Methylmercury exposure alters RNA splicing in human neuroblastoma SK-N-SH cells: Implications from proteomic and post-transcriptional responses

The neurotoxic effects of methylmercury (MeHg) have been intensively studied. However, the molecular mechanisms responsible for the neurotoxicity of MeHg are not fully understood. To decipher these mechanisms, proteomic and high-throughput mRNA sequencing (RNA-seq) technique were utilized, comprehensively evaluating the cellular responses of human neuroblastoma SK-N-SH cells to MeHg exposure. Proteomic results revealed that MeHg exposure interfered with RNA splicing via splicesome, along with the known molecular mechanisms of mercury-related neurotoxicity (e.g. oxidative stress, protein folding, immune system processes, and cytoskeletal organization). The effects of MeHg on RNA splicing were further verified using RNA-seq. Compared to control, a total of 658 aberrant RNA alternative splicing (AS) events were observed after MeHg exposure. Proteomics and RNA-seq results also demonstrated that mercury chloride (HgCl2) influenced the expression levels of several RNA splicing related proteins and 676 AS events compared to control. These results suggested that RNA splicing could be a new molecular mechanism involved in MeHg and HgCl2 neurotoxicity.

Stabilizing cadmium by incorporating it into crystalline products is an effective approach to detoxify cadmium-containing wastes. In this study, two Si-rich matrices in amorphous and crystalline forms (i.e., silica fume and α-quartz, respectively) were employed to incorporate Cd. The processing parameters, namely the type of Si-rich matrix, Cd/Si molar ratio (Г) and sintering temperature, were thoroughly investigated using quantitative X-ray diffraction technique. Cd incorporation was more energetically favored when silica fume was used rather than when α-quartz was used because of the lower Gibbs free energy of formation for silica fume. The sintering temperature and Г values substantially affected the formation of three cadmium silicates, namely monoclinic CdSiO₃, orthorhombic Cd₂SiO₄, and tetragonal Cd₃SiO₅. CdSiO₃ formed only in Г = 1.0 systems. Cd₂SiO₄ was dominant in all reactive systems. In Г = 3.0 systems, Cd₃SiO₅ rather than Cd₂SiO₄ was the predominant Cd-hosting product at temperatures above 850 °C. Leaching test results demonstrated that CdSiO₃ possessed the highest acid resistance among the cadmium silicates. The leachability of Cd₂SiO₄ was very similar to that of Cd₃SiO₅. CdSiO₃ preferred incongruent dissolution, whereas Cd₂SiO₄ and Cd₃SiO₅ favored near-congruent dissolution. This study delineated the feasibility of cadmium incorporation by Si-rich matrices, identifying a promising approach for cadmium detoxification.

Plastic litter in the marine environment is a major global issue. Discarded plastic shotgun ammunition shells and discharged wads are an unwelcome addition and feature among the top ten litter items found on reference beaches in Denmark.To understand this problem, its scale and origins, collections were made by volunteers along Danish coastal shorelines. In all 3669 plastic ammunition items were collected at 68 sites along 44.6 km of shoreline. The collected items were scored for characteristic variables such as gauge and length, shot type, and the legibility of text, the erosion, and the presence of metallic components. Scores for characteristics were related to the site, area, and season and possible influences discussed.The prevalence of collected plastic shotgun litter ranges from zero to 41 items per 100 m with an average of 3.7 items per 100 m. Most ammunition litter on Danish coasts originates from hunting on Danish coastal waterbodies, but a small amount may come from further afield. North Sea coasts are the most distinctive suggesting the possible contribution of long distance drift as well as the likelihood that such litter can persist in marine habitats for decades.The pathway from initial discard to eventual wash-up and collection depends on the physical properties of plastic components, marine tides and currents, coastal topography and shoreline vegetation.Judging from the disintegration of the cartridge and the wear and decomposition of components, we conclude that there is a substantial supply of polluting plastic ammunition materials that has and will accumulate. These plastic items pose a hazard to marine ecosystems and wash up on coasts for many years to come. We recommend that responsible managers, hunters and ammunition manufacturers will take action now to reduce the problem and, thereby, protect ecosystems, wildlife and the sustainability of hunting.

