The contribution of detoxification pathways to pyrethroid resistance in Hyalella azteca Texto completo

Chronic exposure to pyrethroid insecticides can result in strong selective pressures on non-target species in aquatic systems and drive the evolution of resistance and population-level changes. Characterizing the underlying mechanisms of resistance is essential to better understanding the potential consequences of contaminant-driven microevolution. The current study found that multiple mechanisms enhance the overall tolerance of Hyalella azteca to the pyrethroid permethrin. In H. azteca containing mutations in the voltage-gated sodium channel (VGSC), both adaptation and acclimation played a role in mitigating the adverse effects of pyrethroid exposures. Pyrethroid resistance is primarily attributed to the heritable mutation at a single locus of the VGSC, resulting in reduced target-site sensitivity. However, additional pyrethroid tolerance was conferred through enhanced enzyme-mediated detoxification. Cytochrome P450 monooxygenases (CYP450) and general esterases (GE) significantly contributed to the detoxification of permethrin in H. azteca. Over time, VGSC mutated H. azteca retained most of their pyrethroid resistance, though there was some increased sensitivity from parent to offspring when reared in the absence of pyrethroid exposure. Permethrin median lethal concentrations (LC50s) declined from 1809 ng/L in parent (P₀) individuals to 1123 ng/L in the first filial (F₁) generation, and this reduction in tolerance was likely related to alterations in acclimation mechanisms, rather than changes to target-site sensitivity. Enzyme bioassays indicated decreased CYP450 and GE activity from P₀ to F₁, whereas the VGSC mutation was retained. The permethrin LC50s in resistant H. azteca were still two orders-of-magnitude higher than non-resistant populations indicating that the largest proportion of resistance was maintained through the inherited VGSC mutation. Thus, the noted variation in tolerance in H. azteca is likely associated with inducible traits controlling enzyme pathways. A better understanding of the mechanistic and genomic basis of acclimation is necessary to more accurately predict the ecological and evolutionary consequences of contaminant-driven change in H. azteca.

Pollution-tolerant lichens are recognized ecological indicators of air pollution in cities, which can also collect airborne anthropogenic particles in their tissues. Harmful (sub)micron-sized magnetites are a ubiquitous component of air particle pollution, adversely impacting human health. In this work, in situ magnetic susceptibility κᵢₛ of well-characterized ultrafine magnetite and lichen thalli were measured to quantify the amount of airborne magnetic particles (AMP) after calibration and to assess the lichen's decontamination over time. Up to 2850 magnetic measurements were carried out in twenty-nine transplanted lichens (collected in urban and clean areas) from winter 2020 to winter 2021. Before the transplants, their initial κᵢₛ values were 0.23–9.45 × 10⁻⁵ SI, representing AMP contents of 0.1–4.6 mg in lichen thalli. After lichens were transplanted to a shared site, the magnetic signals evidenced short-term increases and long-term decreases. After three, five and nine months, the AMP loss is more pronounced for transplanted lichens from polluted (e.g., AMP_5-months loss = 0.59 mg) than clean (= 0.08 mg) sites. Rainfall influenced the lichen's decontamination between seasons. In situ measurements and lichens offer a valued and time-saving methodology for biomonitoring harmful airborne particles simply and effectively.

China has been famous for its porcelains for millennia, and the combustion processes of porcelain production emit substantial amounts of air pollutants, which have not been well understood. This study provided firsthand data of air pollutant emissions from biomass porcelain kilns. The emission factor of PM₂.₅ was 0.95 ± 1.23 g/kg during the entire combustion cycle, lower than that of biomass burning in residential stoves and coal burning in brick kilns, attributed to the removal effects of the long-distance transport in dragon kilns. The temporal trend of particle pollutants, including particulate matters (PMs) and particulate polycyclic aromatic hydrocarbons (PAHs) (low at ignition phase and high at the end) again indicated the removal effects of the special structure, while gaseous pollutants, such as gaseous PAHs, exhibited the opposite result. The GWC₁₀₀ was estimated as 1.4 × 10⁶ and 0.5 × 10⁶ kg CO₂e/yr for the scenarios in which 50% and 100% of the wood was renewable, respectively. The GWC₁₀₀ of dragon kilns is nearly equal to that of 745 households using wood-fueled stoves. These results indicate the necessity of pollution controls for biomass porcelain kilns to estimate the emission inventory and climate change.

