Gestational PCB52 exposure induces hepatotoxicity and intestinal injury by activating inflammation in dam and offspring mice: A maternal and progeny study

Although Polychlorinated biphenyl (PCB) levels are decreased in the environment, the adverse effects of gestational exposure on the mother and offspring cannot be ignored due to the vulnerability of the fetus. In the present study, pregnant Balb/c mice were administered PCB52 (1 mg/kg BW/day) or corn oil vehicle by gavage until parturition. In the dams, PCB52 caused histopathological changes in the liver, higher serum levels of aminotransferase and alanine aminotransferase, and activated apoptosis and autophagy, suggesting hepatotoxicity. Overexpressed indicators of TLR4 pathway were observed in the liver of PCB52-exposed dams, indicated hepatic inflammation. Moreover, PCB52 exposure weakened the intestinal barrier and triggered inflammatory response, which might contribute to the hepatic inflammation by gut-liver axis. In the pups, prenatal PCB52 exposure affected the sex ratio at birth and reduced birth length and weights. Similar to the dams, prenatal PCB52 exposure induced hepatotoxicity in the pups without gender difference. Consistent with the alteration of gut microbiota, intestinal inflammation was confirmed, accompanying the disruption in the intestinal barrier and the activation of apoptosis and autophagy in the PCB52-exposed pups. Intestinal injury might be responsible for hepatotoxicity at least in part. Taken together, these findings suggested that gestational PCB52 exposure induced hepatic and intestinal injury in both maternal and offspring mice by arousing inflammation.

Polychlorinated biphenyls (PCBs) are a class of persistent organic pollutants (POPs) that have adverse effects on human health. However, the long-term health effects and potential mechanism of neonatal exposure to PCBs are still unclear. In this study, nursing male mice exposed to PCB138 at 0.5, 5, and 50 μg/kg body weight (bw) from postnatal day (PND) 3 to PND 21 exhibited increased serum uric acid levels and liver uric acid synthase activity at 210 days of age. We also found an increased kidney somatic index in the 50 μg/kg group and kidney fibrosis in the 5 and 50 μg/kg groups. Mechanistically, PCB138 induced mitochondrial dysfunction and endoplasmic reticulum (ER) stress, which might have led to inflammatory responses, such as activation of the NF-κB (nuclear factor kappa-B) and NLRP3 (NOD-like receptor protein 3) pathways. The inflammatory response might regulate renal fibrosis and hypertrophy. In summary, this study reports a long-term effect of neonatal PCB exposure on uric acid metabolism and secondary nephrotoxicity and clarifies the underlying mechanism. Our work also indicates that early life pollutant exposure may be an important cause of diseases later in life.

Breast milk samples were collected from 51 mothers in a seaside city Dalian, where the residents usually have higher dietary exposure to polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) and polychlorinated biphenyls (PCBs) due to the larger consumption of seafood. The lipid-basis concentrations of ∑Cl₂–₈DD/Fs, ∑Cl₂–₁₀Bs, and total toxicity equivalent (WHO-TEQ) were measured to be in the ranges of 35.7–2727.8 pg/g, 4.91–52.64 ng/g, and 2.27–36.30 pg/g, respectively. The average proportion of ∑Cl₂–₃DD/Fs was higher than that of ∑Cl₄–₇DD/Fs in the collected human breast milk samples, suggesting that the health risk of Cl₂–₃DD/Fs should be especially concerned. The concentration data of PCDD/Fs and PCBs in human breast milk essentially followed a positive skew probability distribution. Women in high-level exposure scenarios exhibited a higher potential to accumulate homologues Cl₄DFs, Cl₇DFs, Cl₈DF, and Cl₆Bs in breast milk. Three PCDD/F congeners (1,2,3,6,7,8-Cl₆DF, 1,2,3,4,7,8-Cl₆DF, and 1,2,3,4,6,7,8-Cl₇DD) and three PCB congeners (PCB 126, PCB 138, and PCB 169) were identified as good indicators for the accumulation of PCDD/Fs and PCBs in human breast milk, respectively. The food-to-milk accumulation factors (FMAF) were calculated to evaluate the accumulation potentials of different PCDD/F and PCB congeners in human breast milk via dietary exposure. The calculated FMAF value presented a non-monotonic variation with the logarithm of n-octanol–water partition coefficient (log KOW) with a peak at a log KOW value of about 7.3 and a valley at a log KOW value of about 8. The mean value of the estimated daily intake (EDI) of total WHO-TEQ for breast-fed infants in Dalian, predicted by Monte Carlo simulation, was 10 folds higher than the upper range of the tolerable daily intake (TDI) value (4 pg WHO-TEQ/kg bw/d), suggesting continued and enhanced efforts should be made to reduce the exposure risk of infants to PCDD/Fs and PCBs.

