Monitoring of selected estrogenic compounds and estrogenic activity in surface water and sediment of the Yellow River in China using combined chemical and biological tools

We investigated occurrence of selected compounds (4-t-octylphenol: 4-t-OP; 4-nonylphenols: 4-NP; bisphenol-A: BPA; estrone: E1; 17β-estradiol: E2; triclosan: TCS) and estrogenicity in surface water and sediment of the Yellow River in China by using combined chemical analysis and in vitro yeast screen bioassay. Estrogenic compounds 4-t-OP, 4-NP, BPA, E1, E2 and TCS were measured in the water samples, with their average concentrations of 4.7, 577.9, 46.7, 1.3, ND and 6.8 ng/L, respectively. In sediment, the average concentrations of 4-t-OP, 4-NP, BPA and TCS were 35.7, 0.5, 1.7 and 0.7 ng/g while E1 and E2 were not detected in the sediments of all selected sites. In general, the estrogenic compounds in surface water and sediment of the Yellow River were at relatively low levels, thus having medium to minimal estrogenic risks in most sites except for the site of east Lanzhou with high estrogenic risks.

Degradation of heavy oil by bacteria to decompose organic compounds such as aliphatic and aromatic hydrocarbons has been used in bioremediation. However, the biological and environmental effects of the degradation products including intermediates are still not clear. Here, we monitored the degradation of C-heavy oil by analyzing the products formed in cultures with oil-degrading bacteria (complex microbes or a single bacterial strain). Furthermore, proliferation assays using breast cancer MCF-7 cells and gene-expression profiling of MCF-7 cells using oligonucleotide-DNA microarrays were performed to evaluate the estrogenic activity of the degradation products. While the products did not show any significant cell-proliferative activity, the oil samples cultured for longer periods (2–3 months), whether cultured with mixed microbes or a single bacterial strain, showed gene-expression profiles similar to that of 17β-estradiol (E₂). These results suggest that oil-degradation products have estrogenic activity, and estrogen-like components could possibly be produced during the degradation process.

Llobregat River (North-East Spain) is the most important drinking water source for Barcelona and its surrounding area. As one of the only water sources in the area the river water have been overexploited and effluents from more than 30 urban wastewater treatment plants, industries and agriculture runoffs have been discharged into the river. This article reviews the presence of emerging contaminants published during the last decades, emphasizing on the observed effects on ecosystems caused by the contamination. Pesticides, surfactants, estrogens, pharmaceuticals and personal care products and even abuse drugs are the main groups detected in different studies, reporting alterations in species composition, abundance or biomass and endocrine disruption measured by alterations in enzymatic activity or specific protein production. The information available provides an overview of the river status according to the Water Framework Directive.

Great efforts have been made in China to retrofit and upgrade the existing municipal wastewater treatment plants (WWTPs) for enhanced removal of organic substrates and in particular nutrients. However, the removal of trace recalcitrant or hazardous organic chemicals, e.g. steroid estrogens, one group of typical endocrine disrupting chemicals, has long been overlooked. The extensive investigations on estrogen removal rates in global and Chinese WWTPs and the estrogen biodegradation kinetics results in batch laboratory experiments are reviewed in this study. The effects of estrogen initial concentration and nitrifying activated sludge are highlighted. Challenges existing in current estrogen studies are pointed out, which are relevant for researches on fate and behavior of similar down-the-drain chemicals in both Chinese and global WWTPs.

Synthetic and persistent endocrine disrupting chemicals (EDCs) such as 17α-ethinylestradiol (EE2) have been frequently detected in the effluent of wastewater treatment plants and induce hazards to humans and wildlife. In this study, biogenic Mn oxides were tested for the removal of EE2, and factors affecting the reaction were also investigated. The biogenic Mn oxides produced by Pseudomonas putida strain MnB1 were nano-sized and poorly crystallized particles. A concentration of 7.9 mg l−1 biogenic Mn oxides showed 87% EE2 (1 mg l−1) removal efficiency in 2 h, which confirms the excellent potential of biogenic Mn oxides for removal of estrogens. EE2 removal was enhanced at high Mn oxide doses and at low pH. Co-existing heavy metals significantly inhibit EE2 removal, due to their competition for the reactive sites of biogenic Mn oxides. Humic acid (HA) also obstructed EE2 removal, but the adverse effect was alleviated as HA concentration increased, possibly due to the formation of soluble complexes with the released Mn2+, of which adsorption onto Mn oxides reduces surface reactive sites.

The embryos of a marine abalone, Haliotis diversicolor supertexta, were exposed to a typical environmental estrogen, 17α-ethynylestradiol (EE2), for 96 h, to examine the acute toxicity of EE2 to the embryogenesis of the abalone. A marine diatom, Navicula incerta, was used in the test media as settlement substrate and food for the abalone larvae. During the embryo culture, more than 30 % of EE2 could be removed from the test media by the diatom, mainly via biodegradation, leading to a decrease of water-borne exposure dose. Further, the exposure concentrations of EE2 around the living microenvironment of the abalone larvae could be magnified 350–468 times after the larvae settled on the diatom, as indicated by the bioconcentration factors of EE2 in the diatom. Increased bioaccumulation of EE2 in the diatom caused greater inhibition on the metamorphosis of the abalone larvae by enhancing the uptake of EE2 in the larvae via dietary exposure, while declined water-borne exposure dose did not affect the embryonic toxicity of EE2 and its uptake in the abalone larvae. The 96-h median effective concentration of EE2 to the metamorphosis of the abalone larvae was 10.01 μg L⁻¹, when the exposure doses in both the test media and the diatom were controlled stable. The 96-h hazard concentration for 5 % of the species was 1.20 μg L⁻¹, which was still higher than but close to the reported upper contamination level of EE2 and could be employed as the safety threshold for the metamorphosis of the abalone embryos.

