The current study provides an information on the combined effect of pollution with potentially toxic elements (PTEs) and polycyclic aromatic hydrocarbons (PAHs) in hydromorphic soils on the accumulation, growth, functional and morphological-anatomical changes of macrophyte plant, i.e., Phragmites australis Cav., as well as information about their bioindication status on the example of small rivers of the Azov basin. The territory of the lower reaches of the Kagalnik River is one of the small rivers of the Eastern Azov region was examined with different levels of PTEs contamination in soils, where the excess of the lithosphere clarkes and maximum permissible concentrations (MPC) for Mn, Cr, Zn, Pb, Cu, and Cd were found. The features of the 16 priority PAHs quantitative and qualitative composition in hydromorphic soils and P. australis were revealed. The influence of soil pollution on accumulation in P. australis, as well as changes in the morphological parameters were shown. It has been observed that morphometric changes in P. australis at sites experiencing the сontamination and salinity are reflected with the changes in the ultrastructure of plastids, mitochondria, and EPR elements of plant cells. PTEs accumulated in inactive organs and damaged cell structures. At the same time, PAHs penetrated through the biomembranes and violated their integrity, increased permeability, resulted cell disorganization, meristem, and conductive tissues of roots. The nature and extent of the structural alterations found are dependent on the type and extent of pollution in the examined regions and can be utilized as bioindicators for evaluating the degree of soil phytotoxicity characterized by the accumulation of PTE and PAHs.

We investigated the roles of rootstocks in Cu accumulation and tolerance in Malus plants by grafting ‘Hanfu’ (HF) scions onto M. baccata (Mb) and M. prunifolia (Mp) rootstocks, which have different Cu tolerances. The grafts were exposed to basal or excess Cu for 20 d. Excess Cu-treated HF/Mb had less biomass, and pronounced root architecture deformation and leaf ultrastructure damage than excess Cu-challenged HF/Mp. Root Cu concentrations and bio-concentration factor (BCF) were higher in HF/Mp than HF/Mb, whereas HF/Mb had higher stem and leaf Cu concentrations than HF/Mp. Excess Cu lowered root and aerial tissue BCF and translocation factor (Tf) in all plants; however, Tf was markedly higher in HF/Mb than in HF/Mp. The subcellular distribution of Cu in the roots and leaves indicated that excess Cu treatments increased Cu fixation in the root cell walls, which decreased Cu mobility. Compared to HF/Mb, HF/Mp sequestered more Cu in its root cell walls and less Cu in leaf plastids, nuclei, and mitochondria. Moreover, HF/Mp roots and leaves had higher concentrations of water-insoluble Cu compounds than HF/Mb, which reduced Cu mobility and toxicity. Fourier transform infrared spectroscopy analysis showed that the carboxyl, hydroxyl and acylamino groups of the cellulose, hemicellulose, pectin and proteins were the main Cu binding sites in the root cell walls. Excess Cu-induced superoxide anion and malondialdehyde were 28.6% and 5.1% lower, but soluble phenolics, ascorbate and glutathione were 10.5%, 41.9% and 17.7% higher in HF/Mp than HF/Mb leaves. Compared with HF/Mb, certain genes involved in Cu transport were downregulated, while other genes involved in detoxification were upregulated in HF/Mp roots and leaves. Our results show that Mp inhibited Cu translocation and mitigated Cu toxicity in Malus scions by regulating Cu mobility, antioxidant defense mechanisms, and transcription of key genes involved in Cu translocation and detoxification.

Cement industries located nearby limestone outcrops in Brazil have contributed to the coating of cement dust over native plant species. However, little is known about the extent of the response of tropical woody plants to such environmental pollutant particularly during the first stages of plant development and establishment. This work focused on the investigation of possible alterations in leaf structural and ultrastructural traits of 5-month-old Guazuma ulmifolia Lam. (Malvaceae), 6-month-old Myracrodruon urundeuva Allemão (Anacardiaceae), and 9-month-old Trichilia hirta L. (Meliaceae) challenged superficially with cement dust during new leaf development. Leaf surface of plants, the soil or both (leaf plus soil), were treated (or not) for 60 days, under controlled conditions, with cement dust at 2.5 or 5.0 mg cm⁻². After exposure, no significant structural changes were observed in plant leaves. Also, no plant death was recorded by the end of the experiment. There was also some evidence of localized leaf necrosis in G. ulmifolia and T. hirta, leaf curling in M. urundeuva and T. hirta, and bulges formation on epidermal surface of T. hirta, after cement dust contact with plant shoots. All species studied exhibited stomata obliteration while T. hirta, in particular, presented early leaf abscission, changes in cellular relief, and organization and content of midrib cells. No significant ultrastructural alterations were detected under the experimental conditions studied. Indeed, mesophyll cells presented plastids with intact membrane systems. The high plant survival rates, together with mild morphoanatomic traits alterations in leaves, indicate that G. ulmifolia is more resistant to cement dust pollutant, followed by M. urundeuva and T. hirta. Thus, the three plant species are promising for being used to revegetate areas impacted by cement industries activities.
