The subcellular partitioning approach provides useful information on the location of metals within cells and is often used on organisms with high levels of bioaccumulation to establish relationships between the internal concentration and the potential toxicity of metals. Relatively little is known about the subcellular partitioning of metals in wild fish with low bioaccumulation levels in comparison with those from higher contaminated areas. This study aims to examine the subcellular partitioning of various metals considering their chemical affinity and essentiality at relatively low contamination levels. Class A (Y, Sr), class B (Cu, Cd, MeHg), and borderline (Fe, Mn) metal concentrations were measured in livers and subcellular fractions of yellow perch (n = 21) collected in Lake Saint-Pierre, QC, Canada. The results showed that all metals, apart from MeHg, were distributed among subcellular fractions according to their chemical affinity. More than 60% of Y, Sr, Fe, and Mn were found in the metal-sensitive fractions. Cd and Cu were largely associated with the metallothionein-like proteins and peptides (60% and 67% respectively) whereas MeHg was found mainly in the metal-sensitive fractions (86%). In addition, the difference between the subcellular distribution of Cu and other essential metals like Fe and Mn denotes that, although the essentiality of some metals is a determinant of their subcellular distribution, the chemical affinity of metals is also a key driver. The similarity of the subcellular partitioning results with previous studies on yellow perch and other fish species from higher contaminated areas supports the idea that metals are distributed in the cellular environment according to their chemical properties regardless of the bioaccumulation gradient.

Whereas early life stages are usually considered as particularly sensitive to both organic and inorganic contaminants, field studies assessing contaminant bioaccumulation in these stages are scarce. Selenium (Se) is thought to counteract Hg toxic effects when it is found at Se:Hg molar ratios above 1. However, the variation of this ratio in key fish tissues of different early life stages is mostly unknown. The present study therefore aimed to assess Hg and Se content in gravid female tissues (gonads, muscle, liver, gut, and brain) and different life stages (egg masses, newly hatched larvae (NHL), larvae and juvenile) of Yellow Perch (YP) in a large fluvial lake (Lake Saint-Pierre, Québec, Canada). Se:Hg molar ratios were measured for each compartment in order to fill associated knowledge gaps. Total Hg (THg) and methylmercury (MeHg) concentration varied between tissue according to the following trend: Muscle > Liver > Gut > Brain > Gonads. During YP early life stages, MeHg values increased according to an ontogenetic pattern (mg/kg dw) (mean ± SEM): Egg masses (0.01 ± 0.002) < NHL (0.015 ± 0.001) < Larvae (0.14 ± 0.01) < Juveniles (0.18 ± 0.01). Se concentrations in different YP tissues showed the following trend (mg/kg dw) (mean ± SEM): Gut (3.6 ± 0.1) > Liver (2.5 ± 0.1) > Gonads (1.92 ± 0.06) > Brain (1.26 ± 0.03) > Muscle (1.23 ± 0.06). In YP early life stages, Se concentrations were highest in NHL (3.0 ± 0.2), and then decreased as follows: Egg masses (2.8 ± 0.1) > Larvae (1.37 ± 0.04) > Juveniles (0.93 ± 0.05). Se:Hg molar ratios varied considerably and were systematically above 1. This is the first study to simultaneously report Hg and Se bioaccumulation through fish life cycle.

We assessed the risks of mercury in yellow perch, a species important in the trophic transfer of methylmercury, in the Great Lakes region. Mean concentrations in whole perch from 45 (6.5%) of 691 waters equaled or exceeded 0.20 μg/g w.w., a threshold for adverse effects in fish. In whole perch within the size range eaten by common loons (<100 g), mean concentrations exceeded a dietary threshold (0.16 μg/g w.w.) for significant reproductive effects on loons in 19 (7.3%) of 260 waters. Mean concentrations in fillets of perch with length ≥ 15.0 cm, the minimum size retained by anglers, exceeded the USEPA criterion (0.3 μg/g w.w.) in 26 (6.4%) of 404 U.S. waters and exceeded the Ontario guideline (0.26 μg/g w.w.) in 35 (20%) of 179 Ontario waters. Mercury levels in yellow perch in some waters within this region pose risks to perch, to common loons, and to mercury-sensitive human populations.

