Effectiveness of melatonin to restore fish brain activity in face of permethrin induced toxicity

Present study demonstrates permethrin induced oxidative damage in fish brain and explores effectiveness of melatonin to ameliorate brain function. Adult female Notopterus notopterus were exposed to nominal permethrin concentrations at 1/20th (0.34 μg/l) and 1/10th (0.68 μg/l) of LC₅₀ for 15 days. The measured permethrin concentrations using gas chromatography (GC-ECD) were 0.28 μg/l and 0.57 μg/l, respectively. Some fish were sacrificed to collect brain tissue after 15 days of exposure. Remaining fish from both groups were administered exogenous melatonin (50 μg/kg, 100 μg/kg body weight) for 7 days and brain tissues were collected. Brain enzymes, ntioxidant factors, HSP70, HSP90, nuclear factor-kappa binding (NFkB), melatonin receptor (MT1R) proteins were measured. Permethrin treatment significantly (P < 0.05) decreased the levels of glutathione and brain enzymes. Malondialdehyde (MDA), xanthine oxidase (XO), HSPs increased at each concentration of permethrin. However, superoxide dismutase, glutathione s-transferase levels increased at low permethrin concentration followed by sharp decrease at higher concentration. Expression of NFkB and MT1R increased significantly (P < 0.05). Melatonin administration reinstated activity of brain enzymes, reduced MDA, XO levels and modulated HSPs. Melatonin also increased expression of NFkB and MT1R. Exogenous melatonin improves oxidative status in permethrin stressed fish brain. Melatonin modulates expression of HSPs that enables brain to become stress tolerant and survive by initiating NFkB translocation. Melatonin could act through melatonin receptor protein to induce synthesis of antioxidant proteins. Therefore the study successfully evaluates the potential of melatonin application for better culture and management of fish against pesticide toxicity.

Pore water is a crucial storage medium and a key source of sediment phosphorus. A novel equipment based on electrokinetic geosynthetics (EKGs) was used for isolating phosphorus from eutrophic lake sediments through pore water drainage. Three mutually independent indoor group experiments (A, B, and C) were conducted to investigate the effects of voltage gradient (0.00, 0.25, and 0.50 V/cm) on pore water drainage capacity, phosphorus removal performance, sediment physicochemical properties, and phosphorus storage dynamics. The average reduction in the sediment moisture and total phosphorus content was 2.5%, 4.3%, and 4.6% and 28.15, 75.95, and 112.65 mg/kg after 6 days of treatment for A, B and C, respectively. Efficient pore water drainage through gravity and electroosmotic flow and electromigration of phosphate were the main drivers of sediment-dissolved and mobilized phosphorus separation. A high voltage gradient facilitated the migration of pore water and the phosphorus in it. The maximal effluent total phosphorous (TP) concentration was up to 27.9 times that in the initial pore water samples, and negligible effluent TP was detected when the pore water pH was less than 2.5. The TP concentration was exponentially and linearly related to the pH and electronic conductivity of the electroosmotic flow, respectively. The migration of H⁺ within the sediment matrix promoted the liberation of metals bounded to phosphorus, particularly of Ca–P and Fe–P. Pore water drainage through an EKG resulted in Ex–P separation of up to 87.50% and a 13.84 mg/kg decrease in Ca–P and 125.35 mg/kg accumulation of low mobile Fe–P in the weak acid anode zone.

Goethite is a common iron hydroxide, which can be substituted by manganese (Mn) in the goethite structure. It is important to investigate the immobilization of uranium(VI) on Mn-substituted goethite (Mn-Goe) to understand the fate and migration of uranium in soils and sediments. In this study, the sorption of uranium(VI) by Mn-Goe was investigated as a function of pH, adsorbent dosage, contact time, and initial uranium concentration in batch experiments. Several material analysis techniques were used to characterize manganese substituted materials. Results indicated that Mn was successfully introduced into the goethite structure, the length of particles increased gradually, the surface clearly exhibited higher roughness with increasing Mn content, and that uranium(VI) sorption of synthetic Mn-Goe appeared to be higher than that of goethite. The sorption kinetics supported the results presented by the pseudo-second-order model. The sorption capacity of uranium on Mn-Goe was circa 77 mg g⁻¹ at pH = 4.0 and 25 °C. Fourier transform-infrared spectroscopy (FT-IR) analyses revealed that uranium ions were adsorbed through functional groups containing oxygen on the Mn-Goe structure. The enhancement of Mn-substitution for the uranium(VI) sorption capacity of goethite was revealed. This study suggests that goethite and Mn-Goe can both play a significant role in controlling the mobility and transport of uranium(VI) in the subsurface environment, which is helpful for material development in environmental remediation.