The contamination of paddy soils is of great concern since it links to human health via food supply. Limited knowledge is available on PCB residue characteristics and the associated health risks in paddy soils under various environmental conditions. In this study, a soil sampling campaign was conducted in three typical paddy fields, i.e., Sanjiang Plain (SP), Taihu Plain (TP) and Hani Terrace (HT), crossing a transect of 4000 km in China. The concentrations of 29 quantified PCBs varied from 58.6 to 1930 pg/g in paddy soils, with samples at TP showing the highest burden. Tri-CBs were the major homologue group at SP and HT, whereas hexa-CBs at TP. Altitude, temperature, soil organic matter content and soil conductivity well explained the variations in PCB concentrations among sites. The homologue profiles of soil PCBs followed the fractionation theory. In addition, soil conductivity was found to be negatively correlated to low-chlorinated PCBs and positively to high-chlorinated congeners. Furthermore, the toxicities of soil PCBs and the exposure risks through rice intake were estimated. Higher toxicity equivalent quantities and hazard indexes were found at SP than TP and HT, with over one third of the samples displaying health risks. The results of this work highlight the necessity to better understand the occurrence characteristics and the associated health risks of PCBs in soils of rice-growing regions.

The present paper reports on the interrelationships of fish, parasites and the bioaccumulation of hazardous organic compounds in the Zemplínska Šírava water reservoir in eastern Slovakia, which is heavily polluted with polychlorinated biphenyls (PCBs). The concentrations of these contaminants were measured in various fish matrices (dorsal and abdominal muscle tissues, hepatopancreas, intestine wall and adipose tissue) of the freshwater bream, Abramis brama (Cyprinidae), and in its intestinal parasite Caryophyllaeus laticeps (Cestoda), which was used for the first time as a model for a PCB bioaccumulation study. Regarding the fish, the highest concentrations of PCBs were found in the intestine, followed by hepatopancreas and muscle tissues. The amounts of PCBs were higher in abdominal muscles than in their dorsal parts. Concentrations of ∑PCBs above the limits set by European regulations were detected in both muscle parts in the fish, confirming the persistent unfavorable conditions in this locality and high risk for biota and humans. Based on bioconcentration factor values (BCFs), PCBs reached much higher levels in cestodes compared to bream matrices. Some significant differences in PCB amounts between infected and uninfected bream were determined. Fulton's condition factor (CF) significantly differed in infected and non-infected fish (p ˂ 0.05), with CF values surprisingly lower in fish free of parasites compared to parasitized fish, which suggests a “mutualistic” relationship between the parasite and its host.

Considerable amounts of previously deposited persistent organic pollutants (POPs) are stored in the Arctic cryosphere. Transport of freshwater and terrestrial material to the Arctic Ocean is increasing due to ongoing climate change and the impact this has on POPs in marine receiving systems is unknown This study has investigated how secondary sources of POPs from land influence the occurrence and fate of POPs in an Arctic coastal marine system.Passive sampling of water and sampling of riverine suspended particulate matter (SPM) and marine sediments for analysis of polychlorinated biphenyls (PCBs) and hexachlorobenzene (HCB) was carried out in rivers and their receiving fjords in Isfjorden system in Svalbard. Riverine SPM had low contaminant concentrations (<level of detection-28 pg/g dw ΣPCB₁₄, 16–100 pg/g dw HCB) compared to outer marine sediments 630-880 pg/g dw ΣPCB₁₄, 530–770 pg/g dw HCB). There was a strong spatial gradient in sediment PCB and HCB concentrations with lowest concentrations in river estuaries and in front of marine-terminating glaciers and increasing concentrations toward the outer fjord. This suggests that rather than leading to increased concentrations, inputs of SPM from land lead to a dilution of contaminant concentrations in nearshore sediments. Preliminary estimates of SPM:water activity ratios suggest that terrestrial particles (with low contaminant concentrations) may have the potential to act as sorbents of dissolved contaminants in the coastal water column, with implications for bioavailability of POPs to the marine food web. There is concern that ongoing increases in fluxes of freshwater, sediments and associated terrestrial material (including contaminants) from land to the Arctic Ocean will lead to increased mobilization and transport of POPs to coastal ecosystems. However, the results of this study indicate that on Svalbard, inputs from land may in fact have the opposite effect, leading to reduced concentrations in coastal sediments and waters.

Polychlorinated biphenyls (PCBs) may transfer into the neonates through the placental transfer and via breastfeeding after the delivery, thus might be harmful to the infant. Sixty colostrum samples in the Yangtze River Region were collected to investigate the concentration, distribution pattern, and enantiomer characteristic of the PCB exposure. Among all samples, over 90% of pollutants were tetra-to hepta-chlorinated PCBs. The sum concentration of the PCB was 512 (IQR: 322–856) ng g⁻¹ lipid weight. Enantiomer fraction (EF) of PCB 95 and PCB 149 was found lower than the racemic value, while EFs of PCB 45 and PCB 136 were found higher and near-racemic state, respectively. The concentration pattern and enantiomeric properties of the PCBs indicated that the mothers from Mianyang had a recent exposure to PCBs. Among all samples, similar exposure and metabolic pathways of the PCB congeners were observed. PCB exposure showed no significant correlation with the birth outcome of the infants, but 43.3% of the infants have potential health risks via breastfeeding.

