PM2.5 is becoming a worldwide environmental problem, which profoundly endangers public health, thus progressively capturing public attention this decade. As a fragile target of PM2.5, the underlying mechanisms of endothelial cell damage are still obscure. According to the previous microarray data and signaling pathway analysis, a new form of cell death termed ferroptosis in the current study is proposed following PM2.5 exposure. In order to verify the vital role of ferroptosis in PM2.5-induced endothelial lesion and further understand the potential mechanism involved, intracellular iron content, ROS release and lipid peroxidation, as well as biomarkers of ferroptosis were detected, respectively. As a result, uptake of particles increases cellular iron content and ROS production. Meanwhile, GSH depletion, and the decrease of GSH-Px and NADPH play significant roles in PM2.5-induced endothelial cell ferroptosis. Moreover, significantly changed expression of TFRC, FTL and FTH1 hinted that dysfunction of iron uptake and storage is a major inducer of ferroptosis. Importantly, index monitored above can be partially rescued by lipid peroxidation inhibitor ferrostatin-1 and iron chelator deferoxamine mesylate, which mediated antiferroptosis activity mainly depends on the restoration of antioxidant activity and iron metabolism. In conclusion, our data basically show that PM2.5 enhances ferroptosis sensitivity with increased ferroptotic events in endothelial cells, in which iron overload, lipid peroxidation and redox imbalance act pivotal roles.

In recent decades, rapid development of industrialization and urbanization caused adverse impact on the aqueous ecology and environment of the Huaihe River basin, China. In this work, three ²¹⁰Pb-dated sediment cores extracted from the middle reach of Huaihe River in Anhui Province, China were analyzed to elucidate the temporal trends and sources of polybrominated diphenyl ethers (PBDEs). Source diagnostics indicated that commercial Deca-BDE, Penta-BDE and Octa-BDE products and debromination of higher brominated BDE compounds were likely the PBDE sources in the Huaihe River. The prevalence of BDE-47 in the sediment cores was attributed to the extensive use of commercial Bromkal 70-5DE and Bromkal DE-71 in the region. BDE-28 was another congener that was prevalent in all sediment samples, suggesting that reductive debromination occurred in the sediments. Dramatic increase of PBDE concentrations in both three cores since the post-1980s could be attributed to the rapid expansion of production of electronic and telecommunication equipment and household usage in China. PBDE temporal trends in core S1 located at rural area mainly reflected the regional and national inputs deriving from long distance atmospheric transport, and the positive correlations between PBDE concentration in core S1 and gross domestic product (GDP) and household appliances production volumes (HPVs) were observed. PBDE inputs at site S3 mainly include the transport of contaminated water and re-suspended fine sediment particles from the upstream site S2, which was located in the industrial area and adjacent to e-waste recycling area. The government efforts to protect the environment and improve the e-waste management resulted in the progressive decrease trends in PBDE concentrations in cores S2 and S3.

Fine particulate matter (PM₂.₅) pollution in Eastern China (EC) has raised concerns due to its adverse effects on air quality, climate, and human health. This study investigated the long-term variation trend in satellite-derived PM₂.₅ concentrations and how it was related to pollutant emissions and meteorological parameters over EC and seven regions of interest (ROIs) during 1998–2016. Over EC, the annual mean PM₂.₅ increased before 2006 due to the enhanced emissions of primary PM₂.₅, NOₓ and SO₂, but decreased with the reduced SO₂ emissions after 2006 evidently in response to China's clean air policies. In addition, results from statistical analyses indicated that in the North China Plain (NCP), Northeast China (NEC), Sichuan Basin (SCB) and Central China (CC) planetary boundary layer height (PBLH) was the dominant meteorological driver for the PM₂.₅ decadal changes, and in the Pearl River Delta (PRD) wind speed is the leading factor. Overall, the variation in meteorological parameters accounted for 48% of the variances in PM₂.₅ concentrations over EC. The population-weighted PM₂.₅ over EC increased from 36.4 μg/m³ in 1998–2004 (P1) to 49.4 μg/m³ in 2005–2010 (P2) then decreased to 46.5 μg/m³ in 2011–2016 (P3). In the NCP and NEC, the percentages of the population living above the World Health Organization (WHO) Interim Target-1 (IT-1, 35 μg/m³) have risen steadily over the past 20 yr, reaching maxima of 97.3% and 78.8% in P3, respectively, but decreases of ∼30% from P2 to P3 were found for the SCB and PRD.

