Characterisation of thrombocytes in Osteichthyes

Thrombocytes in vertebrates other than mammals, inter alia in fish, are analogues of platelets in mammals. In Osteichthyes, these cells take part in haemostatic processes, including aggregation and release reactions in cases of blood vessel damage, and in the immune response development as well. This paper discusses the development of thrombocytes in Osteichthyes, taking into account the need to make changes to the concept of grouping progenitor cells as suggested in the literature. The following pages present the morphological and cytochemical properties of thrombocytes as well as their defence functions, and also point out differences between thrombocytes in fish and platelets in mammals. The paper further highlights the level of thrombocytes’ immune activity observed in fish and based on an increased proportion of these cells in response to antigenic stimulation, on morphological shifts towards forms characteristic of dendritic cells after antigenic stimulation and on the presence of surface structures and cytokines released through, inter alia, gene expression of TLR receptors, MHC class II protein-coding genes and pro-inflammatory cytokines. The study also points out the need to recognise thrombocytes in Osteichthyes as specialised immune cells conditioning non-specific immune mechanisms and playing an important role in affecting adaptive immune mechanisms.

Histamine is one of the most important and toxic biogenic amines which may be present in food and may cause food poisoning in humans when contained at a high level. It is produced during bacterial decarboxylation of histidine in fish muscles. The aim of the study was to investigate the presence of histamine in fish and fish products available in Poland during 2014–2018. A total of 421 samples of raw (248), smoked (107), canned (50), and marinated fish (16) were analysed by high-performance liquid chromatography with diode array detection. Histamine was detected in 14.1% samples of raw fish, 29% of smoked fish, 22% of canned fish and 93.8% of marinated fish in concentrations ranging from 3.4 to 156.4 mg/kg. Content of this amine above 100 mg/kg was found in four samples: raw Atlantic salmon, smoked European sprat and two samples of marinated Atlantic herring. The study showed that fish and fish products on the Polish market generally meet the food safety criteria for histamine and are safe for consumers.

Introduction: The aim of this study was to evaluate the prevalence of Clostridium botulinum and Clostridium perfringens in food samples purchased from Polish producers. Material and Methods: The analyses were performed on 260 food samples collected in Lublin and Subcarpathian regions: 56 of smoked meat, 21 of pork meat, 20 of dairy products, 26 of vegetable and fruit preserves, 40 of ready-to-eat meals, 27 of fish preserves, and 70 of honey collected directly from apiaries. Results: C. botulinum strains were isolated from 2.3% (6/260) of samples and the isolates were classified as toxin types A (4/260) and B (2/260). C. perfringens strains were isolated from 14% (37/260) of samples. All the isolates were classified as toxin type A, 28 of them were able also to produce α toxin and 9 - β2 toxin. Conclusion: On the basis of the obtained results it could be suggested that risk assessment, especially regarding the entire honey harvesting process, should be provided in order to ensure the microbiological safety of the products to be consumed by infants and people with a weakened immune system.

Introduction: The goal of this study was to assess the influence of vaccination against enteric redmouth disease on oxidative stress biomarkers and antioxidant defence in the muscle tissue of rainbow trout (Oncorhynchus mykiss Walbaum) vaccinated against Yersinia ruckeri in the first and second month after immunisation. Material and Methods: Healthy fish were vaccinated orally with inactivated whole cells of a virulent strain of Y. ruckeri. One and two months after immunisation the muscle samples were collected. Results: No significant difference was noted in lipid peroxidation level in either the first or second month after vaccination, while aldehydic and ketonic derivatives of oxidatively modified proteins (OMB) in the vaccinated group were significantly lower in the second month compared to those in the first month after vaccination (P < 0.05). The content of ketonic derivatives of OMB in muscles in the first month after immunisation was higher compared to untreated control. All these culminated in a depletion of glutathione peroxidase (GPx) activity and low level of total antioxidant capacity (TAC). Conclusion: Correlations between catalase activity and lipid peroxidation and TAC confirmed the pivotal role of catalase in antioxidant defence during immunisation. From a broader perspective, it is suggested that immunisation of fish with Yersinia vaccine is associated with induced free radical formation and oxidative stress. Free radicals would therefore be at least partially responsible for the induction of both humoral and cellular elements of the immunity and increased protective immunity against Y. ruckeri infection.

