Pyrene Degradation by Mycobacterium gilvum: Metabolites and Proteins Involved

Polycyclic aromatic hydrocarbons (PAHs) are toxic organic pollutants and omnipresent in the aquatic and terrestrial ecosystems. A high-efficient pyrene-degrading strain CP13 was isolated from activated sludge and identified as Mycobacterium gilvum based on the analysis of 16S rRNA gene sequence. More than 95% of pyrene (50 mg L⁻¹) was removed by CP13 within 7 days under the alkaline condition. Pyrene metabolites, including 4-phenanthrenecarboxylic acid, 4-phenanthrenol, 1-naphthol, and phthalic acid, were detected and characterized by GC-MS. Results suggested that pyrene was initially attacked at positions C-4 and C-5, then followed by ortho cleavage, and further degraded following the phthalate metabolic pathway. Analysis of pyrene-induced proteins showed that the extradiol dioxygenase, a key enzyme involved in pyrene degradation, was highly up-regulated in pH 9 incubation condition, which illustrated the high efficiency of CP13 under alkaline environment. The present study demonstrated that the isolated bacterial strain CP13 is a good candidate for bioremediation of alkaline PAH-contaminated sites.

Removal of petroleum products from contaminated soil is a long-term process requiring attention and constant monitoring. The aim of this study was to determine the effect of Fyre-Zyme enzyme preparation and/or calcium carbonate on microbiological changes and conversion of n-aliphatic hydrocarbons in soil contaminated of petroleum-derived substances. The material for testing was soil contaminated with petroleum substances. The total concentration of n-alkanes with a C8–C40 chain length was 420.1 mg·kg⁻¹ DM. The pot tests were carried out by introducing stimulators. As a decomposition promoter for n-alkanes in contaminated soil, a 6% water solution of Fyre-Zyme and/or 1% sterile CaCO₃ was used. The pots were incubated at 25 °C for 21 days. The dynamics of changes in the microbiota and concentration of n-alkanes were controlled for 21 days, every 7 days taking soil for testing. Microbiological tests included determining the total number of bacteria and fungi. Chemical analysis was performed by chromatographic method. Stimulating of soil bioremediation of contaminated hydrocarbons with calcium carbonate increased the number of bacteria, and stimulation with Fyre-Zyme and calcium carbonate—the number of filamentous fungi. There was no significant correlation between the concentration of n-alkanes in the soil and the total number of bacteria and fungi but stimulating of soil bioremediation with calcium carbonate increased the number of bacteria, and stimulation with Fyre-Zyme and calcium carbonate—the number of filamentous fungi. The observed correlations indicate that the concentration of n-alkanes in the contaminated soil increases with the addition of Fyre-Zyme.

The most commonly used solution in chrome plating bath is chromic acid (hexavalent Cr), and a considerable amount of mists is released into the air and consequently produce hazards to workers. Thus, the aim of this study was to evaluate whether the biomarker of exposure to metals, specially Cr levels, presents associations with hematological and biochemical parameters and if they can alter the activity of enzymes that contain thiol groups such as pyruvate kinase, creatine kinase, adenylate kinase, and δ-aminolevulinate dehydratase. Fifty male chrome plating workers were used for exposed group and 50 male non-exposed workers for control group. For that, biological monitoring was performed through quantification of metals on total blood and urine by inductively coupled plasma mass spectrometry (ICP-MS) and enzyme activity was performed by spectrometry in erythrocytes. In addition, chromium levels in water was quantified and ecotoxicology assay was performed with Allium cepa test. The results demonstrated that blood and urinary chromium levels in exposed group were higher than the control group (p < 0.0001). Furthermore, decreased activity of enzymes was found in those that contain thiol groups from exposed group when compared with the control group (p < 0.001). The water analysis did not present a statistical difference between control and exposed groups (p > 0.05), demonstrating that water did not seem to be the source of contamination. In summary, our findings indicated some toxicology effects observed in the exposed group, such as thiol enzyme inhibition, mainly associated with occupational exposure in chrome plating and besides the presence of other metals, and Cr demonstrated to influence the activity of the enzymes analyzed in this research.