Due to high global demand, large amounts of abbattoir waste are generated from pork production. Mismanagement of abattoir waste on agricultural lands can result in soil and water contamination with pathogens and contaminants like metals and nutrients. Therefore, possible effects on soil organisms prior to application should be evaluated. Thus, the aim of this study was to determine the effects of fresh pig abattoir waste (PAWf) and waste after stabilization processes on E. andrei through tests of avoidance behavior, acute toxicity and chronic toxicity. In order to do this, the waste was evaluated fresh (i.e., non-treated), and after aerated composting (PAWa), natural composting (PAWn) and vermicomposting (PAWv). In addition, we used a natural soil with no history of agricultural use as control soil. The evaluation was based on avoidance behavior, mortality, initial and final earthworm weight, and reproduction, in addition to a set of enzyme assays formed by acetylcholinesterase, lipid peroxidation, catalase and glutathione S-transferase measured over time. The ecotoxicological results showed that PAWf and PAWa increased AChE activity at different experimental periods, while PAWn decreased activity at 14 days compared to the control. PAWf and PAWa increased TBARS levels at 7 and 14 days, respectively. CAT activity decreased at 3, 7 and 14 days in PAWv, while GST activity increased at 3 days in PAWa and at 3 and 14 days in PAWf compared to the control. In the acute toxicity test, PAWa and PAWn had a toxic effect on E. andrei, resulting in 100% mortality at 14 days of exposure. Based on our findings, pig abattoir waste should undergo vermicomposting prior to agricultural application to soils.

The impacts of microplastic particulates in benthic freshwater organisms have been largely unexplored despite abundant plastic accumulation in the sediments of these systems. We investigated the uptake of plastic particles by benthic filter feeding quagga mussels (Dreissena bugensis) and associated toxicity exhibited through impacts on mortality, filtration rate, reproduction and oxygen consumption. Matrix Assisted Laser Desorption/Ionization Imaging Mass Spectrometry (MALDI-IMS) technology was used to assess the microplastic inclusion. For this purpose, quagga mussels were exposed to four treatments ranging from 0.0 to 0.8 g/L of a high density fluorescent red polyethylene powder in the size range of 10–45 μm for 24-h, and the targeted endpoints were quantified. Identification of several micrograms of microplastics in the digestive tract suggests rapid clearance from the water column by filtering. At the higher concentrations, about 95% of the microplastics ingested remained in the mussels after 24-h. Microplastics were found in the gills which correlated with decreasing filtration rate at higher microplastic concentrations. Despite large-scale ingestion, plastic exposure did not affect survivorship, reproduction rates, or oxygen consumption in the period examined. MALDI-IMS identified unique mass spectra that correlated with microplastic inclusion. This research suggests that microplastics can impair feeding through decreased filtration rates of filter feeding organisms, potentially resulting in a reduction of overall fitness over time and that MALDI-IMS may have the potential to identify microplastics and changes in tissue at the borders of plastic inclusion.

Natural organic matter (NOM) affects the environmental behaviors of AgNPs, which may change their phytotoxicity to plants. However, more evidence can be provided to illustrate how NOM influences AgNPs-induced phytotoxicity. In this study, using rice (Oryza sativa) as a model, the effects of NOM, Suwannee River humic acid (SRHA) and fulvic acid (FA), on the dissolution and phytotoxicity of AgNPs were investigated. Silver ions decreased in both AgNPs and AgNO₃ solution in the presence of NOM, and the effect of SRHA was stronger than FA. Image-XRF (iXRF) results showed that Ag mainly remained in the root rather than the shoot of rice seedling exposed to AgNPs. NOM mitigated the negative effects of AgNPs and AgNO₃ on rice with lower germination inhibition rate, less chlorophyll reduction, more relative biomass and less O₂•⁻ content. Moreover, NOM improved root cell viability according to FDA fluorescent dye as well as maintained the normal root morphology. Interestingly, the neutral sugars content from pectin, hemicellulose 1, hemicellulose 2 and cellulose of root cell wall in AgNPs and AgNO₃ treatments differed from the control, while it was close to the regular content in AgNPs/AgNO₃+SRHA/FA groups, which implied that NOM regulated the changes. Besides, SRHA led to less germination and less relative biomass than FA due to different chemical characters. Thus, NOM needs to be considered when studying the phytotoxicity of AgNPs.

Ingestion of microplastics by marine organisms has been well documented, but their interaction with chemical pollutants has not been sufficiently addressed. The aim of this study was to determine the individual and combined effects of chlorpyrifos (CPF) and polyethylene microplastics (MP) on the survival, fecundity, feeding and egg viability of Acartia tonsa, a calanoid copepod widely distributed in planktonic communities. The median lethal concentration obtained for CPF was higher (LC₅₀ = 1.34 μg/L) than for the combination with MP (LC₅₀ = 0.37 μg/L), or CPF-loaded MP (LC₅₀ = 0.26 μg/L). Significant effects were also observed for feeding and egg production (EC₅₀ = 0.77 and 1.07 μg/L for CPF, 0.03 and 0.05 μg/L for CPF combined with MP, 0.18 and 0.20 μg/L for CPF-loaded MP). No significant effects were observed in the exposure to ‘virgin’ MP. This study confirms the role of MP as vectors of pollutants to marine organisms and supports the increased availability of certain toxicants carried out by MP. The effects observed in fitness-related responses suggest potential damage to A. tonsa populations. The comparison of the results obtained here with environmental concentrations indicates that the combined exposure to CPF and MP could constitute a risk to A. tonsa in the natural environment.

