Effectiveness of melatonin to restore fish brain activity in face of permethrin induced toxicity Texto completo

Present study demonstrates permethrin induced oxidative damage in fish brain and explores effectiveness of melatonin to ameliorate brain function. Adult female Notopterus notopterus were exposed to nominal permethrin concentrations at 1/20th (0.34 μg/l) and 1/10th (0.68 μg/l) of LC₅₀ for 15 days. The measured permethrin concentrations using gas chromatography (GC-ECD) were 0.28 μg/l and 0.57 μg/l, respectively. Some fish were sacrificed to collect brain tissue after 15 days of exposure. Remaining fish from both groups were administered exogenous melatonin (50 μg/kg, 100 μg/kg body weight) for 7 days and brain tissues were collected. Brain enzymes, ntioxidant factors, HSP70, HSP90, nuclear factor-kappa binding (NFkB), melatonin receptor (MT1R) proteins were measured. Permethrin treatment significantly (P < 0.05) decreased the levels of glutathione and brain enzymes. Malondialdehyde (MDA), xanthine oxidase (XO), HSPs increased at each concentration of permethrin. However, superoxide dismutase, glutathione s-transferase levels increased at low permethrin concentration followed by sharp decrease at higher concentration. Expression of NFkB and MT1R increased significantly (P < 0.05). Melatonin administration reinstated activity of brain enzymes, reduced MDA, XO levels and modulated HSPs. Melatonin also increased expression of NFkB and MT1R. Exogenous melatonin improves oxidative status in permethrin stressed fish brain. Melatonin modulates expression of HSPs that enables brain to become stress tolerant and survive by initiating NFkB translocation. Melatonin could act through melatonin receptor protein to induce synthesis of antioxidant proteins. Therefore the study successfully evaluates the potential of melatonin application for better culture and management of fish against pesticide toxicity.

Rivers play an important role in the overall transport of microplastic pollution (1 μm to 5 mm), with fluvial dynamics expected to influence biotic interactions, particularly for fish. So far, there have been few assessments of microplastics in freshwater salmonids. The prevalence (i.e. percentage occurrence) and burden (i.e. abundance per fish) of microplastics were assessed in the gastrointestinal tracts (GITs) and stomach contents (SCs) of 58 brown trout Salmo trutta Linnaeus, 1758 sampled at six sites along the River Slaney catchment in south-east Ireland. Sites were divided into two classifications (high and low exposure) based on proximity to microplastic pollution sources, comprising three sites each. Analysis of biological traits (e.g. fish length) and diet was performed on the same fish to determine possible factors explaining microplastic burden. Microplastics were found in 72% of fish having been recovered from 66% of GITs (1.88 ± 1.53 MPs fish⁻¹) and 28% of SCs (1.31 ± 0.48 MPs fish⁻¹). Fibres were the dominant particle type recovered from GITs (67%) and SCs (57%) followed by fragments. No difference in median microplastic burden was observed between fish collected in high and low exposure sites. Microplastic burden was unrelated to fish fork length, while microplastic size distribution (100 ≤ 350 μm, 350 μm to ≤ 5 mm) was unrelated to S. trutta age class estimates. Furthermore, microplastic burden was not explained by dietary intake. Though further research is necessary, this study showed the presence of microplastics in wild S. trutta collected from an Irish riverine system, which could have further implications for top-level consumers that feed on the species, including humans. Further analysis is required to determine possible trophic linkages for the species, with respect to microplastics, and to assess the suitability of S. trutta for monitoring microplastics in river systems.