Karst ecosystems make an important contribution to the global carbon cycle, in which carbon-fixing microorganisms play a vital role. However, the healthy functioning of karst ecosystems is threatened because pollutants easily diffuse and spread through them due to their strong hydraulic connectivity. The microbiome of a karst river contaminated with antibiotics was studied. Through co-occurrence network analysis, six ecological clusters (MOD 1–MOD 6) with different distribution characteristics were determined, of which four were significantly correlated with antibiotics. The carbon fixation pathways in different ecological clusters were varied, and the dominant hydroxypropionate-hydroxybutyrate cycle and reductive acetyl-CoA pathway were negatively and positively correlated with antibiotics, respectively. Long-term antibiotic contamination altered the selection of carbonic anhydrase (CA) encoding genes in some of the CA-producing mineralization microorganisms. The selection of different carbon fixation pathways is a possible strategy for the microbial community to compensate for the adaptation costs associated with the pressure of antibiotics contamination and emergence of antibiotics resistance. Bayesian network analysis revealed that some carbon sequestration functions (such as β-CA and reductive acetyl-CoA pathway) surpassed certain antibiotic resistance genes in the regulation of environmental factors and microbial networks. An ecological cluster (MOD5) that possibly homologous to antibiotic contamination was the final node of the microbial community in karst river, which indicated that ecological clusters were not only selected by antibiotics, but were also regulated by multiple environmental factors in the karst river system. The carbon sequestration pathway was more directly reflected in the abundance of ecological groups than in the influence of CA. This study provides new insights into the feedback effect of karst system on typical pollutants generated from human activities.

The occurrence and spatial distribution of polybrominated diphenyl ethers (PBDEs) and novel brominated flame retardants (NBFRs) in seawater and surficial sediment samples (N = 19 and 45, respectively) from the South China Sea (SCS) in 2018 were investigated, and the correlation between BFRs and site parameters (total organic carbon, depth, etc.) were assessed by principal component analysis. The concentration ranges of ΣPBDEs in seawater and sediments were 0.90–4.40 ng/L and 0.52–22.67 ng/g dry weight (dw), respectively, while those of ΣNBFRs were 0.49–37.42 ng/L and 0.78–82.29 ng/g dw, respectively. BDE-209 and decabromodiphenyl ethane were the predominant BFRs, accounting for 38.65% and 36.94% in seawater and 26.71% and 68.42% in sediments, respectively. Notably, tris(2,3-dibromopropyl)isocyanurate and 2,4,6-tris(2,4,6-tribromophenoxy)-1,3,5-triazine, seldomly detected in aquatic matrices worldwide, were detected for the first time in the study area, and their relatively high levels and detection frequencies indicate the ubiquitous application of these NBFRs in the Pearl River Delta. Zhuhai and Jiangmen are the main sources of NBFRs in the SCS. Preliminary risk assessment on NBFRs using hazard quotient indicates low to medium risks to marine organisms at some sites. The occurrence of NBFRs in the SCS highlights the prioritization of more toxicological information on these compounds.

Oil spills are a serious environmental problem. To better support risk assessment and pollution control for oil spills, a good understanding of oil transport in the environment is required. This study focused on the numerical simulation of the nearshore oil behaviors based on computational fluid dynamics. Based on the Navier-Stokes momentum equations for an incompressible viscous fluid and volume of fluid (VOF) method, a 3D numerical model of three-phase transient flow was developed. The wave number, averaged flow velocity and oil properties would affect the oil spread extent and the oil volume fraction. The higher the averaged flow velocity and wave number, the lower the oil concentration, and the faster the horizontal movement of the oil. The spilled oil may move to contact the seafloor by increasing the averaged flow velocity at the inlet boundary. Through increasing the wave number, the oil would stay near the water surface. In the nearshore, where the wave is the main seawater motion, the oil containment boom should be set preferentially to the direction of wave transmission for oil cleaning. This study shows that by doubling the wave number and increasing the averaged flow velocity (ten times) at the same time, the maximum oil volume fraction would be reduced by around 32%. Finally, the water temperature had no significant impact on the oil migration, and the impact of evaporation should be considered in the simulation.

Plastic pollution is an emerging problem and is a consequence of the post-consumer plastic waste accumulation in the environment coupled to mismanaged waste programmes. Countries are counteracting the continuous growth of plastic litter with different strategies: introducing bans and limits on both plastic items and materials, promoting plastic recycling and recovery strategies and encouraging voluntary clean up actions, as well as raising public awareness. However, the toxicity of plastics to the environment and organisms is not only related to their polymer chains, but also to the fact that plastic materials contain hazardous additives and can adsorb environmental pollutants (i.e. heavy metals and persistent organic contaminants, respectively). The plastic/additives/pollutants combination may be ingested by marine organisms and then enter in the food chain. Therefore, legislation for additives and contaminants is crucial both to reduce environmental pollution and their toxic effects on organisms, which of course includes humans. In this review, the current policies on plastics and related contaminants are described focusing on current laws. Moreover, recommendations for seafood consumption are suggested, since each fish or mollusc eaten may potentially result in plastic particles, additives or contaminants ingestion.