The presence of pathogens in drinking water can seriously affect human health. Therefore, water disinfection is needed, but conventional processes, such as chlorination, result in the production of dangerous disinfection by-products. In this regard, an alternative solution to tackle the problem of bacterial pollution may be the application of advanced oxidation processes. In this work, the inactivation of total coliforms, naturally present in a Colombian surface water by means of UV/H₂O₂, UV/US, and the UV/US/H₂O₂ advanced oxidation processes, was investigated. Under the investigated conditions, complete bacterial inactivation (detection limit equal to 1 CFU 100 mL⁻¹) was found within 5 min of treatment by UV/H₂O₂ and UV/US/H₂O₂ systems. UV/US oxidation process also resulted in total bacterial load elimination, but after 15 min of treatment. Bacterial reactivation after 24 h and 48 h in the dark was measured and no subsequent regrowth was observed. This phenomenon could be attributed to the high oxidation capacity of the evaluated oxidation systems. However, the process resulting in the highest oxidation potential at the lowest operating cost, in terms of energy consumption, was UV/H₂O₂ system. Therefore, UV/H₂O₂ advanced oxidation system can be used for disinfection purposes, enabling drinking water production meeting the requirements of regulated parameters in terms of water quality, without incurring extremely high energy costs. Nonetheless, further researches are required for minimizing the associated electric costs.

Conventional air pollutants (PM₁₀, CO, NOₓ) gradually increased from fall to winter during 2015 in Istanbul. Several air pollution episodes were observed during this period. This study was made in order to determine polycyclic aromatic hydrocarbon (PAH) levels, identify the sources of air pollution, and make toxicity assessment based on Benzo(a)pyrene equivalent concentrations. The sampling took 14 sequential days during winter. High-pressure weather conditions prevailed at the start of the sampling. The conditions were then changed to low-pressure condition towards the end of the sampling. Strong inversion was effective on the onset of the sampling. Strong inversion was effective at the onset of the sampling. A high-volume sampler was used to collect gas and particle phase samples. Total suspended particle concentrations were between 27 and 252 μg m⁻³. Sixteen PAH species were investigated. Total (gas + particle) PAH concentrations were between 76.4 and 1280.3 ng m⁻³, with an average of 301.4 ng m⁻³. Individual PAH concentrations were between not detected (n.d.) and 99.2 ng m⁻³ in the gaseous phase, and between n.d. and 11.5 ng m⁻³ in the particle phase. Phenanthrene had the highest share among 16 PAH compounds. Benzo(a)pyrene was not detected in 8 days. On the remaining days, its concentration ranged between 5.5 and 14.8 ng m⁻³ with an average of 3.7 ng m⁻³. Low-molecular-weight PAHs dominated gaseous phase; inversely, high-molecular-weight PAHs dominated particle phase. Possible sources were identified by diagnostic ratios. These ratios suggested that coal combustion and diesel vehicle exhaust emissions had a substantial impact on ambient air quality. Benzo(a)pyrene equivalencies were calculated for each PAH compound in order to make toxicity assessment. Total benzo(a)pyrene equivalencies ranged between 0.4 and 30.0 ng m⁻³ with an average of 7.2 ng m⁻³.

Dissolved organic matter (DOM) can become a carrier of soil contaminants. Therefore, an understanding of the evolution and characteristics of DOM produced by Chinese milk vetch during green manuring is crucial. In this study, DOM solutions from 28 days’ manuring with three different organic materials were characterized using three-dimensional fluorescence excitation–emission matrix (3D-EEM) with parallel factor (PARAFAC) analysis, and ultraviolet–visible spectroscopy. With the green manuring milk vetch at flowering period (MVFP), the DOC and water-soluble cadmium (WS-Cd) in soil solution reached 1875 mg/l and 2.64 μg/l, respectively, on day 6 after manuring. The PARAFAC analysis modeled three components: protein-like (tryptophan) and two humic-like components (humic acid and fulvic acid); DOM produced by MVFP was primarily protein-like during the early stage of decomposition. The aromaticity and molecular weight of DOM in the MVFP treatment was lower than in the other treatments, which could promote the release of soil particle-adsorbed Cd to soil solution. Principal components analysis showed that aromaticity was the main factor affecting Cd solubility, and the negative linear correlation of aromaticity with WS-Cd reached 0.4827. The results of this study supported the idea that manuring with MVFP might accelerate Cd infiltration to deep soil with water under gravity.

Recent efforts to aggregate, process, and use biodiversity information have appended novel opportunities and challenges for the field, and a rapid increase in studies that integrate and analyze data in the biological-ecological realm. We developed a web-based GIS system for the wildlife of Khouzestan Province that provides potential distribution maps and other spatial and nonspatial data on the wildlife of Khouzestan Province and its protected areas. We used MaxEnt and a fuzzy inference system to model distributions of species. Our application was structured using a client/server architecture, and the database design and construction was carried out using PostgreSQL/PostGIS, and GeoServer to serve maps. The mapping interface was developed using OpenLayers; ASP.NET was selected for designing the user interface. We used qualitative-quantitative methods to develop, design, refine, and finalize our system particularly as regards usability. The design approach resulted in a user-friendly interface that allows both specialists and non-specialists to quickly and efficiently run models to estimate potential distributions of species. Our application highlights what can be accomplished with a biodiversity-oriented web application.

