Subcellular metal distribution in two deep-sea mollusks: Insight of metal adaptation and detoxification near hydrothermal vents

In this study, we determined the concentrations of Cu, Zn, Ni, Cd, Pb and As and their subcellular distributions within the tissues of mussels (Bathymodiolus marisindicus) and snails (Gigantopelta aegis) from two hydrothermal vent regions, i.e., Tiancheng and Longqi, at Southwest Indian Ridge. Mussels collected from the two venting regions showed comparable concentrations for Ni and Pb, but Cu, Zn, Cd and As concentrations were significantly different in mussel gills between the two vent regions. Similar ranges of metal concentrations were found in the snails as those in the mussels, but most of the metals were mainly accumulated in the viscera, except for Ni. Similar subcellular partitioning of Cu, Zn and Cd was documented in different mussel tissues, with cellular debris (50%) being the predominant fraction, followed by equivalent values in other fractions. Lead was distributed in both cellular debris and metal-rich granules (MRG) fraction, whereas Ni was predominantly distributed in MRG (90%). Arsenic was mainly partitioned in cellular debris and metallothionein-like protein. However, deep-sea snails displayed elevated subcellular partitioning of Cu in the organelles (up to 60%) and may be more susceptible to Cu stress than the mussels. Our results demonstrated the metal-specificity of detoxification strategies in these deep-sea hydrothermal vent mollusks, and the mussels may be more adaptable to high metal exposures than the snails at hydrothermal vent.

Lindane, a lipophilic pollutant, may be toxic to organisms. To explore the toxic effects of lindane and the underlying mechanisms of this toxicity, the animal model Caenorhabditis elegans (C. elegans) was exposed to lindane for 3 d at environmentally relevant concentrations (0.01–100 ng/L) and the physiological, biochemical, and molecular indices were evaluated. Subacute exposure to 10–100 ng/L of lindane caused adverse physiological effects on the development, reproduction, and locomotion behaviors in C. elegans. Exposure to 1–100 ng/L of lindane increased the accumulation of Nile red and blue food dye, which suggested high permeability of the intestine in nematodes. Lindane exposure also significantly influenced the expression of genes related to intestinal development (e.g., mtm-6 and opt-2). Moreover, reactive oxygen species production, lipofuscin accumulation, and expression of oxidation resistance genes (e.g., sod-5 and isp-1) were significantly increased in C. elegans exposed to 10–100 ng/L of lindane, which indicated that lindane exposure induced oxidative stress. According to Pearson correlation analyses, oxidative stress and intestinal damage were significantly correlated with the adverse physiological effects of lindane. Therefore, the adverse effects of lindane may have been induced by intestinal damage and oxidative stress, and mtm-6, opt-2, sod-5, isp-1, and mev-1 might play important roles in the toxicity of lindane.

Significant efforts have been directed towards addressing the adverse health effects of atmospheric particles, emphasizing the relevance of indoor exposure. Homes represent an indoor environment where human spend the majority of their time. Thus, the objective of this work was to concurrently assess different matrix of indoor particles considering both mass (PM₁₀, PM₂.₅) and number (N₂₀₋₁₀₀₀) concentrations in indoor and outdoor air of homes (n = 65). Real-time measurements (PM₁₀, PM₂.₅, UFP) were conducted simultaneously during 48 h in dwellings situated in Oporto, Portugal. In 75% of homes, indoor PM₂.₅ (mean = 53 μg m⁻³) exceeded limit of 25 μg m⁻³, for PM₁₀ (mean = 57 μg m⁻³) 41% of homes demonstrated average levels higher than 50 μg m⁻³, thus indicating potential risks. Indoor PM₁₀ was mostly (82–99%) composed of PM₂.₅, both PM were highly correlated (|rs|>0.9655), thus suggesting the similar origin. Indoor PM originated from infiltrations of outdoor emissions; ∼70% of homes exhibited indoor to outdoor (I/O) ratio < 1. On the contrary, UFP indoors (mean = 13.3 × 10³ # cm⁻³) were higher than outdoors (mean = 10.0 × 10³ # cm⁻³). Indoor UFP spatially varied as follows: kitchens > living rooms > bedrooms. UFP indoors were poorly correlated (|rs| = 0.456) with outdoor concentrations, I/O ratios showed that indoor UFP predominantly originated from indoor emission sources (combustions). Therefore, in order to reduce exposure to UFP and protect public health, the primary concerns should be focused on controlling emissions from indoor sources.