Mercury (Hg) is a pervasive environmental contaminant that can impair avian health, consequently there is a need to gauge exposures. Bird blood provides a measure of recent dietary exposure to Hg, but blood collection and storage can be complex and costly. Dried blood spots (DBS) may help overcome challenges of whole blood analyses, therefore, this study aimed to develop and validate a novel method to assess Hg exposure in birds using DBS. First, accuracy and precision of blood Hg concentrations for entire DBS and DBS punches were determined for white leghorn chicken (Gallus gallus domesticus) dosed with methylmercury (MeHg) via egg injection. Next, we investigated Hg stability in chicken DBS subjected to time, temperature, and humidity treatments. Lastly, we applied the method to DBS created using standard field methods from zebra finch (Taeniopygia guttatato) in the laboratory and American golden-plover (Pluvalis dominica) sampled in the field. All samples were analyzed for total Hg (THg) using direct Hg analysis. Accuracy was determined by comparing DBS concentrations with those of corresponding whole blood and reported as percent recovery. Accuracy for entire chicken DBS was 101.8 ± 5.4%, while DBS punches revealed lower recovery (87.7 ± 4.0 to 92.4 ± 4.1%). There was little effect of time, temperature, and humidity storage treatments on Hg concentrations of DBS, with mean DBS THg concentrations within ±8% of whole blood (n = 10 treatments). For zebra finch, DBS punches were more accurate (93.7 ± 9.7%) compared to entire DBS (126.8 ± 19.4%). While for American golden-plover, entire DBS resulted in the most accurate THg concentrations (111.5 ± 7.6%) compared to DBS punches (edge: 115.4 ± 18.9%, interior: 131.4 ± 16.1%). Overall, results indicate that DBS analysis using direct Hg analysis can accurately evaluate Hg exposure in birds.

Heavy metals deposited on urban road surfaces can be washed-off by stormwater runoff, undermining stormwater reuse safety due to their high toxicity to ecological and human health. Heavy metals on urban road surfaces come from diverse sources and tracking these sources is essential to effectively manage stormwater and hence its reuse safety. This research study developed an innovative approach to tracking sources of heavy metals using data collected in Shenzhen, China. This approach developed was based on a “flag element ratio” theory, where each source generally corresponds to a specific ratio of targeted pollutants to the flag element. It is noted that Cr, Cu, Pb, Ni, and Zn on urban roads were 19.05 mg/kg to 152.01 mg/kg, 25.66 mg/kg to 310.75 mg/kg, 15.61 mg/kg to 220.35 mg/kg, 10.65 mg/kg to 100.28 mg/kg, and 138.14 mg/kg to 1047.05 mg/kg, respectively. Gasoline emission was the main source for Cr, Ni and Pb, while braking wear and tyre wear were the major sources of Cu and Zn, respectively. Furthermore, the rankings of sources of each heavy metal in terms of their contributions were obtained by using this approach. Vehicle exhaust was found as the main contributor for all the heavy metals on urban road surfaces. This highlighted that vehicle exhaust should be seriously considered in terms of controlling heavy metal pollution on urban road surfaces and hence resulting urban road stormwater runoff.

In this paper, results from receptor modelling performed on a well-characterised PM₁ dataset were combined to chemical light extinction data (bₑₓₜ) with the aim of assessing the impact of different PM₁ components and sources on light extinction and visibility at a European polluted urban area. It is noteworthy that, at the state of the art, there are still very few papers estimating the impact of different emission sources on light extinction as we present here, although being among the major environmental challenges at many polluted areas. Following the concept of the well-known IMPROVE algorithm, here a tailored site-specific approach (recently developed by our group) was applied to assess chemical light extinction due to PM₁ components and major sources.PM₁ samples collected separately during daytime and nighttime at the urban area of Milan (Italy) were chemically characterised for elements, major ions, elemental and organic carbon, and levoglucosan. Chemical light extinction was estimated and results showed that at the investigated urban site it is heavily impacted by ammonium nitrate and organic matter. Receptor modelling (i.e. Positive Matrix Factorization, EPA-PMF 5.0) was effective to obtain source apportionment; the most reliable solution was found with 7 factors which were tentatively assigned to nitrates, sulphates, wood burning, traffic, industry, fine dust, and a Pb-rich source. The apportionment of aerosol light extinction (bₑₓₜ,ₐₑᵣ) according to resolved sources showed that considering all samples together nitrate contributed at most (on average 41.6%), followed by sulphate, traffic, and wood burning accounting for 18.3%, 17.8% and 12.4%, respectively.

Ground fine particulate matter (PM2.5) concentrations at high spatial resolution are substantially required for determining the population exposure to PM2.5 over densely populated urban areas. However, most studies for China have generated PM2.5 estimations at a coarse resolution (≥10 km) due to the limitation of satellite aerosol optical depth (AOD) product in spatial resolution. In this study, the 3 km AOD data fused using the Moderate Resolution Imaging Spectroradiometer (MODIS) Collection 6 AOD products were employed to estimate the ground PM2.5 concentrations over the Beijing-Tianjin-Hebei (BTH) region of China from January 2013 to December 2015. An improved geographically and temporally weighted regression (iGTWR) model incorporating seasonal characteristics within the data was developed, which achieved comparable performance to the standard GTWR model for the days with paired PM2.5- AOD samples (Cross-validation (CV) R2 = 0.82) and showed better predictive power for the days without PM2.5- AOD pairs (the R2 increased from 0.24 to 0.46 in CV). Both iGTWR and GTWR (CV R2 = 0.84) significantly outperformed the daily geographically weighted regression model (CV R2 = 0.66). Also, the fused 3 km AODs improved data availability and presented more spatial gradients, thereby enhancing model performance compared with the MODIS original 3/10 km AOD product. As a result, ground PM2.5 concentrations at higher resolution were well represented, allowing, e.g., short-term pollution events and long-term PM2.5 trend to be identified, which, in turn, indicated that concerns about air pollution in the BTH region are justified despite its decreasing trend from 2013 to 2015.