As humans are present in Antarctica only for scientific and tourism-related purposes, it is often described as a pristine region. However, studies have identified measurable levels of Persistent Organic Pollutants (POPs), such as organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs), in the Antarctic region. These are highly toxic anthropogenic compounds with tendency to travel long distances and reach remote environments, where they can bioaccumulate in the biota. Penguins are exposed to POPs mainly through their diet, which they partially eliminate via feathers. Species of the genus Pygoscelis occur around Antarctic continent and its surrounding regions, and can act as indicators of contaminants that reach the continent. Here, we report OCP and PCB levels in feathers of male and female penguins of P. adeliae, P. antarcticus and P. papua from King George Island, South Shetland Islands, Antarctica. Interspecific, sex- and body-size-related differences were investigated in the contamination profiles of PCBs and OCPs. Feather samples were collected from adult penguins (n = 41). Quantification of compounds was performed by gas chromatography-tandem mass spectrometry. The three Pygoscelis species presented similar contamination profiles, with higher concentrations of dichlorodiphenyltrichloroethane (∑DDT; 1.56–3.82 ng g⁻¹ dw), lighter PCB congeners (∑PCB: 11.81–18.65 ng g⁻¹ dw) and HCB (hexachlorobenzene: 1.65–4.06 ng g⁻¹ dw). Amongst the three penguin species, P. antarcticus had lower and P. papua higher concentrations of most of the compounds identified. We found interspecific differences in POPs accumulation as well as sex differences in POP concentrations. Our data indicate a small but significant positive correlation between body size and the concentrations of some compounds. Despite the overall low concentrations found, this study increases knowledge of the occurrence of POPs in Antarctic penguins, thereby reinforcing concerns that Antarctica, although remote and perceived to be protected, is not free from the impact of anthropogenic pollutants.

To assess organochlorine compound (OC) contamination, its possible sources, and adverse health impacts on giant pandas, we collected soil, bamboo, and panda fecal samples from the habitat and research center of the Qinling panda (Ailuropoda melanoleuca qinlingensis)—the rarest recognized panda subspecies. The polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) concentrations were comparatively low which suggests that moderate sources of OC pollution currently. OC levels were lower in samples from nature reserve than in those collected from pandas held in captivity, and OC levels within the reserve increased between functional areas in the order: core, buffer and experimental. The distribution patterns, and correlation analyses, combined with congener distributions suggested PCBs and OCPs originated from similar sources, were dispersed by similar processes, being transported through atmosphere and characterized by historical residues. Backward trajectory analyses results, and detected DRINs (aldrin, dieldrin, endrin and isodrin) both suggest long-range atmospheric transport of pollution source. PCBs pose potential cancer risk, and PCB 126 was the most notable toxicant as assessed be the high carcinogenic risk index. We provide data for health risk assessment that can guide the identification of priority congeners, and recommend a long-term monitoring plan. This study proposes an approach to ecotoxicological threats whereby giant pandas may be used as sentinel species for other threatened or endangered mammals. By highlighting the risks of long-distance transmission of pollutants, the study emphasizes the importance of transboundary cooperation to safeguard biodiversity.

Mammalian polychlorinated biphenyl (PCB) metabolism has not been systematically explored with nontarget high-resolution mass spectrometry (Nt-HRMS). Here we investigated the importance of the gut microbiome in PCB biotransformation by Nt-HRMS analysis of feces from conventional (CV) and germ-free (GF) adult female mice exposed to a single oral dose of an environmental PCB mixture (6 mg/kg or 30 mg/kg in corn oil). Feces were collected for 24 h after PCB administration, PCB metabolites were extracted from pooled samples, and the extracts were analyzed by Nt-HRMS. Twelve classes of PCB metabolites were detected in the feces from CV mice, including PCB sulfates, hydroxylated PCB sulfates (OH-PCB sulfates), PCB sulfonates, and hydroxylated methyl sulfone PCBs (OH-MeSO₂-PCBs) reported previously. We also observed eight additional PCB metabolite classes that were tentatively identified as hydroxylated PCBs (OH-PCBs), dihydroxylated PCBs (DiOH-PCBs), monomethoxylated dihydroxylated PCBs (MeO-OH-PCBs), methoxylated PCB sulfates (MeO-PCB sulfates), mono-to tetra-hydroxylated PCB quinones ((OH)ₓ-quinones, x = 1–4), and hydroxylated polychlorinated benzofurans (OH-PCDF). Most metabolite classes were also detected in the feces from GF mice, except for MeO-OH-PCBs, OH-MeSO₂-PCBs, and OH-PCDFs. Semi-quantitative analyses demonstrate that relative PCB metabolite levels increased with increasing dose and were higher in CV than GF mice, except for PCB sulfates and MeO-PCB sulfates, which were higher in GF mice. These findings demonstrate that the gut microbiome plays a direct or indirect role in the absorption, distribution, metabolism, or excretion of PCB metabolites, which in turn may affect toxic outcomes following PCB exposure.
