Mechanisms and uncertainties of Zn supply on regulating rice Cd uptake

Application of Zinc (Zn) is considered an effective measure to reduce Cadmium (Cd) uptake and toxicity in Cd-contaminated soils for many plant species. However, interaction between Zn and Cd in rice plant is complex and uncertain. In this study, four indica rice cultivars were selected to evaluate the effect of Zn exposure in an EGTA-buffered nutrient solution under varying Zn activities and a field level of Cd activity to characterize the interaction between Zn and Cd in rice. Severe depression in shoots’ biomass, tiller number, and SPAD (Soil and Plant Analyzer Development) value were found at both Zn deficiency and Zn phytotoxicity levels among four tested rice cultivars. There existed a strong antagonism interaction between Zn and Cd in both shoot and root from Zn deficiency to Zn phytotoxicity. The reduction of Cd accumulation in roots and shoots could be explained by the competition between Zn and Cd as well as the dilution effect of increasing biomass. The conflicting effect of Zn supply on Cd uptake may be attributed to the increasing transfer ratio of Cd from root to shoot with the increasing Zn²⁺ activities and the strong depression of Fe and Mn in shoots with the increasing Zn²⁺ activities as well as the variation of genotypes. Balance between Zn and Cd should be considered in field application.

The use of an integrative molecular approach can actively improve the evaluation of environmental health status and impact of chemicals, providing the knowledge to develop sentinel tools that can be integrated in risk assessment studies, since gene and protein expressions represent the first response barriers to anthropogenic stress.This work aimed to determine the mechanisms of toxic action of a widely applied fungicide formulation (chlorothalonil), following a time series approach and using a soil model arthropod, Folsomia candida. To link effects at different levels of biological organization, data were collected on reproduction, gene expression and protein levels, in a time series during exposure to a natural soil.Results showed a mechanistic mode of action for chlorothalonil, affecting pathways of detoxification and excretion, immune response, cellular respiration, protein metabolism and oxidative stress defense, causing irregular cell signaling (JNK and NOD ½ pathways), DNA damage and abnormal cell proliferation, leading to impairment in developmental features such as molting cycle and reproduction. The omics datasets presented highly significant positive correlations between the gene expression levels at a certain time-point and the corresponding protein products 2–3 days later. The integrated omics in this study has provided useful insights into pesticide mechanisms of toxicity, evidencing the relevance of such analyses in toxicological studies, and highlighting the importance of considering a time-series when integrating these datasets.

Antibiotic resistance among gram-negative bacteria is increasingly becoming a problem of global concern. Particularly problematic is the emergence of resistance to last-resort antibiotics such as carbapenems and colistin. The increasing number of reports on the plasmid-mediated colistin resistance gene mcr-1 in isolates worldwide is raising concerns for the future usefulness of this class of antibiotics. Dissemination of mcr-1 is believed to have originated mainly from animal breeding, however, the role of the environment as a transmission source is not yet fully understood. In the current study, 89 extended-spectrum β-lactamase-producing Escherichia coli isolated from 231 samples from different environmental sources in 12 villages in a rural area of Shandong, China, were screened for mcr-1. 17 (19.1%) mcr-1-positive isolates were found from different environmental sources, aggregated in 6 villages. Plasmids of three different Inc-groups carrying mcr-1 were confirmed, indicating that the widespread geographical distribution of mcr-1 in the local area is due to a number of different plasmids. Additionally, almost a third (29.4%) of the isolates carried virulence factors associated to intestinal pathogenic E. coli. These results illustrate the high complexity of the transmission patterns of mcr-1 among different environmental matrices on a local scale and the potential for the environment to facilitate dissemination and emergence of antibiotic-resistant and virulent strains of bacteria.