Biochar sequesters cadmium (Cd) by immobilisation, but the process is often less effective in field trials than in the laboratory. Therefore, the involvement of soil components should be considered for predicting field conditions that could potentially improve this process. Here, we used biochar derived from Spartina alterniflora as the amendment for Cd-contaminated soil. In simulation trials, a mixture of kaolin, a representative soil model component, and S. alterniflora-derived biochar immobilised Cd by forming silicon-aluminium-Cd-containing complexes. Interestingly, the biochar recalcitrance index value increased from 48% to 53%–56% because of the formation of physical barriers consisting of kaolinite minerals and Cd complexes. Pot trials were performed using Brassica chinensis for evaluating the effect of S. alterniflora-derived biochar on plant growth in Cd-contaminated soil. The bio-concentration factor values in B. chinensis were 24%–31% after soil remediation with biochar than in control plants. In summary, these results indicated that soil minerals facilitated Cd sequestration by biochar, which reduced Cd bioavailability and improved the recalcitrance of this soil amendment. Thus, mechanisms for effective Cd remediation should include biochar-soil interactions.

The increasing prevalence and spread of antibiotic resistance genes (ARGs) in intensive aquaculture environments are of great concern to food safety and public health. However, the level of ARGs and their potential propagation factors in an industrial recirculating aquaculture system (RAS) have not previously been comprehensive explored. In this study, the levels of 14 different ARG markers and 2 kinds of mobile genetic elements (MGEs) were investigated in a RAS (including water, fish, feces, pellet feed meal, and biofilm samples) located northern China. qnrA, qnrB, qnrS, qepA, aac(6′)-Ib, and floR were dominant ARGs, which average concentration levels were presented at 4.51–7.74 copies/L and 5.36–13.07 copies/g, respectively, suggesting that ARGs were prevalent in RAS with no recorded history of antibiotic use. Elevated level of ARGs was found in water of RAS even after the final UV treatment compared with its influent. In RAS, Proteobacteria, Verrucomicrobia, Bacteroidetes, and Planctomycetes were the predominant phyla. Notably, elevated levels of potential opportunistic pathogens were observed along with abundant ARGs suggesting an increasing risk of capturing ARGs and MGEs for human pathogens. This study has revealed for the first time that reared fish, their feces, pellet feed meal as the introduction sources and the selection roles of treatment units co-driven the ARG profile, and the co-selection of water environmental factors and their consequently induced bacterial community shifts formed by their influence are the determining drivers for the ARG propagation in RAS.

Short-term exposure to air pollution has pro-thrombotic effects and triggers thrombo-embolic events such as myocardial infarction or stroke in adults. This study evaluates the association between short-term variation in air pollution and treatments for acute thrombo-embolic events among the whole Belgian population. In a bidirectional time-stratified case-crossover design, we included 227,861 events treated with endovascular intervention and 74,942 with antithrombotic enzymes that were reimbursed by the Belgian Social Security between January 1st, 2009 and December 31st, 2013. We compared the concentrations of particulate matter (PM) air pollution (PM₁₀ and PM₂.₅), as estimated at the municipality level on the day of the event (lag 0) and two days earlier (lag 1 and lag 2) with those of control days from the same month, matched by temperature and accounting for day of the week (weekend vs week days). We applied conditional logistic regression models to obtain odds ratios (OR) and their 95% CI for an increase of 10 μg/m³ (PM₁₀) or 5 μg/m³ (PM₂.₅) in pollutant concentrations over three lag days (lag 0, 1 and 2). We observed significant associations of PM₁₀ and PM₂.₅ with treatment of acute thrombo-embolic events at the three lags. The strongest associations were observed for air pollution concentrations on the day of the event (lag0). Increases of 10 μg/m³ PM₁₀ and 5 μg/m³ PM₂.₅ on lag0 increased the odds of events treated with endovascular intervention by 2.7% (95%CI:2.3%–3.2%) and 1.3% (95%CI:1%–1.5%), respectively, and they increased the odds of events treated with antithrombotic enzymes by 1.9% (95%CI:1.1–2.7%) and 1.2% (95%CI:0.7%–1.6%), respectively. The associations were generally stronger during autumn months and among children. Our nationwide study confirms that acute exposure to outdoor air pollutants such as PM₁₀ or PM₂.₅ increase the use of medication and interventions to treat thrombo-embolic events.