In the present work, the effectiveness of conventional wastewater treatments (activated sludge and oxidation ditches) and low-cost wastewater treatments (trickling filter beds, anaerobic lagooning and constructed wetlands) in the removal of pharmaceutically active compounds has been studied. To evaluate the efficiency of removal, 16 pharmaceutically active compounds belonging to seven therapeutic groups (anti-inflammatory drugs, antibiotics, antiepileptic drugs, β-blockers, nervous stimulants, estrogens and lipid regulators) have been monitored during 1-year period in influent and effluent wastewater from 11 wastewater treatment plants of Spain. Mean removal rates of pharmaceutically active compounds achieved in conventional wastewater treatments were slightly higher than those achieved in low-cost treatments, being 64% and 55%, respectively. Ibuprofen, naproxen, salicylic acid and caffeine were the pharmaceutical compounds most efficiently removed, regardless the wastewater treatment applied, with removal rates up to 99%. Anaerobic lagooning was the less effective treatment for the removal of the most persistent compounds: carbamazepine and propranolol.

The behavior of estrone (E1) and 17β-estradiol (E2) in relatively closed water environment was studied by continuous flow experiment using sediments from a freshwater reservoir. For this, four sediment columns (two oxic ones and two anoxic ones) were employed, which were structured by packing 30 cm of undisturbed sediment and 60 cm of overlying water collected from two sites within a reservoir. A mass balance model that considered the influent flux, the effluent flux, mass transfer, sorption, and biodegradation was proposed to describe the behavior of E2 and E1 in the columns. The results indicated that the water–sediment partition coefficient of E1 [Formula: see text] was higher than E2 [Formula: see text]. The degradation rate of E1 (k E1) was smaller than E2 (k E2). Under both oxic and anoxic conditions, E1 was formed from E2. Furthermore, to clarify the impact of the model parameters such as the hydraulic retention time (HRT), K d, and k on the behavior of E2 and E1, variance analysis was performed based on the results of model simulations. The results showed that the concentrations of E2 and E1 in the column effluent were controlled most significantly by the sorption capacity of the natural estrogens onto sediment particles, with the determined contributory ratios changing in the order of sorption > HRT > degradation.

PURPOSE: Resulting from their intensive use as corrosion inhibitors in aircraft deicing and anti-icing fluids (ADAF) and for silver protection in dishwasher detergents benzotriazoles (BTs) are widespread in European surface waters. The current study aimed on an ecotoxicological characterization of 1H-benzotriazole (1H-BT) and 5-methyl-1H-benzotriazole (5MBT). METHODS: Acute and chronic OECD guideline tests were conducted with primary producers (Desmodesmus subspicatus, Lemna minor) and two daphnia species (Daphnia magna, Daphnia galeata) to characterize the hazard of these chemicals. Additionally, the estrogenic activity of both BTs was analyzed in vitro using a recombinant yeast estrogen screen (YES). RESULTS: Both BTs revealed significant effects in acute and chronic experiments, but exhibited no estrogenic activity in the YES. The algal growth test displayed an inhibited cell number increase with effect concentration (EC) values of EC10 1.18 and 2.86 mg l-1 for 1H-BT and 5MBT, respectively. In the Lemna test, EC10 values were 3.94 mg l-1 (1H-BT) and 2.11 mg l-1 (5MBT). D. magna was also affected with EC50 (48 h) values of 107 mg l-1 for 1H-BT and 51.6 mg l-1 for 5MBT. D. galeata was more sensitive with an EC50 (48 h) of 14.7 mg 1H-BT l-1 and 8.13 mg 5MBT l-1. In the 21-day reproduction tests with D. magna, the EC10 for 5MBT was 5.93 mg l-1 while 1H-BT showed no adverse effects. D. galeata turned out to be more sensitive in the chronic study with EC10 values of 0.97 mg l-1 for 1H-BT and 0.40 mg l-1 for 5 MBT. CONCLUSION: Because BTs are regularly found in the aquatic environment at lower μg l-1 concentrations reflecting their persistence and poor elimination during wastewater treatment processes, a preliminary risk assessment was conducted. There is little indication that BTs pose a risk for aquatic ecosystems at current exposure levels during most of the year. However, it cannot be excluded that in winter with a higher usage of ADAFs environmental concentrations may well exceed the level that is considered safe for aquatic organisms.

PURPOSE: The aim of this study is to develop a normalization method for real-time PCR data by analyzing the most stably expressed control genes in mussel (Mytilus edulis) reproductive tissue. METHODS: To facilitate this, six candidate genes, including several commonly used in the literature, were investigated in mussels at different stages of gametogenesis and following experimental exposure to a model estrogen (17b-estradiol). GeNorm and NormFinder softwares were employed to assess the stability of the reference genes. RESULTS: Our results demonstrate that the most stable reference genes are not the same in mussels at different stages of gametogenesis and in experimentally E2-exposed mussels. Interestingly, HEL (helicase) and ACT (actin) mRNA expression levels were most affected by the stage of gametogenesis and yet, in molluscan studies, ACT is possibly the most frequently used reference gene. CONCLUSIONS: We demonstrate that the experimental results are highly dependent on the reference gene chosen and that statistically significant contrasting differences between sample groups are present or absent depending on the reference gene employed.