Polyphenols, pivotal secondary metabolites, are involved in plant adaption to abiotic stresses. Here, we investigated the role and metabolism profile of polyphenols under aluminum (Al) stress in different lettuce genotypes grown in 0.5 mM CaCl₂ solution with AlCl₃ (pH = 4.5). The complementary use of high-resolution mass spectrometry and quantitative biochemical approaches allowed the characterization of total and unique phenols, as well as their roles in Al tolerance. By comparing the most tolerant and sensitive genotype, 8 polyphenols, including 4 phenolic acids, 2 flavonoids, 1 xanthone and 1 unknown compound, were identified in the roots of the tolerant genotype. The total phenolic and flavonoid contents significantly increased in the tolerant genotype under Al stress. Seedlings with more phenolic accumulation usually performed greater Al tolerance. Meanwhile, principal enzymes related to phenolic biosynthesis significantly increased in roots of the tolerance genotype after Al treatment, with phenylalanine ammonia lyase (PAL), cinnamate 4-hydroxylase, and 4-coumarate coenzyme A ligase increased by 16, 18 and 30%, respectively. The elevated total phenolics were significantly suppressed by AIP, a highly specific PAL inhibitor. Consequently, the antioxidant capacity was inhibited, leading to lettuce sensitivity to Al stress. These results clearly suggested the enhancement of unique polyphenolic biosynthesis as an adaptive strategy of lettuce to Al stress by protecting plants from oxidative stress.

TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin) and several other environment/food-borne toxic compounds induce their toxicity via the aryl hydrocarbon receptor (AhR). AhR is also modulated by various endogenous ligands e.g. highly potent tryptophan (Trp)-derivative FICZ (6-formylindolo[3,2-b]carbazole) and natural ligands abundant in the human diet e.g. polyphenols. Therefore, evaluating AhR species-specific responses is crucial for understanding AhR physiological functions, establishing risk assessments, and exploring the applicability of AhR mediators in drug and food industry towards human-based usages. We studied AhR transactivation of FICZ/TCDD in vitro in a time-dependent and species-specific manner using dioxin responsive luciferase reporter gene assays derived from rat (DR-H4IIE) and human (DR-HepG2) hepatoma cells. We observed for the first time that FICZ potency was similar in both cell lines and was 40 times higher than TCDD in DR-HepG2 cells. Depleting Trp-derivative endogenously produced ligands by using culture medium without Trp, resulted in 3-fold higher AhR activation upon adding FICZ in DR-H4IIE cells, in contrast to DR-HepG2 cells which revealed a fast degradation of FICZ induction from 10 h post-exposure to complete disappearance after 24 h. Seven polyphenols and a mixture thereof, chosen based on commercially recommended doses and adjusted to human realistic exposure, caused rat and human species-specific AhR responses. Two isoflavones (daidzein and genistein) induced rat AhR synergistic effects with FICZ and/or TCDD, while quercetin, chrysin, curcumin, resveratrol, and the mixture exerted a strong inhibitory effect on the human AhR. Strikingly, resveratrol and quercetin at their realistic nanomolar concentrations acted additively in the mixture to abolish human AhR activation induced by various TCDD concentrations. Taken together, these results illustrate the species-specific complexity of AhR transcriptional activities modulated by various ligands and highlight the need for studies of human-based approaches.

Decabromodiphenyl ether (DecaBDE) is a brominated flame retardant belonging to the group of polybrominated diphenyl ethers. DecaBDE has been widely used for various applications, such as plastics, textiles, and building and construction materials. Limited information on DecaBDE production and usage inventory has been elaborated, however. Therefore, this work aimed to produce a preliminary emissions inventory of DecaBDE in mainland China by estimating production and consumption amounts of DecaBDE, and characterizing its emission factors during production and usage, based on industrial investigation and theoretical prediction. It was indicated that the total production of DecaBDE reached 464.68 thousand metric tons (kt), of which 62.72 kt were exported, since the beginning of its production. Shandong and Jiangsu provinces dominate the production, with proportions of 77.95% and 18.45%, respectively. The production stage releases most of the DecaBDE to the atmosphere, with an emissions factor of 23 ± 1.9 kg/t, followed by 20 ± 0.9 kg/t DecaBDE to waste water and 16 ± 1.0 kg/t DecaBDE as solid residue. DecaBDE emissions in the consumption stage—namely the plastic production process—are 0.17 ± 0.06–0.23 ± 0.08 kg DecaBDE to the atmosphere and 1.72 ± 0.58–2.29 ± 0.77 kg DecaBDE to solid residue, for each metric ton of plastic produced. The total annual DecaBDE emissions to waste water are 93.98–1140.9 mg—negligible. The results showed that the sources of DecaBDE environmental pollution are its manufacturing and flame-retardant plastic modification plants, which are easily overlooked by both the government and the public. Yet DecaBDE emissions elimination and the environmentally sound management of the DecaBDE waste generated from these two processes are crucial for environmental protection.

