Rusitec vessels were fed a total mixed ration, without oil supplement (control) or with the addition of one vegetable oil (olive, sunflower or linseed oils). Fermentation was maintained in the cultures for three weeks, samples of digesta were collected and bacterial DNA was extracted for studies of microbial diversity using the T-RFLP method. Experimental treatments were significantly clustered from the T-RFLP profiles, so that unsupplemented cultures were clearly differentiated. Using fragment size libraries for a most probable identification of peaks recorded differences between control and supplemented cultures could be established. It was concluded that oil supplementation had a significant and selective effect on ruminal bacteria communities.