Marine sediments are part of the hydrological cycle and the ultimate storage compartment of land-derived organic matter, including pollutants. Since relevant microbially-driven processes occurring at benthic level may affect the quality of the overall aquatic system, the necessity for incorporating information about microbial communities functioning for ecosystem modelling is arising. The aim of this field study was to explore the links occurring between sediment contamination patterns by three selected class of organic pollutants (Polycyclic Aromatic Hydrocarbons, PAHs, Nonylphenols, NPs, Bisphenol A, BPA) and major microbial properties (Prokaryotic Biomass, PB; total living biomass, C-ATP; Prokaryotic C Production rate, PCP; Community Respiration rate, CR) across a gradient of anthropogenic pollution. Sediments were sampled from 34 sites selected along 700 km of the western coastline of the Adriatic Sea. Organic contamination was moderate (PAHs <830 ng g⁻¹; NPs <350 ng g⁻¹; BPA <38 ng g⁻¹) and decreased southward. The amount of PAHs-associated carbon (C-PAHs) increased significantly with sediment organic carbon (OC), along with microbial functional rates. The negative relation between PCP/CR ratio and OC indicated the shift toward oxidative processes in response to organic pollution and potential toxicity, estimated as Toxic Equivalents (TEQs). Our outcomes showed that sediment organic contamination and benthic microbial processes can be intimately linked, with potential repercussions on CO₂ emission rates and C-cycling within the detritus-based trophic web.

The capacity of pharmaceutical pollution to alter behaviour in wildlife is of increasing environmental concern. A major pathway of these pollutants into the environment is the treatment of livestock with hormonal growth promotants (HGPs), which are highly potent veterinary pharmaceuticals that enter aquatic ecosystems via effluent runoff. Hormonal growth promotants are designed to exert biological effects at low doses, can act on physiological pathways that are evolutionarily conserved across taxa, and have been detected in ecosystems worldwide. However, despite being shown to alter key fitness-related processes (e.g., development, reproduction) in various non-target species, relatively little is known about the potential for HGPs to alter ecologically important behaviours, especially across multiple contexts. Here, we investigated the effects of exposure to a field-realistic level of the androgenic HGP metabolite 17β-trenbolone—an endocrine-disrupting chemical that has repeatedly been detected in freshwater systems—on a suite of ecologically important behaviours in wild-caught female eastern mosquitofish (Gambusia holbrooki). First, we found that 17β-trenbolone-exposed fish were more active and exploratory in a novel environment (i.e., maze arena), while boldness (i.e., refuge use) was not significantly affected. Second, when tested for sociability, exposed fish spent less time in close proximity to a shoal of stimulus (i.e., unexposed) conspecific females and were, again, found to be more active. Third, when assayed for foraging behaviour, exposed fish were faster to reach a foraging zone containing prey items (chironomid larvae), quicker to commence feeding, spent more time foraging, and consumed a greater number of prey items, although the effect of exposure on certain foraging behaviours was dependent on fish size. Taken together, these findings highlight the potential for exposure to sub-lethal levels of veterinary pharmaceuticals to alter sensitive behavioural processes in wildlife across multiple contexts, with potential ecological and evolutionary implications for exposed populations.

Stability of silver sulfide nanoparticle (Ag₂S-NP) in the environment has recently drawn considerable attention since it is associated with environmental risk. Although the overestimated stability of Ag₂S-NP in aqueous solution has already been recognized, studies on transformation of Ag₂S-NP in environmental water are still very scarce. Here we reported that Ag₂S-NP could undergo dissolution by manganese(IV) oxide (MnO₂), an important naturally occurring oxidant in the environment, even in environmental water, although the dissolved silver would probably be adsorbed onto the particles (>0.45 μm) in environmental water, mitigating the measurable levels of dissolved silver. The extent and rate of Ag₂S-NP dissolution rose with the increasing concentration of MnO₂. In addition, environmental factors including natural organic matter, inorganic salts and organic acids could accelerate the Ag₂S-NP dissolution by MnO₂, wherein an increase in dissolution extent was also observed. We further documented that Ag₂S-NP dissolution by MnO₂ was highly dependent on O₂ and it was an oxidative dissolution, with the production of SO₄²⁻. Finally, dissolution of Ag₂S-NP by MnO₂ affected zebra fish (Danio rerio) embryo viability, showing significant reduction in embryo survival and hatching rates, compared to embryos exposed to Ag₂S-NP, MnO₂ or dissolved manganese alone. These findings would further shed light on the stability of Ag₂S-NP in the natural environment - essential for comprehensive nano risk assessment.
