Callosobruchus maculatus causes damaging of storage mungbean seeds. This insect feeds on grain product during larval life and depends on its alpha-amylase for survival. Understanding the function of alpha-amylase is essential for a developing method for the insect control. In this studies, C. maculates alpha-amylase was purified and characterized. The alpha-amylase was purified by an affinity chromatography on a selfcoupled beta-cyclodextrin sepharose 6B. The amylase of C. maculatus has one isoform with a molecular weight of 50 kDa. Its specific activity was182.78 U/mg. The optimum temperature of alpha-amylase activity was in the range 50-60 deg C. The pH optimum for alpha-amylase was in the range 5-6.
