Cumulative risk assessment of phthalates associated with birth outcomes in pregnant Chinese women: A prospective cohort study

A prospective cohort study of a Chinese population of mother–neonate pairs (n = 3103) was conducted to investigate the relationship between the cumulative hazard index (HI) of combined diethyl phthalate (DEP), dibutyl phthalate (DBP), dibenzyl phthalate (BBzP) and di(2-ethylhexyl) phthalate (DEHP) exposure and birth outcomes. The estimated HI for phthalates was based on phthalate metabolite concentrations in urine collected between 5th and 14th gestational weeks. The median HI values according to the European Food Safety Authority tolerable daily intake (HITDI) and U.S. Environmental Protection Agency reference dose (HIRfD) were 0.358 and 0.187, respectively. A total of 16.3% and 1.9% of the women exhibited HITDI and HIRfD exceeding the value of one, respectively. In unadjusted models, the categories (low < P25, median P25–P50, high > P75) of HITDI were associated with decreased birth weight (β = −26.34 g, p = 0.021) and head circumference (β = −0.09 cm, p = 0.029), whereas those for HIRfD were negatively associated with birth weight (β = −31.74 g, p = 0.005), birth length (β = −0.11 cm, p = 0.032), head circumference (β = −0.13 cm, p = 0.003) and chest circumference (β = −0.10 cm, p = 0.021) in all neonates. Adjustment for potential confounders revealed that HIRfD was inversely associated with head circumference (β = −0.10 cm, p = 0.020). Stratification by gender indicated that HIRfD was associated with decreased birth length (β = −0.17 cm, p = 0.041) in infant boys and HITDI was associated with decreased birth weight (β = −33.12 g, p = 0.036) and head circumference (β = −0.13 cm, p = 0.027) in girls. This is the first study on the cumulative risk assessment of phthalate exposures in pregnant Chinese women. We found that the HI values of multiple phthalate co-exposure were sex-specifically related to birth outcomes.

Receptor models were typically used in air pollution studies and few publications are available for Positive Matrix Factorization (PMF) that consider the details of parameters and procedures in evaluating the trace organic pollutants in sediments. In this study, environmental fate and source composition of Lake Eymir sediments contaminated by polychlorinated biphenyls (PCBs) were explored by applying two PMF models, Paatero's PMF2 and United States Environmental Protection Agency's (US EPA) EPA-PMF3. PMF2 and EPA-PMF3 rely on different algorithms; Paatero's algorithm and multilinear engine algorithm, respectively. Here, the approaches of two PMF models were compared for the identification of PCB patterns taking into consideration the effects of various uncertainty matrices, residual matrices and goodness-of fit parameters. As a result of the study, it was understood that both models resolved five factors and indicated Clophen A60 as the source of PCBs. These results were consistent with the results resolved by Chemical Mass Balance model applied to the same data set in a previous study. However, source contributions identified by two models differed in quantity, but with similar patterns. This study indicates a way in understanding behavior, fate and global source of persistent organic pollutants in sediment by applying and comparing with a special data including high percentage of below detected value (38.2%) to understand the dynamics of PMF model parameters.

Organic contaminants deposited on glacier snow and ice are subject to partitioning and degradation processes that determine their environmental fate and, consequently, their accumulation in ice bodies. Among these processes, organic compound metabolism by supraglacial bacteria has investigated to a lesser extent than photo- and chemical degradation. We investigated biodegradation of the organophosphorus insecticide chlorpyrifos (CPF), a xenobiotic tracer that accumulates on glaciers after atmospheric medium- and long-range transport, by installing in situ microcosms on an Alpine glacier to simulate cryoconite hole systems. We found that biodegradation contributed to the removal of CPF from the glacier surface more than photo- and chemical degradation. The high concentration of CPF (2–3 μg g−1 w.w.) detected in cryoconite holes and the estimated half-life of this compound (35–69 days in glacier environment) indicated that biodegradation can significantly reduce CPF concentrations on glaciers and its runoff to downstream ecosystems. The metabolic versatility of cryoconite bacteria suggests that these habitats might contribute to the degradation of a wide class of pollutants. We therefore propose that cryoconite acts as a “biofilter” by accumulating both pollutants and biodegradative microbial communities. The contribution of cryoconite to the removal of organic pollutants should be included in models predicting the environmental fate of these compounds in cold areas.

