Silica nanoparticles alleviate cadmium toxicity in rice cells: Mechanisms and size effects

Although it was recently determined that silicon can alleviate cadmium (Cd) toxicity in rice, the effects of silicon properties and the molecular mechanisms are still unclear. Here, the effect of silica nanoparticles (SiNPs) on Cd toxicity in rice was examined using cells cultured in suspension in the presence or absence of SiNPs (19 nm, 48 nm and 202 nm). The results showed that the presence of SiNPs substantially enhanced the proportion of live cells to 95.4%, 78.6% and 66.2%, respectively, suggesting that the extent of alleviation of Cd toxicity decreased gradually with size of SiNPs. The morphological results showed that dramatic damage and severe structural changes in the organelle integrity of cells occurred in the absence of SiNPs, whereas the cells exposed to the SiNPs remained nearly intact even in the presence of high concentrations of Cd. Furthermore, the SiNPs accumulated on the surface of the rice cells. Using inductively coupled plasma mass spectroscopy, Cd accumulated preferentially in plant cells with cell walls. In addition, noninvasive microtest technology showed that the average Cd2+ influx in those treated with SiNPs (19 nm, 48 nm and 202 nm) decreased by 15.7-, 11.1- and 4.6-fold, respectively. The gene expression of Cd uptake and transport (OsLCT1 and OsNramp5) was inhibited by SiNPs, but the gene expression of Cd transport into vacuole (OsHMA3) and Si uptake (OsLsi1) was enhanced by the SiNPs. These results indicate that the presence of SiNPs increased at least 1.87-fold the Si uptake capacity and inhibited the Cd uptake capacity, which together resulted in the alleviation of the toxicity of Cd in rice. This study provided a molecular-scale insight into the understanding of the SiNPs-induced alleviation of the toxicity of Cd in rice.

Opioid drugs, such as morphine (MO), detected in aquatic environments worldwide, may harm fish due to their semi-persistence and ability to potently interact with molecular targets conserved across vertebrates. Here, we established a waterborne bacterial lipopolysaccharide (LPS) challenge assay with zebrafish embryos as a model to investigate chemically-induced disruption of the innate immune system, and used it to study the effects of MO exposure. Exposure to 1 mg/L MO resulted in pronounced immunosuppression, reflected in downregulation of several inflammation-related genes, including myd88, trif, traf6, p38, nfκb2, il-1β, il-8 and ccl34a. Fish exposed to 1 mg/L MO accumulated 11.7 ng/g (wet weight) of MO, a concentration comparable to that reported in blood of chronic drug abusers subject to higher infection rates. Surprisingly, exposure to lower MO concentrations (100 ng/L–100 μg/L) led to exacerbation of LPS-induced inflammation. Two ATP-binding cassette (ABC) transporters known to be involved in the xenobiotic efflux - abcb4 and abcc2, also known as multixenobiotic resistance (MXR) transporters - were downregulated at 100 ng/L MO. We hypothesized that ABC/MXR transporters could modulate the severity of inflammation by being involved in efflux of LPS, thus regulating its accumulation in the organism. Indeed, we could demonstrate that blocking of ABC/MXR transporters by an inhibitor, cyclosporine A, results in stronger inflammation, coinciding with higher LPS accumulation, as visualized with fluorescently labeled LPS. Our work demonstrates that MO can disrupt fish innate immune responses at environmentally relevant concentrations. We also provide evidence for a role of ABC/MXR transporters in LPS efflux in fish. These finding may be applicable across other taxa, as ABC transporters are evolutionary conserved. Since diverse environmentally present chemicals are known to interfere with ABC/MXR transporters' expression or activity, our discovery raises concerns about potential adverse effects of such compounds on the immune system responses in aquatic organisms.

Microwave irradiation (MW) is an effective technique in heating and pyrolysis. This study compared the properties of peanut shell-biochars produced using MW and muffle furnace (FN). At the same pyrolysis temperature, MW biochars preserved more biomass (as indicated by their higher yields and higher abundance of functional groups) and possessed larger surface areas due to the high abundance of micropores. MW biochars generally exhibited higher adsorption of carbamazepine (CBZ) and bisphenol A (BPA) than FN biochars. However, their surface area-normalized sorption was lower, suggesting that the inner pores may not be fully available to CBZ and BPA sorption. We observed significant free radical signals in both types of biochars. Although CBZ and BPA did not degrade in the biochar sorption systems, the potential role of stronger free radical signals in MW biochars for organic contaminant control may not be overlooked in studies with other chemicals.