The aim of this article was to evaluate the influence and effects of chosen bioaccumulative substances i.e. heavy metals, pesticides, and polychlorinated biphenyls (PCBs) on fish, as well as provide information on time trends and potential threat to human health. Chemical substances which pollute water may affect living organisms in two ways. First of all, large amounts of chemical substances may cause sudden death of a significant part of the population of farmed fish, without symptoms (i.e. during breakdown of factories or industrial sewage leaks). However, more frequently, chemical substances accumulate in tissues of living organisms affecting them chronically. Heavy metals, pesticides, and polychlorinated biphenyls are persistent substances with a long-lasting biodegradation process. In a water environment they usually accumulate in sediments, which makes them resistant to biodegradation processes induced by, e.g., the UV light. These substances enter the fish through direct consumption of contaminated water or by contact with skin and gills. Symptoms of intoxication with heavy metals, pesticides, and PCBs may vary and depend on the concentration and bioavailability of these substances, physicochemical parameters of water, and the fish itself.

Introduction: In the present study, some of the commercial fish farms located in the Black Sea region of Turkey, were screened for bacteria between 2012 and 2014.Material and Methods: The bacterial agents isolated from fish were identified by classical biochemical tests and the rapid diagnostic tests (API 20 E and API 20 Strep). All strains were further identified by sequencing of the 16S rRNA genes. The strains were also investigated for resistance to different antimicrobials by the disc diffusion method. Antibiotic resistance genes, including tetracycline (B), β-lactam (ampC, blaTEM, blaPSE), florfenicol (floR), erythromycine (ereA, ereB), sulphonamide (sulI, sulII), and trimethoprim (dhfr1) genes, were determined by the PCR method.Results:Vibrio anguillarum, Vibrio fluvialis, Photobacterium damselae subsp. piscicida, Pseudomonas luteola, Lactococcus garvieae, Streptococcus iniae, Aeromonas hydrophila, and Yersinia ruckeri were isolated from marine and freshwater cultured fish. According to the results of disc diffusion, all isolates were sensitive to florfenicol, trimethoprim+sulfamethoxazole, oxitetracycline, and enrofloxacin, and resistant to lincomycin, penicillin G, and amoxicillin. Also, sulI, sulII, and floR resistance genes were detected in the bacteria.Conclusion: The results of the study open up the opportunity to perform further investigations which could determine the possible role of ARGs in fish pathogens.

Objective: This cross-sectional study was conducted from April to July 2014 in Khartoum state, the Sudan, to investigate aerobic bacteria and fungi of skin lesions of fish in 3 different areas in Khartoum. Material and methods: A total of 50 samples were collected from the skin lesions of different types of fish including Synodontis species (n=17), Tilapia niloticus (n=15), Labeo niloticus (n=10), Hydrocynus species (n=4), and Clarias species (n=4). Liquid, semi-solid, and solid culture media like nutrient broth, blood agar, MacConkey agar, sabouraud dextrose agar (SDA), and Simmons citrate medium were used for the isolation and identification of bacteria and fungi. Besides, Gram staining and biochemical characterization were also conducted.Results: Culturing of the collected samples revealed growth of bacteria from all (100%), and growth of fungi could be found from 32% samples. A number of 188 bacteria were isolated, mainly Staphylococcus species, Bacillus species, Aeromonas species, Pseudomonas species, and Vibrio species. Besides, 16 fungi could be identified containing Aspergillus niger, A. flavus, A. fumigatus, and Phycomycete.Conclusion: Fishes with skin lesions are harboring many pathogenic bacteria and fungi and may act as a source of zoonotic infections and can transmit several pathogens to workers in fish industry and consumers. Therefore, thorough and strict routine inspection of fish is recommended to ensure safety and that there are no serious risks to consumers. [J Adv Vet Anim Res 2016; 3(4.000): 375-385]

