The impact of nanoplastics (NP) using model polystyrene nanoparticles amine functionalized (PS–NH2) has been investigated on pigment and lipid compositions of the marine diatom Chaetoceros neogracile, at two growth phases using a low (0.05 μg mL−1) and a high (5 μg mL−1) concentrations for 96 h. Results evidenced an impact on pigment composition associated to the light-harvesting function and photoprotection mainly at exponential phase. NP also impacted lipid composition of diatoms with a re-adjustment of lipid classes and fatty acids noteworthy. Main changes upon NP exposure were observed in galactolipids and triacylglycerol's at both growth phases affecting the thylakoids membrane structure and cellular energy reserve of diatoms. Particularly, exponential cultures exposed to high NP concentration showed an impairment of long chain fatty acids synthesis. Changes in pigment and lipid content of diatom’ cells revealed that algae physiology is determinant in the way cells adjust their thylakoid membrane composition to cope with NP contamination stress. Compositions of reserve and membrane lipids are proposed as sensitive markers to assess the impact of NP exposure, including at potential predicted environmental doses, on marine organisms.

Eutrophic freshwater lake ecosystems are receiving increasing public attention due to a global increase in large-scale harmful cyanobacterial blooms in surface waters. However, the contribution of phytodetritus accumulation in benthic sediments post-bloom remains unclear. In this study, field investigations were performed using microsensors to evaluate benthic phytodetritus mats by measuring TOC/TN ratios, pigments, biodegradable compounds and odorants as descriptive parameters. Results show that the massive amount of phytodetritus trapped by aquatic plants gradually evolved into benthic cyanobacterial detritus mats, which were characterized as anoxic, reductive and low pH. It was confirmed that the occurrence of odorants is more serious in the detritus mats due to decay and decomposition of the accumulated phytodetritus. The mean odorant content in the vegetated zones was 3–52 times higher than that in the unvegetated zones. The dominant odorants were dimethyl trisulfide (DMTS), β-ionone and β-cyclocitral, with mean contents of 52.38 ng·(g·dw)-1, 162.20 ng·(g·dw)-1 and 307.51 ng·(g·dw)-1, respectively, in the sediment. In addition, odorant production appears to be associated with the distribution of biodegradable compounds in the sediment. This is supported by the marked correlation observed between biodegradable compounds and odorants. Multiple regression analysis showed that biodegradable compounds can be used as indicators to predict odorant content in the sediment. It is noteworthy that the odorant trend in the water column and sediment is symmetrical, indicating a risk of diffusion from the sediment to the water column. This study helps to clarifying the contributions of benthic cyanobacterial detritus mats to odorant production in shallow eutrophic lakes. The information provided herein may also be useful for future management of aquatic ecosystems.

Moro, Isabella | Trentin, Riccardo | Moschin, Emanuela | Dalla Vecchia, Francesca
The pollution of aquatic bodies by pharmaceutical compounds is an emerging environmental problem, with little explored consequences. Oxytetracycline (OTC) is an antibiotic used for treatment of infections caused by a variety of microorganisms and it is widely employed in medicine, livestock husbandry and aquaculture. This pharmaceutical compound may cause deleterious effects on non-target aquatic organisms as microalgae. The objective of this study was to evaluate the effects of OTC on growth, pigment content and morpho-physiology of the microalga Isochrysis galbana Parke. The results highlighted that OTC exposure inhibited the growth of I. galbana in cultures treated with OTC 5.0 and 10.0 mg/L after 3 days and in cultures treated with OTC 5.0, 7.5 and 10.0 mg/L after 5 days. Effects of OTC on cells ultrastructure and physiology consisted in large cytoplasmic lipid inclusions and in a decrease of photosynthetic pigments amount.

