Toxicity of different polycyclic aromatic hydrocarbons (PAHs) to the freshwater planarian Girardia tigrina

Freshwater planarians have been gaining relevance as experimental animals for numerous research areas given their interesting features, such as high regeneration potential, shared features with the vertebrates’ nervous system or the range of endpoints that can be easily evaluated in response to contaminants. Ecotoxicological research using these animals has been steadily increasing in the past decades, as planarians’ potentialities for this research area are being recognized. In this work, we used polycyclic aromatic hydrocarbons (PAHs) as model contaminants and evaluated effects of exposure to phenanthrene, pyrene and benzo[a]pyrene (B[a]P) in planarians. The freshwater planarian Girardia tigrina was chosen and mortality, cephalic regeneration (during and post-exposure), behavioral endpoints and presence of PAHs in tissues, were evaluated. Mortality was only observed in planarians exposed to phenanthrene, with an estimated LC₅₀ of 830 μg L⁻¹. Results indicate that planarian behavioral endpoints were very sensitive in response to sub-lethal concentrations of PAHs, showing a greater sensitivity towards B[a]P and pyrene. Briefly, post-exposure locomotion and post-exposure feeding were significantly impaired by sub-lethal concentrations of all compounds, whereas regeneration of photoreceptors was only significantly delayed in planarians exposed to pyrene. Moreover, levels of PAH-type compounds in planarian tissues followed a concentration-dependent increase, showing uptake of compounds from experimental solutions. The present results highlight the importance of studying alternative and complementary endpoints, such as behavior, not only because these may be able to detect effects at lower levels of contamination, but also due to their ecological relevance. The simplicity of evaluating a wide range of responses to contaminants further demonstrates the utility of freshwater planarians for ecotoxicological research.

The ingestion of microplastics (plastic particles <5 mm) has been observed in a range of marine organisms, and adverse effects have been reported in several species after high concentration exposure. However, the long-term effects of low-dose ingestion remains unclear. The aim of this study was thus to assess the chronic effects of low concentrations of polystyrene microparticles to the intertidal amphipod Echinogammarus marinus, using food consumption, growth, and moulting as endpoints. Amphipods were fed a gelatinous algal feed spiked with microbeads (8 μm) in concentrations of ∼0.9, 9 and 99 microplastics/g for 35 days. E. marinus was also analysed for retention of microplastics, and egestion rate was calculated in a separate high-dose feeding experiment. No significant effects were found in the food consumption or growth assays. There was no accumulation of microplastics in the gut, with only one microbead recorded internally in three (8%) of the exposed amphipods. The low number is likely linked to gastrointestinal functions, allowing for easy egestion of indigestible items. This assumption was supported by the observation that after high-dose exposure, 60% of E. marinus egested all microbeads within 24 h. This study suggests that ingesting low concentrations of 8 μm microplastics do not impair the feeding or growth of amphipods along the exposure period. We hope that negative results such as these may further assist in assessing the impact posed by microplastics to marine organisms.

Oil sands process-affected water (OSPW) — a byproduct of the oil sands industry in Northern Alberta, Canada – is currently stored in on-site tailings ponds. The goal of the present study was to investigate the interaction of OSPW with the olfactory system and olfactory-mediated behaviours of fish upon the first encounter with OSPW. The response of rainbow trout (Oncorhynchus mykiss) to different concentrations (0.1, 1, and 10%) of OSPW was studied using a choice maze and electro-olfactography (EOG), respectively. The results of the present study showed that rainbow trout are capable of detecting and avoiding OSPW at a concentration as low as 0.1%. Exposure to 1% OSPW impaired (i.e. reduced sensitivity) the olfactory response of rainbow trout to alarm and food cues within 5 min or less. The results of the present study demonstrated that fish could detect and avoid minute concentrations of OSPW. However, if fish were exposed to OSPW-contaminated water and unable to escape, their olfaction would be impaired.

For the first time this study reports on the presence of microplastics (1 nm to <5 mm) in the gastrointestinal tracts of small semipelagic fish (Boops boops) in the Balearic Islands (Mediterranean Sea) from March to May 2014. The results show microplastic ingestion in 68% of full stomach samples with an average of 3.75 items per fish. Only filament type microplastics were observed in B. boops full gastrointestinal tracts. The frequency of occurrence of microplastics was high, with values ranging from 42% to 80%, in comparison to the other ingested items. Spatial variability among locations is high, which suggests that this type of contamination is ubiquitously distributed and originates from multiple sources. The results are important and indirectly provide further evidence of the presence of microplastics, which can be ingested by biota, in the marine environment.