Metropolitan areas may suffer with increase of air pollution due to the growth of urbanization, transportation, and industrial sectors. The Metropolitan Area of Vitória (MAV) in Brazil is facing air pollution problems, especially because of the urbanization of past years and of having many industries inside the metropolitan area. Developing air quality system is crucial to understand the air pollution mechanism over these areas. However, having a good input dataset for applying on photochemical models is hard and requires quite of research. One input file for air quality modeling which can play a key role on results is the lateral boundary conditions (LBC). This study aimed to investigate the influence of LBC over CMAQ simulation for particulate matter and ozone over MAV by applying four different methods as LBC during August 2010. The first scenario (M1) is based on a fixed, time-independent boundary conditions with zero concentrations for all pollutants; the second scenario (M2) used a fixed, time-independent concentration values, with average values from local monitoring stations; the third CMAQ nesting scenario (M3) used the nested boundary conditions varying with time from a previous simulation with CMAQ over a larger modeling domain, centered on MAV; and finally, the fourth GEOS-Chem scenario (M4) used the boundary conditions varying with time from simulations of global model GEOS-Chem. All scenarios runs are based on the same meteorology conditions and pollutant emissions. The air quality simulations were made over a domain 61 × 79 km centered on coordinates − 20.25° S, − 40.28° W with a resolution of 1 km. The results were evaluated with the measured data from the local monitoring stations. Overall, significant differences on concentrations and number of chemical species between the LBC scenarios are shown across all LBC scenarios. The M3 and M4 dynamic LBC scenarios showed the best performances over ozone estimates while M1 and M2 had poor performance. Although no LBC scenarios do not seem to have a great influence on total PM₁₀ and PM₂.₅ concentrations, individual PM₂.₅ species like Na, NO₃⁻, and NH₄⁺concentrations are influenced by the dynamic LBC approach, since those hourly individual PM₂.₅ species from CMAQ nesting approach (M3) and GEOS-Chem model (M4) were used as an input to LBC.

To reduce the environmental pollution caused by ammonium paratungstate (APT) production in the Ganzhou area in China, simulated experiments in laboratory and field experiments in cement kilns were performed. The migration characteristics of As in secondary residues (thermometallurgy and hydrometallurgy residues) from APT production in cement kilns were similar, and As in the residues existed in the form of sulfides. When the residues were fed at the kiln inlet, the As in the residues was completely distributed in the clinker after a new mass balance of As was reestablished in a very short time. When the residues were fed at the raw mill, the total input rate of As was far higher than the total output rate. Therefore, a part of As was circulated in the cement kiln, and only a small part of As was distributed in the clinker. In addition, the As concentration in the flue gas and the leaching concentration of As in the clinker were far below the limit value in the Chinese standard. For feeding rates below that are used in the field experiment, co-processing of secondary residues in a cement kiln fed at the kiln inlet is environmentally safe. However, if the secondary residues are consistently fed at the raw mill, the As concentration in the flue gas may gradually increase.

A pot experiment was performed to examine the role of foliar applied mannitol (M) in chromium (Cr) stress alleviation in different maize cultivars. Two maize cultivars, one tolerant (6103) and one sensitive (9108) to chromium stress, were grown in soil treated with three concentrations of Cr (0, 5, and 10 mg kg⁻¹) and three levels of mannitol (0, 50, and 100 mg L⁻¹). Chromium stress decreased the overall growth of plants by reducing the plant height, root/shoot dry weight, chlorophyll contents, and enzymatic activities, while exacerbated the severity of reactive oxygen species in both maize cultivars. Chromium-induced reduction in growth attributes of maize plants was relatively higher in sensitive cultivar than that of tolerant one. Uptake of Cr by the plants and its translocation from roots to shoots increased with increasing concentration in the soil. However, foliar application of mannitol significantly alleviated the Cr stress and improved growth, biomass, and photosynthetic pigments of maize plants. Mannitol also considerably reduced Cr contents in leaves and roots of both cultivars. Hence, it is concluded that mannitol can be helpful for crops grown on heavy metal, especially Cr, contaminated soils for remediation purpose.