Although the coexistence of heavy metals and environmental hormones always occur in aquatic environment, the information of the combined impacts remains unclear. To explore the multi-generational toxicity of cadmium (Cd) and tributyltin (TBT), adult zebrafish (Danio rerio) (F0) were exposed to different treated groups (100 ng/l Cd, 100 ng/l TBT and their mixture) for 90 d, with their offspring (F1 and F2) subsequently reared in the same exposure solutions corresponding to their parents. Both developmental neurotoxicity and thyroid disturbances were examined in the three (F0, F1, and F2) generations. Our results showed that co-exposure to Cd and TBT induced the developmental neurotoxicity in F1 and F2 generations, reflected by the significant lower levels of neurotransmitters (dopamine and serotonin) and the inhibited acetylcholinesterase (AChE) activities. And the thyroid endocrine disruption were observed in the two-generations larval offspring by parental exposure to Cd and/or TBT, including the significantly decreasing levels of thyroid hormones and the down-regulated the expression of genes involved in the hypothalamus-pituitary-thyroid axis, compared to the control. Additional, the embryonic toxicity and growth inhibition were also determined in the fish larvae. Overall, this study examined the impacts of parental co-exposure to Cd and TBT, with regard to developmental inhibition, nervous system damage and endocrine disruption, which highlighted that co-exposure influences are complicated and need to be considered for accurate environmental risk assessment.

Compared to marine microplastics research, few studies have bio-monitored microplastics in inland waters. It is also important to understand the microplastics’ uptake and their potential risks to freshwater species. The Australian glass shrimp Paratya australiensis (Family: Atyidae) is commonly found in fresh waterbodies in eastern Australia, and are sensitive to anthropogenic stressors but have a wide tolerance range to the natural environmental conditions. This study aimed to understand the microplastics’ occurrence and types in water samples and the shrimp P. australiensis, and identify if the shrimp could be a suitable bioindicator for microplastic pollution. Surface water and P. australiensis across ten urban and rural freshwater sites in Victoria were sampled. In total, 30 water samples and 100 shrimp were analysed for microplastic content, and shrimp body weights and sizes were also recorded. Microplastics were picked, photographed and identified using FT-IR microscopy: in water samples, 57.9% of items including suspect items were selected to identify; all microplastics found in shrimp samples were identified. Microplastics were present in the surface waters of all sites, with an average abundance of 0.40 ± 0.27 items/L. A total of 36% of shrimp contained microplastics with an average of 0.52 ± 0.55 items/ind (24 ± 31 items/g). Fibre was the most common shape, and blue was the most frequent colour in both water and shrimp samples. The dominant plastic types were polyester in water samples, and rayon in shrimp samples. Even though results from this study show a relatively low concentration of microplastics in water samples in comparison with global studies, it is worth noticing that microplastics were regularly detected in fresh waterbodies in Victoria, Australia. Compared with water samples, shrimp contained a wider variety of plastic types, suggesting they may potentially behave as passive samplers of microplastics pollution in freshwater environments.

Polybrominated diphenyl ethers (PBDEs) are ubiquitous environmental pollutants with adverse effects on the foetus and infants. This study aimed at assessing in utero exposure levels and transplacental transfer (TPT) characteristics of BDE congeners in primiparous mothers from Kampala, the capital city of Uganda. Paired human samples (30 placenta and 30 cord blood samples) were collected between April and June 2018; and analysed for a suite of 24 tri-to deca-BDE congeners. Extraction was carried out using liquid-liquid extraction and sonication for cord blood and placenta samples, respectively. Clean-up was done on a solid phase (SPE) column and analysis was performed using gas chromatography/mass spectrometry (GC/MS). Total (∑) PBDEs were 0.25–30.9 ng/g lipid weight (lw) (median; 7.11 ng/g lw) in placental tissues and 1.65–34.5 ng/g lw (median; 11.9 ng/g lw) in cord blood serum, with a mean difference of 1.26 ng/g lw between the compartments. Statistical analysis showed no significant difference between the levels of PBDEs in cord blood and placenta samples (Wilcoxon signed rank test, p = 0.665), possibly because foetus and neonates have poorly developed systems to metabolise the pollutants from the mothers. BDE-209 was the dominant congener in both matrices (contributed 40.5% and 51.2% to ∑PBDEs in placenta and cord blood, respectively), suggesting recent and on-going maternal exposure to deca-BDE formulation. Non-significant associations were observed between ∑PBDEs in maternal placenta and maternal age, household income, pre-pregnancy body mass index (BMI), and beef/fish consumption. This suggested on-going exposure to PBDEs through multiple sources such as dust from indoor/outdoor environments and, ingestion of other foods. Based on absolute concentrations, the extent of transplacental transport was greater for higher congeners (BDE-209, -206 and −207) than for lower ones (such as BDE-47), suggesting alternative TPT mechanisms besides passive diffusion. More studies with bigger sample sizes are required to confirm these findings.