The objective of this study was to investigate associations between cardiovascular effects and urban ambient particle constituents using an in vivo crossover experimental design. Ambient particles were introduced to an exposure chamber for whole-body exposure of WKY rats, where the particulate matter with an aerodynamic diameter of <2.5 μm (PM2.5) mass concentration, particle number concentration, and black carbon (BC) were monitored. Organic carbon (OC), elemental carbon (EC), and soluble ions of PM2.5 were determined. In a crossover design, rats were exposed to ambient particles or high-efficiency particle arrestance (HEPA)-filtered control air for 7 days following a 7-day washout interval. The crossover exposure between particles and HEPA-filtered air was repeated 4 times. Radiotelemetric data on blood pressure (BP) [systolic BP (SBP), diastolic BP (DBP), pulse pressure (PP), and mean arterial pressure (MAP)], heart rate (HR), and heart rate viability (HRV) were subsequently obtained during the entire study. Exposure to the PM2.5 mass concentration was associated with decreases in the SBP, DBP, MAP, and HR (p < 0.05), whereas no significant changes in the BP or HR occurred with the particle number or black carbon. For HRV, the ln 5-min standard deviation of the normal-to-normal (NN) interval (LnSDNN) and the ln root mean square of successive differences in adjacent NN intervals (LnRMSSD) were positively associated with the PM2.5 mass concentration (p < 0.05). There were no significant effects of the particle number concentration or BC on HRV. Alterations in the HR were associated with OC, EC, Na⁺, Cl⁻, and NO3⁻. Cl⁻ was associated with the DBP, MAP, HR, SDNN, and RMSSD. NO3⁻ was correlated with the SBP, MAP, HR, SDNN, and RMSSD. In conclusion, we observed cardiovascular responses to ambient particles in vivo using a crossover design which can reduce animal use in future environmental studies.

Thallium (Tl) is a highly toxic rare element. Severe Tl poisoning can cause neurological brain damage or even death. The present study was designed to investigate contents of Tl and other associated heavy metals in arable soils and twelve common vegetables cultivated around a steel plant in South China, a newly-found initiator of Tl pollution. Potential health risks of these metals to exposed population via consumption of vegetables were examined by calculating hazard quotients (HQ). The soils showed a significant contamination with Tl at a mean concentration of 1.34 mg/kg. The Tl levels in most vegetables (such as leaf lettuce, chard and pak choy) surpassed the maximum permissible level (0.5 mg/kg) according to the environmental quality standards for food in Germany. Vegetables like leaf lettuce, chard, pak choy, romaine lettuce and Indian beans all exhibited bioconcentration factors (BCF) and transfer factors (TF) for Tl higher than 1, indicating a hyperaccumulation of Tl in these plants. Although the elevated Tl levels in the vegetables at present will not immediately pose significant non-carcinogenic health risks to residents, it highlights the necessity of a permanent monitoring of Tl contamination in the steel-making areas.

Samples of lichens, snow and particulate matter (PM10, 24 h) are used for the source identification of air pollution in the heavily industrialized region of Ostrava, Upper Silesia, Czech Republic. An integrated approach that uses different environmental samples for metal concentration and Pb isotope analyses was applied. The broad range of isotope ratios in the samples indicates a combination of different pollution sources, the strongest among them being the metallurgical industry, bituminous coal combustion and traffic. Snow samples are proven as the most relevant indicator for tracing metal(loid)s and recent local contamination in the atmosphere. Lichens can be successfully used as tracers of the long-term activity of local and remote sources of contamination. The combination of PM10 with snow can provide very useful information for evaluation of current pollution sources.