Seasonal variation in mercury (Hg) concentrations and food web structure was assessed for eastern Lake Ontario. Hg concentrations, measured in 6 species of invertebrates and 8 species of fishes, tended to be highest in the spring and lowest in the summer for most biota. Yellow perch (Perca flavescens) exhibited significant ontogenetic shifts in diet and Hg, although such patterns were not evident for other species. Food web structure, as indicated by stable isotope values (δ¹⁵N, δ¹³C) was not static. Log-transformed Hg data were strongly and consistently correlated with δ¹⁵N values for the whole food web in each of the three seasons (slopes, 0.17–0.24) and across the entire year (slope, 0.2). While significantly different between seasons, the regression slope values are still consistent with published global Hg biomagnification rates. Our results indicate that the assessment of Hg trends in Great Lakes must take into account seasonal patterns and time of sampling.

We investigated mercury (Hg) concentrations in small fish (mainly yellow perch, Perca flavescens; ∼60% of fish collected) and in blood of common loons (Gavia immer) that prey upon them during the breeding season on lakes in 4 large, widely separated study areas in Canada (>13 lakes per study area; total number of lakes = 93). Although surface sediments from lakes near a base metal smelter in Flin Flon, Manitoba had the highest Hg concentrations, perch and other small fish and blood of common loon chicks sampled from these same lakes had low Hg concentrations similar to those from uncontaminated reference lakes. Multiple regression modeling with AIC analysis indicated that lake pH was by far the most important single factor influencing perch Hg concentrations in lakes across the four study areas (R2 = 0.29). The best model was a three-variable model (pH + alkalinity + sediment Se; Wi = 0.61, R2 = 0.85). A single-variable model (fish Hg) best explained among-lake variability in loon chick blood Hg (Wi = 0.17; R2 = 0.53). From a toxicological risk perspective, all lakes posing a potential Hg health risk for perch and possibly other small pelagic fish species (where mean fish muscle Hg concentrations exceeded 2.4 μg/g dry wt.), and for breeding common loons (where mean fish muscle Hg concentrations exceeded 0.8 μg/g dry wt., and loon chick blood Hg exceeded 1.4 μg/g dry wt.) had pH < 6.7 and were located in eastern Canada.

Yellow perch (Perca flavescens) collected from 11 lakes in the Canadian mining regions of Sudbury (Ontario) and Rouyn-Noranda (Quebec) display wide ranges in the concentrations of cadmium (Cd), nickel (Ni), selenium (Se), and thallium (Tl) in their livers. To determine if these trace elements, as well as copper (Cu) and zinc (Zn), are causing oxidative stress in these fish, we measured three biochemical indicators (glutathione (GSH), glutathione disulfide (GSSG) and thiobarbituric acid-reactive substances (TBARS)) in their livers. We observed that 44% of the yellow perch that we collected were at risk of cellular oxidative stress and lipid peroxidation. Considering all fish from all lakes, higher liver Se concentrations were coincident with both lower proportions of GSSG compared to GSH and lower concentrations of TBARS, suggesting that the essential trace-element Se acts as an antioxidant. Furthermore, fish suffering oxidative stress had higher proportions of Cd, Cu and Zn in potentially sensitive subcellular fractions (organelles and heat-denatured proteins) than did fish not suffering from stress. This result suggests that reactive oxygen species may oxidize metal-binding proteins and thereby reduce the capacity of fish to safely bind trace metals. High Cd concentrations in metal-sensitive subcellular fractions likely further exacerbate the negative effects of lower Se exposure.

