Acute exposure to oil induces age and species-specific transcriptional responses in embryo-larval estuarine fish

Because oil spills frequently occur in coastal regions that serve as spawning habitat, characterizing the effects of oil in estuarine fish carries both economic and environmental importance. There is a breadth of research investigating the effects of crude oil on fish, however few studies have addressed how transcriptional responses to oil change throughout development or how these responses might be conserved across taxa. To investigate these effects, we performed RNA-seq and pathway analysis following oil exposure 1) in a single estuarine species (Cyprinodon variegatus) at three developmental time points (embryos, yolk-sack larvae, free-feeding larvae), and 2) in two ecologically similar species (C. variegatus and Fundulus grandis), immediately post-hatch (yolk-sack stage). Our results indicate that C. variegatus embryos mount a diminished transcriptional response to oil compared to later stages, and that few transcriptional responses are conserved throughout development. Pathway analysis of larval C. variegatus revealed dysregulation of similar biological processes at later larval stages, including alteration of cholesterol biosynthesis pathways, cardiac development processes, and immune functions. Our cross-species comparison showed that F. grandis exhibited a reduced transcriptional response compared to C. variegatus. Pathway analysis revealed that the two species shared similar immune and cardiac responses, however pathways related to cholesterol biosynthesis exhibited a divergent response as they were activated in C. variegatus but inhibited in F. grandis. Our results suggest that examination of larval stages may provide a more sensitive estimate of oil-impacts than examination of embryos, and challenge assumptions that ecologically comparable species respond to oil similarly.

This study evaluates the effects of biological factors of fish and seasonality on Polycyclic Aromatic Hydrocarbon (PAH) accumulation in red mullet (Mullus barbatus) tissue. Specimens were collected monthly with a bottom trawl net in an offshore fishing ground in the Northern and Central Adriatic Sea (Geographical Sub Area 17) throughout 2016.The edible fillets of 380 individuals were analyzed for the concentrations of individual PAH, total PAH, and low, medium and high molecular weight (MW) PAHs. PAH bioaccumulation was related to their physicochemical characteristics (MW, and logarithm of the octanol-water partition coefficient, log Kₒw), some biological parameters of fish (body size, age, sex, reproductive stage and total lipid content), and catch season.The PAH bioaccumulation pattern and the effects of the different factors varied according to PAH MW. The heavier (medium and high MW) PAHs showed higher levels in winter-autumn and in pre-spawners compared with spawners and post-spawners. Our findings suggest that an important detoxification mechanism, albeit limited to the heavier PAHs, acts in the spawning and post-spawning stage. Low MW PAHs appeared to be unaffected by reproductive stage, lipid content and seasonality. Reproductive stage and seasonality seem to play an important role in the accumulation of heavier PAH, whereas total lipid content and age seem to exert a limited influence, and body size no effect at all.

Despite the widespread use of the antiviral drug, Tamiflu®, little is known about the long-term toxic effects of drug or its metabolites in an aquatic ecosystem. This study integrated epidemiological and ecotoxicological methods to determine environmentally relevant concentrations of Tamiflu. A model based on the species medaka (Oryzias latipes) was then used to determine the health status and reproductivity of adults exposed to the drug as well as the embryonic development of offspring. The proposed ecotoxicological model was also used to quantitatively and qualitatively evaluate the toxicodynamic parameters related to egg production, hatchability, and development. Our results revealed that at an environmentally relevant exposure, Tamiflu and its metabolites had no adverse effects on growth, survival, or fecundity of adult medaka. Nonetheless, we observed a reduction in hatchability under exposure to 300 μg L⁻¹ and a reduction in body length under exposure exceeding 90 μg L⁻¹. Under exposure to 300 μg L⁻¹, the estimated spawning time to reach 50% of the maximum percentage of cumulative egg production (ET50) far exceeded that of the control group (without exposure to Tamiflu). We also observed a ∼ 3-fold decrease in maximum egg hatching (Eₘₐₓ). Based on an integrated epidemiological and ecotoxicological model, predictions of environmental concentrations of Tamiflu and its metabolites revealed that the influenza subtypes associated with increases in environmental concentrations: A(H3N2) > A(H1N1) > type B (in order of their effects). We also determined that A(H3N2) posed a potential risk to hatchability and development. Note however, the environmental concentrations of Tamiflu and its metabolites in most countries are lower than the effect concentrations derived in this study, indicating no hazards for aquatic environments. We recommend the use of hatchability and embryonic development as indicators in assessing the effects of long-term parental exposure to Tamiflu metabolites.

European sturgeon Acipenser sturio is an anadromous fish species being classified “critically endangered” with only one remaining population in the Gironde-Garonne-Dordogne basin (France). In the global warming context, this paper aims to determine the sensitivity of A. sturio early life phases to temperature and oxygen saturation. Embryos were experimentally exposed to a combination of temperature (12 to 30 °C) and oxygen (30 to 90% O₂ saturation) conditions. Lethal and sublethal effects were evaluated using embryonic mortality, hatching success, malformation rate, yolk sac resorption, tissue development and swimming speed. Embryonic survival peaked at 20 °C and no survival was recorded at 30 °C regardless of the associated oxygen saturation. No hatching occurred at 50% O₂ sat or below regardless of temperature. Malformation frequency appeared to be minimum at 20 °C and 90% O₂ sat. Swimming speed peaked at 16 °C. The temperature optimum of early life phases of A. sturio was determined to be close to 20 °C. Its upper tolerance limit is between 26 and 30 °C and its lower tolerance limit is below 12 °C. Oxygen depletion induces sublethal effects at 70% O₂ sat and lethal effects at 50% O₂ sat. Within the spawning period in the Gironde-Garonne-Dordogne basin, we identified yearly favourable oxythermal windows. Consequences of climate change would depend of the phenological adaptation of the species for its spawning period.

Endemic fish species of Armenian ichthyofauna—Sevan trout (Salmo ischchan Kessler, 1877)—is registered in the Red Book of Animals of the Republic of Armenia as “Critically Endangered” (corresponds to IUCN category: CR A2cd). Its natural reproduction hardly occurs as a result of numerous problems related to the use and management of water and bio-resources in the Lake Sevan drainage basin. Masrik River is formerly known as a spawning river for two ecological races of the Sevan trout, but there is no up-to-date and in-depth studies of its potential to support natural reproduction. Thus, a set of spawning criteria was arranged as matrix and state of Masrik River as spawning area for the Sevan trout was assessed. The results showed that the potential of different parts of Masrik River to support natural reproduction of Sevan trout varied from quite appropriate to inappropriate. The main limiting factors revealed were water temperature, velocity and substrate conditions.