Plastic waste is continuously introduced not only into marine, but also freshwater environments, where it fragments into microplastics. Organisms may be affected by the particles themselves due to ingestion and indirectly via incorporated additives such as plasticizers, since these substances have the ability to leach out of the polymer matrix. Although it has been indicated that the likelihood of additives leaching out into the gut lumen of organisms exposed to microplastics is low, studies distinguishing between the effects of the synthetic polymer itself and incorporated additives of the same polymer are scarce. Since this is obligatory for risk assessment, we analyzed the chronic effects of flexible polyvinylchloride (PVC), a widely used polymer, containing the plasticizer diisononylphthalate (DiNP) on morphology and life history of the freshwater crustacean Daphnia magna and compared these effects with the effects of rigid PVC, lacking DiNP, as well as a glass bead control. After up to 31 days of exposure, rigid PVC and glass beads did not affect body length and relative tail spine length of D. magna, whereas flexible PVC led to an increased body length and a reduced number of offspring. None of the treatments increased the mortality significantly. We were able to show that 2.67μg/L DiNP leached out of the flexible PVC into the surrounding medium using GC-MS. Yet, we were not able to measure leachate inside the gut lumen of D. magna. The effects emerged towards the end of the experiment, due to the time dependent process of leaching. Therefore, the results highlight the relevance of long-term chronic exposure experiments, especially as leaching of additives takes time. Further, our study shows the importance to distinguish between microplastics containing leachable additives and the raw polymer in ecotoxicological testing.

Microcystin-leucine-arginine (MC-LR) can cause male reproductive disorder. However, the underlying mechanism are not yet entirely elucidated. In this study, we aimed to investigated the effects of MC-LR on the integrity of blood-testis barrier (BTB) and the related molecular mechanisms. Both in vivo and in vitro experiments revealed that MC-LR caused disruption of BTB and gap junctions between Sertoli cells respectively, which was paralleled by the alteration of connexin43 (Cx43). Our data demonstrated that MC-LR decreased gap junction intercellular communication (GJIC) and impaired Cx43 expression by activating the phosphatidylinositol 3-kinase/Akt cascades. In addition, a possible protective effect of Icariin (ICA), a flavonoid isolated from Chinese medicinal herb, against MC-LR toxicity was investigated. The ICA prevented the degradation of GJIC and impairment of Cx43 induced by MC-LR via suppressing the Akt pathway. Together, our results confirmed that the expression of Cx43 induced by MC-LR was regulated in vivo and in vitro, which was involved in the destruction of BTB. Additionally, ICA seems to be able to mitigate the MC-LR toxic effects.

Urban and highway surfaces discharge polluted runoff during storm events. To mitigate environmental risks, stormwater retention ponds are commonly constructed to treat the runoff water. This study is the first to quantify the retention of microplastics in the sediments of such ponds. It applied state-of-art FTIR-methods to analyse the composition, size, shape, and mass of microplastics in the range 10–2000 μm. Seven ponds serving four land uses were investigated, and the results are related to catchment characteristics, sediment organic matter content, and hydraulic loading. We have not found a correlation between the microplastics abundance, polymer composition, size distribution and the land use in the catchment, as well as the sediment organic matter content. Both the highest (127,986 items kg⁻¹; 28,732 μg kg⁻¹) and the lowest (1511 items kg⁻¹; 115 μg kg⁻¹) accumulation of microplastics were found in the sediments of ponds serving industrial areas. There was, however, a correlation to the hydraulic loading of the ponds, where the sediments of the highest-loaded ponds held the most microplastics. This study shows that sediments in stormwater retention ponds can trap some of the microplastics and prevent them from being transported downstream. These systems need to be considered when assessing the fate of microplastics from urban and highway areas.

The accumulation of phthalate acid esters (PAEs) in the environment has aroused a global concern. Microbial degradation is the most promising method for removing PAEs from polluted environment. Di-n-butyl phthalate (DBP) is one of the most widely used PAEs. In this study, a highly efficient DBP-degrading strain, Enterobacter sp. DNB-S2 was isolated from Mollisol in northeast China, and the degradation rate of 500 mg L⁻¹ DBP reached 44.10% at 5 °C and 91.08% at 50 °C within 7 days. A new intermediate, n-butyl benzoate BP, was detected, implying a new degradation pathway. The complete genome of the strain DNB-S2 was successfully sequenced to comprehensively understand of the entire DBP catabolic process. Key genes were proposed to be involved in DBP degradation, such as esterases, 3,4-dihydroxybenzoate decarboxylase and catechol 2,3-dioxygenase genes. Intermediate-utilization tests and real-time quantitative polymerase chain reaction (RT-qPCR) validated the proposed DBP catabolic pathway. The aboriginal bacterium DNB-S2 is a promising germplasm for restoring PAE-contaminated Mollisol regions at low temperature. This study provides novel insight into the catabolic mechanisms and abundant gene resources of PAE biodegradation.