Soils, especially permafrost in the Arctic and the Tibetan Plateau, are one of the largest reservoirs of mercury (Hg) in the global environment. The Hg concentration in the grassland soils over the Tibetan Plateau and its driving factors have been less studied. This study analyzes soil total mercury (STHg) concentrations and its vertical distribution in grassland soil samples collected from the Tibetan Plateau. We adopt a nested-grid high-resolution GEOS-Chem model to simulate atmospheric Hg deposition. The relationship between STHg and soil organic carbon (SOC), as well as atmospheric deposition, are explored. Our results show that the STHg concentrations in the Tibetan Plateau are 19.8 ± 12.2 ng/g. The concentrations are higher in the south and lower in the north in the Tibetan Plateau, consistent with the previous results. Our model shows that the average deposition flux of Hg is 3.3 μg m⁻² yr⁻¹, with 57% contributed by dry deposition of elemental mercury (Hg⁰), followed by dry (19%) and wet (24%) deposition of divalent mercury. We calculate the Hg to carbon ratio (RHg:C) as 5.6 ± 6.5 μg Hg/g C, and the estimated STHg is 86.6 ± 101.2 Gg in alpine grasslands in the Tibetan Plateau. We find a positive relationship between STHg and SOC in the Tibetan Plateau (r² = 0.36) and a similar positive relationship between STHg and atmospheric total Hg deposition (r² = 0.24). A multiple linear regression involving both variables better model the observed STHg (r² = 0.42). We conclude that SOC and atmospheric deposition influence STHg simultaneously in this region. The data provides information to quantify the size of the soil Hg pool in the Tibetan Plateau further, which has important implications for the Hg cycles in the permafrost regions as well as on the global scale.

Fluoride has been considered as a risk factor of cardiovascular disease due to its endothelial toxicology. However, the mechanism underlying the endothelial toxicity of fluoride has not been clearly illustrated. MiR-200c-3p was strongly linked with endothelial function and its level is increased in serum of fluorosis patients, but it is unclear the role of miR-200c-3p in the fluoride induced endothelial dysfunction. In this study, we confirmed that fluoride exposure induced the apoptosis of endothelial cells both in established rats model and cultured human umbilical vein endothelial cells (HUVECs). And miR-200c-3p was found to be upregulated in NaF treated HUVECs. Fluoride stimulation increased caspase-dependent apoptosis through miR-200c-3p upregulation, with repressing expression of its target gene Fas-associated phosphatase 1 (Fap-1), which functioned as Fas inhibitor. This resulted in activation of Fas-associated extrinsic apoptosis via interaction with increased Fas, Fadd, Cleaved Caspase-8 and Cleaved Caspase-3. The activation of Fas-associated extrinsic apoptosis was abrogated by miR-200c-3p inhibitor. Furthermore, the antiapoptotic effect of downregulated miR-200c-3p was restored by Fap-1 siRNA. These results suggested a determinant role of the miR-200c-3p/Fap-1 axis in fluoride induced endothelial apoptosis.

Marine fishes are exposed to great human-induced alterations due to the indiscriminate discharges into the sea, increasing marine pollution. For this study, 324 specimens of Sardina pilchardus from the Canary Islands were analized during a period of 2 years (June 2016 to May 2018). The concentration of 11 metals and trace elements (Al, B, Cd, Cr, Cu, Fe, Li, Ni, Pb, V and Zn) was determined in each individual using the Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES) technique. Statistical analyses were carried out considering the following factors: oceanographic season, maturity of the gonads in the seasons, size of the specimens. Sardina pilchardus specimens captured in the hot season presented higher Ni, Li and Pb levels than the specimens caught in the cold season. Immature specimens had higher concentration in more metals than the mature specimens. This fact may be due to the fact that these specimens require a much higher metabolic rate due to their growth and do not detoxify like mature specimens. Significant differences were found in the concentration of metals in all the analyzes performed.

Kaolinite supported CoFe₂O₄ (KCF) was synthesized and employed to adsorb doxycycline (DOX), an antibiotic and Congo red (CR), a dye from aqueous solution. The prepared KCF nanocomposite was treated in a muffle furnace at 300, 500 and 700 °C, and thereafter characterized. X-ray diffractogram revealed structural damage of kaolinite and appearance of distinct peaks of CoFe₂O₄ with an increase in calcination temperature, while transmission electron microscopy (TEM) images showed that CoFe₂O₄ nanoparticles were supported on the lamellar surface of kaolinites. Comparative adsorption mechanism of the two targeted contaminants showed that adsorption of DOX was influenced by hydrogen bond and n-π interaction, while that of CR was due to hydrophobic interaction and hydrogen bond. However, the adsorption of the two contaminants was best fitted to the isotherm that was proposed by Langmuir, with a monolayer maximum adsorption capacity of 400 mg g⁻¹ at 333 K for DOX, and 547 mg g⁻¹ at 298 K for CR. The removal of DOX from aqueous solution was favored by an increase in temperature (endothermic), while that of CR was exothermic. Thermodynamics studies confirmed that the adsorption of the two contaminants is feasible and spontaneous. The presence of natural organic matter (NOM) did not affect the removal of the two contaminants. Regeneration and reusability study showed that KCF is economically viable. Therefore, introducing inorganic particles like cobalt ferrite into the matrix of kaolinites provides a composite with promising adsorption capacity.