It is of environmental significance to study the leaching performance of additives from microplastics (MPs) and further evaluate the toxicity of leachate to microalgae. Here, we investigated the effects of accelerated aging on characteristics, leaching, and toxicity of commercial lead chromate pigmented MPs. Results show that aging of MPs caused surface cracks and fragmentation, increased their surface area and carbonyl contents, and promoted the release of lead chromate pigment. Chromium (Cr) and lead (Pb) tend to leach under acidic condition, rather than neutral and alkali environment. Aging treatment facilitates the leaching performance and a high concentration of NaCl solution also favors the leaching process. Toxicology experiments demonstrate that only high concentration of leachate (>10 μg L⁻¹) exerted significant inhibitory influence (p < 0.005) on cell photosynthesis of Microcystis aeruginosa. The growth inhibition of algal cells remarkably increased with increasing leachate concentrations. We observed more inhibiting effects on cell growth and photosynthesis using the leachates of aged MPs. Longer aging time leads to more release of Cr and Pb, rendering higher toxicity to microalgae. These novel findings will benefit us from assessing the leaching behavior of additives in MPs and their toxicological risks to aquatic organisms.

2020
A field study was conducted to understand the physiological responses, yield and grain quality of an old (HUW234) and a modern (HD3118) wheat cultivar exposed to elevated ozone (O₃). The cultivars were grown under ambient O₃ (NF) and ambient +20 ppb O₃ (NF+) conditions using open-top chambers (OTCs). The comparative study of an old and a modern cultivar showed variable physiological responses under elevated O₃ exposure. Elevated O₃ in old cultivar caused high reductions in Rubisco activity (Vcₘₐₓ) and electron transport rate (J) compared to modern cultivar with simultaneous reductions in the rate of photosynthesis and chlorophyll fluorescence. In modern cultivar, high stomatal density and conductance caused higher O₃ uptake thereby triggering more damage to the adjacent stomatal cells and photosynthetic pigments coupled with reductions in photosynthetic rate and photosynthetic nitrogen use efficiency (PNUE). Modern cultivar also showed relatively high reduction in grain yield compared to old one under NF + treatment. Furthermore, grain quality traits (such as starch, protein and amino acids) of modern cultivar were better than old cultivar under ambient O₃, but showed more deterioration under NF + treatment. Results thus indicated that modern cultivar is relatively more susceptible to O₃ and showed more negative impacts on plant performance, yield and quality of grains compared to old cultivar.

The present study investigated the beneficial role of selenium-nanoparticles (Se-NPs) in mitigating the adverse effects of soil-salinity on growth and yield of strawberry (Fragaria × ananassa Duch.) plants by maneuvering physiological and biochemical mechanisms. The foliar spray of Se-NPs (10 and 20 mg L⁻¹) improved the growth and yield parameters of strawberry plants grown on non-saline and different saline soils (0, 25, 50 and 75 mM NaCl), which was attributed to their ability to protect photosynthetic pigments. Se-NPs-treated strawberry plants exhibited higher levels of key osmolytes, including total soluble carbohydrates and free proline, compared with untreated plants under saline conditions. Foliar application of Se-NPs improved salinity tolerance in strawberry by reducing stress-induced lipid peroxidation and H₂O₂ content through enhancing activities of antioxidant enzymes like superoxide dismutase and peroxidase. Additionally, Se-NPs-treated strawberry plants showed accumulation of indole-3-acetic acid and abscisic acid, the vital stress signaling molecules, which are involved in regulating different morphological, physiological and molecular responses of plants to salinity. Moreover, the enhanced levels of organic acids (e.g., malic, citric and succinic acids) and sugars (e.g., glucose, fructose and sucrose) in the fruits of Se-NPs-treated strawberry plants under saline conditions indicated the positive impacts of Se-NPs on the improvement of fruit quality and nutritional values. Our results collectively demonstrate the definite roles of Se-NPs in management of soil salinity-induced adverse effects on not only strawberry plants but also other crops.

