International audience | Organochlorinated compounds including PolyChloroBiphenyles, Dichloro-DiphenylTrichloroethan and metabolites are determinated in Stenella coeruleoalba (n = 37) stranded on the french Mediterranean coasts from 2007 till 2009. Studies are carried out on lung, muscle, kidney, liver, and blubber. The sought-after compounds are all detected to variable levels in each tissue and organ. In general, total PCBs are the most abondant, followed by total DDTs. The concentration (in ng g(-1) of lipid weight) in blubber of S. coeruleoalba, varied from 2,052 to 158,992 for PCBs and from 1,120 to 45,779 for DDTs. The ratios DDE/tDDTs are higher than 80% in almost all samples. The overall results of this work, compared to previous studies concerning the Mediterranean Sea, seems to confirm the tendency to a decrease of the contamination by organics compounds for the cetaceans in the Western Mediterranean Sea. The fetus is prenatally exposed to a mixture of organochlorine pesticides (OCPs), mercury (Hg), docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA) and selenium (Se) through maternal seafood consumption in real-life scenario. Prenatal exposure to these contaminants and nutrients has been suggested to affect thyroid hormone (TH) status in newborns, but the potential relationships between them are unclear and the joint effects of the mixture are seldom analyzed. The aim of the study is to investigate the associations of prenatal exposure to a mixture of OCPs, Hg, DHA, EPA and Se with TH parameters in newborns. 228 mother-infant pairs in Shanghai, China were included. We measured 20 OCPs, total Hg, DHA, EPA and Se in cord blood samples as exposure variables. The total thyroxine (TT4), free thyroxine (FT4), total triiodothyronine (TT3), free triiodothyronine (FT3), and thyroid-stimulating hormone (TSH) levels and the FT3/FT4 ratio in cord serum were determined as outcomes. Using linear regression models, generalized additive models and Bayesian kernel machine regression, we found dose-response relationships of the mixture component with outcomes: among the contaminants, p,p'-DDE was the most important positive predictor of TT3, while HCB was predominantly positively associated with FT3 and the FT3/FT4 ratio, indicating different mechanisms underlying these relationships; among the nutrients, EPA was first found to be positively related to the FT3/FT4 ratio. Additionally, we found suggestive evidence of interactions between p,p'-DDE and HCB on both TT3 and FT3, and EPA by HCB interactions for TT3, FT3 and FT3/FT4 ratio. However, the overall effects of the mixture on thyroid hormone parameters were not significant. Our result suggests that prenatal exposure to p,p’-DDE, HCB and EPA as part of a mixture might affect thyroid function of newborns in independent and interactive ways. The potential biological mechanisms merit further investigation.

Agricultural use of organochlorine pesticides (OCPs) increased during the twentieth century but many of them have been progressively banned several decades after their introduction. Nevertheless, these lipophilic chemical compounds may persist in soils and sediments. From sediment deposits, it is possible to reconstruct the chronology of OCP releases in relation to former applications through time. Nevertheless, long-term fate of OCPs i.e. source, transfer, and storage through the watershed, is also related to the OCPs-sediment characteristics interactions, and our study showed the significant links between OCPs and labile or refractory organic matter. From sediment cores collected in a mainly agricultural watershed, the Eure River watershed (France), aldrin and lindane widespread applications during the 1950s–1970s have been recorded. While lindane applications declined after that date, according to the temporal trend of the stable isomer of hexachlorocyclohexane (β-HCH), α-, and γ-HCH have been recorded at significant levels in the 2000s, suggesting first local post-ban applications. Nevertheless, the relationships between these OCPs and labile organic matter resulted in an overestimation of the post-ban releases. Also, the detection of stable metabolites of dichlorodiphenyltrichloroethane (DDT) (i.e. 4,4′-DDE) and heptachlor (i.e. heptachlor epoxide) several decades after their ban, revealed the role of old deep soils erosion in the chronology of OCP releases and thus the reemergence of stable transformation products from historical OCPs.