In the Kebili region of southern Tunisia, there is increasing demand of water from the Lower Cretaceous Continental Intercalaire (CI) aquifer and the Upper Cretaceous-Miocene Complex Terminal (CT) aquifer. The CI aquifer, given limited low recharge of water and increasing amounts of water extraction, has suffered intense overexploitation since the year 2000. Currently, the sustainability of CI resources is threatened by oil and brine contamination detected at a number of water wells in the Kebili region.Hydrocarbon pollution of the aquifers seems to be ubiquitous because the groundwaters sampled in El Fedjej and Nefzaoua basins exhibit bad water quality according to a number of toxicity indices. Geochemical data indicate that, on a regional scale, groundwater quality, salinity, and dissolved element concentrations are best correlated to petroleum contamination-extraction and to aquifer vulnerability to human perturbations rather than to multiple interactions within the hydrogeological system of the region.The analyses of petroleum compounds in sampled waters indicate that these waters are most consistent with increasing concentrations of organic pollutants; the organic matter is crude and unaltered, testifying to continuous flows of contaminants. The brine contamination, however, is limited to the Djemna water well where the water exhibits a salinity of 20 g L⁻¹. Combined, these findings suggest that groundwater composition in CI water wells in Kebili field is influenced by the migration of hydrocarbons and brine-enriched waters through fractures, and (or) by aquifer decompression.

Polycyclic aromatic hydrocarbons (PAHs) and aliphatic hydrocarbons (AHs), including petroleum biomarkers, were studied in four sediment cores collected around Deception and Penguin Islands, Antarctica. Total PAHs in Deception Island (DCP) samples ranged from 2.0 to 26.8 ng g⁻¹, and in Penguin Island (PGI) varied between 13.2 and 60.3 ng g⁻¹. Multiple sources of PAHs were verified in DCP, with petrogenic-derived compounds being predominant over the last 10 years. In PGI, PAHs related to natural contributions from the erosion of coal deposits were reported. Total AHs in DCP ranged from 4.5 to 19 μg g⁻¹ and in PGI varied between 5.3 and 21.9 μg g⁻¹. In DCP, the n-alkanes distribution pattern showed the presence of petroleum residues in the top sections and both terpanes and hopanes were detected, related to the use of fossil fuels for power generation and in different types of vessels. In PGI, the main source of n-alkanes was marine inputs and only terpanes were detected. The slight increase in hydrocarbon levels observed from 1980 onward in DCP was assumed to be due to the development of tourism in the region and to the scientific station activities. In PGI, anthropogenic-related hydrocarbons were detected in the recent sections and were linked to the development of tourism near the island, scientific activities and the increase in vessel traffic. In general, the concentrations of hydrocarbons found around both islands were comparable to those found in uncontaminated Antarctic regions.

Ecosystem services are present everywhere, green vegetation coverage (or green areas) is one of the primary sources of ecosystem services considering urban areas sustainability and peoples urban life quality. Urban vegetation cover loss decreases the capacity of nature to provision ecosystem services; the loss of urban vegetation is also observed within the Amazon. This study aims at identifying urban vegetation loss and relate it to the provision of ecosystem services of reduction of air quality, reduction of air pollution, and climate regulation. Urban vegetation coverage loss was calculated using NDVI on LANDSAT 5 imagery over a 23-year period from 1986 to 2009. NDVI thresholds were arbitrarily selected, and complemented by in locus observation, to establish guidelines for quantitative (area) and qualitative (density) evolution of green cover, divided in six different categories, named as water, bare soil, poor vegetation, moderate vegetation, dense vegetation and very dense vegetation. Data on air pollution, noise pollution and temperature were outsourced from previous works. Measurement show a significant loss of very dense, dense and moderate vegetation coverage and an increase in poor vegetation and bare soil areas, in accordance to increase in air and noise pollution, and local temperature, and provides positive refashions between the loss of urban green coverage and decrease in ecosystem services.