Tris (2-ethylhexyl) phosphate (TEHP) is one of the most commonly used organophosphorus flame retardant (OPFR) analogues and is commonly detected in surface water and sediments. Limited information is available about the metabolic pathway or metabolite formation related to TEHP in fish. In this study, rare minnows (Gobiocyprisrarus) were exposed to TEHP in static water for 30 d to investigate the bioaccumulation and metabolite distribution in the fish muscle, liver, kidney, gill, GI-tract, ovary and testis. Based on the estimated kᵤₚ,ₚₐᵣₑₙₜ and kd,ₚₐᵣₑₙₜ values, the bioconcentration factors (BCFₚₐᵣₑₙₜ) of TEHP in fish tissues were calculated in the order of kidney > ovary ≈ liver ≈ testis > gill ≈ GI-tract > muscle; this finding was consistent with the results of our previous study on other alkyl-substituted OPFRs. In addition, this study identified the metabolic profiles of TEHP in the liver. TEHP was oxidatively metabolized by the fish to a dealkylated metabolite (di 2-ethylhexyl phosphate; DEHP) and hydroxylated TEHP (OH-TEHP). OH-TEHP further underwent extensive phase II metabolism to yield glucuronic acid conjugates. DEHP was mainly distributed in rare minnow in the following order: liver > GI-tract > kidney ≫ other tissues. However, the metabolite showed lower accumulation potential in fish tissues than TEHP, with metabolite parent concentration factors (MPCFs) for DEHP of less than 0.1 in all the investigated tissues. The BCFₚₐᵣₑₙₜ values of TEHP in various fish tissues were only 9.0 × 10⁻³-7.2 × 10⁻⁴ times its estimated tissue-water partition coefficient (Kₜᵢₛₛᵤₑ₋wₐₜₑᵣ) values based on tissue lipid, protein and water contents, which indicated the significance of biotransformation in reducing the bioaccumulation potential of TEHP in fish. The toxicokinetic data in the present study help in understanding the tissue-specific bioaccumulation and metabolism pathways of TEHP in fish and highlight the importance of toxicology research on TEHP metabolites in aquatic organisms.

Pollution by antimony (Sb) and arsenic (As) in soil can pose a great threat to human health. Straw-returning is widely applied to paddy fields for improving and remediating soil. A pot experiment was conducted to investigate the effect of straw-returning on Sb and As transformation and translocation in a soil–rice system. In this study, Sb and As co-contaminated soil was thoroughly mixed with different proportions (0, 0.5, 1, and 2%) of straw and used for growing rice plants through the entire growing stage in a pot experiment and 4 weeks in a microcosm experiment. The straw application significantly increased Sb and As mobility. The concentrations of total Sb and As in soil-pore water increased after the application of straw in most growing stages. The Sb volatilization in the pot and microcosm experiments was also stimulated by straw application. With the high dose of straw application (2%), the concentration of Sb in brown grain was reduced by 72% compared with the control, but As concentrations increased by around 77%. These findings provide a new perspective in that straw-returning could affect the behavior of both Sb and As in soil and reduce the Sb accumulation in brown grain and some guidance in the use of straw-returning in Sb-contaminated paddy soil.