Diet is considered as the most important human exposure pathway for polybrominated diphenyl ethers (PBDEs). Metabolism and accumulation patterns of PBDEs in different growth periods of chickens are helpful for evaluating human dietary exposure, but such information is scarce. In this study, female chickens were fed with food spiked with BDE-209 at 85 mg kg⁻¹, and the intake, accumulation, and excretion of BDE-209 and its main metabolites in various tissues were examined. Concentrations of BDE-209 in chicken tissues increased over time in a tissue-specific manner; they were the greatest in liver and generally the lowest in breast meat during the entire exposure period. The kinetic patterns were dependent on both growth-dilution effects and accumulated concentrations of BDE-209. Tissue concentrations of ∑8PBDE (sum of BDE-28, 47, 99, 100, 153, 154, 183, and 209) followed the sequence of liver > blood > skin > intestine > stomach > leg meat > breast meat. Different tissue partition coefficients and perfusion rates for blood may have resulted in different PBDE concentrations in tissues. The absorption efficiency of BDE-209 in chicken tissues followed the sequence of liver (0.15 ± 0.032%) > skin (0.14 ± 0.038%) > intestine (0.071 ± 0.021%) > breast meat (0.062 ± 0.020%) > leg meat (0.059 ± 0.016%) > stomach (0.021 ± 0.0095%), likely due in part to facilitated absorption of BDE-209 by transport proteins (P-glycoproteins). On average, 9.3 ± 1.7% of BDE-209 was excreted in feces. Estimated human average dietary intake via the consumption of chicken tissues of ∑8PBDE for adults and children was 319 and 1380 ng day⁻¹ for liver, 211 and 632 ng day⁻¹ for leg meat, and 104 and 311 ng day⁻¹ for breast meat from the contaminated group. Liver clearly poses the highest exposure risk for human consumption, particularly if chickens are fed with contaminated feed.

Many antibiotics, including sulfonamides, are being frequently detected in soil and groundwater. Livestock waste is an important source of antibiotic pollution, and sulfonamides may be present along with organic-rich substances. This study aims to investigate the sorption reaction of two sulfonamides, sulfamethoxazole (SMZ) and sulfapyridine (SPY) in two organic-rich sorbents: a commercial peat soil (38.41% carbon content) and a composted manure (24.33% carbon content). Batch reactions were conducted to evaluate the impacts of pH (4.5–9.5) and background ions (0.001 M–0.1 M CaCl2) on their sorption. Both linear partitioning and Freundlich sorption isotherms fit the reaction well. The n values of Freundlich isotherm were close to 1 in most conditions suggesting that the hydrophobic partition is the major adsorption mechanism. In terms of SMZ, Kd declined with increases in the pH. SPY has a pyridine group that is responsible for adsorption at high pH values, and thus, no significant trend between Kd and pH was observed. At high pH ranges, SPY sorption deviated significantly from linear partitioning. The results suggested the sorption mechanism of these two sulfonamide antibiotics tended to be hydrophobic partitioning under most of the experimental conditions, especially at pH values lower than their corresponding pKa2. The fluorescence excitation emission matrix and dissolved organic carbon leaching test suggested composted manure has higher fulvic acid organics and that peat soil has higher humus-like organics. Small organic molecules showed stronger affinity toward sulfonamide antibiotics and cause the composted manure to exhibit higher sorption capacity. Overall, this study suggests that the chemical structure and properties of sulfonamides antibiotics and the type of organic matter in soils will greatly influence the fate and transport of these contaminants into the environment.

Habitat loss and environmental pollution are among the main causes responsible for worldwide biodiversity loss. The resulting species and population declines affect all vertebrates including reptiles. Especially in industrialized countries, pollution by agrochemicals is of remarkable importance. Here, habitat loss has historically been associated with expansion of agriculture. Species persisting in such environments do not only need to cope with habitat loss, but more recently, also with chemical intensification, namely pesticide exposure. In this study, we examined effects of different fungicide and herbicide applications on the common wall lizard (Podarcis muralis) in grape-growing areas. We used three enzymatic biomarkers (GST, GR, AChE) and for the first time saliva from buccal swabs as a minimal-invasive sampling method for detection. Our results demonstrate absorption of substances by lizards and effects of pesticide exposure on enzymatic activities. Our findings are in accordance with those of previous laboratory studies, although samples were retrieved from natural habitats. We conclude that buccal swabs could become a useful tool for the detection of pesticide exposure in reptiles and have the potential to replace more invasive methods, such as organ extraction or cardiac puncture. This is an important finding, as reptiles are non-target organisms of pesticide applications, and there is a strong need to integrate them into pesticide risk assessments.

China produces approximately 20%–30% of the total global chlorinated paraffins (CPs). The establishment of a short-chain CP (SCCP) emission inventory is a significant step toward risk assessment and regulation of SCCPs in China and throughout the globe. This study developed a gridded SCCPs emission inventory with a 1/4° longitude by 1/4° latitude resolution from 2008 to 2012 for China, which was based on the total annual CPs emissions for the nation. The total national SCCPs emission during this 5-year period was 5651.5 tons. An additive in metal cutting fluids was a major emission source in China, contributing 2680.2 tons to the total atmospheric emissions of SCCPs from 2008 to 2012, followed by the production of CPs (2281.8 tons), plasticizers (514.3 tons), flame retardants (108.6 tons), and net import (66.6 tons). Most of these emission sources are located along the eastern seaboard of China and southern China. A coupled atmospheric transport model was employed to simulate environmental contamination by SCCPs using the gridded emission inventory of SCCPs from 2008 to 2012 as the model initial conditions. Simulated atmospheric and soil concentrations were compared with field monitoring data to validate the emission inventory. The results showed good consistency between modeled and field sampling data, supporting the reliability and credibility of the gridded SCCPs emission inventory that was developed in the present study.