The bioaccumulation and biomagnification of 22 major perfluoroalkyl substances (PFAS) were investigated in tissues of polar bears (Ursus maritimus) and their major prey species, the ringed seal (Pusa hispida), from the Scoresby Sound region of East Greenland. In polar bear liver the mean Σ4PFSA (perfluoroalkyl sulfonic acid) concentration (C4, C6, C8 and C10) was 2611 ± 202 ng/g wet weight (ww; 99% perfluorooctane sulfonate (PFOS)) and two orders of magnitude higher than the 20 ± 3 ng/g ww (89% PFOS) concentration in fat. The mean Σ4PFSAs in seal liver was 111 ± 5 ng/g ww (98% PFOS) and three orders of magnitude higher relative to the 0.05 ± 0.01 ng/g ww concentration in blubber (100% perfluorohexane sulfonate). Perfluoro-1-octane sulfonamide (FOSA) was quantifiable in bear (mean 10 ± 1.4 ng/g ww) and seal (mean 0.6 ± 0.1 ng/g ww) liver but not in fat or blubber. The mean Σ13PFCAs (C4–C18; perfluoroalkyl carboxylic acids) in bear liver (924 ± 71 ng/g ww) was much greater than in seal liver (74 ± 6 ng/g ww). In bear fat and seal blubber, the mean Σ13PFCAs were 15 ± 1.9 and 0.9 ± 0.1 ng/g ww, respectively. Longer chain C11 to C14 PFCAs dominated in bear fat and seal blubber (60–80% of Σ13PFCA), whereas shorter-chain C9 to C11 PFCAs dominated in the liver (85–90% of Σ13PFCA). Biomagnification factors (BMFs) were orders of magnitude greater for PFHxS and C9 to C13 PFCAs when based on bear liver to seal blubber rather than bear liver to seal liver, and PFCA (C9 to C13) BMFs decreased with increasing chain length. Seal blubber to bear liver BMFs better reflects the dietary exposure relationship of PFAS between bears and seals.

This study aimed to determine the effects of FeMn modified biochar composite (FMBC) treatment on the pH, redox properties, enzyme activities, and bacterial communities of As-polluted paddy soil. The two utilized FMBCs (FMBC₁ and FMBC₂) exhibited markedly different effects on soil pH, and treatment with biochar (BC) or FMBCs increased the soil redox potential and reduced the content of available As, facilitating the conversion of originally present non-specifically sorbed and specifically bound As forms to residual, amorphous hydrous oxide–bound, and crystalline hydrous oxide–bound ones. In general, the activities of soil enzymes increased after the above treatments, with the exception of that of alkaline phosphatase, which decreased upon supplementation with FMBC₂. Supplementation with BC or FMBCs increased the abundance of Proteobacteria and Firmicutes, decreasing that of Bacteroidetes. Notably, FMBC₁ and FMBC₂ affected soil properties in different ways, although the mechanisms of the corresponding influence were similar. Thus, treatment with BC-based materials changed the distribution of As and the activities of soil enzymes, additionally affecting a variety of other physicochemical soil properties to make it suitable for microbial growth.

The occurrence, level, and distribution of multiclass emerging organic contaminants (EOCs) in fish and mollusks from the Klang River estuary were examined. The targeted EOCs for this assessment were phenolic endocrine disrupting compounds (bisphenol A, 4-OP, and 4-NP), organophosphorous pesticides (quinalphos, chlorpyrifos, and diazinon), estrogenic hormones (E2, E1, and EE2), and pharmaceutically active chemicals (primidone, sulfamethoxazole, dexamethasone, diclofenac, amoxicillin, progesterone, and testosterone). Results from this study showed that the prevalent contamination of the Klang River estuary by EOCs with diclofenac, bisphenol A, progesterone, and amoxicillin were predominantly detected in fish and mollusks. Among the EOCs, diclofenac and progesterone had the highest concentrations in fish and mollusk samples, respectively. The concentrations of diclofenac and progesterone in fish and mollusk samples range from 1.42 ng/g to 10.76 ng/g and from 0.73 ng/g to 9.57 ng/g, respectively. Bisphenol A should also be highlighted because of its significant presence in both fish and mollusks. The concentration of bisphenol A in both matrices range from 0.92 ng/g to 5.79 ng/g. The calculated hazard quotient (HQ) for diclofenac, bisphenol A, and progesterone without consideration to their degradation byproduct were less than one, thus suggesting that the consumption of fish and mollusks from the Klang River estuary will unlikely pose any health risk to consumers on the basis of the current assessment. Nonetheless, this preliminary result is an important finding for pollution studies in Malaysian tropical coastal ecosystems, particularly for organic micropollutant EOCs, and can serve as a baseline database for future reference.