A nationwide emission estimate of perfluoroalkyl substances (PFASs) from wastewater treatment plants (WWTPs) is required to understand the source–receptor relationship of PFASs and to manage major types of WWTPs. In this study, the concentrations of 13 PFASs (8 perfluorocarboxylic acids, 3 perfluoroalkane sulfonates, and 2 intermediates) in wastewater and sludge from 81 WWTPs in South Korea were collected. The emission pathways of PFASs were redefined, and then the national emission of PFASs from WWTPs was rigorously updated. In addition to the direct calculations, Monte Carlo simulations were also used to calculate the likely range of PFAS emissions. The total (Σ13PFAS) emission (wastewater + sludge) calculated from the direct calculation with mean concentrations was 4.03 ton/y. The emissions of perfluorooctanoic acid (PFOA, 1.19 ton/y) and perfluorooctane sulfonate (PFOS, 1.01 ton/y) were dominant. The Monte Carlo simulations suggested that the realistic national emission of Σ13PFASs is between 2 ton/y and 20 ton/y. Combined WWTPs treating municipal wastewater from residential and commercial areas were identified as a major emission source, contributing 65% to the total PFAS emissions. The Han and Nakdong Rivers were the primary contaminated rivers, receiving 89% of the total PFAS discharge from WWTPs. The results and methodologies in this study can be useful to establish a management policy for PFASs.

Hexachlorobenzene (HCB) and pentachlorobenzene (PeCBz) have been listed as unintentional POPs in the annex of the Stockholm Convention and thus, attracted attention by government and researchers. Since the intentional production and use has ceased in most countries, the unintentional releases to the environment have increased. This study gathered 206 and 78 emission factors (EFs) of unintentional HCB and PeCBz from scientific publications and governmental reports, respectively. Most of the EFs referred to the release vector “air” (EFAir) and to a less extent to “product” (EFProduct). EFs were proposed for different source categories/classes used in the Toolkit according to the technologies that released the HCB or PeCBz. Overall, lowest and highest EFAir for HCB were found in the metallurgical industry range from 1 μg/t in well controlled plants (coke, iron and steel) up to 40,000 μg/t (secondary zinc). EFs for PeCBz were in similar order of magnitude. Due to lack of data, EFs to water, land or residue cannot be proposed. Using linear regression and statistical analysis such as Pearson correlation, we found strongest correlation of EFAir between HCB and PeCBz (R2 = 0.79, P < 0.01) and weaker, but still significant, correlations for EFAir between PCDD/PCDFTEQ and HCB (R2 = 0.56; P < 0.01) or PeCBz (R2 = 0.31 P < 0.01) for various thermal processes.

Arsenic (As) in soils is of major environmental concern due to its ubiquity and carcinogenicity. Pteris vittata (Chinese brake fern) is the first known As-hyperaccumulator, which is highly efficient in extracting As from soils and translocating it to the fronds, making it possible to be used for phytoremediation of As-contaminated soils. In addition, P. vittata has served as a model plant to study As metabolisms in plants. Based on the recent advances, we reviewed the mechanisms of efficient As solubilization and transformation in rhizosphere soils of P. vittata and effective As uptake, translocation and detoxification in P. vittata. We also provided future research perspectives to further improve As hyperaccumulation by P. vittata.

Radiocesium (137Cs) migration from headwater forested areas to downstream rivers has been investigated in many studies since the Fukushima Dai-ichi Nuclear Power Plant (FDNPP) accident, which was triggered by a catastrophic earthquake and tsunami on 11 March 2011. The accident resulted in the release of a huge amount of radioactivity and its subsequent deposition in the environment. A large part of the radiocesium released has been shown to remain in the forest. The dissolved 137Cs concentration and its temporal dynamics in river water, stream water, and groundwater have been reported, but reports of dissolved 137Cs concentration in soil water remain sparse.In this study, soil water was sampled, and the dissolved 137Cs concentrations were measured at five locations with different land-use types (mature/young cedar forest, broadleaf forest, meadow land, and pasture land) in Yamakiya District, located 35 km northwest of FDNPP from July 2011 to October 2012. Soil water samples were collected by suction lysimeters installed at three different depths at each site. Dissolved 137Cs concentrations were analyzed using a germanium gamma ray detector. The dissolved 137Cs concentrations in soil water were high, with a maximum value of 2.5 Bq/L in July 2011, and declined to less than 0.32 Bq/L by 2012. The declining trend of dissolved 137Cs concentrations in soil water was fitted to a two-component exponential model. The rate of decline in dissolved 137Cs concentrations in soil water (k1) showed a good correlation with the radiocesium interception potential (RIP) of topsoil (0–5 cm) at the same site. Accounting for the difference of 137Cs deposition density, we found that normalized dissolved 137Cs concentrations of soil water in forest (mature/young cedar forest and broadleaf forest) were higher than those in grassland (meadow land and pasture land).