Polyhalogenated carbazoles (PHCZs) have attracted mounting environmental concerns since they were recently discovered in sediments and soil. Current knowledge on their occurrence, environmental behavior and fate remains very limited in general. In the present study, 11 PHCZ congeners were screened in surface sediments of Lake Tai, an important freshwater system located in the Yangtze River Delta, China. Total concentrations of PHCZs (∑PHCZs) ranged up to 15.8 ng/g dry weight (median: 1.54 ng/g dw), rivaling those of polybrominated diphenyl ethers (∑PBDEs, 0.07–15.9 ng/g dw) in the same sediments. The PHCZ congener composition profiles revealed a dominance of 3,6-dichlorocarbazole and 3,6-dibromocarbazole with comparable concentrations. These two dominant congeners differed in spatial distribution patterns in Lake Tai, indicating different sources or origins. Potential toxic effects associated with the levels of PHCZs in the sediments were evaluated via the toxic equivalent (TEQ) approach. The TEQs of PHCZs in Lake Tai sediments ranged up to 1.36 pg TEQ/g dw. As the first report on the occurrence of PHCZs in an Asian waterbody, our findings suggest that PHCZs should be given more attention during environmental monitoring and risk assessments of hazardous chemicals, as they may represent another group of persistent organic pollutants with dioxin-like effects and wide distributions.

Metal exposure can produce oxidative stress by disrupting the prooxidant/antioxidant balance. It has been suggested that calcium (Ca) may provide protection against metal toxicity in the organism. The objective of this study is to explore the effects of Ca availability and metal pollution on oxidative stress biomarkers in pied flycatcher (Ficedula hypoleuca) nestlings. For this purpose, we performed a Ca-supplementation experiment with birds inhabiting a Ca-poor and metal-polluted area in SW Finland. An array of oxidative stress biomarkers (GSH, GSH:GSSG ratio, GPx, GST, CAT, SOD, lipid peroxidation and protein carbonylation) was measured in red blood cells. The effects of antioxidant molecules and oxidative damage on nestling size, growth, fledging success and fledgling number were evaluated. We observed an up-regulation of GST activity and increased protein carbonyl content in the polluted zone, probably related to a combination of higher metal exposure and reduced food quantity and quality in this area. As expected, birds from the unpolluted zone showed higher GSH:GSSG ratio but, unexpectedly, also showed signs of higher lipid peroxidation (not statistically significant, p = 0.056), both responses likely being related with the lower Ca availability. Our study suggests that different measures of oxidative damage are affected by different factors: while damage to proteins was the target of metal exposure/food limitation, poor Ca availability may enhance damage to lipids in growing birds. The intercorrelations found between Ca in plasma, metal exposure and the different oxidative stress biomarkers show that the antioxidant defense is finely regulated to cope with increased oxidative challenges. Finally, our results suggest that the antioxidant status during early development, conditioned by environmental pollution and Ca availability, is one factor affecting nestling survival and fledgling number.

On-road emissions vary widely on time scales as short as minutes and length scales as short as tens of meters. Detailed data on emissions at these scales are a prerequisite to accurately quantifying ambient pollution concentrations and identifying hotspots of human exposure within urban areas. We construct a highly resolved inventory of hourly fluxes of CO, NO2, NOx, PM2.5 and CO2 from road vehicles on 280,000 road segments in eastern Massachusetts for the year 2012. Our inventory integrates a large database of hourly vehicle speeds derived from mobile phone and vehicle GPS data with multiple regional datasets of vehicle flows, fleet characteristics, and local meteorology. We quantify the ‘excess’ emissions from traffic congestion, finding modest congestion enhancement (3–6%) at regional scales, but hundreds of local hotspots with highly elevated annual emissions (up to 75% for individual roadways in key corridors). Congestion-driven reductions in vehicle fuel economy necessitated ‘excess’ consumption of 113 million gallons of motor fuel, worth ∼ $415M, but this accounted for only 3.5% of the total fuel consumed in Massachusetts, as over 80% of vehicle travel occurs in uncongested conditions. Across our study domain, emissions are highly spatially concentrated, with 70% of pollution originating from only 10% of the roads. The 2011 EPA National Emissions Inventory (NEI) understates our aggregate emissions of NOx, PM2.5, and CO2 by 46%, 38%, and 18%, respectively. However, CO emissions agree within 5% for the two inventories, suggesting that the large biases in NOx and PM2.5 emissions arise from differences in estimates of diesel vehicle activity. By providing fine-scale information on local emission hotspots and regional emissions patterns, our inventory framework supports targeted traffic interventions, transparent benchmarking, and improvements in overall urban air quality.
