Study of spore preservation ability of representatives of the Bacillus genus in water and spoil at different pH values and temperatures | Изучение сохраняемости спор представителей рода Bacillus в воде и почве при различных значениях pH и температуры

anglais. There has been studied of spore inactivation dynamics of bacillus anthracix agents and closely related saprophytes of the g. Bacillus in different environmental conditions. The vaccine strain Bacillus anthracis (BA) STI-1 and 10 filed isolates recovered from homoithermic animals and from soil, as well as 5 strains of saprophytes antigenically related to BA were used. BA in the dose of 10E3-10E4 spores/cm3 were introduced into a sterile normal saline and stored at t20+-0.2 deg. C. The samples were periodically sown on agar. For 970 days of observation 97.7% spores were inactivated at pH 3.0; 66% - at pH 5.0 and 5.4 – at pH 7.0. Typical chernozem (TC) with pH 6.86 and swampy boggy soil (SBS) with pH 3.05 were disseminated with BA spores based on 1.0*10E4 spore/g, stored at 20 and 4 deg C. In the SBS samples after 7 days 39.5% spores died, after 970 days – TC samples (pH 7.0) – 0 and 54%, respectively. The inactivation rate of saprophyte spores was higher by 20%, after 970 days only 0.1% remained viable. In TC (the humus content 8-12%, pH 6.86, T20+-1 deg. C, the moisture content 60%) BA passes a full biological cycle. The 40 times increment of microbial mass occurred during the first 30-50 days, for the following 2 years the population of viable cells gradually reduced, but remained 7-12 times higher as compared to the initial one. At t 4 deg. C in TC 30-40% spores were inactivated in the 1-st week. After 970 days in the samples 8.5% of the initial population of spores was detected. In SBS at t20 and 4 deg. C the viability of spores in the first week reduced to 62.6 and 74%m respectively, by 970 days – up to 3.1 and 3.6%. It is suggested in evaluating the time of epizootic danger of anthrax burial places considering the soil climatic factors promoting the inactivation of BA spores.

russe. Исследована динамика инактивации спор возбудителя сибирской язвы и близкородственных сапрофитов р. Bacillus в разных условиях окружающей среды. Использовали вакцинный штамм Bacillus anthracis (BA) СТИ-1 и 10 полевых изолятов, выделенных от теплокровных животных и из почвы, а также 5 штаммов сапрофитов, антигенно родственных BA. В стерильный физраствор с pH 3,0; 5,0 и 7,0 вносили ВА в дозе 10E3-10E4 спор/см3, хранили при t 20+-0,2 град. C. Пробы периодически высевали на агар. За 970 сут наблюдений произошла инактивация 97,7% спор при pH 3,0; 66% - при pH 5,0 и 54% - при pH 7,0. Чернозем типичный (ЧТ) с pH 6,86 и болотно-торфяную почву (БТП) с pH 3,05 обсеменяли спорами BA из расчета 1,0*10E4 спор/г, хранили при t 20 и 4 град. C. В пробах БТП через 7 сут погибло 39,5% спор, через 970 сут - 97,7%, в пробах ЧТ (pH 7,0) - 0 и 54% соответственно. Скорость инактивации спор сапрофитов была на 20% выше, через 970 сут жизнеспособными остались лишь 0,1%. В ЧТ (содержание гумуса 8-12%, pH 6,86, t 20+-1 град. C, влажность 60%) BA проходят полный биологический цикл. Нарастание микробной массы в 40 раз происходило в течение первых 30-50 дн., за следующие 2 года количество жизнеспособных клеток постепенно снижалось, но оставалось в 7-12 раз выше по сравнению с исходным. При t 4 град. C в ЧТ в 1-ю нед было инактивировано 30-40% спор. Через 970 сут в пробах выявляли 8,5% от исходного количества спор. В БТП при t20 и 4 град. C жизнеспособность спор в 1-ю неделю снизилась до 62,6 и 74% соответственно, к 970 сут - до 3,1 и 3,6%. Предлагается при оценке срока эпизоотической опасности сибиреязвенных захоронений учитывать почвенно-климатические факторы, способствующие инактивации спор BA.