Bisphenol-A (BPA) is an environmental contaminant used in the manufacturing of polycarbonate plastics and epoxy resins, which has been discovered in freshwater systemsworldwide as a result of effluent from manufacturing. This bioactive molecule is an estrogen mimic and has become a concern for exposure, especially during development, resulting in its removal from baby bottles and other consumer products. BPA is an endocrine disruptor in a variety of species and has been classified as a toxic substance in multiple countries. In this study, we examined the effect of BPA exposure on leukocyte counts in wild yellow perch, Perca flavescens. Yellow perch were exposed to either 2, 4, and 8 ppb BPA; Saprolegnia; or a blank control for a period of 7 days. Leukocyte blood counts were significantly higher in Saprolegnia, 4 ppb BPA, and 8 ppb BPA treatments compared to control. To test compound effects of BPA and Saprolegnia on leukocyte counts over a 7-day period, perch were exposed to either 4 ppb BPA, 4 ppb BPA+Saprolegnia, or control. Leukocyte counts were significantly higher in the 4 ppb BPA treatment relative to control. The 4 ppb BPA+Saprolegnia treatment was numerically elevated from the control, exhibiting a 153 % increase relative to control. BPA represents a contaminant with immunomodulatory properties that remain to be determined. © Springer Science+Business Media Dordrecht 2013.

We sampled and analyzed individually, edible dorsal muscle from largemouth bass (LMB), Micropterus salmoides (n = 138) and yellow perch (YP), Perca flavescens (n = 97) from 15 lakes to investigate potential local impacts of mercury emission point sources in northeastern Massachusetts (MA), USA. This area was identified in three separate modeling exercises as a mercury deposition hotspot. In 1995, 55% of mercury emissions to the environment from all MA sources came from three municipal solid waste combustors (trash incinerators) and one large regional medical waste incinerator in the study area. We determined the mercury accumulation history in sediments of a lake centrally located in the study area. Recent maximum mercury accumulation rates in the sediment of the lake of ~ 88 μg/m²/year were highly elevated on a watershed area adjusted basis compared to other lakes in the Northeast and Minnesota. Fish from the study area lakes had significantly (p = 0.05) greater total mercury concentrations than fish from 24 more rural, non-source-impacted lakes in other regions of the state (LMB n = 238, YP n = 381) (LMB: 1.5–2.5 x; YP: 1.5 x). The integration of this extensive fish tissue data set, depositional modeling projections, historical record of mercury accumulation in sediments of a lake in the area, and knowledge of substantial mercury emissions to the atmosphere in the area support designation of this area as a mercury depositional and biological concentration hotspot in the late 1990s, and provides further evidence that major mercury point sources may be associated with significant local impacts on fisheries resources.

This article identifies environmental factors that explain most of the dynamic year-to-year changes in mercury concentrations of young-of-year (YOY) yellow perch (Perca flavescens) in study reservoirs. Mercury concentrations in fish, collected each fall, were measured for 9 years in four reservoirs in northeastern Minnesota. Three to 4 years of data were also obtained for two natural lakes and one other reservoir. Average annual concentrations varied considerably from year to year with a mean change of 39% between consecutive years across all lakes. Those averages show a similar time trend for each lake over the years and suggest that important factors influencing mercury bioaccumulation change annually and are also experienced in common over the study region. Three factors satisfying that description are precipitation depth, water level, and average air temperature. This article reveals that all three have statistically significant correlations with observed mercury concentrations. Moreover, multiple regressions indicate that maximum water levels and average air temperatures explain most of the observed variations. Regressions employing precipitation depth and temperature are less significant.

Mercury (Hg) contamination is a global issue due to its anthropogenic release, long-range transport, and deposition in remote areas. In Kejimkujik National Park and National Historic Site, Nova Scotia, Canada, high concentrations of total mercury (THg) were found in tissues of yellow perch (Perca flavescens). The aim of this study was to evaluate a possible relationship between THg concentrations and the morphology of perch liver as a main site of metal storage and toxicity. Yellow perch were sampled from five lakes known to contain fish representing a wide range in Hg concentrations in fall 2013. The ultrastructure of hepatocytes and the distribution of Hg within the liver parenchyma were analyzed by transmission electron microscopy (TEM) and electron energy loss spectrometry (EELS). The relative area of macrophage aggregates (MAs) in the liver was determined using image analysis software and fluorescence microscopy. No relation between general health indicators (Fulton’s condition index) and THg was observed. In line with this, TEM examination of the liver ultrastructure revealed no prominent pathologies related to THg accumulation. However, a morphological parameter that appeared to increase with muscle THg was the relative area of MAs in the liver. The hepatic lysosomes appeared to be enlarged in samples with the highest THg concentrations. Interestingly, EELS analysis revealed that the MAs and hepatic lysosomes contained Hg.
