ng g⁻¹, respectively. Thiamethoxam was more frequently detected than imidacloprid. Thiamethoxam and imidacloprid were detected up to 336 and 230 days after treatment, respectively. The highest detection frequencies (100%) and highest thiamethoxam and imidacloprid mean concentrations (26.0 ± 7.3 ng g⁻¹ and 11.0 ± 10.6 ng g⁻¹, respectively) occurred in wildflowers collected 9 days after the treatments. Since application, a clear decrease in the concentration of both compounds and differences in the accumulation depending on wildflower species were observed. Cross contamination was detected, indicating a transport from adjacent treated plots. Maintaining a cover crop in citrus orchards may lead to detrimental effects on non-target arthropods if these neonicotinoid compounds are used for pest control since they can entail a chronic exposure during at least 230 days for imidacloprid and 336 days for thiamethoxam.

A comparative study of indicator bacteria concentrations obtained by laboratory analysis of grab samples of storm water, which were collected manually or by automatic samplers, was carried out in two urban catchments. Samples were analyzed for four types of indicator bacteria, total coliforms, Escherichia coli (E. coli), enterococci, and Clostridium perfringens and further documented by measurements of total suspended solids (TSS) and turbidity. Analysis of complete data sets (N = 198) indicated no statistically significant differences in the geometric means of all the constituent samples collected automatically or manually, but there were some small differences between the results produced by the two sampling methods applied. Total coliform concentrations were positively biased in samples collected by automatic samplers, but for the three remaining indicator bacteria (E. coli, enterococci, and C. perfringens), the opposite was true. Risk of sample cross-contamination in automatic samplers was assessed in the laboratory by sampling consecutively synthetic storm water with high and low concentrations of E. coli and enterococci. The first low-concentration samples preceded by high-concentration samples were cross-contaminated and the measured concentrations were positively biased. This cross-contamination was explained by storm-water residue in the sampling line. Such a residue remained in place even after line purging by compressed air, and its mass depended on the sampling line length (tested up to 5 m), as verified by measurements in the laboratory. The study findings should be helpful for improving field protocols for indicator bacteria sampling.

The scope of this study was to investigate the uptake of chromium and nickel by onions (Allium cepa) and potatoes (Solanum tuberosum) and their impact on plant enzymes catalase (CAT, E.C. 1.11.1.6) and peroxidase (POX, E.C. 1.11.1.7). A greenhouse experiment was conducted, simulating the irrigating conditions existing in the two biggest tuber-producing regions of Greece (Asopos and Messapia). Plants were cultivated for 4 months in six irrigation lines, each one supplied by an aqueous solution, containing levels of Cr(VI) and Ni(II) ranging from 0 μg/L (control) to 1,000 μg/L. Significant statistical correlations were observed between (i) the levels of heavy metals in plants, (ii) the levels of heavy metals in plants and in irrigation water, and (iii) the levels of heavy metals and the enzymatic activities in plants. The existing EU legislation has no legal limits for Ni and Cr in food, and the nutritional implications of this study are discussed.

For class II, type A2 biological safety cabinets (BSC), NSF/ANSI Standard 49 should be conformed in cabinet airflow velocity derivation, particle contamination, and aerodynamic flow properties. However, there exists a potential problem. It has been built that the cabinet air flow stabilize is influenced by the quantity of downflow of air and the height above the cabinet exhaust opening. Three air downflow quantities were compared as an operating apparatus was placed from 20 to 40 cm above the bench of the cabinet. The results show that the BSC air downflow velocity is a function of increased sampling height, displaying that containment is improvingly permitted over product protection as the sampling height decreases. This study investigated the concentration gradient of particles at various heights and downflow air quantity from the bench of the BSC. Experiment results indicate that performance near the bench was better than in the rest of the BSC. In terms of height, the best cleanliness was measured at a height of 10 cm over the bench; it reduced actually with add in height. The empirical curves accommodate, founded on the concentration gradient of particle created was elaborated for evaluating the particle concentration at different heights and downflow air quantity from the source of the bench of the BSC. The particle image velocimetry system applied for BSC airflow research to fix amount of airflow patterns and air distribution measurement and results of measurements show how obstructions can greatly influence the airflow and contaminant transportation in a BSC.