Cadmium as a common environmental stressor may exert highly toxic effects on herbivorous insects. The question was whether possible elevation of an oxidative stress and imbalance of energetic reserves in insects may depend on developmental stage, sex and insect population’s multigenerational history of exposure to cadmium. So, the aim of this study was to compare of the development traits, total antioxidant capacity, lipid peroxidation, RSSR to RSH ratio and the concentration of carbohydrates, glycogen, lipids and proteins in whole individuals (larvae or pupae) of Spodoptera exigua originating from two strains: control and selected over 120 generations with sublethal metal concentration (44 Cd mg per dry weight of diet). Generally, the increase of the protein, carbohydrates, glycogen concentration and lipid peroxidation decrease with age of the larvae were found. Revealed cases of a higher mobilisation of carbohydrates and proteins, and changes in total antioxidant capacity or lipid peroxidation, in individuals being under metal exposure, occurred in strain-depended mode. Short-term Cd exposure effect was connected with possible higher engagement of proteins and glycogen in detoxification processes, but also higher concentration of lipid peroxidation. In turn, for long-term Cd exposure effect lower lipids concentration and higher thiols usage seemed to be more specific.

Liming is a safe and effective remediation practice for Cd contaminated acid paddy soil. The fate of Cd can also be strongly influenced by redox chemistry of sulfur. But it is unclear if, to what extent and how the combination of liming and sulfur mediation could further control Cd uptake by paddy rice. A rice cultivation pot experiment was conducted to evaluate the impact of different sulfur forms (S⁰ and SO₄²⁻ in K₂SO₄) on the solubility, uptake and accumulation of Cd in the soil-paddy rice system and how liming and reducing organic carbon mediate the process. Results showed that under neutral soil circumstances achieved by liming, co-application of K₂SO₄ and glucose significantly reduced brown rice Cd by 33%, compared to liming alone. They made it more readily for Cd²⁺ to be precipitated into CdS/CdS₂ or co-precipitate with newly formed FeS/FeS₂/iron oxides. The higher pH balancing capability of K₂SO₄ as well as liming kept the newly formed sulfide or iron containing minerals negatively charged to be more prone to adsorb Cd²⁺, that kept the porewater Cd²⁺ the lowest among all the treatments. Individual K₂SO₄ showed significant promoting effect on soil Cd solubility due to SO₄²⁻ chelation effect. Furthermore, K₂SO₄ had much weaker inhibiting effect on Cd translocation from root to grain, it showed no significant attenuating effect on brown rice Cd. S⁰ containing treatments displayed weaker or no attenuating effect on brown rice Cd due to its strong soil acidification effect. On the basis of liming, organic carbon induced sulfur (K₂SO₄) mediation showed great application potential for safe production on large areas of acid paddy soil contaminated by Cd.

Previous studies have developed a stable weather index (SWI) based on meteorological elements that adequately represent PM₂.₅ pollution over the North China Plain (NCP). However, the SWI performs poorly over the Yangtze River Delta (YRD) region because air pollution over this region is affected not only by stagnant weather (STAG) but also by transport (TRANS). For example, air pollutants can be transported from the NCP to the YRD by cold fronts. In this study, an obliquely rotated principal component analysis in the T-model is applied to classify the synoptic patterns of winter weather over the YRD region from 2013 to 2018. Among the four identified synoptic patterns, two of which cause TRANS, one pattern is most likely to cause STAG, and one pattern can lead to either STAG or TRANS depending on the location of high pressure around Shandong province. Due to the large contribution (63%) of TRANS to the total PM₂.₅ pollution events, a transport pollution index (TPI) is constructed to describe the transport features of PM₂.₅ pollution over the YRD region. Our results show that, when considering the SWI alone, the correlation coefficients between the SWI and ln(PM₂.₅) range from 0.50 to 0.57 in the main cities of the YRD. Excitingly, when considering both the TPI and SWI (TPI+SWI), the correlation coefficients increase significantly to 0.63–0.78, suggesting that TPI+SWI better reflects the wintertime PM₂.₅ pollution level over the YRD region. In addition, satisfactory performance in validation also suggests that TPI+SWI can increase the accuracy of evaluating and forecasting of PM₂.₅ pollution episodes over regions downstream of source emissions.