The strict environmental management has been implemented in Taihu Basin to reduce the surface water contamination; however, the effectiveness of the management actions has not been comprehensively evaluated. In the present study, 364 samples were collected during four campaigns over a span of one year from surface water, municipal wastewater treatment plants (MWWTPs), industrial wastewater treatment plants (IWWTPs), industrial enterprises, and aquaculture in a typical region in the Upper Taihu Basin. Overall concentration, temporal variation and spatial distribution of 16 PAHs in surface water and various pollution sources were evaluated and the potential pollution sources were identified. Results showed that concentrations of individual PAHs in the surface water ranged from less than the limit of quantification (LOQ) to 949 ng L⁻¹, indicating a reduction of PAH contamination level after the implementation of environmental management actions. Influent of MWWTPs and wastewater from industrial enterprises exhibited relatively high ∑PAHs concentrations (mean: 880 ng L⁻¹ and 642 ng L⁻¹, respectively); these samples also exhibited a similar seasonal variation as well as composition of PAH congeners to those found in surface water, and therefore were designated as the main emission sources of PAHs in the studied region. Additional source apportionment using principal component analysis was also conducted to verify the proposed sources and diagnose other pollution sources. The findings provided a thorough understanding of PAH pollution, especially its major emission sources, in a typical region with pollution-intensive industries after the implementation of strict environmental management.

This study investigates the characteristics of PCN emission and removal from two secondary copper metallurgical processes (plants A and B) equipped with different air pollution control devices (APCDs). Different operating conditions and feeding materials result in varying emission factors of PCNs from two plants. The average PCN concentration emitted from plant B (7597 ng Nm⁻³) is significantly higher than that emitted from plant A (32.5 ng Nm⁻³) and those reported in China (5.8–2845 ng Nm⁻³). Similar trend is found for fly ash samples collected from two plants. Low chlorinated homologues (Mono-to Tri-CNs) are the major contributors to total PCNs measured in flue gas, fly ash and slag samples. Combination of semi-dry absorber, activated carbon injection and baghouse is effective for PCN removal in plant A, with the overall removal efficiency of 98%. The overall removal efficiency of PCNs achieved with APCDs equipped in plant B is 90%, however, increases of some homologues as the flue gases passing through baghouse and wet scrubber are found, suggesting the occurrence of memory effect within baghouse and wet scrubber.

High concentration of fluoride (up to 20.9 mg/L) in groundwater with significant variation (p = 5.9E-128) among samples was reported from Birbhum district, an acknowledged fluoride endemic region in India. The groundwater samples (N = 368) were grouped based on their hydrochemical properties and aquifer geology for hydro-geochemical characterization. Friedman’s test showed p < 0.0001 confidence level which indicates that fluoride concentration among geological groups and water groups are independent. Bland-Altman plot was used to study the inter-relationships among the groups through bias value (∂) and limit of agreement (LoA). Among the geological groups, laterites and granite-gneiss groups exhibited statistically significantly difference in fluoride geochemistry; whereas the younger and older alluvium groups displayed similar characteristics. The fluoride concentration was found to be in the order Lateritic > Granite-gneiss > Older alluvium ≥ Younger alluvium. Dissolution of minerals (such as fluorite, biotite) in laterite sheeted basalt, and granite-gneiss is the main source of groundwater fluoride in the region. Fluoride concentration is also influenced by depth of water table. Hydrochemical study indicated that fluoride concentration was higher in Na–HCO₃ than in Ca–SO₄ and Ca–HCO₃ type of groundwater. The fluoride concentration were positively correlated with Na⁺ and pH and negatively correlated with the Ca²⁺ and Mg²⁺ signifying linkage with halite dissolution and calcite, dolomite precipitation. Geostatistical mapping of WQI through empirical bayesian kriging (EBK) with respect to regional optimal guideline value (0.73 mg/L) classified that groundwater in some parts of the district are unfit for drinking purpose. Health survey (N = 1767) based on Dean’s criteria for dental fluorosis indicated presence of slight to moderate dental hazard. Besides, providing baseline data for management of groundwater quality in the study area, the study demonstrated the applicability of Bland-Altman analysis and empirical bayesian kriging (EBK) in delineation and interpolation of fluoride contaminated region.