Studies have documented that exposure to organochlorine pesticides (OCPs) is linked with breast cancer, but the underlying biological mechanisms are still unknown. This study included 313 women diagnosed with breast cancer and 313 controls in Wuhan, China, and measured 18 OCPs in serum and 3 oxidative stress biomarkers in urine. Multivariable adjusted regression models were used to evaluate the associations among OCPs, oxidative stress biomarkers, and breast cancer. The mediating effect of oxidative stress was assessed by mediation analysis. We observed that most OCPs were positively associated with risk of breast cancer (all FDR-P values < 0.05 or 0.10). Moreover, we found that p,p'-DDT, p,p'-DDD, dieldrin, heptachlor, and heptachlor epoxide were positively associated with 4-hydroxy-2-nonenal-mercapturic acid (HNE-MA) and 8-iso-prostaglandin F₂α (8-isoPGF₂α), which in turn were positively associated with risk of breast cancer. Mediation analysis indicated that HNE-MA and 8-isoPGF₂ɑ mediated the positive associations between these OCPs and risk of breast cancer, with mediating proportion ranging from 6.23% to 19.9%. Our results suggest that lipid peroxidation may mediate the positive associations between OCP exposures and risk of breast cancer.

In the general population, chronic exposure to low-dose persistent organic pollutants (POPs), particularly organochlorine pesticides (OCPs), has been recently linked to many chronic diseases. Widespread contamination of the food chain and human adipose tissue has made avoiding exposure to these chemicals impossible; thus, alternative strategies for decreasing the chemical burden must be investigated. Recently, macronutrient intake was found to significantly modify the toxicokinetics of POPs in animal experimental studies. Thus, we evaluated whether macronutrient intake was related to serum concentrations of OCPs in healthy adults without cardio-metabolic diseases. Subjects included 1,764 adults, aged 20 years or above, who participated in the National Health and Nutrition Examination Survey 1999–2004. Macronutrient intake was assessed based on a 24-h dietary recall interview. Six individual OCPs commonly detected among the general population were evaluated as markers of OCPs and other coexisting lipophilic chemicals stored in adipose tissue and released into circulation. High fat intake was associated with lower concentrations of OCPs, while high carbohydrate intake showed the opposite result. When three types of fats were individually evaluated, both saturated fatty acids and monounsaturated fatty acids, but not polyunsaturated fatty acids, were inversely associated with serum concentrations of OCPs. Adjustment for possible confounders did not change the results. When stratified by age, gender, body mass index, and physical activity, these associations were similar in most subgroups. Thus, similar to the findings observed in animal experimental studies, a moderate-fat diet with low carbohydrate intake was related to low serum concentrations of OCPs in humans. Although these findings need to be replicated, changing dietary macronutrient intake can be investigated as a practical strategy for dealing with unavoidable lipophilic chemical mixtures such as OCPs in modern society.

We conducted the first comprehensive assessment of the presence, source, and ecotoxicological implication of 13 banned and restricted organochlorine pesticides (OCPs) in the surface water along the Ganga River for two different seasons. Surface water samples were collected along the 2525 km stretch of the Ganga through 43 sites representing five zones of diverse land-use pattern, pesticide consumption rate, and varied flow. The mean concentrations of ΣOCPs were significantly higher (∼2–5 times) in the post-monsoon or wet season [range: 0.126 to 10.402 μg/L (mean: 2.482 μg/L ± 3.589 and median: 1.433)] than in the post-winter or dry season [range: 0.053 to 3.010 μg/L (mean: 0.765 μg/L±1.033 and median: 0.399)]. Lindane (γ-HCH) was the dominant and most frequently detected pesticide at all the sites, indicating possible continued use of this banned pesticide in agricultural practices. The spatial distribution of OCPs revealed non-significant difference amongst different zones and indicate that point source pollution from the open drains along the Ganga could be responsible for observed trend. Ratio diagnostic analysis highlighted the fresh inputs and potential illegal use of lindane and chlordane at all the zones whereas, historical use of DDT was revealed at the majority of sites. Interestingly, fresh inputs of DDT were observed in the relatively pristine high altitude Upper zone (UZ) suggesting long-range atmospheric transfer and its continued use in the zone. Risk quotient (RQ) analysis revealed high ecotoxicological risks (>1), at all the studied sites for p, p’ DDE. The lower zone (LZ) emerged as a high ecological risk zone. The study highlights that though the OCPs analysed in this study are banned/restricted in India, still the implementation of the ban is poor and delayed and the country requires stricter adherence to its National Implementation Plan (NIP) on pesticides.