Ground-level ozone (O3) concentrations have been elevating in the last century. While there has been a notable progress in understanding O3 effects on vegetation, O3 effects on ecological stoichiometry remain unclear, especially early in the oxidative stress. Ethyelenediurea (EDU) is a chemical compound widely applied in research projects as protectant of plants against O3 injury, however its mode of action remains unclear. To investigate O3 and EDU effects early in the stress, we sprayed willow (Salix sachalinensis) plants with 0, 200 or 400 mg EDU L−1, and exposed them to either low ambient O3 (AOZ) or elevated O3 (EOZ) levels during the daytime, for about one month, in a free air O3 controlled exposure (FACE); EDU treatment was repeated every nine days. We collected samples for analyses from basal, top, and shed leaves, before leaves develop visible O3 symptoms. We found that O3 altered the ecological stoichiometry, including impacts in nutrient resorption efficiency, early in the stress. The relation between P content and Fe content seemed to have a critical role in maintaining homeostasis in an effort to prevent O3-induced damage. Photosynthetic pigments and P content appeared to play an important role in EDU mode of action. This study provides novel insights on the stress biology which are of ecological and toxicological importance.

2017

To evaluate the ozone (O3) sensitivity among peach tree (Prunus persica) cultivars widely planted in Beijing region and explore the possible eco-physiological response mechanisms, thirteen cultivars of peach seedlings were exposed to either charcoal-filtered air or elevated O3 (E-O3, non-filtered ambient air plus 60 ppb) for one growing season in open-top chambers. Leaf structure, stomatal structure, gas exchange and chlorophyll a fluorescence, photosynthetic pigments, antioxidant defense system and lipid peroxidation were measured in three replicated chambers. Results showed that E-O3 significantly reduced abaxial epidemis thickness, but no effects on the thicknesses of adaxial epidemis, palisade parenchyma and spongy parenchyma. Stomatal area, density and conductance were not significantly affected by E-O3. E-O3 significantly accelerated leaf senescence, as indicated by increased lipid peroxidation and more declines in light-saturated photosynthetic rate and pigments contents. The reduced ascorbate content (ASC) was decreased but antioxidant enzyme activity (CAT, APX and SOD) and total antioxidant capacity (TAC) were significantly increased by E-O3 among cultivars. The cultivars with visible symptoms also had more reductions in net photosynthetic rate than those without visible symptoms. Ozone sensitivity among cultivars was strongly linked to leaf mass per area (LMA), antioxidant enzymes activity e.g. SOD, APX rather than stomatal parameters (stomatal area, density and conductance) and ASC. Results could provide a theoretical basis for selecting and breeding the ozone-resistant cultivars of peach trees grown in high O3-polluted regions.

In recent years, some pigments and chemical products have been reported to contain polychlorinated biphenyl (PCB) congeners as unintentional byproducts, and these have also been detected in residential environments from indoor air and house dust. In this study, using in vitro reporter gene assays, we characterized the agonistic and antagonistic activities of a total of 25 PCB congeners contained in pigments (PCB-1 to -16, -20, -35, -40, -52, -56, -77, -101, -126, and -153) against five nuclear hormone receptors, (estrogen receptor (ER) α/β, glucocorticoid receptor (GR), androgen receptor (AR), thyroid hormone receptor (TR) α1) and aryl hydrocarbon receptor (AhR). In the ERα/β assays, 19 and 13 of the 25 PCBs tested showed ERα/β agonistic and/or antagonistic activities, respectively. Relatively potent agonistic activities against ERα/β were found in PCB congeners possessing chlorides at positions 2 and 3. In the GR and AR assays, five and all of the 25 PCB congeners showed antagonistic activity, respectively. Among the anti-androgenic PCB congeners, the activities were more potent in PCB congeners possessing more than three chlorides including consecutive ortho- and meta- or meta- and para-chlorides. In the AhR assay using a sensitive DR-EcoScreen cell line, five of the 25 PCB congeners showed agonistic activity. We newly found that PCB-1, -35 and -56 can act as AhR agonists. Despite these activities among the PCBs, the effects of PCB-11, mainly detected in pigments and chemical products, against these receptors were found to be weaker than those of other tested PCBs. These results suggest that unintentional PCBs in pigments and chemical products might act as agonists and/or antagonists against ERα/β, AR, GR, and AhR, and some of the PCBs might disrupt endocrine functions via multiple receptors and/or simultaneously induce dioxin-like activity via AhR.