Total mercury (THg) and δ¹⁵N were analysed in liver and muscle of juvenile green turtles (Chelonia mydas) from a foraging site in southeastern Brazil (~23°S) to evaluate contamination levels and make inferences about the timescale that animals were recruited from oceanic to coastal waters. The values for THg and δ¹⁵N in liver and muscle were 432.2 and 72.8 μg kg⁻¹ and 10.0‰ and 9.2‰, respectively. THg concentrations in tissues indirectly indicated that the main food resources for this consumer (i.e., macroalgae) had negligible levels of this element. Thus, this foraging site could be considered a pristine area in terms of Hg contamination. The THg and δ¹⁵N in liver were derived by more recent food ingestion than those in muscle. The juvenile green turtles that died because of incidental captures in fisheries practiced along this foraging site were individuals newly recruited from oceanic waters.

The ingestion of microplastics and natural fibres (<5 mm) was assessed for two commercial fish species in the western Mediterranean Sea: Sardina pilchardus and Engraulis encrasicolus. Gastrointestinal tracts from 210 individuals from 14 stations were examined with 14.28–15.24% of the small pelagic fish S. pilchardus and E. encrasicolus having ingested microplastics and natural fibres. A latitudinal increase in condition index (Fulton's K) of S. pilchardus gave an indication that larger individuals with better physical condition are less likely to ingest microplastics and natural fibres. Fibres were the most frequent particle type (83%) and Fourier Transform Infrared spectroscopy (FT-IR) analysis indicated polyethylene terephthalate was the most common microplastics material (30%). Results from this study show that both microplastics and natural fibres of anthropogenic origin are common throughout the pelagic environment along the Spanish Mediterranean coast.

Microplastics are present in marine habitats worldwide and laboratory studies show this material can be ingested, yet data on abundance in natural populations is limited. This study documents microplastics in 10 species of fish from the English Channel. 504 Fish were examined and plastics found in the gastrointestinal tracts of 36.5%. All five pelagic species and all five demersal species had ingested plastic. Of the 184 fish that had ingested plastic the average number of pieces per fish was 1.90±0.10. A total of 351 pieces of plastic were identified using FT-IR Spectroscopy; polyamide (35.6%) and the semi-synthetic cellulosic material, rayon (57.8%) were most common. There was no significant difference between the abundance of plastic ingested by pelagic and demersal fish. Hence, microplastic ingestion appears to be common, in relatively small quantities, across a range of fish species irrespective of feeding habitat. Further work is needed to establish the potential consequences.

The application of chemical dispersants is one option of oil spill response (OSR). Here, Northern shrimp (Pandalus borealis) larvae were experimentally exposed for short periods (6 h and 1 h) to a realistic concentration of chemically dispersed oil (CDO) (~10 mg L⁻¹ THC), mechanically dispersed oil (MDO) (~7 mg L⁻¹ THC), and dispersant only (D). A control (C) with seawater served as reference. Short-term effects on survival and feeding were examined right after exposure and longer-term consequences on survival, feeding, growth and development following 30 days of recovery. Both exposure durations provoked long lasting effects on larval fitness, with 1 h exposure leading to minor effects on most of the selected endpoints. The 6 h exposure affected all endpoints with more adverse impacts after exposure to CDO. This study provides important data for assessing the best OSR option relevant to NEBA (Net Environmental Benefit Analysis).

Strongylocentrotus intermedius cultured in the northern Yellow Sea in China was utilized to evaluate the effects of chronic CO₂-driven ocean acidification (OA) on adult sea urchins. Based on the projection of the Intergovernmental Panel on Climate Change (IPCC), present natural seawater conditions (pHNBS = 8.10 ± 0.03) and three laboratory-controlled OA conditions (OA₁, ΔpHNBS = − 0.3 units; OA₂, ΔpHNBS = − 0.4 units; OA₃, ΔpHNBS = − 0.5 units) were employed. After 60-day incubation, our results showed that (1) OA significantly repressed the growth of adult S. intermedius; (2) food consumption tended to be decreased with pH decline; (3) intestinal morphology was changed, and activities of intestinal cellulase and lipase were decreased under acidified conditions; (4) expression levels of two immune-related genes (SiTNF14 and SiTGF-β) were altered; (5) rate-limiting enzyme activities of the glycolytic pathway and tricarboxylic acid cycle (TAC) were changed in all OA treatments compared to those of controls.

Crown-of-thorns starfish, Acanthaster planci (COTS), predation is a major cause of coral reef decline, but the factors behind their population outbreaks remain unclear. Increased phytoplankton food resulting from eutrophication is suggested to enhance larval survival. We addressed the hypothesis that larval success is associated with particular chl-a levels in tightly controlled larval:algal conditions. We used chl-a conditions found on coral reefs (0.1–5.0μgchl-aL−1), including nominal threshold levels for disproportionate larval success (≥1.0μgchl-aL−1). High success to the juvenile occurred across an order of magnitude of chl-a concentrations (0.5–5.0μgchl-aL−1), suggesting there may not be a narrow value for optimal success. Oligotrophic conditions (0.1μgchl-aL−1) appeared to be a critical limit. With a review of the evidence, we suggest that opportunistic COTS larvae may be more resilient to low food levels than previously appreciated. Initiation of outbreak populations need not require eutrophic conditions.