The response of sediment bacterial communities in subtropical freshwater benthic microcosms to sediment-associated triclosan (TCS; 28 d exposure) was analysed using Illumina high-throughput sequencing. This study highlights the interactive effects of TCS and the presence of benthic macroinvertebrates (Limnodrilus hoffmeisteri and Viviparidae bellamya) on sediment bacterial communities. Our results show that TCS alone significantly altered the taxonomic composition and decreased alpha diversity of sediment bacterial communities at concentrations ≥80 μg TCS/g dry weight (dw) sediment (sed). Regarding dominant phyla, TCS significantly reduced the relative abundance of Bacteroidetes and Firmicutes at these concentrations, whereas the relative abundance of Chloroflexi and Cyanobacteria increased. In the presence of benthic macroinvertebrates, the sediment bacterial community was affected by 8 μg TCS/g dw sed as well. However, the presence of benthic macroinvertebrates did not cause measurable changes to bacterial community in unspiked (i.e., control) sediment. These results indicate that TCS alone would not alter the sediment bacterial community at environmentally relevant concentrations (up till 8 μg/g dw sed), but may have an effect in combination with the presence of benthic macroinvertebrates. Therefore, we recommend to include benthic macroinvertebrates when assessing the response of sediment bacterial communities during exposure to environmental stress such as organic contaminants.

Although fertilization plays an important role in determining the contents of soil dissolved organic matters or water-extractable organic matter (DOM, WEOM), knowledge regarding the dynamics, biodegradability, and microbial community shifts of WEOM in response to different fertilization treatments is very limited, particularly in rice–wheat cropping soil. Thus, in the present study, we performed biodegradation experiments using WEOM extracted from samples of soil that had been subjected to four different fertilization treatments: unfertilized control (CK), chemical fertilizer (CF), 50% chemical fertilizer plus pig manure (PMCF), and 100% chemical fertilizer plus rice straw (SRCF). UV spectrum and fluorescence 3D excitation–emission matrix analyses applied to investigate the chemical composition of WEOM revealed that all examined WEOMs were derived from microbial activity and the dominant portion comprised humic acid-like compounds. After the incubation, 31.17, 31.63, 43.47, and 33.01% of soil WEOM from CK, CF, PMCF, and SRCF treatments, respectively, were biodegraded. PMCF- derived WEOM had the highest biodegradation rate. High-throughput sequencing analyses performed to determine the microbial community before and after the incubation indicated that Sphingomonas, Bacillus, and Flavisolibacter were the predominant bacterial genera in the original inoculum derived from the four fertilization treatments. Following biodegradation, we observed that the dominant bacteria differed according to fertilization treatments: Curvibacter (43.25%) and Sphingobium (10.47%) for CK, Curvibacter (29.68%) and Caulobacter (20.00%) for CF, Azospirillum (23.68%) and Caulobacter (13.29%) for PMCF, and Ralstonia (51.75%) for SRCF. Canonical correspondence analysis revealed that, shifts in the microbial community were closely correlated with pH and specific UV absorbance at 254 nm. We speculated that the inherent traits of different WEOM and the properties of soil solutions under different fertilization treatments shaped the soil microbial community structure, thereby influencing the biodegradation of WEOM.

In early nineteen century, a gas field was operational in southern part of Sindh, Pakistan for power production. The plant was completely un-operational for last three decades, whereas all wastage and raw materials are still dumped there, which might be the source to contaminate the ground water. The most of the workers population still living in different villages nearby the gas field. In present study, evaluated the undesirable effects of the toxic metals (lead and cadmium) via consuming groundwater for drinking and other domestic purpose especially in children of ≤5.0 years. For comparative purpose groundwater of nonindustrial area (nonexposed) was also analysed and their impact on age matched children was carried out. Biological samples (scalp hair and blood) were collected from children of exposed and nonexposed areas. The Cd and Pb in scalp hair and blood samples were carried out by graphite furnace atomic absorption spectrometry. Whereas, Cd and Pb in groundwater obtained from both areas were determined prior to applied preconcentration method as reported in our previous works. The Cd and Pb contents in the groundwater of villages of exposed area were found in the range of 5.18–10.9 and 19.9–69.5 μg/L, respectively. Whereas, the groundwater of nonexposed area contains Cd and Pb in the range of 1.79–3.78 and 5.07–24.3 μg/L, respectively. It was observed that the concentrations of Cd and Pb in scalp hair and blood samples of children belongs to exposed area have ≥2.0 fold higher than the resulted data attained for age matched control children, indicating as the exposure biomarkers of toxic metals. The children belong to exposed area have poor health, anemic and low body mass index (<13 kg/m2). A significant positive correlations among Cd and Pb concentrations in biological samples of exposed subjects and groundwater was observed (p < 0.01).