Fish can be considered as an ideal food as it contains high protein content, minerals, vitamins and omega 3 fatty acids. Organochlorine pesticides (OCPs) are characterized by their low cost, severe toxicity against a wide array of pests, long duration of action and stability in the environment. OCPs have been used for many years in African countries, particularly in Egypt. Despite being outlawed everywhere, OCPs are still being used illegally. One of the main responsibilities of the food safety and public health sectors is to guarantee the safety and wholesomeness of such food products before they are made available to the general public. In order to determine the residual OCP contents in retailed tilapia, catfish, mullet, saurus, and pagrus, this study was conducted. The obtained results in the current investigation revealed the detection of OCPs at 80%, 50%, 20%, 15%, and 15% in the examined catfish, tilapia, mullet, saurus, and pagrus, respectively. Different OCPs were detected at variable concentrations; however, such concentrations were within the established permissible limits in Egypt. In conclusion, OCPs are still detected in different aquatic species and residual concentrations can be detected in different fish species retailed in the Egyptian fish markets. Therefore, it is highly advised to continuously check for OCP residues in fish.

In this study, the detection of biogenic amines in fresh and products of fish is focused and biological trials have been used to reduce their levels. 120 random samples of fresh fish (Lates niloticus), sardine salted, herring smoked, and tuna canned (for every thirty samples) were collected from different markets in Shibine, Menoufia, Egypt. The biogenic amine residues have examined in all samples that were collected (histamine, putrescine, cadaverine and tyramine). The average levels of biogenic amines in the samples under investigation of fresh fish, canned tuna, herring smoked, and sardine salted are 10.74 ± 0.54, 15.08 ± 0.76, 17.93 ± 0.81 and 26.12 ± 0.89 mg % for histamine, 7.61 ± 0.49, 11.13 ± 0.57, 14.35 ± 0.64 and 19.59 ± 0.81 mg% for putrescine, 5.96 ± 0.43, 10.22 ± 0.49, 12.41 ± 0.58 and 16.85 ± 0.62 mg% for cadaverine each, accordingly. Finally, the tyramine mean value was 2.05 ± 0.10 mg % in fresh fish, 6.37 ± 0.19 mg % in tuna canned, 7.9 ± 0.24mg % for herring smoked and10.44 ± 0.27 mg % for sardine salted. Fish fillets were experimentally inoculated with B. polymyxa culture (2x107), the effect was ideal, the level of histamine reduced to 31.4 mg/kg after eight hours, 19.5 mg/kg after 12 hrs and 12.8 mg/kg 24 hours later with reduction percentages of 37.2%, 61.0%, and 74.4%, respectively and cadaverine level reduced to 22.1 mg/kg after 8 hours, 15.4 mg/kg after 12 hrs and 9.7 mg/kg 24 hrs later with reductions percentages were 26.3%, 48.7%, and 67.7%, respectively.

Fish is one of the most important foods because of its high nutritional value, high palatability, and easy digestion. At the same time, it acts as a vehicle for many types of pathogenic microorganisms especially Aeromonas species, which results in public health hazards. Therefore, the present study was conducted to evaluate the prevalence of Aeromonas species in fresh fish (catfish, mullet, lizardfish, and coralfish) marketed in Zagazig city, Sharkia Governorate, Egypt. In addition, multiplex PCR was performed to detect some virulence-associated genes in A. hydrophila isolates. Furthermore, antimicrobial susceptibility testing was carried out on A. hydrophila isolates using commonly used antimicrobials in Egypt through the disc diffusion method. The achieved results indicated contamination of fish with different species of Aeromonas such as A. veronii, A. sobria, A. caviae and A. hydrophila. The results revealed that Aeromonas species isolated with an overall percentage of 55% of all examined fish. Bacteriological examinations revealed 20% A. hydrophila, 20% A. sobria, 10% A. caviae and 5% A. veronii. Antibiotic sensitivity declared high resistance of the isolates to different antimicrobial agents used in Egypt, including penicillin (100%), Ampicillin (90.0%), Streptomycin (90.9%), Cephalothin (72.7%), Tetracycline (72.7%), Cefotaxime (63.6%), and Sulfamethoxazole (54.5%). Therefore, hygienic measures should be adopted to control microbial contamination either in the aquatic environment or in fish markets.
