Molecular effects and bioaccumulation of levonorgestrel in the non-target organism Dreissena polymorpha

Bioaccumulation and effects of the contraceptive hormone levonorgestrel were examined in the non-target organism Dreissena polymorpha. Molecular biomarkers of biotransformation, elimination, antioxidant defence and protein damage were analyzed after exposure to increasing concentrations of levonorgestrel in a flow-through system. The lowest concentration (0.312 μg L⁻¹) was 100-fold bioconcentrated within four days. A decrease of the bioconcentration factor was observed within one week for the highest test concentrations (3.12 and 6.24 μg L⁻¹) suggesting enhanced excretory processes. The immediate mRNA up-regulation of pi class glutathione S-transferase proved that phase II biotransformation processes were induced. Disturbance of fundamental cell functions was assumed since the aryl hydrocarbon receptor has been permanently down-regulated. mRNA up-regulation of P-glycoprotein, superoxide dismutase and metallothioneine suggested enhanced elimination processes and ongoing oxidative stress. mRNA up-regulation of heat shock protein 70 in mussels exposed to the two highest concentrations clearly indicated impacts on protein damage.

Comparative studies of biomonitors of trace metal contamination are relatively scarce. We took advantage of a point source pollution in a reservoir (Flix, Spain) to compare trace metal (Hg, Pb, Cd, Se, As, Zn, Cu, Cr) bioaccumulation patterns among 16 food web components. Our results indicate that most organisms are suitable for Hg biomonitoring, whereas other metals are better monitored by only some of them. Biofilms and zebra mussel were the organisms with larger and more diverse biomonitoring capacity. However, we show that using groups of biomonitors increase the scope and strengths of the conclusions and specific goals can be better addressed. We conclude providing an overview of the strengths and weaknesses of the main organisms considered for biomonitoring trace metals in rivers and reservoirs.

Development of stress markers for the invader freshwater zebra mussel (Dreissena polymorpha) is of great interest for both conservation and biomonitoring purposes. Gene expression profiles of several putative or already established gene expression stress markers (Metallothionein, Superoxide dismutase, Catalase, Glutathione S transferase, Glutathione peroxidase, Cytochrome c oxidase, the multixenobiotic resistance P-gp1, and heat shock proteins HSP70 and HSP90) were analyzed by quantitative Real-Time PCR in adults and pediveliger larvae after exposure to metals (Hg, Cu, Cd). A defined pattern of coordinated responses to metal exposure and, presumably, to oxidative stress was observed in gills and digestive gland from adults. A similar, albeit partial response was observed in larvae, indicating an early development of stress-related gene responses in zebra mussel. The tools developed in this study may be useful both for future control strategies and for the use of zebra mussel as sentinel species in water courses with stable populations.

The biocide triclosan (TCS, 5-chloro-2-(2,4-dichlorophenoxy)phenol) is commonly used in several personal care products, textiles, and children’s toys. Because the removal of TCS by wastewater treatment plants is incomplete, its environmental fate is to be discharged into freshwater ecosystems, where its ecological impact is largely unknown. The aim of this study was to determine the effect of TCS on the antioxidant enzymatic chain of the freshwater mollusk zebra mussel (Dreissena polymorpha). We measured the activity of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx), as well as the phase II detoxifying enzyme glutathione S-transferase (GST) in zebra mussel specimens exposed to 1Â nM, 2Â nM, and 3Â nM TCS in vivo. The mussels were exposed for 96Â h, and the enzyme activities were measured every 24Â h. We measured clear activation of GST alone at all three dose levels, which shows a poor induction of the antioxidant enzymatic chain by TCS. CAT and SOD were activated only at 3Â nM, while GPx values overlapped the baseline levels.

Synthetic musks are used in many consumer products for their pleasant odor and their binding affinity for fabrics. In the early 1990s, polycyclic musks were reported to occur in air, water, sediment, wildlife, and humans from many European countries. Concentrations of polycyclic musks, particularly 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethyl-cyclopenta-[γ]-2-benzopyran (HHCB) and 7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4-tetrahydronapthalene (AHTN), have been reported to increase over time in the environment. In this study, concentrations of musks in water, sediment, fish, and mussel were determined from three locations along the upper Hudson River. HHCB and AHTN were detected in water (n = 5; 3.95-25.8 and 5.09-22.8 ng/L, respectively), sediment (n = 3; 72.8-388 and 113-544 ng/g, dry weight), fish (n = 30; <1-125 and <1-32.8 ng/g, lipid weight), and zebra mussel (n = 4; 10.3-19.3 and 42.2-65.9 ng/g, lipid weight) samples. Bioaccumulation factors of HHCB calculated for white perch, catfish, smallmouth bass, and largemouth bass were in the range of 18 to 371, when the concentrations in fish were expressed on a wet weight basis; the factors were in the range of 261 to 12,900, when the concentrations in fish were expressed on a lipid weight basis.