Research on the atmospheric occurrence of short chain chlorinated paraffins (SCCPs) in industrialized areas is scarce. In this study, we investigated the concentrations, profile, and spatial distribution of SCCPs in the highly industrialized and developed areas of the Yangtze River Delta (YRD) in China using polyurethane foam passive air samplers. Sampling was performed during two separate periods in 2011. The concentrations of atmospheric SCCPs ranged from 6.1 to 63 ng m⁻³ in summer and 6.2–42 ng m⁻³ in winter. The C₁₀ and C₁₁ groups were the predominant carbon groups in all the samples. Different congener patterns between summer and winter were found, indicating that congeners in the air in winter may be influenced by local and external sources. The highest level of SCCPs was found in Suzhou, which is a highly industrialized area with many manufactories including electronic industries and plastic factories. Higher levels of SCCPs were found in the air than polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), and polybrominated diphenyl ether (PBDEs), suggesting that the production and use of SCCPs were much higher than prohibited or restricted persistent organic pollutants (e.g., PCBs, OCPs, and PBDEs). Future studies should investigate the different sources of atmospheric SCCPs by conducting a comprehensive assessment of SCCP exposure.
Mostrar más [+] Menos [-]Performance of a novel magnetic solid-phase-extraction microsphere and its application in the detection of organic micropollutants in the Huai River, China

Solid phase extraction has been increasingly applied for the detection of organic micropollutants (OMPs). However, time-consuming and high-cost disadvantages also limit the widespread use of this method, especially for the extraction of large-volume field water samples. In this study, a gas chromatography-mass spectrometry (GC-MS) method based on the magnetic microsphere (M150) solid-phase-extraction (MSPE) was established to investigate the OMPs in source water throughout the whole Huai River. In brief, the results demonstrated that the extraction efficiency of the M150 was superior to that of C₁₈ and HLB for the selected OMPs, including species of polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs), phthalate esters (PAEs) and nitrobenzenes (NBs), and the method detection limits of M150 for these OMPs were comparable to those of C₁₈ and HLB. The optimized conditions of extraction and elution were the 100 mg/L dosages of microspheres, extraction time of 60 min and pH of 2, and the eluent with a similar polarity, hydrophobicity and molecular structure to the OMPs rendered higher elution efficiencies. A total of 21 types of OMPs affiliating to PAHs, OCPs, PAEs and NBs were detected by the established method, with the total concentrations of 505–2310 ng/L in source water of the Huai River. Spatial differences of the OMPs were also observed, demonstrating the link between pollutant profiles and geographical locations. This study provides an alternative to enrich OMPs in filed water samples, and it reveals pollutant profiles of source water throughout the whole Huai River.

Persistent organic pollutants (POPs) including PCDD/Fs, PCBs and organochlorine pesticides (OCPs) are among the most important and hazardous pollutants of soil. Food producing animals such as chicken, beef, sheep and goats can take up soil while grazing or living outdoors (free-range) and this can result in contamination.In recent decades, large quantities of brominated flame retardants such as polybrominated diphenyl ethers (PBDEs), short-chain chlorinated paraffins (SCCPs) and per- and polyfluorinated alkylated substances (PFAS) have been produced and released into the environment and this has resulted in widespread contamination of soils and other environmental matrices. These POPs also bioaccumulate and can contaminate food of animal origin resulting in indirect exposure of humans.Recent assessments of chicken and beef have shown that surprisingly low concentrations of PCBs and PCDD/Fs in soil can result in exceedances of regulatory limits in food. Soil contamination limits have been established in a number of countries for PCDD/Fs but it has been shown that the contamination levels which result in regulatory limits in food (the maximum levels in the European Union) being exceeded, are below all the existing soil regulatory limits. ‘Safe’ soil levels are exceeded in many areas around emission sources of PCDD/Fs and PCBs. On the other hand, PCDD/F and dioxin-like PCB levels in soil in rural areas, without a contamination source, are normally safe for food producing animals housed outdoors resulting in healthy food (e.g. meat, eggs, milk).For the majority of POPs (e.g. PBDEs, PFOS, PFOA, SCCP) no regulatory limits in soils exist.There is, therefore, an urgent need to develop appropriate and protective soil standards minimising human exposure from food producing animals housed outdoors. Furthermore, there is an urgent need to eliminate POPs pollution sources for soils and to control, secure and remediate contaminated sites and reservoirs, in order to reduce exposure and guarantee food safety.
